Effect of Cnidium officinale Extract on Recovery Capability of Allergic Contact-Dermatitis in Rat (흰쥐의 알레르기성 접촉피부염의 회복에 미치는 천궁추출물의 영향)

  • The epidermal recovering-effects of the Cnidium officinale extract after inducing contact-dermatitis by 1-chloro-2,4-dinitrochlorobenzene (DNCB) showed by measuring thickness of skin, identifying IgE amounts in plasma, and activities of anti-oxidant enzymes using histochemical and biochemical methods. By three-time applications of the extract, morphological changes of dermatitis in impaired region were recovered dramatically and shape of skin surface and thickness of epidermis were restored to be normal. Also, lipid content was recovered to the level of normal state. We suggested that extract treatment lowered a hypersensitive reaction by decreasing of IgE level in blood and restored activities of superoxide dismutase and catalase. In conclusion, we proposed that C. officinale extract might be used as natural resources for treating effectively allergic contact-dermatitis.

Pharmacokinetics and Bioequivalence of Haloperidol Tablet by Liquid Chromatographic Mass Spectrometry with Electrospray Ionization

  • The purpose of this study is to investigate the bioequivalence of two haloperidol 5 mg tablets, Myung In haloperidol (Myung In Pharm. Co., Ltd., test drug) and $Peridol^{R}$(Whanin Pharm. Co., Ltd., reference drug), and also to estimate the pharmacokinetic parameters of haloperidol in Korean volunteers. The bioavailability and pharmacokinetics of haloperidol tablets were examined on 24 healthy volunteers who received a single oral dose of each preparation in the fasting state in a randomized balanced 2 way crossover design. After an oral dosing, blood samples were collected for a period of 60 h. Plasma concentrations of haloperidol were determined using a liquid chromatographic electrospray mass spectrometric (LC-MS) method. The pharmacokinetic parameters were calculated with noncompartmental pharmacokinetic analysis. The geometric means of $AUC_{0-60h} and C_{max}$ between test and reference formulations were $17.21\pm8.26 ng\cdot/mL vs 17.31\pm13.24 ng\cdot/mL and 0.87\pm0.74 ng/mL vs 0.85\pm0.62 ng/mL$. respectively. The $90\%$ confidence intervals of mean difference of logarithmic transformed $AUC_{0-60h} and C_{max} were log0.9677{\sim}log1.1201 and log0.8208{\sim}log1.1981$, respectively. It shows that the bioavailability of test drug is equivalent with that of reference drug. The geometric means of other pharmacokinetic parameters ($AUC_{inf}. t_{1/2}, V_{d}/F, and CL/F$) between test drug and reference drug were $21.75\pm8.50 ng{\cdot}h/mL vs 21.77\pm15.63 ng{\cdot}h/mL, 29.87\pm8.25 h vs 29.60\pm7.56 h, 11.51\pm5.45 L vs 12.90\pm6.12 L and 0.26\pm0.09 L/h vs 0.31\pm0.17 L/h$, respectively. These observations indicate that the two formulation for haloperidol was bioequivalent and, thus, may be clinically interchangeable.

The Changes of NO, nNOS, Norepinephrine by Acupucture at LU7, HT5, PC6 Acupoints in Rats (수삼음경의 락혈 침자가 백서의 혈위 조직내 nNOS. NO와 조직 및 혈장 Norepinephrine의 변화에 미치는 영향)

  • Objectives : A previous study demonstrated that the connecting points of three yang meridians attenuated changes of nNOS, and Norepinephrine(NE) in rats. The current study investigated the changes in nNOS, NO and NE upon the needle insertion at varying depths at the connecting point of three yin meridians of the hand. Methods : Needles were inserted into rats, on both left and right sides of the connecting point, including the LU7, HT5 and PC6 acupoints which are three yin meridians of the hand. After insertion, needles were retained for five minutes. Each acupuncture groups were treated acupuncture at each acupoint and at the depths of superficial, middle and deep layer. After the retention, blood was drawn via cardiac puncture, and tissues of each point near meridian vessel was extracted to examine the changes in the expression of nNOS, NO and NE. Results : Compared with the superficial layer group, nNOS production significantly decreased in the middle and deep layer at LU7 acupoint group and in the deep layer at HT5, PC6 acupoint group. The tissue NE decreased in the deep layer on PC6 acupoint and the plasma NE increased at the middle layer at LU7 acupoint group but decreased at the deep layer on at LU7 acupoint group. Conclusions : Acupuncture at connecting points of three yin meridians of the hand can regulate the activities of nNOS, and NE.

Bioequivalence of Paroxetine Tablet to Seroxat Tablet (Paroxetine 20 mg) (세로자트정(파록세틴 20 mg)에 대한 삼천리파록세틴정의 생물학적동등성)

  • Paroxetine, a potent and selective serotonine reuptake inhibitor, has been used for the treatment of depression, obsessive-compulsive disorder, panic disorder and social phobia. The bioequivalence of two paroxetine preparations was evaluated according to the guidelines of Korea Food & Drug Administration (KFDA). The test product was Samchully Paroxetine $tablet^{\circledR}$ made by Samchully Pharm. Co. and the reference product was Seroxat $tablet^{\circledR}$ made by GlaxoSmithKline. Twenty healthy male subjects, $22.4{\pm}2.6$ years old and $63.8{\pm}4.2\;kg$, were divided into two groups and a randomized $2{\times}2$ cross-over study was employed. After one tablet containing 20 mg paroxetine was orally administered, blood was taken at predetermined time intervals and the concentration of paroxetine in plasma was determined using a validated HPLC method with fluorescence detector. Two pharmacokinetic parameters, $AUC_t$ and $C_{max}$, were calculated and analyzed statistically for the evaluation of bioequivalence of two products. Analysis of variance was carried out using logarithmically transformed parameter values. The 90% confidence intervals of $AUC_t$ and $C_{max}$ were log 0.84-log 1.16 and log 0.85-log 1.14, respectively. These values were within the acceptable bioequivalence intervals of log 0.8 to log 1.25. Thus, the criteria of the KFDA guidelines for the bioequivalence was satisfied, indicating that Samchully Paroxetine tablet is bioequivalent to Seroxat tablet.

Antioxidant and Antimicrobial Activities of Camellia Oleifera Seed Oils

  • The antioxidant and antimicrobial activities of Camellia oleifera seed oil were studied. Four kinds of seed oil samples were prepared, crude oil and refined oil, extracted by cold pressing method (CPC, CPR), and organic solvent extraction (OSC, OSR). Antioxidant activity analysis was measured in 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid)-diammonium salt, ferric reducing Ability of Plasma, and 2,2-diphenyl-1-picrylhydrazyl assays. Besides, the percentage of inhibition of red blood cells hemolysis induced by 2,2'-azobis(2-amidnopropane) dihydrochlorid, the lag time of LDL conjugated dienes formation in vitro, and the inhibitors of loss in tryptophan fluorescence were all used to estimate the antioxidant activity of the samples. The total phenolic contents (TPC) were detemined by Folin-Ciocalteu method. The TPC of the C. oleifera seed oils can be arranged in descending order: CPC ($1.9172{\mu}g/mL$) > OSC ($1.5218{\mu}g/mL$) > CPR ($1.0611{\mu}g/mL$) > OSR ($0.6782{\mu}g/mL$). And the oils were investigated for activity against Escherichia coli, Bacillus subtilis, Saccharomyces cerevisiae and Aspergillus niger. The results showed the antioxidant activity of crude oil by cold pressing method was stronger than others, and all oils did inhibit activity of the top three bacteria expert A. niger. The further significance of the study contributes to measure the antioxidant and antimicrobial activity of the potential health benefits by the different methods of preparation and the oil of C. oleifera seeds acting as free radical scavenger, pharmaceuticals and preservatives may offer some information in medicine and cosmetic not just in food field.

Pharmacological Effects of the Allium victorialis var. platyphyllum Extracts on the Rats Induced by Streptozotocin, Poloxamer-407, $CCl_4$ and D-Galactosamine (Streptozotocin, Ploxamer 407, $CCl_4$ 및 D-Galactosamine으로 유도한 병태모델 흰쥐에 대한 산마늘 추출물의 약리효과)

  • To develop Allium victorialis var. platyphyllum (Liliaceae) for an available functional food, pharmacological experiments on the extracts of this plant were undertaken in the rat treated with streptozotocin, poloxamer-407 and $CCl_4$. The two MeOH extracts were obtained from the leaves and the bulbs, respectively. The three agents, streptozotocin, poloxamer-407 and $CCl_4$, were treated to induce diabetes mellitus, hyperlipidemia and hepatic injury, respectively. Treatment with the leaf extract lowered blood glucose by 24.9% at 200 mg/kg (p.o.) in the STZ-treated rat and prohibited the increase of body weight, water consumption and food intake. This exσact also significantly decreased not only thε plasma cholesterol and triglyceride in the poloxamer-407-treated rat by 35.3% but also serum ALT by 49.0%. The potency was found by overall estimation on the experimental results as followings: 200 mg/kg leaf extract> 200 mg/kg bulb extract> 100 mg/kg leaf exσact> 100 mg/kg bulb extract. These results suggested that this plant might have the availability for a functional food. It was also suggested that the leaves are more effective than the bulbs for the functional vegetable, especially in hyperlipidemia and hepatic injury.

Anti-hypertensive Effects of ethanol extract of Phyllostachys Pubescens via Antioxidant Activity (맹종죽의 항산화활성을 통한 항고혈압 효능)

  • Phyllostachys pubescens (Maengiong-Juk), a kind of the bamboo, was reported to have many beneficial pharmacological actions. in this study, of using 70% ethanol extract of Phyllostachys pubescens we investigated its efficacy on angiotensin converting enzyme (ACE) and antioxidant enzyme activities. In addition, vasorelaxant effect was examined in rat aortic rings. The inhibitory effect of ACE activity by Phyllostachys pubescens extract (PPE) was dose-dependently increased by 61.42% at 10mg/ml. PPE relaxed the pre-contracted rat aortic rings with 10$^{-6}$M phenylephrine, showing about 88% at 4.0mg/ml. Sprague Dawley (SD) rats were given different concentrations of PPE mixed in the drinking water for 10 weeks. PPE did not show any difference with control group in blood pressure, body weight (BW) and food intake. However, it revealed the highest total antioxidative effect at dose of 1.0 g/100 g BW in plasma by TEAC assay. Thiobarbituric acid reactive substance (TBARS) and protein carbonyl levels which are markers of tissue peroxidation, were significantly lowed at the same dosage. Furthermore, hepatic antioxidant enzymes such as total superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and catalase activities were also significantly increased by PPE (1.0 g/100 g BW). In conclusion, we suggest that PPE might have antihypertensive effect through increasing antioxidant activities.

Clinical Findings and Gene Analysis of BH4 Responsive PKU Patients in Korea (BH4 responsive PKU 환자들의 임상적 특성과 유전자분석)

  • Purpose: Phenylketonuria (PKU) is the first inherited metabolic disease of which treatment is known. We performed this study to find out clinical symptoms and prognosis of tetrahydrobiopterin (BH4) responsive PKU patients and genetic relation. Methods: Clinical, biochemical, genetic analysis were done retrospectively in 23 patients diagnosed BH4 responsive PKU in Soonchunhyang University Hospital from March 2000 to September 2012. Results: Patients were classified to mild hyperphenylalaninemia and mild phenylketonuria with initial plasma phenylalanine level below 20 mg/dL. After BH4 loading, blood phenylalanine decrease level ranged between 37% and 99%. Initial treatment with low phenylalanine formula or BH4 was started before 2 month after birth except 2 patients. And one of them resulted in developmental delay in language and social activity. The others showed satisfactory progress without developmental delay. In genetic analysis, of 46 allele, R241C allele mutation was identified most commonly (41%). R241C/A259T, R241C/R243Q, R241C/V388M, R241C/T278I was detected in 5 (21.7%), 3 (13%), 2 (8%), 2 (8%) patients, respectively. Conclusion: R241C mutation was detected most frequently in this study group and R243Q mutation which is known to be prevalent in Korean PKU patients was found in 4 patients (8.6%). Early diagnosis and treatment is important in PKU patients.

The Effect of Endogenous and Exogenous Estrogen on Risk Factors of Coronary Heart Disease -Part II : On Platelet Aggregation- (내인성 및 외인성 Estogen이 관상동맥질환 위험인자에 미치는 영향 -제2보: 내인성 및 외인성 Estrogen이 혈소판 응집에 미치는 영향-)

  • In the atherosclerotic subjects, arterial endothelial cell injury and plaque formation are suspected to be strong causable factors in developing acute coronary syndrome, and it was revealed that platelets have a very important role in this case. Women are exposed to atherosclerosis at a different degree after menopause or oral contraception. The purpose of this study was to determine the effects of endogenous and exogenous estrogen on the degree of platelet aggregation in platelet rich plasma(PRP) in twenty nonsmoking healthy Korean women for 12 weeks. The subjects were assigned to three groups: (1) eight women aged 49 to 60(yr) for the postmenopausel(Pst) group, (2) eight, aged 22 to 30(yr) for the premenopausa(Pre) group, (3) four, aged 23 to 30(yr) for the oral contraceptive (OC) group which used triphasic OC formulation. Fasting blood sample were obtained from the subjects, (1) once per 6 weeks in the Pst group, (2) every phase of the menstrual cycle in the Pre group, (3) each once during and after OC administration in the OC group. ADP, collagen and epinephrine were used as aggregating reagents, and platelet aggregation and time(Δt: time reaching to the maximum point of aggregation) in PRP were measured at the maximum point of aggregation in five minutes. All the data were adjusted for dietary effects, personality type and body mass index(BMI) by using analysis of covariation(ANCOVA). Platelet aggregation to ADP and collagen(MADP and MCOLL) were not significantly different among the three groups, and Δt to ADP and collagen(TADP and TCOLL) were not either. But maximum platelet aggregability and Δt to epinephrine(MEPIN and TEPIN) were significantly different among the three groups, and the OC group showed the lowest value (p<0.01). Maxtimum platelet aggregability and Δt during the menstrual cycle were not significantly different in the Pre group. Any other significant differences in the maximum platelet aggregability and Δt were found between oral contraception phase and washing out phase(menstruation) in the OC group. In results, maximum platelet aggregability and aggregation time to ADP and collagen seemed not to be affected by endogenous and exogenous estrogen, even though MEPIN and TEPIN showed significantly low in the OC group among the three groups.

Effect of Smoking and Carotenoid-rich Food Consumption on Lipid Peroxidation and Antioxidant Status (흡연과 Carotenoid 함유 식품 섭취빈도에 따른 지질과산화 및 항산화능 비교)

  • This study was performed to determine the effect of carotenoid-rich food consumption and smoking on the lipid peroxidation and antioxidant status in human. The subjects consisted of the health 210 middle-aged adults who visited health care center in the Inha University hospital. The blood and urine samples of the subjects were taken to analyze serum lipid profiles, plasma TBARS, total antioxidant status (TAS) and urinary 8-isoprostanes concentration. The anthropometric indices of the subjects were measured. The nutritional intake and the frequency of carotenoid-rich food consumption was determined by semi-quantitative food frequency questionnaire survey. HDL-cholesterol level of the smoking subjects was significantly lower than that of the non-smoking subjects in men. In the analysis of the carotenoid-rich food consumption, the frequency of pepper and tomato consumption of the non-smoking subjects was significantly higher than that of the smoking subjects in men. In women, the Sequency of pear and peach consumption of the non-smoking subjects was significantly higher than that of the smoking subjects. HDL-cholesterol level of the high carotenoid rich food consumption group was significantly higher than that of medium and low group in women. TAS of the high carotenoid rich food consumption group was significantly higher than that of medium and low group. In conclusion, carotenoid intakes seemed to be effective to reduce lipid peroxidation and increase antioxidant status in the body. The frequency of the carotenoid-rich food consumption seemed to be lower in the smokers than in the non-smokers. However, further researches consisted of large-scaled and randomized clinical trials are required to determine whether carotenoids have any other beneficial effect in human.
