The aim of the present study was to investigate the ovulation rate and its relationship to fertilization ability in Landrace, Durock and Crossbred pigs. Gilts were natural mated at a body weight of at least 120 kg under the same hormone treatment. Embryos were surgically collected 1 day after natural mating (Day 0). Embryos derived from in vivo-fertilized oocytes were cultured in medium PZM-3. The ovaries were examined and the pathological findings were recorded. The number of corpus hemorrhagicum was counted, and was assumed to equal the ovulation rate. There was no difference in the number of corpus hemorrhagicum (20.4, 28.8 and 23.2) and ovulation (13.5, 26.8 and 17.2) in the Landrace, Durock and Crossbred pigs. The two pronucleus formation was 76.0, 80.0 and 86.9%. The Day-7 embryos had blastocyst rates of 68.0, 75.0 and 73.9%. There was no difference in the number of total cells and apoptotic cells. In the future, more studies require determining relationships between ovulation and fertilization rate in different species of pigs.
The ability to preselect the sex of piglets is advantageous in the pig industry. The objective of this study was to examine the feasibility of using intracytoplasmic sperm injection (ICSI) with sorted spermatozoa to produce piglets with a preselected sex. Pig embryos were produced by ICSI of frozen X- and Y-sperm that had been separated by flow cytometry. The developmental competence of the embryos was investigated in vitro and in vivo. The populations of X- and Y-spermatozoa were 52.7% and 47.3%, respectively in our samples. The in vitro development of ICSI embryos was enhanced by longer of in vitro maturation of oocytes ($44{\sim}48\;h$ vs. $40{\sim}43\;h$). Their cleavage ($65{\sim}70%$) and blastocyst formation ($9{\sim}12%$) rates were not significantly different between male and female ICSI embryos, or between sorted and unsorted sperm-derived embryos. One pregnancy was established in a recipient that was transferred with 110 female ICSI embryos, but the pregnancy was terminated on Day 89 of gestation. Our results suggest that the separation X- and Y-spermatozoa by flow cytometric sorting can be a useful tool in combination with ICSI for the production of pig embryos and piglets of preselected sex.
Purpose: These experiments were undertaken to evaluate the effect of administration of Palmulgunjatang gamibang on ovarian functions in female mice. Methods: We administered the Palmulgunjatang gamibang to 6-week-old female CF-1 mice for 4, 8, or 12 days. After administration of Palmulgunjatang gamibang with different concentration, the female mice were injected PMSG and hCG for ovarian hyperstimulation. The mice were divided into 3 different groups for each experiment. To compare the differences, we set a control group treated with plain water at the same volume by the same way. Results: In case of 4-day, 8-day, 12-day administration of Palmulgunjatang gamibang, the mean number of total ovulated oocytes and the number of morphologically normal oocytes were increased compared with control group. We were also examined the embryonic developmental competence in vitro. In case of 4-day administration of Palmulgunjatang gamibang, the rates of blastocyst formation from 2-cell stages were higher than control group. Conclusion: From our results suggested that the medication of Palmulgunjatang gamibang has beneficial effect on reproductive functions of female mice via promotion of cell proliferation.
Embryonic stem (ES) cells are derived from the inner cell mass of the blastocyst-stage embryos that can be propagated indefinitely and, at the same time, can be differentiated into all the cell types that constitute the body. Current research using ES cells is mainly focused on the efficient generation of specific cell types by employing optimal differentiation conditions, which often requires the genetic manipulation of ES cells. As a way of developing an efficient system to regulate foreign gene expression in ES cells, we have inserted the gene encoding Corynebacterium diphtheria toxin-A (DTA) into an autonomously induced plasmid under positive doxycycline control ('Tet-on' system). In this study, we demonstrate that this system can lead to the cell death of mouse ES cells by the induction of DTA expression when exposed to the tetracycline derivative, doxycycline. MTT assay showed that this induction resulted in the apoptosis of ES cells.
This study was conducted using inbred ICR mice to investigate the sex-ratio of preimplantation mouse embryos. For the investigation of sex-ratio of mouse embryos, the karyotype of embryos collected at 70-72, 74-76, 78-80 and 82-84 hr after HCG injection was analyzed by chromosomal analysis. Eight-cell embryos were cultrued up to blastocyst stage, then divided them into three groups(fast-, intermediate- and slow-) according to the blastocoel formation. The sex-ratio was also investigated by chromosomal analysis. 1. The highest apperance of eight-cell and morula was observed at the embryos collected respectively at 66-68 hr(84.6%) and 82-84 hr(79.3%) compared to any other group. 2. The successful rate of embryos sexing at 4-, 8-cell and morula stage were 23.1% (3/13), 42.1%(138/328) and 32.6%(47/141), respectively. The respective sex ratios (female vs male) of 4-, 8-cell and morula were 66.7:33.3, 49.3:50.7 and 39.5:60.5. 3. Of the 476 eight-cell embryos cultured in vitro, 427(89.7%) embryos were developed to the blastocysts and the number of fast-, intermediate- and show-developing embryos were 139, 144 and 144, respectively. 4. Female to male ratios fo fast-, intermediate- and slow-developing group were 23.0:77.0, 55.2:44.8 and 73.8:26.2, respectively. Significantly higher (P<0.05) number of female (48/65;73.8%) was observed in the group of slow-developing embryo than that out of total number of embryos(82/188;43.6%).
Single nuclei from two-, four- and eight-cell mouse embryos were transplanted into enucleated two-cell mouse embryos by micromanipulation and sendai virum mediated fusion. no significant difference in successful injectin rate and fusion rate was found between the cell stages of nuclear donor embryos. There nuclear transplant embryos receiving different cell stage nuclei were cultured in vitro for 96 hours. 75.3% of 255 embryos receiving 2-cell nuclei, 68.2% of 236 embryos reciving 4-cell nuceli and 46.9% of 228 embryos receiving 8-cell nuclei were developed to blastocyst, respectively. The number of blastomeres was significantly(P<0.05) reduced in the embryos receiving 8-cell nuclei, compared with the embryos receiving 2-cell, 4-cell nuclei or the intact embryos. Also the size of blastocysts was significantly(P<0.05) smaller in the embryos receiving 8-cell nuclei, compared with the intact or other nuclear transplant embryos. These results suggest that single nuclei introduced into the enucleated two-cell embryos are able to support the in vitro development of the reconstituted embryos to blastocysts. The prominant retardation of blastocoele formation and cell division was shown in nuclear transplant embryos receiving eight-cell nuclei when they were cultured in vitro.
Transcription factor called activating enhancer binding protein 2C (AP2-gamma) is found in a variety of species and expressed from oocyte stage onwards, particularly restricted to the trophectoderm. Recent studies demonstrated that ablation of Tfap2c led to failure of tight junction biogenesis, particularly the knock-down embryos of Tfap2c did not form cavity from morula to blastocyst in mouse and pig. We speculated that the Tfa2pc may also be involved in desmosome biogenesis because blastocoel formation is coincident with the establishment of desmosome. To determine this, we depleted Tfap2c injecting siRNA into one-cell zygote and analysed the expression levels of genes that are required for desmosome complex such as PkP2, Pkp3, Dsc2, and Dsg2. We found only Pkp3 was up-regulated in the knockdowned morula embryos. Interestingly, upstream region of Pkp3 had putative Tfap2c binding sites. In conclusion, our results suggest that Tfap2c is not a crucial factor but somehow it might be involved in desmosome biogenesis directly or indirectly via Pkp3.
Alpha lipoic acid (ALA) is a biological membranes compound. As the antioxidant, it decreases the oxidized forms of other antioxidant substances such as vitamin C, vitamin E, and glutathione (GSH). To examine the effect of ALA on the in vitro maturation (IVM) of porcine oocytes, we investigated intracellular GSH and reactive oxygen species (ROS) levels, and subsequent embryonic development after parthenogenetic activation (PA). Intracellular GSH levels in oocytes treated with 50uM ALA increased significantly (P < 0.05) and exhibited a significant (P < 0.05) decrease in intracellular ROS levels compared with the control group. Oocytes matured with 50 uM of ALA during IVM displayed significantly higher cleavage rates (67.8% vs. 83.4%, respectively), and higher blastocyst formation rates and total cell number of blastocysts after PA (31.6%, 58.49 vs. 46.8%, 68.58, respectively) than the control group. In conclusion, these results suggest that treatment with ALA during IVM improves the cytoplasmic maturation of porcine oocytes by increasing the intracellular GSH levels, thereby decreasing the intracellular ROS levels and subsequent embryonic developmental potential of PA.
To investigate the effect of extracellular matrix proteins on the in vitro development of blastomeres isolated from 2, 4, and 8-cell embryos(termed 1/2, 1/4 and 1/8 blastomeres, respectively) of ICR strain mice, those were cultured in fibronectin, gelatin, or collagen precoated culture dishes containing 1.5ml of NaHCO3-BMOC-3 medium at 37$^{\circ}C$ for 72 hrs, under the atmosphere at 5% CO2. and 95% air. Fibronectin, gelatin, or collagen significantly(P<0.01) increased and blastocyst formation rate compared with controls in 1/2(65.3, 59.2, 60.7% vs. 21.6%), 1/4(63.7, 53.4, 57.1% vs. 26.3%), and 1/8 blastomeres(61.1, 52.3, 53.7% vs. 19.1%). Both the nuclear number(P<0.05) and diameter of blastocysts(P<0.01) developed from balstomeres were significantly affected by the origin of blastomeres. The nuclear number of blastocysts developed from 1/2, 1/4, and 1/8 blastomeres ranged 29.3$\pm$1.6, 24.5$\pm$1.3, and 20..$\pm$1.2, respectively. And the diameter of those blastocysts was 88.3$\pm$2.4, 57.6$\pm$2.1, 39.8$\pm$1.9${\mu}{\textrm}{m}$, respectively.
This study was conducted to investigate the efficacy of vitrification procedure for the cryopreservation of porcine oocytes and the utilization of vitrified oocytes as recipient cytoplasts for somatic cell nuclear transfer (NT), and observed that porcine oocytes are evaluated by pronuclear formation, and parthenogenetic development. Single fetal donor cells were deposited into the perivitelline space of vitrified enucleation oocytes, followed by electrical fusion and activation. NT embryos were cultured in NCSU-23 medium supplemented with 5% FBS, at $38.5^{\circ}C$ in 5% $CO_2$ and air. 1. When the developmental rates of the oocytes after being culture for $0{\sim}10$ hours vitrified with EDS and ETS were 42.0%, 38.0%, respectively. This results were lower than the control group(62.2%). 2. When the developmental rates of the oocytes after being culture for $0{\sim}10$ hours vitrified-thawed with sucrose and glucose, 5% PVP, NCSU-23 supplemented with 10% FBS were 33.3%, 25.9%, respectively. This results were lower than the control group(55.6%). 3. The fusion and development to the blastocyst stage between the NT embryos constructed with the vitrified and non-vitrified oocytes were significant differences. Developmental rate of oocytes and NT embryos constructed with the vitrified or non-vitrified oocytes were $13.0{\pm}2.4%\;and\;23.2{\pm}2.4%$, respectively.
