도축혈액 혈장 단백질의 특성 및 접착제로의 응용 (Characterization of Plasma Proteins from Bloods of Slaughtered Cow and Pig and Utilization of the Proteins as Adhesives)

  • 도축동물의 대부분 폐기되는 혈액으로부터 기능성 소재를 발굴하고 수질오염을 방지하기 위해서 소, 돼지 도축혈액으로부터 간단하고 신속한 방법으로 혈장 단백질을 분리하는 방법을 개발했고 그것을 혈분접착제로 응용하였다. 혈액을 원심분리한후 혈장단백질은 2% TCA 또는 0.6N HCl을 최종농도가 되게 첨가하여 침전시킴으로써 얻어졌다. 소, 돼지 혈장단백질을 비교 분석하기 위하여 SDA-PAGE를 하였고 얻어진 혈장단백질을 이용하여 혈분접착제로서 합판제조에 사용하여 다른 합성접착제와 접착성 시험을 한 결과 다른 합성 접착제와 비교해서 만족한 결과를 얻었다.

  • This research deals with an issue of preventive medicine in bioinformatics. We can diagnose liver conditions reasonably well to prevent Liver Cirrhosis by classifying liver disorder patients into fatty liver and high risk groups. The classification proceeds in two steps. Classification rules are first built by clustering five attributes (MCV, ALP, ALT, ASP, and GGT) of blood test dataset provided by the UCI Repository. The clusters can be formed by the K-mean method that analyzes multi dimensional attributes. We analyze the properties of each cluster divided into fatty liver, high risk and normal classes. The classification rules are generated by the analysis. In this paper, we suggest a method to diagnosis and predict liver condition to alcoholic patient according to risk levels using the classification rule from the new results of blood test. The K-mean classifier has been found to be more accurate for the result of blood test and provides the risk of fatty liver to normal liver conditions.

PLGA Microspheres in Hyaluronic Acid Gel as a Potential Bulking Agent for Urologic and Dermatologic Injection Therapies

  • In this study, we investigated whether PLGA microspheres in combination with hyaluronic acid (HA) gel have appropriate properties as a bulking agent for urologic injection therapies and whether the implantation of PLGA microspheres and HA gel induces angiogenesis in the newly formed tissues. In order to investigate whether this bulking agent is injectable, this material was injected through 24-gauge needles into the subcutaneous dorsum of the mouse. The bulking agent was easily injected without needle obstruction. Histological analyses of the hybrid tissues at 2 weeks showed that host cells at the surrounding tissues migrated into the spaces between the implanted PLGA microspheres and formed tissue-like structures. An inflammatory response to the implants was mild at 2 weeks and diminished at 8 weeks. Importantly, extensive ingrowth of blood vessels was observed in the hybrid tissues formed by the injection of PLGA microspheres and HA, whereas blood vessels rarely formed in the hybrid tissues formed by the injection of PLGA microspheres only. The implant volume was conserved for almost the entire implantation period. Histological analyses of the distant organs of the bulking agent-implanted animals, such as the lungs, liver, heart, brain, kidney, and spleen, showed no evidence of the injected microsphere migration. These results show that PLGA microspheres in combination with HA possess the appropriate characteristics for a bulking agent for urologic injection therapies and induce extensive blood vessel formation in the hybrid tissues.

Proteomic Analysis of the Hydrophobic Fraction of Mesenchymal Stem Cells Derived from Human Umbilical Cord Blood

  • Mesenchymal stem cells (MSCs) are promising candidates for cell therapy and tissue engineering, but their application has been impeded by lack of knowledge of their core biological properties. In order to identify MSC-specific proteins, the hydrophobic protein fraction was individually prepared from two different umbilical cord blood (UCB)-derived MSC populations; these were then subjected to two-dimensional (2D) gel electrophoresis and peptide mass fingerprinting matrix-assisted laser desorption/ionization (MALDI)-time of flight (TOF)-mass spectrometry (MS). Although the 2D gel patterns differed somewhat between the two samples, computer-assisted image analysis identified shared protein spots. 35 spots were reliably identified corresponding to 32 different proteins, many of which were chaperones. Based on their primary sub-cellular locations the proteins could be grouped into 6 categories: extracellular, cell surface, endoplasmic reticular, mitochondrial, cytoplasmic and cytoskeletal proteins. This map of the water-insoluble proteome may provide valuable insights into the biology of the cell surface and other compartments of human MSCs.

  • Poly(ethylene glycol)s (PEGs) are unique in their material properties, such as biocompatibility, non-toxicity, and water-solublizing ability, which are extremely useful for a variety of biomedical applications. In addition, a variety of functional PEGs with specific functionality at one or both chain ends have been synthesized for many specialized applications. Surface modifications using PEG have been demonstrated to decrease protein adsorption and platelet or cell adhesion on biomaterials. Furthermore, PEGs having anionic sulfonate terminal units have been proven to enhance the blood compatibility of materials, which has been demonstrated by the negative cilia concept. The preparation of telechelic PEGs having a sulfonic acid group at one end and a polymerizable methacryloyl group at the other is an interesting undertaking for providing macromers that can be used in various vinyl copolymerization and gel systems. In this paper, preliminary results on the synthesis and polymerization behavior of a novel PEG macromer is described with the aim of identifying a biocompatible material for applications in various blood-contacting devices.

  • 현재 암 환자들을 대상으로 하는 항암치료법은 나노입자, 폴리머 중합체, 지질, 리포솜 등을 치료 전달체로 이용하여 항암치료를 진행하는 방법들이 주로 활발하게 사용되고 있다. 이러한 전달체는 항암 치료제를 직접 암세포로 정확하게 표적 운반하는 정확성, 정확하게 운반한 후 선택적으로 항암 치료제를 방출해야하는 유출제어, 다른 일반 세포들을 약물로부터 안전하게 보호하는 기능 등을 동시에 가지고 있어야 하지만, 대부분 생산과정에서 많은 유해한 화학약품을 사용하며 이로 인한 독성을 유발하는 많은 사례가 빈번하게 발생하고 있다. 따라서 본 논문에서는 겸형 적혈구를 응용한 새로운 항암 전달체로서의 가능성을 타진하고 새로운 항암치료의 방법을 제시하고자 한다.

The Effect of Cortisol on Proliferative Properties of Flounder (Paralychthys Olivaceus) B Lymphocytes

  • Flounder B lymphocytes isolated from different tissues were studied in terms of cell proliferation, apoptosis and the effects of cortisol on these processes. B lymphocytes, isolated from the flounder head kidney and spleen, were characterized by higher proliferation and lower intracellular calcium ($Ca^2$) response to lgcrosslinking compared with peripheral blood B lymphocytes. Cortisol induced high levels of apoptosis (150% of control levels) in peripheral blood B lymphocytes, in combination with a stimulatory LPS signal. Head kidney and to a lesser extent spleen B lymphocytes, although less sensitive than their equivalent in peripheral blood, underwent cortisol-induced apoptosis irrespective of extra stimulation up to 142% of control levels. Also proliferation with and without LPS stimulation was suppressed by cortisol (compared to plasma values measured during stress conditions) that is effective in inducing a significant increase in apoptosis in all three populations of B-cells, suggesting that cortisol may be important for immunoregulation in both stressed and non-stressed conditions. This implies possible severe impact of stress on lymphocyte development and activity, Different sensitivity of B-cells to the corticosteroid, with respect to developmental stage and activity, may prevent excessive and long lasting depletion of B-lymphocytes.

Sedation with dexmedetomidine in elderly patients during dental surgery: a retrospective case series

  • Objectives: The number of elderly patients with systemic basal disease requiring invasive dental treatment has increased. Appropriate prediction of surgical invasiveness and combined use of psychosedation are thought to contribute to safe whole-body management. Dexmedetomidine (DEX) exhibits analgesic and anti-anxiolytic properties and causes mild respiratory depression. Studies regarding DEX use in elderly non-intubated patients are scarce. We aimed to use retrospective data to determine an effective dose of DEX to induce adequate sedation in elderly patients undergoing invasive dental surgery under local anesthesia. Materials and Methods: One hundred two patients aged 70 to 96 years were presumably appropriately controlled with sedation. DEX was administered at an initial loading dose of 2.0 to $3.1{\mu}g/kg/hr$ for 10 minutes. We divided the patients into five groups by age and compared their blood pressures and heart rates. Results: In all five groups, blood pressure decreased suddenly at approximately 15 and 20 minutes after DEX administration. A marked decrease in blood pressure was noted in patients aged 75 to 79 years. Conclusion: For elderly patients aged 75 years and above, the initial loading dose of DEX needs to be reduced to lower than half that required for young and middle-age adults.

Isolation of Peripheral Blood-Derived Mesenchymal Stem Cells in Mares and Foals

  • Peripheral blood-derived mesenchymal stem cells (PB-MSCs) have shown promise in cell-based therapy, as they can be harvested with ease through minimally invasive procedures. This study aimed to isolate PB-MSCs from foals and mares and to compare the proliferation and cellular characteristics of the PB-MSCs between the two groups. Six pairs of mares and their foals were used in this study. MSCs were isolated from PB by direct plating in a tissue culture medium, and cell proliferation (population doubling time [PDT], and colony-forming unit-fibroblast assay [CFU-F]), and characterization (morphology, plastic adhesiveness, colony formation, trilineage differentiation) were examined. There was no significant difference in the PB-MSC yield, CFU-F, and PDT between the mares and foals. PB-MSCs from both mares and foals showed typical MSC characteristics in terms of spindle-shaped morphology, plastic adhesive properties, formation of colonies, trilineage differentiation. These results suggest that PB-MSCs isolated from horses, both adult horses, and foals, can be used for equine cell-based therapy.

MTA의 Myths & Facts (The Myths and Facts of MTA)

  • Since MTA has many beneficial properties such as biocompatibility, great sealing capacity, antibacterial effects, low cytotoxicity, and stimulation of formation of mineralized tissue, it has been widely used as the material of choice in root-end filling, apexification, pulpotomy, perforation repair and so on. However, despite its favorable characteristics, MTA presents working properties which are less than ideal. The resulting cement from the mixing of powder and water is difficult to manipulate, and its setting time has been reported to be 2 h 45 min whereas the working time is <4 minutes. Additional moisture is also required to activate the setting of the cement. Moreover, according to recent studies, the physical properties of MT A may be hampered by acidic environment or blood contamination. Therefore, practitioners may have surprisingly worse results than they expected when they are not fully acquainted with the characteristics and manipulation method of MTA.
