Effects of Jerusalem Artichoke (Helianthus tuberosus L.) Extracts on Blood Glucose and Lipid Metabolism in STZ-induced Diabetic Rats (돼지감자 추출액이 Streptozotocin으로 유발된 당뇨쥐에서 혈당 및 지질대사에 미치는 효과)

  • This study investigated the effect of Jerusalem artichoke (Helianthus tuberosus L.) extract on Blood Glucose and Lipid Metabolism in Streptozotocin (70 mg/kg B.W., i.p.)-induced Diabetic Rats. Sprague-Dawley male rats (200~220 g) were divided into normal control group (NC), diabetic control group (DC) and Jerusalem artichoke treated diabetic group (DJ). Diabetes was induced by single intraperitoneal injection of streptozotocin as a dose of 70 mg/kg body weight. Food (p<0.001) and water (p<0.05) intakes were higher in diabetic groups than the normal group. Body weight gain and food efficiency ratio were significantly lower in diabetic groups than normal group (p<0.01). However, they were higher in the DJ group than in the DC group. The serum levels of AST and ALT were significantly lower in the DJ group than in the DC group (p<0.05). The serum level of HDL-C was significantly higher in the DJ group than in the DC group (p<0.001). The serum levels of Triglyceride (p<0.05), LDL-C (p<0.001), and glucose (p<0.001) were significantly lower in the DJ group than in the DC group. At 3 and 4 weeks after the experiment, blood glucose level in the DJ group was significantly lower than the DC group (p<0.05). In conclusion, these results indicated that Jerusalem artichoke can prevent or retard the development of diabetic complications via its beneficial effects for alleviating the hyperglycemia and improved lipid metabolism.

Effect of Combined Exercise on Lung Function, Blood Vitamin D, Calcium and Bone Metabolism Hormones in Elderly Women (복합운동이 여성노인의 폐기능, 혈중 비타민 D, 칼슘 및 골대사호르몬에 미치는 영향)

  • The purpose of this study was to investigate the effect of combined exercise on lung function, blood vitamin D, calcium and bone metabolism hormones in elderly women who are over 65 years by dividing them into a combined exercise group(n=13), control group(n=17). The combined exercise program included 60-minutes sessions 3times each week. Aerobic exercise intensity was 40-50%HRR(RPE 12-13) for 1-4 week, 50-60%HRR(RPE 13-14) for 5-8 week, and 60-70%HRR(RPE 14-15) for 9-12 week and resistance exercise intensity was set at OMNI-RES 3-4 for 1-4 week, OMNI-RES 5-6 for 5-8 week, OMNI-RES 7-8 for 9-12 week. As a result, lung function indicated that FEV1 showed an interaction effect between group and time and FVC/FEV1 levels significantly increased in combined exercise group. Blood vitamin D showed an interaction effect between group and time, also, significantly increased in combined exercise group and control group. Calcium showed an interaction effect between group and time, and significantly decreased in control group. Bone metabolism hormones indicated that both calcitonin and osteocalcin showed an interaction effect between group and time, osteocalcin significantly decreased in control group. For the following this conclusion, elderly women can be improved their lung function through 12 weeks combined exercise and also mitigate the blood vitamin D but there was no meaningful results of calcium and bone metabolism hormones.

Inhibitory Effect of Aqueous Extracts from the Fruit Body of Lentinus edodes on Rat Intestinal Mucosa $\alpha$-glucosidase Activity and Reducing the Increase of Blood Glucose after Streptozotocin-induced Diabetic Rats

  • The aqueous extract from the fruit body of Lentinus edodes was evaluated for inhibitory activities against $\alpha$-glucosidase isolated from Spargue-Dawley male rats. A aqueous extract of Lentinus edodes exhibited 13.8% inhibitory activity on using 2 mM p-nitrophenyl $\alpha$-D-glucopyranoside as a substrate ($IC_{50}$ 75.3 mg/ml). The aqueous extract of Lentinus edodes inhibition type on $\alpha$-glucosidase was determined to be competitive inhibition. When it was oral administered to increase of blood glucose levels after STZ-induced in a dose dependent dietary. These results suggest that aqueous extract of Lentinus edodes effect a metabolism of intestine, and thereby reducing the increase of blood glucose after STZ-induced.

The Effects of Chromium Picolinate Supplementation on Glucose and Lipid Level in Rats Fed in High Fat Diet (Chromium Picolinate 보충이 고지방 식이를 섭취한 흰쥐의 체내 지질 농도에 미치는 영향)

  • This study aimed to investigate the effects of chromium picolinatec (CrP) supplementation on the glucose and lipid metabolism of rats. Five-week-old male Sprague-Dawley rats were fed a high-fat diet fir 110 days and further treated with CrP or placebos for 6 weeks. The effect of CrP supplementation on body weight, blood glucose, insulin and lipid levels was examined. The results suggested that body weight gain was not significantly different between the control and the CrP supplemented group. Plasma glucose, triglyceride and cholesterol levels in the CrP treated group significantly decreased compared to those of the control group (p < 0.05). The total lipid, triglyceride and cholesterol concentrations in the livers of the CrP treated group significantly decreased compared to those of the control group (p < 0.05). These results indicate that supplementation of chromium picolinate ran reduce triglyceride and glucose concentrations in the blood and total lipid and total cholesterol levels in the liver.

Effect of Cimetidine Pretreatment on the Pharmacokinetics of Sulfisomidine Administered Intravenously in Rabbits (시메티딘이 설프이소미딘의 약물동태에 미치는 영향)

  • These paper was attempted to investigate the mechanism of increased blood level of sulfisomidine by cimetidine pretreatment pharmacokinetically. Especially, effect of cimetidine pretreatment on both renal clearance and biliary clearance of sulfisomidine was studied in rabbits. The results are as follows. The blood level of sulfisomidine administered intravenously in dose of 25mg/kg was elevated significantly by cimetidine pretreatment. Relative bioavailability and biological half-life were increased significantly by cimetidine pretreatment. Overall elimination rate constant ($betha$) and distribution rate constant ($K_{13}$) of sulfisomidine were decreased significantly by cimetidine pretreatment. The renal and biliary clearance of sulfisomidine were decreased significantly compared with those of control rabbits by cimetidine pretreatment. The results may be also related to the inhibition of sulfisomidine metabolism enzyme activity or reduction of blood flow in the liver.

  • Pharmacokinetic interaction of cimetidine and isoniazid was investigated in the rabbits. Isoniazid was administered orally at a dose of 30mg/kg to six rabbits after 10, 20, and 30mg/kg pretreatment of cimetidine twice a day for 10days. Concentration of the free and the total isoniazid in the blood and the urine was determined by spectrophotometer. Relative bioavailability and biological half-life($t\frac{1}{2}{\beta}$) were increased significantly by cimetidine pretreatment. Overall elimination rate constant and total clearance of isoniazid were decreased significantly by cimetidine pretreatment. The ratio of metabolites to isoniazid in the blood and the urine was decreased significantly by cimetidine pretreatment. Relative bioavailability, INAH to metabolites ratio in the blood and decrease in total clearance were highly correlated with the does of cimetidine pretreated. This result might be due to the inhibition of isoniazid metabolism in the liver by cimetidine pretreatment.

  • Effects of a single administration of dichloromethane (DCM) on the hepatic drug metabollzing activity were determined using adult female rats. Rats were treated with DCM (3 mmol/kg, ip) and the disappearance of antipyrine (100 mg/kg, iv) or ethanol (2 g/kg, ip) from blood was measured. The blood concentration and half-life of antipyrine was not influenced by DCM administration. And DCM did not alter the blood concentration of ethanol measured for 240 min after the treatment. The effect of DCM treatment on in vitro cytochrome P-450-dependent enzyme activities was examined as well. No significant difference in either aniline hydroxylase or aminopyrine N-demethylase was observed in hepatic microsomal fractiorts of rats treated with DCM 24 hr prior to sacrifice. The present study indicates that acutely given DCM does not alter the metabolism of xenobiotics in vivo. The failure of DCM to alter the in vitro hepatic microsomal drug metabolizing activity was also noted.

  • Hangover is associated with alcohol metabolism in body after the ingestion of an alcoholic beverage. It has been known that hangover is caused by increasing blood acetaldehyde concentration. This study was carried out to evaluate effect against blood ethanol(EtOH) and acetaidehyde(AcH) on the seven natural products samples(Lotus seed, Sweet chest nut rose, Kohki, Gurume-K, Gurume-J, Phytic acid and Chlorophyll). Also, samples which were selected as good products were mixed. Then, effect against blood EtOH and AcH formulas were tested in vivo. (omitted)

Relationships of Obesity in Childhood to Plasma Lipids, Blood Pressure and Blood Glucose (아동기 비만이 혈장 지질, 혈압 및 혈당에 미치는 영향)

  • In order to investigate the relationships of obesity in childhood to plasma lipid, blood pressure and blood glucose concentration, we selected 21 subjects for the moderate obese group(MO), 9 for mild obese group(MI), and 19 for the control group(C) among children aged 10~12. While the level of plasma triglyceride and VLDL-cholesterol of the MO group was much higher than that of group C, a significantly lower percentage of HDL-cholesterol was found in the MO compared to the percentage found in group C. Also the level of the total cholesterol and LDL-cholesterol of the MI group as well as the MO was much higher than that of C. The elevated total-cholesterol level of the Mi group was due to increased LDL-cholesterol and that of the MO was due to increases in both VLDL-cholesterol and LDL-cholesterol. As the result of these differences, the atherogenic index of the MO was significantly higher than that of C. The incidence of hypercholesterolemia( 200mg/dl) of the MI and MO was 60.0% and 77.8% respectively, All of the physical parameters and indexes except height were positively correlated with plasma lipid levels, systolic blood pressure, and blood glucose concentration. The analyses of the correlated with plasma lipid levels, systolic blood pressure, and blood glucose concentration. The analyses of the correlation indicated that central fat to peripheral fat ratio and waist to thigh girth ratio seemed to be closely associated to plasma lipid levels and atherogenic index. The MO had significantly higher systolic blood pressure than C and significantly higher blood glucose concentration was found in both MI and MO than in C. These results confirmed that obesity in childhood may be relevant to chronic metabolic diseases such as abnormal lipid metabolism, atherosclerosis, high blood pressure and diabetes mellitus.

The Effect of Silk Amino Acid Supplementation on the Level of Blood Energy Substrates and Hormones during Prolonged Exercise

  • The silk amino acid supplementation is unknown to affect the release of several hormones related to energy production and metabolism during prolonged exercise. This study examined the effects of silk amino acid supplementation on the level of blood amino acid, energy substrates and hormones level during prolonged treadmill exercise in college taekwondo player. A prolonged treadmill test was carried out 60 min at 65% of maximal heart rate on 8 athletics. Blood samples were obtained form antecubital vein of subjects at rest bed 30 minute before test, after exercise and rest 1 hour. The subjects were supplemented silk amino acid (6,390 mg/day) fur 4 week. The silk amino acid supplementation did not produce significant changes on the levels of blood lactate, ammonia, amino acid, glucose, triglyceride, total cholesterol, HDL, LDL, seratonin and leptin at rest bed 30 minute before test, after exercise and rest 30 minute. The silk amino acid 4 week supplementation did not affect the levels of blood amino acid, energy substrates and hormones during prolonged treadmill exercise.
