The absorption of blood onto the surfaces of white cotton, polyester, rayon, and nylon fabrics was studied. Different categories of fabrics (woven and knitted) with diverse thickness, were manually folded twice to obtain four fabric layers, and $100{\mu}L$ of human blood was dropped onto the surface of the fabrics. The amount of blood that penetrated the fabric layer and the shape of bloodstain observed on the fabrics were influenced by the chemistry, thickness, and texture of the fabric. The blood bearing fabrics were left to dry for 3 days, washed by hand using tap water, and Lumiscene was then sprayed onto the fabrics to enhance the latent bloodstain for comparison of the shape of the bloodstain before and after washing by hand. The features of the bloodstain after washing varied greatly with the recipient fabrics. Additionally, stronger luminescence was observed at the surface where the blood was deposited compared to the background. However, it was confirmed that physical contact during the washing can deform the original shape of the bloodstain. The effect of the drying time on the bloodstain after hand washing was also studied. $100{\mu}L$ of blood was dropped on the surfaces of the fabrics and dried for 0, 1, 12, 24, 72 h, and 7 days, then washed by hand, before the bloodstain was enhanced with Lumiscene. The results of this experiment indicated that the increased drying time induces stronger chemiluminescence of Lumiscene. However, after drying of the bloodstain for 7 days, the luminescence of the bloodstain was decreased at the blood deposited site and increased around the blood deposited site.
The most common sources of biological trace material which are found in crime scene are the human bloodstains. Reliable identification in the forensic casework is important as it provides crucial insights into crime scene reconstruction and can thus contribute towards solving crimes. Blood-stains are routinely tested in forensic practise using various methods including the leucomalachite green (LMG) test, Kastle-Meyer phenolphthalein test, tetramethylbenzidine test, orthotolidine test, or the luminol chemoluminescence test with the latter cleaning attempts. All these presumptive thus indicative but not identifying tests take advantage of the peroxidase-like activity of the heme unit of the hemoglobin molecule in human blood. Therefore, false-positive results can be caused by the presence of strong oxidants, such as chlorine-containing detergents or by true peroxidases (e.g., from plants). In this study, composition for Gum guaiac was evaluated for the forensic identification of bloodstain and compared with the LMG. The sensitivity and specificity of the composition for Gum guaiac were examined more stable in bloodstain. The positive of Composition for Gum guaiac shown even with the 100,000-fold diluted bloodstain, which was no difference in comparison with LMG test. It was shown that composition for Gum guaiac was very stable to resist boiling for 20 minutes and the effect of bacteria did not affect the genetic analysis as well. The above result of the crime scene investigation, composition for Gum guaiac is easily expected to help identifying bloodstain in the evidences.
DNA isolation for PCR-based short tandem repeat (STR) analysis is essential to recover high yields of amplifiable DNA from low copy number (LCN) DNA samples. There are different methods developed for DNA extraction from the small bloodstain and gloves, commonly found at crime scenes. In order to obtain STR profiles from LCN DNA samples, DNA extraction protocols, namely the automated $iPrep^{TM}$$ChargeSwitch^{(R)}$ method, the automated $QIAcube^{TM}$ method, the automated $Maxwell^{(R)}$ 16 DNA $IQ^{TM}$ Resin method, and the manual $QIAamp^{(R)}$ DNA Micro Kit method, were evaluated. Extracted DNA was quantified by the $Quantifiler^{TM}$ Human DNA Quantification Kit and DNA profiled by $AmpFISTR^{(R)}$$Identifiler^{(R)}$ Kit. Results were compared based on the amount of DNA obtained and the completeness of the STR profiles produced. The automated $iPrep^{TM}$$ChargeSwitch^{(R)}$ and $QIAcube^{TM}$ methoas produced reproducible DNA of sufficient quantity and quality trom the dried blood spot. This two automated methods showed a quantity and quality comparable to those of the forensic manual standard protocols normally used in our laboratory. In our hands, the automated DNA extraction method is another obvious choice when the forensic case sample available is bloodstain. The findings of this study indicate that the manual simple modified $QIAamp^{(R)}$ DNA Micro Kit method is best method to recover high yields of amplifiable DNA from the numerous potential sources of LCN DNA samples.
Lueco-crystal violet(LCV) is a very effective reagent used to enhance bloodstain. However, when the LCV is treated to the bloody fingerprint, the bubbling occurs, so that the minutiae and features of the fingerprint can be damaged. In this study, we studied a method to reduce the bubbling of LCV. As a result, the most effective method for reduction bubbling and enhancement bloodstain was to treat ethanol-based aminotriazole(AT) solution before LCV treatment.

Blood is a commonly found body fluid at crime scenes, and plays an important role in identifying suspects and in the reconstruction of crime scenes. Although serological detection of blood has been widely used in the field of forensic science, research on the detection of old bloodstains is scarce. This work aimed to compare various methods for the detection of old bloodstains and validate the reliability of their results. Four presumptive tests-Tetramethylbenzidine, $Bluestar^{(R)}$, Leucomalachite Green, Kastle-Meyer tests-and two confirmatory tests-Fecal Occult Blood (FOB) and Rapid Stain $Identification^{(TM)}-Blood$ ($RSID^{TM}-Blood$) tests-were compared. Bloodstain samples from post-mortem cases were collected on gauzes and then stored at room temperature for periods from 7 to 30 years. All the presumptive tests were positive, even for the 30-year-old sample. However, FOB and $RSID^{TM}-Blood$ provided false negative results for some samples stored for 17 years or more (1988 to 2001). The results indicate that FOB and $RSID^{TM}-Blood$ are not reliable for the detection of old bloodstains. These findings can be useful in the selection of an appropriate detection method for serological testing of old bloodstains. In addition, the information will be useful background knowledge when applied in the field of forensic practice.
Bloody fingerprints are important in many cases - especially in murder case. This study compared effectiveness of some reagents which had better quality of developing bloody fingerprints from human skin. Leuco crystal violet(LCV), hungarian red, amido black, coomassie blue, acid violet17, and crowle's double stain were used. Fingerprints were generated in the depletion series, and tissue method was used in applying all reagents. LCV showed the best result in this study.
Objectives In the theory of Korean Medicine, Cicadae Periostracum (CP) has been used to treat skin diseases such as inflammatory dermatitis, tetanus and pruritus. CP can reduce heat and disperse wind. In prior studies, anti-allergic effect and anti-inflammatory effect of CP were reported. However, there has been no report regarding the correlation of CP and allergic contact dermatitis (ACD). This study was performed to show the effects of CP in ACD induced by 2,4-dinitrochlorobenzene (DNCB) in mice. Methods In this experiment, the effects of CP on biological changes were measured, such as changes in ear and spleen weight, ear and dorsum skin thickness, clinical aspect on the dorsum skin and histological changes. The effect of proliferation rate of splenocytes was also investigated in vitro and vivo study. Results In results, CP application (CPA) group and CP application and administration (CPAA) group were significantly restrained in ear weight gain and increase in ear and dorsum skin thickness compared to the control group. In addition, CPA and CPAA group showed diminished erythema, desquamation, bloodstain, and marks. Also, the histological assessment showed that CP treatement diminished thickness of epidermis, hyperkeratosis, pigmentation and parakeratosis. Conclusions In conclusion, these data suggest that CP can decrease symptoms of ACD.

In this study, we used a chemiluminescence reaction to investigate the removal effect of pet soil, such as dog blood, urine and feces. The soiled fabrics were washed with a standard laundry course of 30℃ and a washing time of 30 min and a pet care laundry course of 40-60℃ and a washing time of 100 min. The detergency was evaluated by the surface reflectance and chemiluminescence reaction (bloodstain detection by luminol test and urine-stain and feces-stain detection by UV blacklight test) before and after washing. The surface reflectance results did not show any difference in detergency for both courses, whereas the chemiluminescence reaction did. The detergency of the pet care course compared to the standard course was 101% according to the surface reflectance and 120% according to the chemiluminescence reaction. Therefore, residual stains not detected by surface reflectance can be evaluated through chemiluminescence reaction, and it was confirmed that pet stains can be managed more hygienically by washing for a long time at a high temperature.
Commercial one-step rapid fecal occult blood (FOB) kit which was used as a screening test to detect traces of blood in stool samples was evaluated for the feasibility of the forensic identification of human blood. The sensitivity was determined and compared with the conventional Leucomalichite green (LMG) method. In addition, the specificity of the kit and the effects of various chemicals and environmental factors were examined. FOB kit was specific for human hemoglobin and more sensitive than LMG test (approximately 100 times). FOB kit showed positive band using at least 1,000,000-fold diluted human blood. The antigen was very stable regardless of storage temperature and boiling. The positive reaction was not affected by LMG and Luminol, the traditional tests for identification of bloodstain. As a results, FOB test kit could be effectively applied to identification of human blood at crime scene and crime laboratories.

The detection of blood at a crime scene is an important process for identification and case reconstruction. However, blood may be difficult to observe with the naked eye on dark or multi-colored surfaces. Acidic hydrogen peroxide (AHP) is a recently reported blood enhancement reagent that can enhance blood with high sensitivity by increasing the exposure time of the camera. However, it has never been compared to previously known techniques on dark or multi-colored surfaces. For this purpose, the method of observation/photographing (UV and IR photography), alginate casting, leuco rhodamine 6G (LR6G), and AHP were applied to bloody impression on dark or multi-colored surfaces and the results were compared. As a result, blood treated with AHP had a higher contrast to the surfaces than UV and IR photography, and it was applicable on all surfaces, opposed to alginate casting. In addition, AHP successfully enhanced blood on dark or multi-colored surfaces, similar to LR6G.
