Morin alleviates fructose-induced metabolic syndrome in rats via ameliorating oxidative stress, inflammatory and fibrotic markers

  • Metabolic syndrome (MBS) is a widespread disease that has strongly related to unhealthy diet and low physical activity, which initiate more serious conditions such as obesity, cardiovascular diseases and type 2 diabetes mellitus. This study aimed to examine the therapeutic effects of morin, as one of the flavonoids constituents, which widely exists in many herbs and fruits, against some metabolic and hepatic manifestations observed in MBS rats and the feasible related mechanisms. MBS was induced in rats by high fructose diet feeding for 12 weeks. Morin (30 mg/kg) was administered orally to both normal and MBS rats for 4 weeks. Liver tissues were used for determination of liver index, hepatic expression of glucose transporter 2 (GLUT2) as well as both inflammatory and fibrotic markers. The fat/muscle ratio, metabolic parameters, systolic blood pressure, and oxidative stress markers were also determined. Our data confirmed that the administration of morin in fructose diet rats significantly reduced the elevated systolic blood pressure. The altered levels of metabolic parameters such as blood glucose, serum insulin, serum lipid profile, and oxidative stress markers were also reversed approximately to the normal values. In addition, morin treatment decreased liver index, serum liver enzyme activities, and fat/muscle ratio. Furthermore, morin relatively up-regulated GLUT2 expression, however, down-regulated NF-κB, TNF-α, and TGF-β expressions in the hepatic tissues. Here, we revealed that morin has an exquisite effect against metabolic disorders in the experimental model through, at least in part, antioxidant, anti-inflammatory, and anti-fibrotic mechanisms.

왕둥굴레의 생약학적연구(生藥學的硏究) (Pharmacognostical Studies on the Rhizome of Polygonum robustum Nakai)

  • The rhizome of Polygonatum robustum Nakai (Liliaceae) has been used as a crude drug for the purpose of tonic and thirst cure in Korea. The dried rhizomes were extracted with hot ether, then the residue was extracted with hot alcohol. 1) The three kinds of chemical constituents, substance, I, II and III were isolated by silicagel column chromatography from ether and alcohol extracts of the rhizomes. Substance I was confirmed as ${\beta}-sitosterol$, $C_{29}H_{50}O$ by mass and IR spectroscopy. Substance II was identified as stigmasterol by GLC. 2) Substance III was obtained by crystallization from the column chromatography of alcohol fraction. It was suggested as diosgenin based on chemical and spectral discussions. 3) The concentration of blood sugar was significantly decreased in the group administered the ether extract with 20% dextrose and adrenaline in comparison to that of 20% dextrose and adrenaline along.

  • During the serial attempts to identify the chemical and biological characteristics of Ailanthi Cortex Radicis, the root bark of Ailanthus altissima (Simaroubaceae), we find out the serious toxic effect on kidney by chloroform fraction of the methanolic extract of the herb drug. The toxicities were revealed as the increase of urea nitrogen amount in blood and lactate dehydrogenase and ${\gamma}-glutamyltransferase$ activities in urine and the decrease of the concentration of glutathione and both of protein bound and non-protein bound -SH in kidney tissue.

Hepatoprotective and Anti-diabetic Effects of Pelvetia siliquosa, a Marine Algae, in Rats

  • The effects of various fractions from the whole plant of Pelvetia siliquosa Tseng et Chang (Fucaceae) on the $CCl_4$-induced hepatotoxicity as well as on streptozotocin (STZ)-induced diabetes in rats were investigated. The ether fraction exhibited a potent rat lens aldose reductase (RLAR) inhibition in vitro and showed a significant inhibition of not only serum glucose concentrations but also sorbitol accumulations in the lens, red blood cells and sciatic nerves in the STZ-induced diabetic rats. When administered orally in Sprague-Dawley rats, $H_{2}O$ fraction was found to cause a significant inhibition of the rise in the serum transaminase activities in $CCl_4$-intoxicated rats. These results suggested that this plant might possess constituents with hepatoprotective, anti-diabetic effects and those effects on diabetic complications.

Effect of Niacin Supplementation on Growth, Nutrient Utilization and Blood Biochemical Profile in Male Buffalo Calves

  • In order to investigate the effect of different levels of niacin supplementation on growth, nutrient utilization, their balance and blood biochemical profile, 15 male buffalo calves (9-10 months of age, $88.4{\pm}4.37kg$ average body weight) were divided into 3 equal groups each of 5 calves, following a completely randomized design, and fed individually for 120 days with wheat straw and concentrate mixture to meet their nutrient requirements. In addition calves were supplemented with 0 ppm (control, group I), 100 ppm (group II) and 200 ppm (group III) niacin. After 90 days of experimental feeding a metabolism trial was conducted to estimate the digestibility of nutrients and their balance. Fortnightly body weights were recorded to assess their growth rate and blood was collected from the jugular vein at day 0 and subsequently at 30-day intervals from all the experimental buffalo calves to study blood biochemical parameters. Results showed that intake and digestibility of dry matter, organic matter, crude protein, ether extract, total carbohydrates, neutral detergent fibre, acid detergent fibre, cellulose and hemicelluloses were statistically similar in the 3 groups. Buffalo calves in all three groups were in positive nitrogen, calcium and phosphorus balance, without showing any significant effect of the treatments. Dry matter, crude protein, digestible crude protein and total digestible nutrient intake ($g/d/kgW^{0.75}$) were similar in the control and niacin supplemented groups. Digestible crude protein (%) and total digestible nutrients (%) in the ration of the 3 groups were 8.07, 7.99, 7.92 and 56.70, 56.63, 56.74, respectively, and were comparable among the groups. The average daily gain (g) in-group II (567.50) was not significantly (p>0.05) higher than group I (500.0) and group III (510.0). Blood biochemical constituents (glucose, total protein, albumin, globulin, urea-N, insulin) showed no significant effect of niacin supplementation. However, serum cholesterol (mg/100 ml) was significantly (p<0.01) lower in the 200 ppm niacin-supplemented group than in the control and 100 ppm niacin-supplemented groups. It can be concluded that supplementation of niacin at 100 and 200 ppm in the diet of buffalo calves had no significant beneficial effect on their growth and nutrient utilization.

  • Dangguisu-san (DGSS) is widely known traditional herbal medicinal formula in Korea for treatment of traumatic injury by traffic accident, ecchymosis, abdominal distension and anti-thrombosis of blood. This study was conducted to develop the simultaneous analyze method using high performance liquid chromatography (HPLC) and examine the effect of anti-inflammatory activity of DGSS-dry extract (DGSS-DE) and DGSS-mix extract powder (DGSS-MEP). Physicochemical characteristics of DGSS-DE and DGSS-MEP showed that there is no significant difference in pH, titratable acidity, total soluble solid content and browning degree except for color value (L, a, b). 15 functional constituents of DGSS were identified and the correlation coefficient values of DGSS-DE and DGSS-MEP were conformed 0.950. Also, DGSS-DE and DGSS-MEP significantly decreased the secretion of nitric oxide (NO), prostaglandin E2 (PGE2), interleukin-1β (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) through inhibited expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), IL-1β, IL-6, and TNF-α. From these result, DGSS-MEP showed similar chemical composition and anti-inflammatory effect to DGSS-DE. Therefore, DGSS-DE and DGSS-MEP may be useful as potential source of drug to prevent inflammation.

주요 양식어류의 하절기 혈액성분 비교 (Comparison in Serum Constituents of Cultured Marine Fishes in Early Summer Season)

  • 주요 해산 양식어류의 혈액 성분에 관한 기초 자료를 얻기위하여, 적수온기(수온 16.5도씨)에 조피볼락(Sebastes schlegeli, 평균체중 553g), 농어(Lateolabrax japonicus, 평균체중 325g), 넙치(Paralichthys olivaceus, 평균체중 390g), 돌돔(Oplegnathus fasciatus, 평균체중 490g), 은연어(Oncorhynchus kisutch, 평균체중 1,750g) 및 전갱이(Trachurus japonicus, 평균체중 438g)를 사용하여 꼬리 정맥에서 채혈하여 혈청 중의 총단백질(TP), 알부민(ALB), 중성 지방(TRIG), 콜레스테롤(CHOL), 포도당(GLC), 리파제(LIPA), 아밀라제(AMYL), 아미노산전이효소(AST), 나트륨(Na), 칼륨(K), 염소(Cl), 인(PHOS) 등을 측정하였다. TP 농도는 4.4~6.0 g/dl의 수준이었으며, ALB 농도는 1.7~2.4g/dl의 수준이었다. A/G비는 은연어가 0.9로 가장 높았고 나머지는 0.6정도로 비슷하였다. 지질 성분 중 TRIG 농도는 어종에 따라 유의적인 차이를 보여, 전갱이와 조피볼락(178~180 mg/dl)이 가장 높았고, 은연어와 돌돔(102~114 mg/dl)이 가장 낮았으며, 넙치와 농어(126~159 mg/dl)는 그 중간 수준이었다. 그리고 CHOL 농도도 전갱이가 255 mg/dl로 가장 높았고 이어서 농어 (230 mg/dl)와 넙치, 은연어(206~217 mg/dl), 조피볼락(154 mg/dl)의 순서였으며 돌돔이 가장 낮아 88 mg/dl의 수준을 보여 유의적인 차이가 있었다. 어종별 혈당(GLC)의 농도는 전갱이(138 mg/dl)가 가장 높았고 돌돔 (111b mg/dl), 은연어나 농어 (64~78 mg/dl), 조피볼락(47 mg/dl), 넙치(14 mg/dl)의 순서였다. 지질분해효소(LIPA)와 당질분해효소(AMYL)의 어종별 활성은 어종간에 유의적인 차이를 보여, 은연어 LIPA와 AMYL 활성이 모두 가장 높았지만 농어는 가장 낮았으며, 돌돔과 넙치와 조피볼락에서 AMYL 활성은 검출 한계치(5 U/dl) 이하였다. 그리고 무기질의 Na와 Cl의 농도는 유사한 수준이었으며, Na 농도와 K 농도는 서로 상반되는 경향이었다. 각종 혈중 성분의 계절적인 차이를 조사하였더니, PHOS와 CHOL 농도는 계절적인 영향이 컷지만, K와 GLC 농도는 큰 차이를 보이지 않았고, 어류 중에서는 넙치가 큰 차이를 보였다. 즉, 넙치나 전갱이는 적수온기에서 혈중농도가 높았지만 조피볼락이나 돌돔은 저수온에서 혈중 농도가 높았다. 전해질과 LIPA 활성은 대체로 저수온기에서 농도가 높았으나, TP와 ALB등은 적수온기에서 더 높았고, 어너지로 쓰이는 TRIG나 CHOL, GLC 농도의 계절에 따른 차이는 어종에 따라 달랐다.

Nutrient Intake, Its Utilization, Rumen Fermentation Pattern and Blood Bio-Chemical Constituents of Sheep Fed Urea Treated Mustard (Brassica campestris) Straw

  • A study was conducted to compare the feeding value of urea treated and untreated mustard straw (MS) for sheep. Treated MS was prepared by adding urea-N at 1.84% and followed by packing in a pit silo for 21 days. Two groups of six empty Avikaline ewes were fed untreated (UTMS) and treated (TMS) mustard straw along with 200 g concentrate per head daily for 90 days. Untreated MS had 0.41% N and the urea treatment increased its N value to 1.58 %. The cell wall constituents were decreased in the TMS except for cellulose which remained unaffected. Dry matter intake of TMS was consistently higher than that of UTMS. Digestibility of DM, OM and fibre fractions of MS improved by the urea treatment. Ewes in both groups were in positive N balance while % N retention was lower in UTMS (26.30%) than in TMS (52.14%). The TMS fed group on average consumed 30.2 g DM, 2.9 g digestible crude protein and $0.2MJ\;DE\;per\;kg\;BW\;day^{-1}$ and maintained their weight whereas, the UTMS fed ewes lost weight. The VFA concentration in rumen liquor was higher in TMS than in UTMS. Total-N, ammonia-N and TCA-precipitable-N were also higher in TMS fed ewes. Blood glucose concentrations in the two groups were similar at initiation of the study. However the glucose concentration of UTMS fed group was significantly (p<0.01) lower than those fed UTMS at the termination of the study. Urea-N concentration was also higher in TMS fed group after 90 days of feeding period. It is concluded that urea treatment of MS improved N value of MS from 0.41% to 1.58% along with sizable improvement in nutritive value and in conjunction with 200 g concentrate, TMS can serve as maintenance ration for sheep. Effect of Chromium Dietary Supplementation on the Immune Response and Some Blood Biochemical Parameters of Transport-stressed Lambs

  • Forty-eight Naemi lambs (avg. BW 31.7 kg) were transported by truck for a distance of 1,450 km from Al-Jouf to Riyadh, Saudi Arabia. On arrival day, the lambs were randomly allocated to four groups receiving diets supplemented with 0.0, 0.3, 0.6 and 0.9 ppm organic chromium (Cr). Each group consisted of four separately housed replicates of three lambs each. The animals were fed ad libitum on a grower diet for 84 days. Blood samples were obtained shortly before transportation, upon arrival and at weekly intervals thereafter from all lambs for analysis of plasma and serum. Plasma glucose and serum cortisol, total protein, albumin, urea-N and total cholesterol concentrations were determined. A cursory clinical examination of the lambs, along with rectal temperature, was undertaken at different intervals during the experiment. The lambs were inoculated each with 2 ml i.v. chicken red blood cells (CRBC) on days 0, 21, and 42. Serum total, IgG and IgM antibody titers were determined at weekly intervals post-immunization. An in vivo intradermal hypersensitivity test was carried out on 6 lambs from each group on days 10 and 70. Transportation of the lambs resulted in a significant (p<0.001) elevation of serum cortisol, total protein and albumin levels, as well as increased plasma glucose concentration, with corresponding decrease in total cholesterol, while blood urea-N remained largely unchanged. These constituents returned to normal levels during subsequent weeks, with no significant differences in their concentrations being observed between the Cr-supplemented groups and controls. Rise in rectal temperature after transportation was reduced to a greater extent (p<0.05) in Cr-supplemented versus control lambs. Total, IgG and IgM antibody titers against CRBC rose significantly (p<0.05) during immunizations in all groups, with significantly and linearly higher (p<0.05) total and IgG titers in Cr-supplemented versus control lambs. By contrast, no significant effect due to Cr supplementation was recorded in IgG titers, which increased equally in Cr-fed and control groups. Skin thickness in response to intradermal inoculation of phytohaemagglutinin (PHA) was also significantly (p<0.01) increased as a result of Cr supplementation. These results indicate that dietary Cr supplementation might be useful during stress especially for enhancing immune responses in transport-stressed lambs.
