Enzymatic Determination of Somatic Cells by Using Transparisation in Raw Milk

  • The transparisation technology for milk and milk products could be applied widely and very importantly to various determination because transparisation can economize the cost and increase with precision in the milk payment system. Component of butanone or Triton in transparisation solvent would inhibit the growth of bacteria and method. Enzymatic determination of leucocytes were proposed to evaluate milk quality as mastitis in the milk payment system, this can be easily applied to simplify automation of the determation with the lowest investment cost in milk pay system. The significance of this technique, it can be used in the quality control of raw milk and milk products, milk payment system, and programming of national dairy project. Transparisation technology is used in somatic cell counting by enzymic methods. The range of deviation for this method is 16% in 74 samples. But the deviation is increased to 20% when the Infoss method is used. It is affected by the percentage of epithelial cells and white blood cells in somatic cells from different animals and the stages of aging. NAgase activity has an obvious correlation with white-blood cells in milk. In the case of mastitis the white-blood cells is 90-95% in somatic cells in milk, it is showing greater precision in measuring the state of mastitis. In conclusion the enzymic method of somatic cell counting is a relatively simple and easy method of measurement and can be easily practiced. And the importance of this method is also worth utilizing for indirect counting of Somatic cells by use of synthetic substrates to NAgase. In the future, with the further development of the research in this field, it will b possible to automatize the measurement.

Protective Effects of Leaf and Flower Extracts from Cirsium japonicum var. ussuriense on Oxidative Damage in Normal Human Erythrocytes and Plasma (엉겅퀴(Cirsium japonicum var. ussuriense) 잎 및 꽃 추출물이 정상인 적혈구와 혈장의 산화적 손상에 대한 보호효과)

  • Cirsium japonicum var. ussuriense is often used in treatment of human disease such as hemorrhage, blood congestion and inflammation. This study was accomplished to evaluate the antioxidant properties of the leaf (CLE) and flower (CFE) extracts of C. japonicum var. ussuriense to protect normal human red blood cells (RBC) and plasma samples against oxidative damage in vitro. CLE and CFE were prepared by extracting with hot water. In red blood cells and plasma, oxidative hemolysis and lipid peroxidation induced by the aqueous peroxyl radical generator [2,2'-azobis (2-amidinopropane) dihydrochloride, AAPH] were significantly suppressed by CLE or CFE in a dose-dependent manner at the same time. CLE and CFE also prevented the depletion of cytosolic antioxidant glutathione (GSH) in RBC. These results suggest that the leaves and flowers of C. japonicum var. ussuriense may have the antioxidant properties.

Effects of porcine blood plasma on the emulsion stability, physicochemical characteristics and textural attributes of emulsified pork batter

  • This study was conducted to determine the effects of addition of porcine blood plasma (PBP) to the emulsified pork batter as a substitute for the soy protein isolate (SPI) or sodium caseinate (SC) on the emulsion stability and physicochemical and textural properties of the emulsified pork batter. A total of 10 treatments were no addition and 0.5%, 1.0%, and 1.5% addition with each of SPI, SC, and PBP. The moisture and fat losses of the pork emulsion after cooking decreased with increasing percentage of any of SPI, SC, and PBP (p < 0.05). Further, moisture loss was less for the PBP treatment than for SPI and SC (p < 0.05). The lightness, redness, and whiteness of the emulsified pork batter decreased (p < 0.05) due to any of the SPI, SC, and PBP treatments whereas the yellowness and the chroma and hue values increased. The lightness, redness, yellowness, and chroma and hue values differed also among the SPI, SC, and PBP treatments (p < 0.05); however, the numerical difference between any two types of substitutes was less than 8% of the two corresponding means in all of these variables. Textural properties, including the hardness, cohesiveness, springiness, gumminess, chewiness, and adhesiveness, were not influenced by any of the SPI, SC, and PBP treatments (p > 0.05), except for greater gumminess and chewiness for the PBP treatment than for SC. The present results indicate that PBP is comparable or even superior to SPI or SC in its emulsion-stabilizing effect and therefore could be used a substitute for the latter as a non-protein ingredient of pork emulsion batter.

Structure and properties of silk sericin obtained from different silkworm varieties

  • Recently, many researchers have studied silk sericin because of its high water retention, good wound healing, good cyto-compatibility, and blood-glucose- and cholesterol-lowering effects. Although sericin film can be used in wound dressing and cosmetic packs, its poor mechanical properties have prevented its use in industrial fields. In the present study, sericin was obtained from different silkworm varieties, and the effect of silkworm variety on the structure and properties of sericin was examined. Except for a small difference in serine content, no significant difference in sericin was noted among the silkworm varieties. In addition, silkworm variety almost had no effect on solution viscosity, implying that it does not influence the molecular weight of sericin. Mechanical properties of sericin film were strongly affected by silkworm variety. Wonwon 126 showed the best mechanical properties, while N74 and Geumgwangju displayed the worst properties.

Transfer of Orally Administered Terpenes in Goat Milk and Cheese

  • The objective of the present study was to investigate the relationships between terpenes… intake and their presence in animal tissues (blood and milk) as well as in the final product (cheese). Eight dairy goats were divided in two balanced groups, representing control (C) and treatment (T) group. In T group oral administration of a mixture of terpenes (${\alpha}$-pinene, limonene and ${\beta}$-caryophyllene) was applied over a period of 18 d. Cheese was produced, from C and T groups separately, on three time points, twice during the period of terpenes… oral administration and once after the end of experiment. Terpenes were identified in blood by extraction using petroleum ether and in milk and cheese by the use of solid phase micro-extraction (SPME) method, followed by GC-MS analysis. Chemical properties of the milk and the produced cheeses were analyzed and found not differing between the two groups. Limonene and ${\alpha}$-pinene were found in all blood and milk samples of the T group after a lag-phase of 3 d, while ${\beta}$-caryophyllene was determined only in few milk samples. Moreover, none of the terpenes were traced in blood and milk of C animals. In cheese, terpenes' concentrations presented a more complicated pattern implying that terpenes may not be reliable feed tracers. We concluded that monoterpenes can be regarded as potential feed tracers for authentification of goat milk, but further research is required on factors affecting their transfer.

Effect of the Erigeron annuus In Vitro Antioxidant Properties and Extract on Serum Lipid in Mice (개망초 추출 분말의 In Vitro에서의 항산화 특성과 고지방식이로 유도된 Mice의 혈청지질 수준에 미치는 영향)

  • This study was conducted to verify the effects of Erigeron annuus powder on serum lipid levels of high-fat diet-induced mice from a nutritional viewpoint. Erigeron annuus powder has been used as a folk remedy since ancient times in Korea. There was no significant difference in the weight of the kidneys and spleens of the mice. The high-fat diet group had a significantly higher kidney weight compared to other groups (p<0.05). In the group of mice fed 20% Erigeron annuus powder with a high-fat diet, the concentration of serum LDL-cholesterol was high (p<0.05), whereas the concentration of triglyceride was remarkably lower compared to other groups (p<0.05). The group fed 10% Erigeron annuus with a high-fat diet had the lowest concentration of blood phospholipids (p<0.05) as well as the highest alkaline phosphatase and alanine aminotransferase levels in blood (p<0.05). There was no difference in blood insulin concentration. However, blood leptin concentration was significantly higher ($5.88{\pm}3.53ng/dL$) in mice fed a high-fat diet compared to other groups (p<0.05). Measurements of Erigeron annuus revealed that TPC, $ABTS^+$ radical scavenging activity of trolox, DPPH radical scavenging activity, and the measured value of FRAP were higher in the ethanol extract than in the water extract. Especially, the antioxidant activity effects were excellent for the $ABTS^+$ radical scavenging activities of trolox and FRAP values of Erigeron annuus. Therefore, Erigeron annuus powder showed antioxidant activity. Hence, Erigeron annuus powder drastically lessened triglyceride concentration in blood in high-fat diet-induced mice. Thus, the powder is considered to have utility in the food processing industry. Additional related experiments are ongoing.

  • This study was carried out to understand the effects of Bambusae Caulis in Liquamen manufactured by different production facilities on blood glucose of the mice induced with streptozotocin. Filtered Bambusae Caulis in Liquamen manufactured by two different facilitiesproduction process were used. The physiochemical properties of filtered Bambusae Caulis in Liquamen manufactured by two different facilities(300L×300W×400H and 700L×700W×800H electric furnace) were compared. Furthermore, the effects of filtered Bambusae Caulis in Liquamen were observed in terms of blood glucose, BUN and AL T in the mice induced with streptozotocin. The results were as follows : The pH value was the highest in L-BCL(E)B(pH 2.83) between manufactured by two different facilities. The L-BCL(E)B which observed low values of Hunter's color values show decreased concentration of methanol and phenolic compounds. The blood glucose contents was decreased in the experimental groups compared with the control. The amount of BUN and ALT did not show any differences between control and experimental group.

  • Conventional drug carriers such as liposomes, nanoparticles, polymer micelles, polymeric conjugate and lipid microemulsion for cancer chemotherapy shield normal tissues from toxic drugs to treat cancer cells in tumors. However, inaccurate tumor targeting uncontrolled drug release from the carriers and unwanted accumulation in healthy sites can limit treatment efficacy with current conventional drug carriers with insufficient concentrations of drugs in the tumors and unexpected side effects as a result. Sickle red blood cells show natural tumor preferential accumulation without any manipulation due to the adhesive interaction between molecular receptors on the membrane surface and counter-receptor on endothelial cells. In addition, structural changes of microvascular in tumor sites enhances polymerization of sickle red blood cells. In this research, we examined the use of sickle red blood cells as a new drug carrier with novel tumor targeting and controlled release properties to quantify its therapeutic effects.

Effect of age, season and sex on the blood properties in the Cheju horse (제주마(濟州馬)의 혈액성분(血液成分)에 미치는 연령(年齡), 계절(季節) 및 성(性)의 영향(影響))

  • The values of blood composition were analyzed by age, season, and sex from Chejuhorse in Korea. The results obtained were summarized as follow; 1. The values of white blood cell(WBC), packed cell volume(PCV), mean corpuscular volume (MCV), and mean corpuscular hemoglobin concentration(MCHC) were significantly different between age. The WBC value was decreased with the progress of age and the PCV value was lower in the early age of 1,2 to 3 years than in the higher age of 4 to 5, and over 6 years. 2. The mean value of MCV was increased with the progress of age but the adverse was true in MCHC. 3. In the case of seasons, the values of red blood cell(RBC), WBC, hemoglobin(Hb) PCV, MCV, mean corpuscular hemoglobin(MCH), MCHC, globulin(Glo), albumin/globulin(A/G) ratio, inorganic phosphorus(Pi), cholesterol(Chol), and chlorine(Cl) showed significant differences. The number of RBC was not varied from winter to summer but decreased in autumn. The number of WBC was high in summer and autumn, but low in winter and spring. 4. The values of MCV were high in autumn and winter, but low in spring and summer. 5. The values of MCH and MCHC were similar in spring and summer, but strikingly increased in autumn. 6. The values of Glo and A/G ratio were negatively correlated with the season; The value of A/G ratio was lowest in summer but no differences were observed in spring, autumn and winter. On the other hand, the value of Glo was highest in summer but no differences were observed in spring, autumn, and winter. 7. The values of Pi, Chol, and CI were high in summer and winter, but low in spring and autumn. 8. The values of RBC, MCV, and MCH were significantly different between sex. The value of the RBC count in male was higer than the female, but the adverse is true in MCV, and MCH.

Antioxidant Activity of Rosa rugosa Thunberg and Effect on Serum Lipid Level in High Fat Diet-induced Mice (해당화 열매 추출 분말의 항산화 활성과 고지방 식이로 유도된 Mice의 혈청지질 수준에 미치는 영향)

  • Since Korean ancient times, powder of Rosa rugosa Thunberg has been used as a folk remedy. This study was conducted to verify the effects of Rosa rugosa Thunberg powder on in vitro antioxidant properties and serum lipid levels of high-fat diet-induced mice from a nutritional viewpoint. In the case of Rosa rugosa Thunberg powders, measurement of TPC, ABTS radical scavenging activity of trolox, DPPH radical scavenging activity, and measured value of FRAP were higher in ethanol extract than water extracts. For LDL-cholesterol concentration, mice fed 10% powder of Rosa rugosa Thunberg with high-fat diets showed a high numerical value compared with other groups (p<0.05). When testing for triglyceride concentrations in blood, mice fed 10% and 20% powder of Rosa rugosa Thunberg with high-fat diets showed the lowest numerical values (p<0.05). When testing for blood insulin concentrations, the high-fat diet group showed higher levels than compared to the control group (p<0.05). When testing for blood leptin concentrations, the high-fat diet group was $5.88{\pm}3.53ng/dL$, whereas mice fed 10% powder of Rosa rugosa Thunberg with high-fat diets showed a blood leptin level of $10.36{\pm}5.96ng/dL$ (p<0.05). Therefore, results prove that powder of Rosa rugosa Thunberg reduces triglyceride concentrations in the blood, and could be used as an excellent natural antioxidant in the future.
