Effect of Crude saponin from Red-ginseng efflux on Blood biochemical parameters in Rats Acutely Exposed to 2,3,7,8-tetrachlorodibenzo-ρ-dioxin (TCDD) (홍삼유출액으로부터 분리한 조사포닌이 TCDD (2,3,7,8-Tetrachlorodibenzo-ρ-dioxin)로 급성독성을 유도한 흰쥐의 혈액 생화학지수에 미치는 영향)

  • This study was carried out to investigate the protective effect of crude saponin from red ginseng efflux (RGE-CS) on biochemical parameters in male rats acutely exposed to 2,3,7,8-tetrachlorodibenzo-$\rho$-dioxin (TCDD). Forty male rats ($200{\pm}20g$) were divided into 4 groups. Normal control group (NC) received vehicle and saline; only TCDD-treated group (TT) received TCDD ($5{\mu}g/kg$, single dose) intrperitoneally; RGE-CS 20 received 20 mg/kg of crude saponin i.p. for 4 weeks from 1 week before TCDD-exposure; RGE-CS 40 also received 40 mg/kg of crude saponin i.p. for 4 weeks from 1 week before TCDD-exposure. Body weight of TT group was significantly decreased after TCDD-exposure. However, body weight of crude saponin groups increased throughout the experimental period, although the increasing rate was slower than that of NC group. Decrease in body weight was not observed during the experimental period in RGE-CS 40. Increases in triglyceride (TG), total cholesterol (TC), low-density lipoprotein (LDL), AST, ALT and $Fe^{2+}$ levels by TCDD intoxication were significantly attenuated by the RGE-CS treatment. Decrease in glucose, amylase, lactate dehydrogenase (LDH) and creatinine kinase (CK) by TCDD also were inhibited by the RGE-CS. These results suggest that saponin from red-ginseng efflux might be a useful protective agent against TCDD, an endocrine disrupter.

Assessment of Allograft Function in Dog Single Lung Transplantation on CT (한국산 잡견에서 컴퓨터단층촬영을 이용한 이식폐의 기능평가)

  • In the field of the experimental lung transplantation, we analyzed the CT findings of acute rejection, infection in the left single allotransplanted lung of adult mongrel dogs, and the CT findings were compared with the histological findings obtained by the lung biopsy Twenty two adult mongrel dogs were divided into two groups(Donor and recipient group). Donor lungs were flushed with LPDG(low pota,ilium dextral glucose) solution(n=4) or modified Euro-collins solution(n=7) and preserved over 20 hours with $10^{\circ}C(1$ case preservation for 4hours). After left single lung transplantation, the chest X-ray and sequential computed tomogram were performed with concomitant hemodynamic study and arterial blood gas analysis on immediate postoperative period, postoperative 3rd day and postoperative 7th day. Two of eleven transplanted lungs had acute rejection which was represented as moderate infiltration at immediate or 1st postoperative d y but became extensive infiltration at postoperative 3rd day on CT. There were showed one case of bronchopleural fistula, six cases of pneumonia and two cases of pulmonary infarction. In one rejection cases, the opacity of transplanted lung was improved by injection of methylprednisolone 500mg daily during 3 days. We concluded that CT was a useful noninvasive evaluation parameter after lung transplantation and the serial CT scan enabled early detection of acute rejection.

Prediction of Surgical Anesthesia in Medetomidine/Tiletamine/Zolazepam Anesthetized Dogs using Electroencephalography (Medetomidine/Tiletamine/Zolazepam 마취견에서 뇌전도를 이용한 외과마취의 평가)

  • Changes of electroencephalogram (EEG), mean arterial blood pressure (MAP) and heart rate under surgical anesthesia were investigated in medetomidine (MED) and tiletamine/zolazepam (ZT)-anesthetized dogs. To determine the level of surgical anesthesia, pedal withdrawal reflex was regularly tested after ZT injection. The first time point without pain response was regarded as the beginning of surgical anesthesia (SSA). After SSA, the first time point showing positive pain response was considered the end of surgical anesthesia (ESA). Comparing the control, an additional significant decrease of ${\delta}2$ and ${\alpha}2$ was observed at SSA. Comparing the control, ${\delta}2$ was significantly decreased at ESA. Significant reductions of MAP were observed at pre-ESA and ESA. Heart rate significantly decreased in all stages. These results suggest that ${\delta}2$ band power is a valuable parameter for correlating surgical anesthesia in dogs anesthetized with MED and ZT.

Inhibitory Effect of PME88 MelonSOD on the Ultraviolet-Induced Photo-aging (PME88 멜론SOD의 자외선으로 인한 피부 광노화 억제 효과)

  • PME88 (gliadin-combined) melon superoxide dismutase (SOD) is known to promote the production of the body‘s own natural antioxidants including superoxide dismutase, catalase and glutathione peroxidase. In this study, we investigated the inhibitory effects of PME88 melonSOD on the ultraviolet-induced photo-aging by the evolution of minimal erythemal dose (MED), erythema quotation and spectrocolorimetric measurements of erythema. The analysis of the evolution of the MED showed a significant increase 28 days after the daily taken of the PME88 melonSOD. The analysis of the erythema quotation showed that on D29, for the dose 1.25 MED, erythema intensity is significantly higher for placebo group than for PME88 melonSOD group. At doses 0.64 MED$_{D14}$, 0.80 MED$_{D14}$ and 1 MED$_{D14}$ the value of parameter $a^*$ (the most sensitive to the colour changes bound to the variations of blood flow. It permits to assess the evolution of erythema) is significantly higher for placebo group. No significant difference has been observed between groups (PME88 melonSOD and placebo) on the evolution of the number and consistency of feces after 4 weeks of treatment. No intolerance has been observed during the 4 weeks of treatment. These results mean that PME88 melonSOD as a dietary supplement could be useful to attenuate ultraviolet-induced skin photo-aging.

Maintenance of Platelet Counts with Low Level QC Materials and the Change in P-LCR according to Hemolysis with XN-9000 (XN-9000장비에서 Low Level QC물질에서의 혈소판 수 관리와 용혈에 따른 P-LCR의 변화)

  • The platelet count in clinical laboratories is essential for the diagnosis and treatment of hemostasis abnormalities, and accurate platelet counting in the low count range is of prime importance for deciding if a platelet transfusion is needed and for monitoring after chemotherapy. Quality control is designed to reduce and correct any deficiencies in the internal analytical process of a clinical laboratory prior to the release of patient results. Fragmented erythrocytes are the major confusing factors for platelet counting because of their similar size to platelets. The authors found that the low range QC values were out of 2SD with a Sysmex automatic analyzer in internal quality control process. Thus far, there has been little discussion on the relationship between hemolysis and the platelet parameters. Therefore, this study focused on the performance of automated platelet counts, including the PLT-F, the PLT-I, and PLT-O methods at the low platelet range using the low level QC materials and compared the 5 platelet parameters with the hemolyzed samples. The results showed that the CV was the smallest with PLT-F and P-LCR increased from 18.4 to 31.9% in the hemolysis samples. These results indicate that a more accurate estimation of the platelet counts can be achieved using the PLT-F method than the PLT-I method at the low platelet range. The use of the PLT-F system improves the confidence of results in low platelets samples in a routine hematology laboratory. The results suggest that P-LCR is a new parameter in assessing samples when the specimen is suspected of hemolysis and deterioration. Nevertheless, further studies will be needed to establish the relationship with P-LCR and hemolysis using human blood specimens.

Effect of Salinity on Survival, Growth and Physiological respone of Juvenile Chum Salmon (Oncorhynchus keta) (연어(Oncorhynchus keta) 치어의 생존, 성장 및 생리학적 반응에 미치는 염분의 영향)

  • This study was conducted to investigate the effects of different salinity on growth, survival and hematological parameters of juvenile chum salmon (Oncorhynchus keta). The fish were tested at salinity 0, 10, 20 and 30‰ for 30 days with three replicated groups. After the 30 days, the survival rate was 87.5% or more for 0, 10, 20‰ and significantly decreased to 75% at 30‰. Weight gain, specific growth rate, feed efficiency, daily feed intake, daily protein intake and protein efficiency ratio were the highest at 10‰. In the plasma components, the alanine aminotransferase (ALT) was significantly decreased at 10, 20 and 30‰ in compared with 0‰, whereas sodium (Na+) and chloride (Cl-) was significant increased and there was no significant change in the total protein (TP) and potassium (K+). In conclusion, the optimal salt concentration for the growth of juvenile chum salmon (11~33 g) was confirmed to be 10‰, but the association with the change in blood composition should be further studied.

Added Value of Contrast Leakage Information over the CBV Value of DSC Perfusion MRI to Differentiate between Pseudoprogression and True Progression after Concurrent Chemoradiotherapy in Glioblastoma Patients

  • Purpose: To evaluate whether the added value of contrast leakage information from dynamic susceptibility contrast magnetic resonance imaging (DSC MRI) is a better prognostic imaging biomarker than the cerebral blood volume (CBV) value in distinguishing true progression from pseudoprogression in glioblastoma patients. Materials and Methods: Forty-nine glioblastoma patients who had undergone MRI after concurrent chemoradiotherapy with temozolomide were enrolled in this retrospective study. Twenty features were extracted from the normalized relative CBV (nCBV) and extraction fraction (EF) map of the contrast-enhancing region in each patient. After univariable analysis, we used multivariable stepwise logistic regression analysis to identify significant predictors for differentiating between pseudoprogression and true progression. Receiver operating characteristic (ROC) analysis was employed to determine the best cutoff values for the nCBV and EF features. Finally, leave-one-out cross-validation was used to validate the best predictor in differentiating between true progression and pseudoprogression. Results: Multivariable stepwise logistic regression analysis showed that MGMT (O6-methylguanine-DNA methyltransferase) and EF max were independent differentiating variables (P = 0.004 and P = 0.02, respectively). ROC analysis yielded the best cutoff value of 95.75 for the EF max value for differentiating the two groups (sensitivity, 61%; specificity, 84.6%; AUC, 0.681 ± 0.08; 95% CI, 0.524-0.837; P = 0.03). In the leave-one-out cross-validation of the EF max value, the cross-validated values for predicting true progression and pseudoprogression accuracies were 69.4% and 71.4%, respectively. Conclusion: We demonstrated that contrast leakage information parameter from DSC MRI showed significance in differentiating true progression from pseudoprogression in glioblastoma patients.

A Study on the Drug Classification Using Machine Learning Techniques (머신러닝 기법을 이용한 약물 분류 방법 연구)

  • This paper shows the system of drug classification, the goal of this is to foretell the apt drug for the patients based on their demographic and physiological traits. The dataset consists of various attributes like Age, Sex, BP (Blood Pressure), Cholesterol Level, and Na_to_K (Sodium to Potassium ratio), with the objective to determine the kind of drug being given. The models used in this paper are K-Nearest Neighbors (KNN), Logistic Regression and Random Forest. Further to fine-tune hyper parameters using 5-fold cross-validation, GridSearchCV was used and each model was trained and tested on the dataset. To assess the performance of each model both with and without hyper parameter tuning evaluation metrics like accuracy, confusion matrices, and classification reports were used and the accuracy of the models without GridSearchCV was 0.7, 0.875, 0.975 and with GridSearchCV was 0.75, 1.0, 0.975. According to GridSearchCV Logistic Regression is the most suitable model for drug classification among the three-model used followed by the K-Nearest Neighbors. Also, Na_to_K is an essential feature in predicting the outcome.

Ascitic Fluid Analysis for the Differentiation of Malignancy-Related and Nonmalignant Ascites (악성 복수와 비악성 복수의 감별을 위한 복수액 분석)

  • The differentiation between malignancy-related ascites(MRA) and non-malignant ascites (NMA) is important for further diagnostic and therapeutic purposes. Although many parameters were investigated, none has provided a complete distinction between MRA and NMA. We investigated several ascitic fluid parameters to determine the differential power, and to differentiate malignant-related from nonmalignant-related ascites with a sequence of sensitive parameters followed by specific parameters. For the present study, 80 patients with ascites were divided into two groups: MRA and NMA, The MRA group was consisted of 27 patients with proven malignancy by image study, biopsy, and follow up: 21 of these patients had peritoneal carcinomatosis, but the remaining 6 showed no evidence of peritoneal carcinomatosis. The NMA group was consisted of 53 patients with no evidence of malignancy: among these patients, one had SLE, and others had liver cirrhosis, The samples of blood and ascites were obtained simultaneously, and then the levels of ascites cholesterol, CEA. protein and LDH, cytology, albumin gradient, ascites/serum concen-tration ratios of LDH(LDH A/S), and ascites/serum concentration ratios of protein(protein A/S) were measured. Applying cut-off limits for determined parameters, we estimated the diagnostic efficacy of each parameter, Among the eight parameters investigated, ascites fluid cholesterol yielded the best sensitive value of 93%(cut-off value 30mg/dl), and cytologic examination and the protein A/S(cut-off value 0.5) showed the most specific value of 100% and 96%, respectively. Based on the above results, the diagnostic sequence with cholesterol as a sensitive parameter followed by the combination of cytologic examination and protein A/S as specific parameters, was tested in 80 patients. This diagnostic sequence identified 81.5% of patients with malignancy, and all patients with peritoneal carcinomatosis were classified as malignancy-related ascites. In spite of many limitations, this proposed diagnostic sequence may permit a cost-effective and simple differentiation of malignancy-related ascites from nonmalignant ascites.

Effect of Peking-Duck By-Product Extracts Supplemented with Medicinal Herbs on Serum Heavy Metal Levels and Blood Parameters of Rats Exposed to Lead and Mercury (한약재를 첨가한 오리부산물 추출액이 납과 수은에 노출된 흰쥐 혈청의 중금속 및 혈액지표에 미치는 영향)

  • This experiment was planned to develop a functional supplement by food resources to prevent and lessen the deleterious effects caused by environmental pollutants such as polluted food, air, water and heavy metals. The goal of this study was to investigate the effects of peking-duck extracts supplemented with six kinds of medicinal herbs (DJ) on the intoxication of lead and mercury in rats. Sprague-Dawley rat weighing $150g\pm15g$ g, were randomly assigned to 5 groups, basal diet only (NCG), heavy metal without DJ injection (HCG), heavy metals and DJ (3 mg/mL) injection (HMLD), heavy metal and DJ (30 mg/mL) injection (HMMD), heavy metal and D] (300 mg/mL) injection (HMHD). Mecury (Hg) and lead (Pb) injected at the level of 50 ppm for 17 days. Also DJ oral feeding was continued for 31 days. The result of this study were as follows; Food intake and body weight gain in heavy metal administered groups were lower than those of control group (NCG). The activities of GOT, GPT and BUN level were significantly reduced in DJ-treated groups as compared to HCG. DJ was shown to suppress the accumulation of Hg and Pb in serum. The results suggest that DJ might have protective effect on Hg and Pb intoxication.
