Correlations between the Fatty Acid Composition of Serum Phospholipids and Blood Pressure

  • Studies of the relationship between the composition of serum fatty acids and blood pressure are complex and controversial. Fatty acids, important constituents of biological membranes, could potentially affect vasoreactivities including blood pressure. In this study the compositions of fatty acids in serum phospholipids were compared between three types of hypertensive subjects (men, pre-menopausal women, and post-menopausal women) and their respective nrmotensive controls. Serum lipids were extracted and phospholipids were separated by thin layer chromatography. The percentage of palmitic acid (16 : 0) in serum phospholipids was significantly higher and the percentage of stearic acid (18 : 0) was significantly lower in all three hypertensive groups, compared with their corresponding control groups. Only in the group of post-menopausal women, palmitic acid was closely associated wish increases in both systolic (SBP) and diastolic blood pressure (DBP), while stearic acid was associated with decreases in both SBP and DBP. The polyunsaturated fatty acids in serum phospholipids behaved differently from saturated fatty acids. The ratios of products / precursor fatty acids, such as $\sumLCPUFA\omega6/18 : 2\omega$6, 20 : 4$\omega$6/18 : 2$\omega$6, ∑LCPUFA$\omega$3/18 : 3$\omega$3 and 22 : 6$\omega$3/20 : 5$\omega$3, were all clearly associated with both SBP and DBP in hypertensive, post-menopausal women. Desaturation and elongation in fatty acid metabolism could affect the bioavailability of eicosanoid precursors. Changes in the constituent fatty acids of phospholipids and eicosanoid precursors may also influence fluidity, ionic transport, hormone receptors and enzyme activities in biological membranes. In conclusion, both systolic and diastolic blood pressure in post-menopausal women was positively associated with the level of palmitic acid, and negatively associated with the level of stearic acid, in serum phospholipids. The relationships between serum phospholipid-$\omega$6 and $\omega$3 series fatty acids and blood pressure in women, especially in post-menopausal women, require further investigation by taking into consideration hormonal status and eicosanoid metabolism. Funker study is needed to determine the value of dietary manipulation of fatty acid constituents of serum phospholipids, relating to hypertension in women.

Effects of IgY Supplementation on Hanwoo Calves Fed withHigh Quality Roughage (양질 조사료 급여에 따른 한우 송아지의 IgY 처리효과)

  • This study was conducted to examine the effect of IgY supplementation on growth performance, blood metabolism, and disease occurrence in the weaned calves at 3 or 4 month of age. Average daily gain (ADG) and feed intakes were not affected by IgY supplementation, regardless of weaning months. White blood cell (WBC) count of the control group was higher in the final stage than in the initial stage (p<0.05), while that of the IgY supplemented group was lower in the final stage compared to the initial stage (p<0.05). regardless of treatments. Hematocrit (HCT) and red blood cell (RBC) counts were lower in the final stage than in the initial stage (p<0.05), but mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH) counts were higher in the final stage than in the initial stage, regardless of treatments. In the control group, concentrations of albumin and globulin were higher in the final stage compared to the initial stage in the weaned calves at 3 month of age (p<0.05), and concentration of blood urea nitrogen (BUN) was increased in the IgY supplemented group. In the control group, concentrations of aspartate aminotransferase (AST) and gamma (${\gamma}$)-glutamyl transferase (GGT) were higher in the final stage compared to the initial stage (p<0.05). In contrast, concentrations of aspartate aminotransferase (AST) and gamma(${\gamma}$)-glutamyl transferase (GGT) were lower in the final stage compared to the initial stage for the IgY supplemented group. A diarrhea calf was only found in the control group. Manure score was higher in the control group compared to IgY supplemented group (p<0.05). Thus, these results indicates that IgY supplementation had positive effects on some blood metabolites, fecal condition score and diarrhea without negative effect on growth performance of the weaned Hanwoo calves.

Muscle oxygenation, endocrine and metabolic regulation during low-intensity endurance exercise with blood flow restriction

  • [Purpose] The present study investigated the effect of endurance exercise with blood flow restriction (BFR) performed at either 25% maximal oxygen uptake (${\dot{V}}O_2$ max) or 40% ${\dot{V}}O_2$ max) on muscle oxygenation, energy metabolism, and endocrine responses. [Methods] Ten males were recruited in the present study. The subjects performed three trials: (1) endurance exercise at 40% ${\dot{V}}O_2$ max without BFR (NBFR40), (2) endurance exercise at 25% ${\dot{V}}O_2$ max with BFR (BFR25), and (3) endurance exercise at 40% ${\dot{V}}O_2$ max with BFR (BFR40). The exercises were performed for 15 min during which the pedaling frequency was set at 70 rpm. In BFR25 and BFR40, 2 min of pressure phase (equivalent to 160 mmHg) followed by 1 min of release phase were repeated five times (5 × 3 min) throughout 15 minutes of exercise. During exercise, muscle oxygenation and concentration of respiratory gases were measured. The blood samples were collected before exercise, immediately after 15 min of exercise, and at 15, 30, and 60 minutes after completion of exercise. [Results] Deoxygenated hemoglobin (deoxy-Hb) level during exercise was significantly higher with BFR25 and BFR40 than that with NBFR40. BFR40 showed significantly higher total-hemoglobin (total-Hb) than NBFR40 during 2 min of pressure phase. Moreover, exercise-induced lactate elevation and pH reduction were significantly augmented in BFR40, with concomitant increase in serum cortisol concentration after exercise. Carbohydrate (CHO) oxidation was significantly higher with BFR40 than that with NBFR40 and BFR25, whereas fat oxidation was lower with BFR40. [Conclusion] Deoxy-Hb and total Hb levels were significantly increased during 15 min of pedaling exercise in BFR25 and BFR40, indicating augmented local hypoxia and blood volume (blood perfusion) in the muscle. Moreover, low-and moderate-intensity exercise with BFR facilitated CHO oxidation.

Effects of Water Extracts of Endocarps and Seeds of Omija(Schizandra chinensis Baillon)on Alcohol metabolism (오미자 과육과 종자의 물추출물이 알콜대사에 미치는 효과)

  • In this study the effects of water extracts of endocarps and seeds of omija(Schizandra chinensis Baillon)on alcohol metabolism in rats were investigated. 25% alcoholic solution(v/v,0.75g/200g B.W.)and 40% alcoholic solution(v/v,0.80g/200g B.W.)were orally administered to rats for 2 hours. The levels of metabolites and enzyme activities both on serum and liver were not changed by acute oral adminiatration of 25% and 40% alcoholic solution. Blood alcohol levels were significantly lowered by treatment of water extracts of endocarps and seeds of omija in rats administered with 25% and 40% alcohol. In 25% alcohol treated group, serum GPT level was decreased but hepatic G-6-P DH and pyruvate levels were increased. In 40% alcohol treated group, water extracts of endocarps and seeds of omija resulted in a significant decreased in serum levels of FFA, GPT but increased in serum glucose level. Hepatic levels of cytosolic protein, glycogen and pyruvate were increased by water extracts of omija parts in 40% alcohol treated group.

  • The positron emission tomography (PET) has been used for the evaluation of the characteristics of various tumors. The role of PET in oncology has been evolved from a pure research tool to a methodology of enormous clinical potential. The unique characteristics of PET imaging make sophisticated quantitation possible. Several quantitative methods, such as standardized uptake values (SUV), simplified quantitative method, Patlak graphical analysis, and Sokoloff's glucose metabolism measurement, have been used in the field of oncology. However, each quantitative method has limitations of its own. For example, the SUV has been used as a quantitative index of glucose metabolism for tumor classification and monitoring response to treatment, even though it depends on blood glucose level, body configuration of patient, and scanning time. The quantitative methods of PET are reviewed and strategy for implementing these methods are presented.

  • Urine contains protein and nucleic acid(urea, uric acid, creatinine, ammonia, amino acids), various organic and inorganic materials, vitamin, hormone, enzyme etc. The examination of gualitative or quantitative change of the above mentioned materials and picking up the abnormal materials are useful to diagnose diseases. The test strip for examination of urine is applied to the routine test, monitoring of medical therapy and recurrence, self monitoring, and screeing in preventive medicine. We have been using multitest strip for checking the bacterial infection(nitrite), PH, protein, glucose, ketone body, urobilinogen, bilirubin and occult blood. So it is possible to diagnose three groups of diseases as follows, abnormal metabolism of the carbohydrate, diseases of kidney and other urogenital system, diseases of hepatobiliary system and hemolytic disease causing abnormal metabolism of bile juice.

Identification of Novel Metabolic Proteins Released by Insulin Signaling of the Rat Hypothalmus Using Liquid Chromatography-Mass Spectrometry (LC-MS)

  • Objective : The brain is dependent on glucose as an energy source. Intricate homeostatic mechanisms have been implicated in maintaining the blood glucose concentration in the brain. The aim of this study is to find the way to identify the metabolic proteins regulating the glucose in rat hypothalamus. Methods : In this study, we analysed the secretome from rat hypothalamus in vivo. We introduced 500 nM of insulin into the rat hypothalamus. The chromatographic patterns of the secretome were identified, after which Mass Spectrometry-Mass Spectrometry (MS-MS) analysis was performed. Results : In Liquid Chromatography-Mass Spectrometry (LC-MS) analysis, 60 proteins were identified in the secretome. Among them, 8 novel proteins were unveiled and were associated with the energy metabolism of insulin signaling in mitochondria of rat hypothalamic neuron. Nineteen other proteins have unknown functions. These ligands were confirmed to be secreting from the rat hypothalmus on insulin signaling by western blotting. Conclusion : The hypothalamus is the master endocrine gland responsible for the regulation of various physiological and metabolic processes. Proteomics using LC-MS analysis offer a efficient means for generating a comprehensive analysis of hypothalamic protein expression by insulin signaling.

  • The ubiquitous presence of lactoferrin(LF) receptor in human as reported by the research group of Prof. Bo Lonnerdal, Univ. California (Suzuki, Y. A.,2001) encouraged us to search for the unknown physiological roles of Lf. Under the collaboration with Prof. Etsumori Harada, Tottori Univ., and his research group, we have found two novel biological activities of LF as the control of the lipid metabolism and the effect on the central nervous system. Relating to the lipid metabolism, LF could, in animal experiments, reduce triglyceride and total cholesterol both in blood and liver (Takeuchi, T et αl., 2003). LF increased plasms HDL-C and lowered LDL-C. In the central nervous system, LF showed anti-nociceptive activity mediated by ${\mu}$-opioid receptor in the rat spinal cord (Hayashida, K. et al., 2003). LF enhanced analgesic action of morphine synergistically via nitric oxide synthesis (Hayashida, K., et al., 2003) LF showed opioid-mediated suppressive effect on distress induced by maternal separation in rat pups (Takeuchi, T., et al., 2003).

Effects of Chronic Ethanol Administration on Folate Metabolism and Plasma Homocysteine Concentration in the Rats (만성적인 에탄올 섭취가 흰쥐의 엽산대사 및 혈장 호모시스테인 농도에 미치는 영향)

  • The present study was conducted to investigate the effect of different levels of ethanol ingestion on 131ate metabolism and plasma homocysteine concentration in Sprague-Dawley male rats receiving 0, 10, 30% of their caloric intake as ethanol for S weeks. Diets containing 10% ethanol had no effect on plasma and red blood cell(rbc) 131a1e. However, in rats fed a 30% ethanol diet, rbc folate increased and plasma 131ate decreased significantly, In the rats maintained first on a 30% ethanol diet for S weeks and then on a control diet for 2 weeks, the levels of plasma and rbc f31ate were normalized by withdrawal of ethanol. Urinary fo1ate excretion increased markedly in rats fed 10% and 30% ethanol diets and decreased to 51% of controls by withdrawal of ethanol. Plasma homocysteine concentration increased significantly in rats fed a 30% ethanol diet. The results suggest that chronic ingestion of ethanol increased urinary 131ate excretion markedly, which may decrease plasma 131ate and deplete liver folate.

  • Effects of compounds isolated from Ainsliaea acerifolia on alcohol metabolism in rats were examined and the results were as follows: Apigenin $7-O-{\beta}-D-glucoside$, after a single oral administration to rats, was found to cause a significant decrease in the serum ethanol concentration as well as enhancement of liver cytosolic alcohol dehydrogenase(ADH) activity.
