Comparison of Oxidative Stress in Red Blood Cells Induced by Hair Dyeing Application to Young Women (염모제 도포방법에 따른 인체 적혈구의 산화스트레스 비교)

  • To ascertain the effects of hair dyeing application on oxidative stress in human, a mixture of permanent black colored hair dye with the same amount of oxidant containing 6% hydrogen peroxide was used. A hair dyeing with contacting the scalp (conventional dyeing) and a hair dyeing with 3 to 4mm away from the scalp (alternative dyeing) were applied to each 15 young healthy women. Blood was taken from the brachial vein at two sampling times, just before and 6 hours alter the hair dyeing, and antioxidant enzyme activities and antioxidant contents were measured in red blood cells. After dyeing, malondialdehyde(MDA) contents for conventional dyeing group was shown to a tendence of more increased than alternative dyeing group. After dyeing, reduced glutathione (GSH) contents for conventional dyeing group was shown to a tendence of more decreased than alternative dyeing group. After dyeing, superoxide dismutase (SOD) and catalase (CAT) activities were significantly decreased in conventional dyeing group (p < 0.01), however, SOD and CAT activities were not significantly decreased in alternative dyeing group. After dyeing, there was no significant decrease in glutathione peroxidase (GPX) activity both for conventional dyeing group and alternative dyeing group. Therefore, after dyeing, the degree of oxidative stress in red blood cells for alternative dyeing group was appeared to be lower than conventional dyeing group.

Effects of Korean Ginseng(Panax Ginseng, C. A. Meyer) Extracts on Rat Exposed to Heat Environment (고려인삼 추출물이 고온환경에 노출된 흰쥐에 미치는 영향)

  • It was investigated that how the periodic exposure to heat environment, and the treatment of korean white and red ginseng extracts had effects on the weight, diet uptake, blood components, organ weight, and the lipid peroxidation of liver in male S.D. rats. In the result of experiments using rats, chronic heat environment for 7 days at $38^{\circ}C$, 5 hrs per day, induced significant decrease of an average increase rate of body weight, but diet uptake was not affected clearly. In heat environment, the number of red blood cells and hemoglobin were not changed, but the number of white blood cells was significantly increased. The liver weight against body weight was decreased in rats. Also, MDA contents, related to lipid peroxidation, were remarkably increased in rat liver by heat environment. These physiological changes were attenuated by treatments of white and red ginseng extracts before and after exposure to heat environment, particularly in growth rate and lipid peroxidation of liver in rats. Also, red ginseng extracts had a better effect, though it was not that significant, than the white ginseng on the inhibition of lipid peroxidation and the weight change of liver. Although the investigation on the effective components and the study on the activity changes of associated materials are needed to perform, these present results imply that Korean ginseng may contribute to protection of body homeostasis against drastic climate changes.

Intragastrically Applicated CCl4-Thiopental Sodium Enhanced Lipid Peroxidation and Liver Fibrosis (Cirrhosis) in Rat: Malonedialdehyde as a Parameter of Lipid Peroxidation Correlated with Hydroxyproline as a Parameter of Collagen Synthesis (Deposition)

  • We investigated the pathogenesis of liver tissue damage during the lipid peroxidation and fibrogenesis with the observation of correlations between the parameters of collagen synthesis (and deposition) and lipid peroxidation in liver fibrosis (cirrhosis) rats. Rats were randomly divided into two groups, normal and $CCl_4$-thiopental sod. intoxicated group. And the one group was treated intragastrically with the mixture of $CCl_4$-thiopental sod. 3 times per week for 3 weeks. The liver tissue and sera were used for the measurement of hydroxyproline (HYP), malonedialdehyde (MDA) and superoxide dismutase (SOD). Biochemical parameters such as aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), total-bilirubin and blood urea nitrogen (BUN) were measured. Additionally, the expression of collagen ${\alpha}1$(III) and $\beta$-actin mRNA was observed by RTPCR. The histological change in liver tissue was also observed by Masson's trichrome and H&E staining. Correlation analysis was carried by Spearman's rho method. All biochemical parameters except total-bilirubin were significantly higher in the $CCl_4$-thiopental sod. treated group than that of the normal group (p < 0.01). In the $CCl_4$-thiopental sod. treated group, Hyp as a parameter of collagen synthesis (deposition) and MDA as a metabolite of lipid peroxidation, were significantly elevated by 1.98 and 2.11 times higher than that of the normal group (p < 0.001) respectively. The activity of SOD in the $CCl_4$-thiopental sod. treated group is decreased significantly by 44.8% (p < 0.001). And collagen ${\alpha}1$(III) mRNA was more expressed in the $CCl_4$-thiopental sod. treated group than that of the normal group. However, the expression of $\beta$-actin mRNA is showed similar in both of groups. A good correlation was observed between the content of hyp and MDA concentration (r = 0.70, n = 40) in the two groups. And the correlation between the levels of hyp and SOD (r = -0.71, n = 25) is also reliable. However, no correlation were observed between MDA concentration and SOD (r = -0.40, n = 25) in the two groups. Elevated levels of MDA in $CCl_4$-thiopental sod. treated rats indicated enhancement of lipid peroxidation, which is accompanied by a decrease in SOD activity. Moreover, we could confirm that the parameters of collagen synthesis (and deposition) is in good correlation with the metabolite of lipid peroxidation (MDA) and the lipid peroxidation antagonizing enzyme (SOD). Hence, we propose that (1) lipid peroxidation and collagen synthesis (and deposition) could be enhanced by intragastrically application of $CCl_4$-thiopental sod. during a short terms. And (2) the intoxication of $CCl_4$-thiopental sod. could be used for monitoring of lipid peroxidation and collagen synthesis (and deposition) for test of antioxidant and antifibrotic agent.

Effect of Low Ambient Temperature on the Concentration of Free Radicals Related to Ascites in Broiler Chickens

  • A flock of Arbor Acres chickens were reared in cages and provided with high energy pelleted feed. At 14 d of age, a total of 350 birds were separated into 3 groups randomly as follows: 100 birds were exposed to ambient temperature of 20$^{\circ}C$ as a control group, 150 birds were exposed to lower ambient temperature of 11$^{\circ}C$ to induce ascites (group I), and another group of 100 birds were exposed to lower ambient temperature of 11$^{\circ}C$ and fed diet containing 1% L-arginine for ascitic prophylactic treatment (group II). Blood and tissue samples (lung and liver) were collected from chickens at 3, 4, 5, 6 and 7 wk of age subsequently, to analyze the concentration and activities of free radicals, mononaldehyde (MDA), superoxide dismutase (SOD), Nitric Oxide (NO) and Nitric oxide synthase (NOS). The results showed that the prevalence of ascites in the control, group I and group II was 3%, 9.33% and 3% respectively (p<0.01). The concentration of free radicals in the lungs of 3 wks old preascitic broilers in group I was significantly higher than in the corresponding control group (p<0.05). The concentrations of free radicals in lung and liver in the 7 wk period, and that of NO and SOD in the plasma were significantly lower in group I than in the control group (p<0.01). However, the accumulated MDA contents in group I were higher than in the control group and group II (p<0.05), respectively. In the same way, the activity of NOS in group II was higher than both group I and control group (p<0.01) during the 7 wk period. There was no significant difference between SOD activities of group II and the control group (p>0.05), and also insignificant difference between NOS in group I and the control group (p>0.05). The results of this study indicate that there was a significant decrease in the concentration of MDA in group II. On the other hand, the concentration of free radicals decreased and MDA concentration increased in group I during the 7 wk period. The reduction in concentration of MDA in group II, following arginine supplementation may be associated with the scavenging activity of NO.

Antioxidative Activity and Lipid Composition from Different Part and Supplement of Codonopsis lanceolata in Rat (더덕 부위별, 첨가수준이 실험쥐의 항산화활성과 지질조성에 미치는 효과)

  • This study was conducted to examine antioxidative activity and lipid composition from different parts and supplement flesh and skin of Codonopsis lanceolata in vivo. Forty six-week-old white Sprague Dawley rats were divided into 5 groups and fed with experimental diet for six weeks to measure antioxidant enzymes activities and lipid composition in blood and liver microsome. The activity of glutathione peroxidase in blood was high in all groups supplemented with Condonopsis lanceolata and the difference was observed in accordance with the supplemented part rather than the supplemented level. However, glutathione reductase activity and the content of malondialdehyde (MDA) in blood showed difference depending on the level of supplementation rather than the supplemented part. The content of liver MDA in all groups supplemented with Condonopsis lanceolata was lower than that in the control group. As the level of skin supplementation increased, an increase in glutathione peroxidase activity was also observed. Only in the group that 5% of Condonopsis lanceolata skin was supplemented, the glutathione reductase activity was higher than in the control group. Total cholesterol and LDL-cholesterol of blood in the group supplemented with Condonopsis lanceolata flesh or skin were significantly lower than those in control group. HDL-cholesterol in blood was high when the flesh of Condonopsis lanceolata was supplemented. Total cholesterol and triglyceride in liver of the group supplemented with Condonopsis lanceolata flesh or skin were significantly lower than those in control group. In summary, this animal test showed that the supplementation of Condonopsis lanceolata, flesh or skin, generally improved the antioxidative effect of diet and lipid composition.

Effect of Broussonetia papyrifera L. silage on blood biochemical parameters, growth performance, meat amino acids and fatty acids compositions in beef cattle

  • Objective: The study was conducted to investigate the effects of Broussonetia papyrifera L. (B. papyrifera) silage on growth performance, serum biochemical parameters, meat quality, and meat amino acids and fatty acids compositions in beef cattle. Methods: Sixty-four male Angus beef cattle were assigned to 4 groups with 4 pens in each group and 4 beef cattle in each pen, and fed with the total mixed ration supplemented with 0%, 5%, 10%, or 15% B. papyrifera silage for 100 days (control group, 5% group, 10% group and 15% group) separately. Results: Beef cattle had significantly higher final body weight (BW) in 15% group, higher average daily gain (ADG) and dry matter intake (DMI) in 5% group, 10% group and 15% group, and higher feed conversion ratio (FCR) in 10% group and 15% group. Significantly higher blood superoxide dismutase (SOD) concentration was noted in 15% group, higher blood total antioxidant capacity (TAC) in 10% group and 15% group, lower 8-hydroxydeoxyguanosine (8-OHdG) and malondialdehyde (MDA) in 15% group. Meat had lower pH in 15% group, higher Commission International DeI'Eclairage (CIE) L in 5% group, 10% group, and 15% group, and lower drip loss in 15% group. Greater concentration of meat polyunsaturated fatty acids (PUFA) was observed in 10% group and 15% group, and docosahexaenoic acid (DHA) in 15% group. Conclusion: Diet with 15% B. papyrifera silage could improve performance and increase final BW, ADG, DMI, and FCR, enhance the antioxidant functions by decreasing blood 8-OHdG and MDA and increasing blood SOD and TAC, improve the meat quality by lowing pH and drip loss and increasing CIE L, increase the meat PUFA and DHA concentration. Polyphenols and flavonoids might be the main components responsible for the antioxidant activity and anti-biohydrogenation in the B. papyrifera silage. And B. papyrifera silage could be used as a new feedstuff in beef cattle nutrition.

The Effects of GONGCHENGWHAN on the Blood Cell, Serum and Antioxidant Effects on Serum, Brain Tissue of Mouse (공청환(拱淸丸)이 노화(老化) 백서(白鼠)의 혈액변화(血液變化) 및 혈청(血淸)과 뇌조직(腦組織)의 항산화물활성(抗酸化物活性)에 미치는 영향(影響))

  • This study was to investigate the effects of GONGCHENGWHAN(GCW) on the blood and brain tissues of aged rats. For the experiment, the aged rats were divided into three groups : Non treated group (NC), distilled water feeding group(PC), GCW feeding group(GCW).Each group was treated as the procedure for ten days before administration of scopolamine. After this treating, we injected scopolamine to Pc and GCW group their blood cell(WBc, RBC, platelet), blood serum (BUN, creatinine, glucose, uric acid), erythrocyte hemolysis, and the activities of cholinesterase as well as measured the amounts malondialdehysde in the blood serum, catalase, and SOD in the brain tissue.The main results of this investigation are as follows.1. In respect of the number of WBC, GCW group exhibited the administrated incerase in comparison with PC. In respect of the number of RBC· and platelet, however, the changes was not administrated.2. In respect of the number of BUN, creatinine, glucose and uric and the blood, GCW group exhibited the administrated decrease in comparison with PC.3. In respect of erythrocyte hemolysis, GCW group exhibited the administrated repression in comparison with PC.4. In respect of the activity of cholinestease in the blood serum, GCW group exhibited the administrated improvement in comparison with PC.5. In respect of the amounts of malondialdehyde in the blood serum, GCW group exhibited the administrated dexrease in comparison with PC.6. In respect of the activity of catalase in brain tissue, GCW group didn't exhibit the administrated change in comparison with PC7. In respect of the activity of SOD in brain tissue, GCW group exhibited the administrated improvement in comparison with PC.Results of this study indicates that GCW improves the activities of cholinesterase and SOD, but represses the formation of the free radical and the accumulation of the antioxidant materials such as MDA. It implies that GCW has some effects on antiaging.

  • This experiment was done to investigate the effects of GAMIKWYBICHONGTANG(GKCT) on the blood and brain tissues of aged rats. The experimental groups were divided into three groups and treated as follows for ten days before administration of scopolamine ; Non treated group(Normal), Distilled water feeding group (Control), GKCT feeding group(GKCT). After feeding them each, Control and GKCT were injected scopolamine for 5 days.We examined the changes of blood cell(WBC, RBC, platelet), blood serum(BUN, creatinine, glucose, uricacid), erythrocyte hemolysis, the activities of cholinesterase, and measured the amounts of malondialdehyde of the blood serum and checked the activities of catalase, SOD of the brain tissues.The results were as follows;1. GKCT showed significant increase of the number of WBC, but those of RBC and platelet didn't significantly changed in comparison with Control.2. GKCT showed significant decrease of BUN, creatinin, glucose, uric acid in blood serum in comparison with Control.3. Erythrocyte hemolysis were decreased significantly in GKCT in comparison with Control.4. About the activity of cholinesterase of blood serum, GKCT showed no significant increase in comparison with Control.5. In TBA reaction to measure the amount of MDA, oxidant material of blood serum of rats, GKCT showed significant decrease in comparison with Control.6. About the activity of catalase of brain tissue, GKCT showed no significant change in comparison with Control.7. About the activity of SOD of brain tissue, GKCT showed significant increse in comparison with Control.According to the above results, GKCT can reduce the formation of free radical and the accumulation of antioxidant materials, it is suggested that GAMIKWYBICHONGTANG(GKCT) has some effects on antiaging. It is also needed more following studies.

The Antioxidant Effect of Vitamin C and Deferoxamine on Paraquat-induced Cytotoxicity in Cultured Lymphocytes (배양림프구에서 Paraquat의 세포독성에 대한 Vitamin C와 Deferoxamine의 항산화 효과)

  • Purpose: As basic information of antioxidant treatments for the patient with paraquat intoxication, in human peripheral lymphocytes, the cytotoxicity of paraquat was measured, and to evaluate the antioxidant effect of vitamin C and deferoxamine against this cytotoxicity, malondialdehyde (MDA), superoxide dismutase (SOD) activity and total antioxidant status (TAS) were measured. Methods: From 10 healthy adults, after obtaining a consent, 20ml peripheral blood was collected. Experimental groups were divided to (1) control group, the group treated with an identical amount of saline, (2) P group: the group treated with paraquat only, (3) PV group: the group treated with paraquat followed by vitamin C 30 minutes later, (4) PD group: the group treated with paraquat followed by deferoxamine 30 minutes later, (5) PVD group: the group treated with paraquat followed by vitamin C 30 minutes later and subsequently deferoxamine one hour later, and (6) PDV group: the group treated with paraquat followed by deferoxamine 30 minutes later and subsequently vitamin C 1 hour later, and thus to total 6 groups. In each group, 10 samples of peripheral blood was assigned and $100{\mu}M\;paraquat,\;100{\mu}M$ vitamin C, and $100{\mu}M$ deferoxamine were used as reagent. Lymphocytes were isolated, cultured, and cytotoxicity was measured by the Microculture Tetrazolium method (MTT assay), MDA and SOD activity, and TAS concentration were measured. Results: In regard to the cytotoxicity measured in each group, their cytotoxicity was decreased in the group treated with antioxidants, in comparison with the group treated with paraquat only. In the cases that the order of the treatment of these two antioxidants was altered, viability in the PDV group $(1.077{\pm}0.121)$ was increased more that the PVD group $(0.888{\pm}0.152)$ statistically significantly (p=0.018). Concerning the amount of MDA, in comparison with the P group $(6.78{\pm}0.93{\mu}mol/L)$, after the treatment of each antioxidant, the concentration of MDA was decreased statistically significantly (p<0.05). In the group treated with two antioxidants together, in comparison with the group treated only with one antioxidant, the amount of MDA was increased statistically significantly $(PV:\;3.96{\pm}0.98{\mu}mol/L,\;PD:\;4.92{\pm}1.50{\mu}mol/L,\;PVD:\;3.22{\pm}0.83{\mu}mol/L,\;and\;PDV:\;3.42{\pm}0.95{\mu}mol/L,\;p=0.007)$. The concentration of SOD measured in the blood in each group after the administration of paraquat, in comparison with the control group, a pattern of the elevation of SOD activity and subsequent decrease was detected, however, it was not statistically significant. In the comparison of the groups treated with antioxidants, in comparison with the P group $(1419.9{\pm}265.9{\mu}mol/L)$, SOD activity was decreased statistically significantly in only the PDV group $(1176.4{\pm}238.9{\mu}mol/L)$ (p=0.017). In regard to TAS measured in each group, in comparison with the P group $(0.87{\pm}0.05{\mu}mol/L)$, in all groups treated with the antioxidants, the PV group was $1.00{\pm}0.03{\mu}mol/L$ (p=0.005), the PD group was $9.01{\pm}0.24{\mu}mol/L$ was $4.64{\pm}3.98{\mu}mol/L$ (P=0.005), and the PDV group was $9.41{\pm}0.27{\mu}mol/La$ (p=0.005), and thus total antioxidant activity was increased statistically significantly In a multiple comparison test, the PDV group showed the highest total antioxidant activity (p<0.0001). Conclusion: The result of the assessment of the antioxidant effect of vitamin C and deferoxamine on paraquat-induced cytotoxicity showed that in regard to cytotoxicity, SOD activity and TAS measurement, the best result was observed in the PDV group. Therefore, it was found that vitamin C and deferoxamine were effective antioxidants for the paraquat-induced cytotoxicity, and it suggests that the administration of deferoxamine followed by vitamin C may improve their antioxidant effect more.

Investigation of the Protective Effect of Kefir against Isoproterenol Induced Myocardial Infarction in Rats

  • This study aims to investigate the protective effects of kefir against myocardial infarction induced by isoproterenol (ISO). The rats were randomly divided into 4 groups, each group consisting of 8 rats. The control group, the kefir group (5 mL/kg/d kefir administered to rats as intra-gastric gavage for 60 d), the ISO group (100 mg/kg ISO was administered to rats, s.c. on 61. and 62. d), and kefir+ISO group (5 mL/kg/d kefir was administered to rats intra gastric gavage for 60 days prior to ISO, 100 mg/kg in two doses on day 61 and 62). 12 h after the last ISO dose, all rats were decapitated and their blood samples were collected. Cardiac tissue was reserved for histopathological examination. creatine kinase (CK), alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), triglycerides, total cholesterol,very low density lipoprotein (VLDL), low density lipoprotein (LDL), high density lipoprotein (HDL) and glucose were measured by autoanalyzer, whole blood malondialdehyde (MDA), glutathione (GSH) and plasma advanced oxidation protein products (AOPP) levels were measured spectrophotometrically. It was determined that in the group of kefir+ISO, the levels of AST (p<0.001), CK (p<0.001), LDH (p<0.001), MDA (p<0.001) and AOPP (p<0.001) were decreased, while the GSH (p<0.05) increased, compared to ISO group. There were no significant changes in lipid profile and glucose levels between these two groups. In conclusion, by examining cardiac enzymes and histopathological changes in cardiac tissue, it can be concluded that the administration of kefir in myocardial infarction induced by ISO can protect the heart with its antioxidant characteristic and minimize the toxic damage created by ISO.
