Mineral extraction from by-products of brown rice using electrodialysis and production of mineral salt containing lower sodium (전기투석을 이용한 현미부산물로부터의 미네랄성분 추출 및 나트륨감량형 미네랄 소금 제조)

  • The purpose of this study was to develop a supplemental healthy food that can help prevent high blood pressure-related diseases caused due to the excessive consumption of sodium in salt. This was achieved by using ion-displacement techniques to produce mineral salt with lower sodium content by using fermented brown rice by-products rich in minerals. Mineral salt containing 2019.2 mg/100 g of potassium, 678.5 mg/100 g of magnesium, 48.7 mg/100 g of calcium, and 19.5 mg/100 g of sodium was obtained by fermenting brown rice by-products to create a culture medium for the mineral salt. Mineral salt containing 1769.7 mg/100 g of potassium, 573.6 mg/100 g of magnesium, 35.3 mg/100 g of calcium, and 19.5 mg/100 g of sodium was obtained by filtering and refining the by-product extract of fermented brown rice. The results showed that when the stream velocity of the instrument used for electrolysis was 200 mL/min and the current and the concentration of the reactive liquid in the purified water chamber were higher, the effect of electrolysis was greater. Ion hot water extraction of the fermented brown rice by-products improved by up to 95% and was collected as purified water within 90 min of the reaction time. Chloride ions with pH 7.4 were produced by mixing sodium hydroxide in a purified saline water chamber with electro-analyzed water. The salt produced in this study contained low sodium, 5.7~30%, as compared to 40% sodium content of the normal salt.

Prediction of Surgical Anesthesia in Medetomidine/Tiletamine/Zolazepam Anesthetized Dogs using Electroencephalography (Medetomidine/Tiletamine/Zolazepam 마취견에서 뇌전도를 이용한 외과마취의 평가)

  • Changes of electroencephalogram (EEG), mean arterial blood pressure (MAP) and heart rate under surgical anesthesia were investigated in medetomidine (MED) and tiletamine/zolazepam (ZT)-anesthetized dogs. To determine the level of surgical anesthesia, pedal withdrawal reflex was regularly tested after ZT injection. The first time point without pain response was regarded as the beginning of surgical anesthesia (SSA). After SSA, the first time point showing positive pain response was considered the end of surgical anesthesia (ESA). Comparing the control, an additional significant decrease of ${\delta}2$ and ${\alpha}2$ was observed at SSA. Comparing the control, ${\delta}2$ was significantly decreased at ESA. Significant reductions of MAP were observed at pre-ESA and ESA. Heart rate significantly decreased in all stages. These results suggest that ${\delta}2$ band power is a valuable parameter for correlating surgical anesthesia in dogs anesthetized with MED and ZT.

Angiotensin II-Induced Generation of Reactive Oxygen Species Is Regulated by a Phosphatidylinositol 3-Kinase/L-Type Calcium Channel Signaling Pathway (Angiotensin II에 의해 유도되는 활성산소발생 기전에 대한 연구)

  • Angiotensin II (AngII) is an essential hormone that affects vascular physiology. For example, stimulation of vascular smooth muscle cells (VSMCs) rapidly induces vasoconstriction and results in the up-regulation of blood pressure. Chronic stimulation of VSMCs with AngII also results in hypertrophy. In this study, we confirmed an involvement of phosphatidylinositol 3-kinase (PI3K)-dependent calcium mobilization in AngII-induced generation of reactive oxygen species (ROS). Stimulation of rat aortic smooth muscle cells (RASMCs) with AngII significantly induced the generation of ROS in a dose- and time-dependent manner. AngII-induced generation of ROS was completely abolished by pharmacological inhibition of PI3K (with LY294002), but inhibition of the ERK signaling pathway had no effect. AngII-induced calcium mobilization was completely blocked by inhibition of PI3K, whereas inhibition of the ERK signaling pathway by PD98059 was ineffective. Depletion of extracellular calcium or inhibition of the L-type calcium channel by nifedipine completely blocked AngII-induced calcium mobilization. Depletion of extracellular calcium by EGTA and incubation of RASMCs with calcium-free medium both significantly blocked AngII-induced ROS generation. Inhibition of the L-type calcium channel also significantly blocked AngII-induced ROS generation. These results suggest that AngII-induced ROS generation is regulated by calcium mobilization, which, in turn, is modulated by a PI3K/L-type calcium channel signaling pathway.

Eleutheroside E Content in Eleutherococcus spp. (오갈피속(屬) 식물(植物)의 Eleutheroside E 함양(含量))

  • The root and stem bark of Eleutherococcus spp. is used for the treatment of paralysis, rheumatism, arthritis, diabetes, stamina failure, high blood pressure, etc. This study was con­ducted to identify botanical origins and to select standard variety of Eleutherococcus spp. with analyzing eleutheroside E content(%) of root bark and that of Eleutherococcus senticosus at different collected area, time, part and grown area. The content of eleutheroside E in the root bark was determined by HPLC, and it was 0.1925% in Eleutherococcus senticosus, 0.1103% in E. chiisanensis, 0.0348% in E. koreanum and 0.0236% in E. sieboldianum. In seasonal change of eleutheroside E content, it was higher in autumn product than summer product in Eleutherococcus senticosus. It also was higher in root bark than stem bark in different part. To investigate eleutheroside E content of root bark among different collected local area in E. sen­ticosus, it was high order to Mt. Deokyu, Hokkaido and Mt. Seolak as a 0.1625%. 0.0493% and 0.027 %, respectively. The growth of Eleutherococcus senticosus was good at grown Unbong (altitude 10m), but eleu­theroside E content was high at grown Nogodan (altitude 1, 450m).

Effects of Iksujisundan on Renal Function, Peroxynitrite Scavenging Activity and Polyol Pathway in Streptozotocin-induced Diabetic Rats (익수지선단(益壽地仙丹)이 Streptozotocin으로 유발된 당뇨병 흰쥐의 신기능, 활성산소, 활성질소 및 Polyol Pathway에 미치는 영향)

  • Objectives : Diabetes is a disease in which the body does not produce or properly use insulin. Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Iksujisundan has been known to be effective for the treatment of diabetes. The present study was carried out to investigate the effect of Iksujisundan on renal function, peroxynitrite(ONOO-) scavenging activity and polyol pathway in streptozotocin-induced diabetic rats. Methods : The crushed Iksujisundan was extracted 3 times, each time with 3 volumes of methyl alcohol at 60$^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 87.8g. Iksujisundan extract was orally administreted at 100 mg per 1 kg of body weight for 20 days to the diabetic rats induced by streptozotocin(60mg/kg). The effects of Iksujisundan extract on the streptozotocin-induced diabetic rats were observed by measuring the serum level of glucose, insulin, lipid components, creatinine and BUN, and also the kidney levels of superoxide anion radical(${\cdot}$O2-), nitric oxide(NO) and ONOO-, and also the enzyme activities involved in the polyol pathway. Results : The effects of Iksujisundan on the streptozotocin-induced diabetic rats with regards to body weight, blood glucose and indulin levels, creatinine and BUN levels, total cholesterol and triglyceride lavels, and HDL-cholesterol levels were all shown to be good enough to prevent and cure the diabetes and its complications. Iksujisundan inhibited the generation of ${\cdot}$O2-,NO and ONOO- in the kidney of streptozotocin-induced diabetic rats. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the streptozotocin-induced diabetic rats were reversed toward natural activities. Conclusions : Iksujisundan might inhibit the development of diabetes and its complications by scavenging reactive oxygen and nitrogen species, thereby by reducing oxidative stresses and also by regulating the activities of polyol pathway enzymes, all of which could help to recover kidney function.

Effect of Ethanol Extract of Diospyros Kaki Thunberg on the Motility of Isolated Rabbit Duodenum (감나무의 Ethanol 추출액(抽出液)이 척출가토(剔出家兎) 장관운동(腸管運動)에 미치는 영향(影響))

  • Diospyros Kaki Thunberg is the species of persimmon tree that grows in Korea. Although its fresh or dried fruits are often served as a desert, it has little been known if persimmon tree has any specific pharmacological action. The leaves and branches of persimmon tree has long been used as folk remedies for palsy and frostbite in the southern part of Korea and it is also in use for the treatment of hiccup and nocturnal enuresis in chinese herbal medicine. Recently it was reported that an intravenous administration of Diospyros Kaki Thunberg ethanol extract (KTEE) into the animals lowered arterial blood pressure. Lee concluded from his study on the mechanism of depressor action of KTEE that at least a part of depressor response he observed was caused by acetylcholine-like action of KTEE. On the other hand little study has been made on the effect of KTEE on the motility of isolated animal intestines. Therefore the present study was undertaken to investigate effect of KTEE and the mechanism of its action on the motility of isolated rabbit duodenum. Ethanol extract of Diospyros Kaki Thunberg was prepared by boiling 1 kg of dried branches of persimmon tree in 1 liter of ethanol and the motility of isolated rabbit duodenum was recorded on physiograph by means of force transducer connected with Magnus apparatus. Doses of KTEE used were $5{\times}10^{-4}gm/ml,\;1{\times}10^{-3}gm/ml,\;and\;2{\times}10^{-3}gm/ml$. And the isolated duodenum was separately pretreated with acetylcholine $(5{\times}10^{-7}gm/ml)$, pilocarpine $(2.5{\times}10^{-6}gm/ml)$, histamine $(5{\times}10^{-6}gm/ml)$ and barium chloride $(2.5{\times}10^{-5}gm/ml)$ in order to find out interactions of these drugs with KTEE. The results obtained are as follows: 1. At doses of $5{\times}10^{-4}gm/ml,\;1{\times}10^{-3}gm/ml$ KTEE reduced contractions of isolated duodenum, while tonus as well as contaction of duodenum were depressed with $2{\times}10^{-3}gm/ml$ of KTEE. 2. Since the inhibitory effect of KTEE on the intestinal motility was not blocked by pretreatment with acetylcholine, pilocarpine, and barium chloride, it was strongly suggested that the inhibitory action of KTEE on intestinal motility is mainly Caused by its antihistamine effect. 3. It is also concluded that the principal substance of KTEE responsible for inhibition of intestinal motility may also have a vasodilating activity and would not be an acetylcholine-like substance in case it is same substance as that cause depressor responses.

Effect of hydrothermal processing on ginseng extract

  • Background: Panax ginseng Meyer is cultivated because of its medicinal effects on the immune system, blood pressure, and cancer. Major ginsenosides in fresh ginseng are converted to minor ginsenosides by structural changes such as hydrolysis and dehydration. The transformed ginsenosides are generally more bioavailable and bioactive than the primary ginsenosides. Therefore, in this study, hydrothermal processing was applied to ginseng preparation to increase the yields of the transformed ginsenosides, such as 20(S)-Rg3, Rk1, and Rg5, and enhance antioxidant activities in an effective way. Methods: Ginseng extract was hydrothermally processed using batch reactors at $100-160^{\circ}C$ with differing reaction times. Quantitative analysis of the ginsenoside yields was performed using HPLC, and the antioxidant activity was qualitatively analyzed by evaluating 2,2'-azino-bis radical cation scavenging, 2,2-diphenyl-1-picrylhydrazyl radical scavenging, and phenolic antioxidants. Red ginseng and sun ginseng were prepared by conventional steaming as the control group. Results: Unlike steaming, the hydrothermal process was performed under homogeneous conditions. Chemical reaction, heat transfer, and mass transfer are generally more efficient in homogeneous reactions. Therefore, maximum yields for the hydrothermal process were 2.5-25 times higher than those for steaming, and the antioxidant activities showed 1.6-4-fold increases for the hydrothermal process. Moreover, the reaction time was decreased from 3 h to 15-35 min using hydrothermal processing. Conclusion: Therefore, hydrothermal processing offers significant improvements over the conventional steaming process. In particular, at temperatures over $140^{\circ}C$, high yields of the transformed ginsenosides and increased antioxidant activities were obtained in tens of minutes.

Effect of 12 Hours Fasting Resistance Exercise on Metabolic Efficiency and Fatigue in Convergence Ages (융복합 시대의 12시간 공복저항운동이 대사효율성 및 피로물질에 미치는 영향)

  • The purpose of this study to examine the effect of 12 hours fasting resistance exercise on metabolic efficiency and fatigue of middle-aged female for 8 weeks. 50 middle-aged female target group pre-test exercise group(EX) and the control over the homogeneity in the control group(CON) were selected for this purpose divided into two groups. Exercise group(EX) is maintained after 8 weeks four times weeks 12 hours fasting resistance was performed for 60 minutes, the control group(CON) were compared to a normal life with the group of the exercise group(EX). Resistance applied to the exercise group(EX) were using free weights and a fixed mechanism applied around the upper and lower body exercise program for heart muscle, exercise intensity based on the 70% 1RM until 4 weeks, 5-8, which was performed by the aid program set to 80%. Through this procedure were as follows. First, 12 hours fasting resistance exercise showed the significance of the interaction effect in metabolic efficiency, blood pressure(SBP, DBP), fat, fasting glucose, fasting insulin(p<.01). Second, 12 hours fasting resistance exercise showed the significance of the interaction effect in the treatment program in which lactic acid, uric acid(p<.01).

  • The purpose of this study was to identify the risk factors influencing stroke. The research method was used two-stage stratified cluster sampling using data from the National Health and Nutrition Survey (2016-2018). This study subjects performed a complex sample logistic regression analysis on adults aged 19 or older. Risk factors affecting stroke were shown as follows: in age groups, odds ratio were conformed when people are in their 70s, the rate is 8.861 times higher; when they are in their subjective health conditions, the odds ratio are 4.501 times higher in the bad groups; odds ratio was 3.158 times higher in the group diagnosed with hypertension and 1.598 times higher in the group diagnosed with diabetes. The result of this study found that chronic diseases such as high blood pressure and diabetes and the management of senior citizens are important because stroke is dangerous and can be managed through prevention.

Clinical Observation on Effects and Adverse Effects of Choweseuncheng-tang on Obesity Patients (비만 환자에 대한 조위승청탕의 효과 및 부작용에 관한 임상적 고찰)

  • Objective : This study was designed to investigate the effects and adverse effects of Choweseuncheng-tang on obesity patients Methods : Choweseuncheng-tang was given to 20 obesity patients who have no abnormality of GOT, GPT and past medical history. Additionally, electroacupuncture was performed on Fuchao(腑巢) with 60Hz. We compared body weight, body fat and laboratoy test(GOT, GPT, BUN, Creatinine) between before and after treatment. Blood pressure, heart rate and clinical symptoms were checked to investigate the side effects. Decrease of body weight was checked in accordance with Sasang constitutional type, appetite type, and BMI. Results : 6 patients(BMI 23-24.9), 10 patiets(BMI 25-29.9), and 4 patients(BMI over 30) were showed decrease of body weight. We found body fat was decerased $3.16{\pm}1.25%$. BUN, Creatinine, GOT and GPT were increased within normal range. There were adverse event of autonomic hyperactivity(11 patients), gastrointestinal symptoms(7 patients) and others(3 patients). Conclusions : According to above results, Choweseuncheng-tang could be used effectively, but should be taken cautiously on obese patients.
