Effects of Dietary Carbohydrases on Growth Performance, Nutrient Digestibility and Blood Characteristics in Finishing Pigs

  • The objective of this study was to assess the effects of dietary carbohydrases on growth performance, nutrient digestibility and blood characteristics in finishing pigs. A total of 90 pigs [(Landrace ${\times}$ Yorkshire) ${\times}$ Duroc] (initial BW = $56.15{\pm}1.26kg$) were used for a 35 d feeding trial. The dietary treatments included: 1) CON (control diet), 2) MIX (CON + mixture with ${\alpha}$-galactosidase and ${\beta}$-mannanase 0.05%) and 3) MAN (CON + ${\beta}$-mannanase 0.05%). There were six replications per treatment with five pigs per pen. The average daily gain (ADG) in MIX was higher than in CON (p<0.05). No significant differences were noted in the average daily feed intake (ADFI) and feed efficiency (G:F) among dietary treatments (p>0.05). Apparent total tract digestibility (ATTD) of dry matter (DM) and energy (E) in MIX increased (p<0.05) relative to CON and MAN. The ATTD of nitrogen (N) in MIX was higher (p<0.05) than in CON. No differences in red blood cells (RBC), white blood cells (WBC), lymphocytes and IgG concentrations were observed among dietary treatments (p>0.05). In conclusion, the addition of the mixture of carbohydrases (${\alpha}$-galactosidase and ${\beta}$-mannanase 0.05%) increased ADG and nutrient digestibility in finishing pigs.

Micronucleus Test of DHU001, a Polyherbal Formula, in Bone Marrow Cells of Male ICR Mice

  • The genotoxic effects of DHU001, a polyherbal formula were evaluated using the mouse micronucleus test. DHU001 was administered once a day for 2 continuous days by oral gavage to male ICR mice at doses of 2000, 1000 and 500 mg/kg. Cyclophosphamide was used as a known genotoxic agent in a positive control. The appearance of a micronucleus is used as an index for genotoxic potential. In addition, the changes on the total white blood cells and differential counts on the lymphocytes, neutrophils, eosinophils, basophils and monocytes in the prepared blood smears were also conducted to observe the possible immunosuppression. The results indicats that DHU001 showed no genotoxicity effects up to 2000 mg/kg dosing levels and did not influenced on the total white blood cells and differential counts. In addition, it is also considered that there were no problems from cytotoxicity of DHU001 tested in this study because the polychromatic erythrocyte ratio was detected as > 0.41 in all tested groups.

The results obtained indicated that KJD shows no genotoxicity effects up to 2000 mg/kg dosing levels, but KJD shows slight increased trends in the blood total leukocyte numbers as pharmacological effects of immune stimulation. In addition, it is also considered that there were no problems from cytotoxicity of KJD tested in this study because the polychromatic erythrocyte ratio was detected as > 0.42 in all tested groups.

가미홍화탕의 방사선 부작용 억제 효능연구 (Efficacy Study of Kami-honghwa-tang on the Reduction of Side Effects of Radiotherapy)

  • In this study, Kami-honghwa-tang (KH-19) was designed and animal study was conducted to evaluate its efficacy on the reduction of the side effect of radiotherapy. Bone marrow toxicity is one of the major side effect of radiotherapy which cause the reduction of blood cells, and KH-19 was designed to protect and enforce blood. C57BL/6 mice were irradiated with 4 Gy of gamma ray, and divided into control group which was treated with water and KH-19 group which was treated with 1.5g/Kg of KH-19 up to 4 weeks. KH-19 group showed significantly increased white blood cells, lymphocytes and platelet count compared with control group (p<0.05). When bone marrows were examined, KH-19 group showed higher cell densities than control group (p=0.06). KH-19 may increase blood cell count after radiation by its protective effects on bone marrow.

NC/Nga 마우스 전혈을 이용한 항 아토피 피부염 물질 탐색 (Screening of Anti-Atopic Dermatitis Material by Using NC/Nga Mouse Whole Blood System)

  • Background: Allergic inflammation was induced by activated Th2 lymphocytes, leading to IgE production and eosinophil activation. A Th2 disproportion was shown in atopic children soon after birth. During specific allergen stimulation, an increase of Th2 cells was observed in most cases. In this study, we prepared new screening "whole blood" system for searching the anti-atopic materials. Cytokine production and IgE secretion from whole blood system were assessed and we confirmed the results by using animal system. Methods: Pathological features in NC/Nga mice are similar to those observed in human atopic dermatitis. Whole blood from NC/Nga mouse was stimulated by using TNCB (Th2 activator) or candidate materials of anti-atopic dermatitis, and the production of cytokines (IL-4, IL-12, and IFN-${\gamma}$) were measured by ELISA. In order to confirm the results of whole blood system, in vivo test was done by using NC/Nga mice. Results: In whole blood system, LPS and extracts of green tea, hardy orange and onion induced the production of IL-12 and IFN-${\gamma}$ while they reduced the production of IL-4. Also, LPS and extracts of onion reduced IgE production. Though atopic dermatitis was observed from a mouse stimulated with TNCB, it was not when a mouse was co-stimulated in LPS or extracts of onion. The results are same as those observed in whole blood system. Conclusion: Whole blood system was simple and speedy methods for searching a materials compared with the conventional high-cost animal system. And the results using whole blood system was proved to be reliable in our experiments for screening anti-atopic material. We expect that the system can be applied to other experiments for searching similar materials.

유도훈련이 경호전공 비만대학생들의 신체구성, 혈중지질 및 면역력에 미치는 영향 (The Effect of Judo Training on Body Composition, Blood lipids and Immunity of Security Majoring Obese University Students)

  • 경호전공 대학생들은 경호대상자의 생명과 재산을 보호하기 위해 경호무도를 익히고 습득하고 있다. 또한 경호무도는 경호원의 건강한 몸과 정신을 향상시키고 유지하는데 도움이 되고 있다. 하지만 현재 대학에서 경호를 전공하는 학생 중 경호무도훈련 참여시간 부족과 과도한 음식섭취로 인해 비만해 지는 경우가 늘고 있는 실정이며, 이는 본인의 건강상태뿐만 아니라 추후 경호업무에도 좋지 않은 영향을 초래할 수 있을 것이다. 경호무도 종목 중 유도는 신체구성에 긍정적인 영향을 미치며, 근력과 순발력은 향상시키는 것으로 알려져 있다. 따라서 본 연구는 경호전공 비만남자대학생을 대상으로 10주간의 유도훈련이 신체구성, 혈중지질 및 면역력에 미치는 영향을 규명하였다. 본 연구의 결과를 종합해 볼 때, 유도훈련은 체지방 감소로 인해 비만을 방지하고 중성지방과 콜레스테롤을 감소시켜 동맥경화성 질환을 예방할 수 있을 것으로 판단된다. 또한 면역기능에 있어 인체 세균 감염에 대한 방어기능과 염증반응, 외부항원 대항에 긍정적으로 작용할 것으로 생각된다. 이는 경호원의 비만을 방지할 뿐만아니라 건강한 삶을 유지하여 경호업무 능률을 향상시킬 수 있을 것으로 사료된다.

  • A routine hematological observation in the course of starvation was carried out on eight experimentel1y starved rabbits. They were strictly selected and restricted all of food intake with the exception of optional water intake until death. The body weight of each rabbit on the day before starvation was about 2 kilograms. The results are summarized as follows. 1. The average decrememt ratio of body weight on the terminal day before death was $34.3{\pm}7.5$ per cent with the range from 24.5 to 46.3 per cent. The average life duration until death was $10.25{\pm}2.6$ days, the range being from 6 to 14 days. 2. The decrease in number of reticulocytes with a parallel disappearance of polychromatic erythrocytes in peripheral blood in the course of starvation Was the most remarkable change in erythrocytic series, an evidence suggesting marked restriction of the erythropoietic function on 3rd to 4th day and almost complete suspension in about a week of starvation. 3. Erythrocyte count, hemoglobin content and haematocrit value of peripheral blood, were normal or indicative of slight hemoconcentration. 4. Mean Corpuscular Hemogloin Concentration was slightly higher than normal and Mean Corpuscular Volume tended to be low and no appreciable shifts were observed in Mean Corpuscular Diameter and Price-Jones curve of erythrocytes, while fewer macrocytes than normal were seen. These changes were considered to have resulted from a marked decrease in young erythrocytes in peripheral blood in the course of starvation. 5. Neither poikilccytoses or anisosytosis was observed. 6. Leukopenia was observed in all of 8 starved rabbits. The decrement ratio on the terminal day of starvation was between 13 to 64 per cent. The leukopenia was mainly due to fall of lymphocytes in 6 cases and to fall of neutrophilic leukocytes in the other 2 cases. In many cases, irregular fluctuation of neutrophilic leukocytes in its biological curve were seen in contrast to the relatively smooth changes of lymphocytes. Eosinophilic leukocytes tended to decrease in absolute number especially in later stage of starvation. Little significance in regard to monocytes and basophilic leukocytes in this study was discussed. 7. Proplasma cells, rarely plasma cells, appeared with a tendency to increase in number at later stage of starvation. 8. The most characteristic changes on circulating blood cells in complete starvation of rabbits were the leukoponia and failure of regeneration of erythroctes. These changes were considered as adaptive phenomena in response to the catabolic consumption of body constituents.

Partial Purification and Characterization of a Glycoprotein Factor from Fresh Ginseng

  • The aqueous extract of fresh ginseng (Panax ginseng C.A. Meyer) contains a macromolecular fraction that showed mitogenic and comitogenic activities in human peripheral blood Iymphocytes. Purification of the crude extract by size (ultrafiltration, Sephadex G-200) and charge (DEAE-cellulose. DEAE-Sepharose) yielded a semi-purified fraction (DS-3). This fraction contains at least three subgroups of anionic macromolecules with apparent molecular weight greater than 600 kilodaltons. It is a glycoprotein with a large amount of glucuronic acid. It acts as a mitogen in both T and B cells of human peripheral blood lymphocytes. It could also potentiate the mitogenic action of Concanavalin A in Iymphocyte T cells. Such potentiation is not due to increased binding of Concanavalin A to the cell surface. Its mitogenic and co-mitogenic effects do depend on the presence of extracellular Ca2+.
