Effects of Lycium chinense Miller Fruit and its Constituent Betaine on Immunomodulation in Balb/c Mice

  • BACKGROUND: The aim of this study is to investigate the effects of Lycium chinense Miller fruit and its representative constituent betaine on reduction of immobility time and blood parameters in balb/c mice. METHODS AND RESULTS: We investigated the immobility time and the changes in aspects of blood biochemical parameters by the administration of L. chinense Miller fruit and its representative constituent betaine, after the forced swimming test. The immobility time was significantly reduced about 41.3% and 53.6%, respectively, in the animal of L. chinense Miller fruit and its representative constituent betaine-administrated group for 7 days, in comparison with that of the control group. The level of blood urea nitrogen was significantly decreased in L. chinense Miller fruit and its representative constituent betaine-treated group compared with the control group (P < 0.05), respectively. In addition, the interlukin-2 levels of mice in L. chinense Miller fruit and betaine treated group was increased compared with the control group. CONCLUSION: These results indicate that L. chinense Miller fruit and betaine might be helpful in the immune function improvement, enhance physical stamina, and fatigue recovery.

The Effects of Isokinetic Exercise Program on the Muscle Strength and Blood Constituent by the Relatively Angular Velocities

  • The purpose of this study was to investigate the effects of angular velocity on muscle strength and blood constituent. Subjects was classified into two groups, which were $60^{\circ}/sec$ angular velocity group (n=8), and $240^{\circ}/sec$ angular velocity group (n=8). Each group was applied to perform the isokinetic exercises on extensor muscle group for each 7 times in 10 set (3 days per a week for 3 weeks). Muscle strength was measured using peak torque of quadriceps femoris and blood constituent was measured using RBC, WBC and Hb. The peak torque was more significantly increased after 3 weeks application of $60^{\circ}/sec$ angular velocity group than $240^{\circ}/sec$ angular velocity group. The RBC and Hb were more significantly increased after 3 weeks application of $60^{\circ}/sec$ angular velocity group than $240^{\circ}/sec$ angular velocity group. But WBC was more significantly increased after 3 weeks application of $240^{\circ}/sec$ angular velocity group than $60^{\circ}/sec$ angular velocity group. These results indicate that $60^{\circ}/sec$ angular velocity isokinetic exercise application were effective treatment strategy on increase of muscle strength. But $240^{\circ}/sec$ angular velocity isokinetic exercise application were effective treatment strategy on immune protect system.

A Study on Application of Hwangnyeonhaedok-tang Blended Prescriptions in Dongeuibogam (동의보감(東醫寶鑑) 중(中) 황연해독탕(黃連解毒湯)의 활용(活用)에 대한 연구(硏究))

  • The results of documentary survey of 81 prescriptions of Hwangnyeonhaedok-tang and Hwangnyeonhaedok-tang blended ones introduced in Dongeuibogam in which there are 42 of prescriptions using Hwangnyeonhaedok-tang as main constituent and 39 of prescriptions using Hwangnyeonhaedok-tang as assistant constituent can be summarized as followings, 1. Hwangnyeonhaedok-tang blended prescriptions using Hwangnyeonhaedok-tang as main constituent are applied to 20 categories of disease such as 7 of external disease from the chapters of various dermatos is, various injury, tumor(16.6%), 5 of fire disease from the chapter of fire(12%), 5 of eye diseases from the chapter of eye(12%), 4 of disease originated from damage by coldness from the chapter of coldness(9%), etc. 2, It has been found that mainly considered causative agents in diseases, when making up Hwangnyeonhaedok-tang blended prescriptions using Hwangnyeonhaedok-tang as main constituent, are feverish toxin in internal organs, followed by exogenous damp and fever, feverish toxin from damage by coldness and more such as extravasated blood, damp toxin, lack of ki and blood, lack of yin, congestion, the seven passions, blood overflow and excess liquor and sex.

  • The objectives of this investigation were to produce artificially an antitumor constituent by submerged culture of the mycelium of Coriolus versicolor (Fr.) Quel., to characterize the influence of various modifications of the nutrient and culture conditions with respect to the pro­duction, to determine chemical composition of the antitumor constituent, and to examine effects of the constituent on the immune response of mice. Submerged agitation of the mycelium in flasks containing a nutrient solution showed its adequate growth. Especially the mycelial growth in the medium containing glucose and yeast extract was abundant. The addition of cotton seed flour or ginseng waste to the medium increased the yield of mycelial growth and the production of the antitumor constituent. The replacement of glucose with starch also yielded the adequate growth. The antitumor constituent extracted from the mycelium and isolated from the culture filtrate was a protein-bound polysaccharide. The analyses of this constituent by GLC and amino acid autoanalysis showed that it contained four monosaccharides and fifteen amino acids. The protein-free polysaccharide of the constituent was also found to exert greater antitumor activity against sarcoma-180 in mice than the entire constituent. The antitumor constituent was found to potentiate the immune response of mice against sheep red blood cell. The protein-bound polysaccharide exerted more favorable influence on the immunity than the protein-free moiety.

A Study on the Changes of Blood Constituent in Male Students of TAE-EUM-IN (태음인(太陰人) 남학생(男學生)의 혈액변화(血液變化)에 대한 연구(硏究))

  • Comparative blood analysis was carried out to investigate the change of blood constituent in TAE-EUM-IN group and control group. RBC, WBC, hemoglobin, hematocrit, total protein, triglyceride, phospholipid, total cholesterol, LDL-cholesterol, HDL-cholesterol, BUN, creatinine, ACTH, cortisol and prostaglandin E and $F_2{\alpha}$ were measured. And the following results were obtained: 1. In the change of blood cell, the value of hematocrit showed significant difference, while that of RBC, WBC and himoglobin didn't. 2. In the change of protein, the value of total protein showed significant difference, but that of albumin didn't. 3. In the change of lipid and cholesterol in serum, the value of triglyceride, phospholipid, total cholesterol and LDL-cholesterol indicated significant difference, while that of HDL-cholesterol indicated significant difference, while that of HDL-cholesterol didn't. 4. The value of BUN in serum represented significant difference, but that of creatinine didn't. 5. The value of cortisol in plasma showed significant difference, but that of ACTH didn't. 6. The value of prostaglandin E and $F_2{\alpha}$ in plasma showed significant difference. These results suggest that investigation on hormones and blood constituents enables objectification in differentiation of four types of physical constitution.

  • Comparative blook analysis was carried out to investigate the difference of blood constituent in four types of physical constitution. Isoleucine(Ile), Alanine(Ala), Cysteine(Cys), Tyrosine(Tyr), Methionine(Met) were measured. And the following results were obtained: 1. The value of Isoleucine(Ile) on the constitution didn't shown significant difference, but between the value of SO-YANG-IN and SO-EUM-IN showed significant difference. 2. The value of Alanine(Ala) on the constitution didn't shown significant difference. 3. The value of Cysteine(Cys) on the constitution didn't shown significant difference, but between the value of SO-YANG-IN and SO-EUM-IN showed significant difference. 4. Between the value of control group and SO-YANG-IN showed significant difference, and between the value of SO-YANG-IN and TAE-EUM-IN, SO-YANG-IN and SO-EUM-IN showed significant difference. 5. Between the value of control group and SO-YANG-IN showed significant difference.

Identification of Carotenoids from Green Alga Haematococcus pluvialis by HPLC and LC-MS (APCI) and Their Antioxidant Properties

  • Haematococcus pluvial is, a green alga, accumulates astaxanthin (3,3'-dihydroxy-$\beta$,$\beta$'-carotene-4,4'-dione) upto 2-3% on a dry weight basis. In the present study, identification of carotenoids from Haematococcus cyst cell extract by HPLC and LC-MS (APCI) and their antioxidant properties were evaluated in in vitro model systems. The extract exhibited 89% and 78% antioxidant activities in the $\beta$-carotene linoleate model and the hydroxyl radical scavenging model, at 9 ppm of total carotenoid, respectively. The extract also showed 80%, 85%, and 79% antioxidant activities against lipid peroxidation in the kidney, brain, and liver of rats. Low-density lipoprotein oxidation induced by $Cu^{2+}$ ions was also protected (45%, 64%, and 75%) by the extract in a dose-dependent manner with different carotenoid levels. Thiobarbituric acid reactive substances concentration in the blood, liver, and kidney of rats were also significantly (p<0.005) decreased in H. pluvialis-treated rats. The potent antioxidant activity is attributable to various carotenoids present in the extract.

  • An extract of Alnus japonica (Betulaceae) cortex has been traditionally used for purifying blood, and curing feces containing blood, enteritis, diarrhea, alcoholism and cut wounds. In the preliminary test was carried out for determining whether it has the novel pharmacological activity, the butanol fraction showed significant inhibitory effect on carrageenan-induced paw edema as an acute inflammation, HCI.ethanol-induced gastric lesion and aspirin-ligation gastric ulcer. EFFECT OF EXERCISE ON MILK YIELD, MILK COMPOSITION AND BLOOD METABOLITE CONCENTRATIONS IN HEREFORD × FRIESIAN CATTLE

  • Three experiments were carried out in which lactating Hereford ${\times}$ Friesian cattle walked up to ten kilometers a day for three periods of five days with two non-walking days between each walking period and in which the animals were fed different diets. Measurements were made of milk yield, milk constituent yields and concentrations and blood metabolite concentrations. Exercise caused significant reductions in milk yield and in the yields of lactose and milk protein. Milk fat yield was not reduced when animals were exercised. During exercise the concentrations of ${\beta}-OH$ butyrate and free fatty acids increased, whereas the concentrations of glucose, magnesium and inorganic phosphorus decreased. Diet influenced the effect of exercise on some blood metabolite concentrations.

Ultrasonic treatment of waste livestock blood for enhancement of solubilization

  • The aim of this study was to recycle the waste livestock blood as one of the waste biomass by turning proteins, the main constituent of blood, into effective biological resources like amino acid. Ultrasonic technology was applied to solubilize the proteins in the waste livestock blood. And of the multiple ultrasonic frequencies tested, 20 kHz was confirmed to yield the highest solubilization rate. The optimum pretreatment conditions were determined to be 30-min treatment at an ultrasonic irradiation density of 0.5 W/mL, which resulted in a solubilization rate of 96.01%. Also, a gel permeation chromatography (GPC) confirmed that a large amount of proteins were solubilized, and in an experiment where ultrasonic treatment was applied to kill bacteria, death rates of general bacteria and total coliforms were found to be reduced by 99.93% and 100%, respectively. Based on these results, ultrasonic technology was confirmed to be a crucial part of treating and recycling the proteins in waste livestock blood.
