Objective: This study was carried out to assess the status of working environments, to improve the working environments, and to be helpful to prevent occupational disease. Methods: The authors measured the noise level, concentration of dust, heavy metals, organic solvents, and other chemicals at 95 industries (22 working processes) by type of working process in manufacturing industry of parts and accessories for motor vehicles and its engines from January to December 2003. Samples were measured and analyzed by regulations of Korea Ministry of Labor, manuals of National Institute for Occupational Safety and Health (NIOSH) and Occupational Safety and Health Administration (OSHA). Results: Results are as follows. 1. Major processes exceeding mean noise levels provided the American Conference of Governmental Industrial Hygienists (ACGIH) Threshold Limit Value (TLV) were shakeout [97.5 dB(A)]. shot-blast [94.2 dB(A)], pressing [92.9 dB(A)], crushing [91.2 dB(A)], and cleaning [90.6 dB(A)]. 2. Mean concentrations of dusts were not exceeded to the TLV. But concentration on some points of processes as like welding (6.50 mg/m3), foundry (5.24 mg/m3) were exceeded. 3. Mean concentrations of heavy metals were not exceeded to the TLV. 4. Mean concentrations of organic solvents and chemicals were not exceeded to the TLV. 5. Improving rate for working environment were significantly higher in industries with health manager than without (p<0.05), and by increasing the year of working environmental measurement (p<0.01). Conclusions: The results suggest that working processes of exceeding to the TLV will be needed rapid improvement of the working environment, and also the others will be needed positive management of the working environment. Health managers must be recommended to employ in the workplace, and further studies for relationship between working environment and health effects for the workers must be carried out.
In the continued research on natural fungicides for control of plant diseases by using plant-derived products, we found that Spatholobus suberectus Dunn had a strong fungicidal activity against several plant pathogens. S. suberectus (1 kg) was extracted with 80% aqueous MeOH and then the concentrated extract was partitioned with n-hexane, EtOAc, n-BuOH and $H_2O$ successively. The four layers were tested their disease contron efficacies against 6 plant diseases such as rice blast (RCB), rice sheath blight (RSB), tomato grey mold (TGM), tomato late blight (TLB), wheat leaf rust (WLR), and barley powdery mildew (BPM). The EtOAc fraction was highly active showing over 80% control against RCB, TGM, TLB, and BPM. By using silica gel chromatography, preparative TLC and HPLC, six compounds that were expected to have antifungal activity were separated. Their chemical structures were identified as ethanone, hydroxytyrosol, epicatechin, procyanidin B2, dimethoxy daizein and formononetin by ESI-MS, $^1H$-NMR, $^{13}C$-NMR, and 2D-NMR spectroscopic analyses. The chemicals except epicatechin were first reported in S. suberectus. Study on in vitro and in vivo antifungal activities of the isolated compounds is in progress.
This study was investigated for the purpose of the isolation and identification of antagonistic bacteria with antifungal activity against plant pathogens. This bacteria denominated Bacillus subtilis KYS-10 and the optimum growth condition were 4% sucrose, 1% yeast extract, 0.2% $K_2HPO_4$, pH 7, 150 rpm, $30^{\circ}C$, 8 day. The antifungal activities against nine plant pathogens determined inhibition zone size by diffusion methods. The results, G. zeae (scab) 70 mm and P. grisea KACC 40439 (blast), P. capsici KACC 40177 (phytophthora blight) and C. destructans KACC 41077 (root rot of ginseng) 40~43 mm, and C. gloeosporioides KACC 43520 (ripe rot), C. gloesporioides KACC 40003 (anthracnose), S. shiraiana KACC 41065 (stem rot) and S. shiraiana (mulberry sclerotial disease) 35~39 mm and F. Oxysporum KACC 44452 (bulb rot of ginseng) 28 mm. From these experiment results, author suggest that Bacillus subtilis KYS-10 would be developed as a biological control agent thorough the field experimet in the near future.
A Pseudomonas strain SG3 producing biosurfactant and showing antifungal and insecticidal activities was isolated from agricultural soil severely contaminated with machine oils. The antagonistic bacterium inhibited mycelial growth of all of the tested fungal pathogens. The fermentation broth of SG3 also effectively suppressed the development of various plant diseases including rice blast, tomato gray mold, tomato late blight, wheat leaf rust, barley powdery mildew and red pepper anthracnose. An antifungal substance was isolated from the fermentation broth of SG3 by ethyl acetate partitioning, silica gel column chromatography and preparative HPLC under the guide of bioassay. The chemical structure of the antifungal substance was determined to be rhamnolipid B by mass and NMR spectral analyses. The antifungal biosurfactant showed a potent in vivo antifungal activity against gray mold and late blight on tomato plants. In addition, rhamnolipid B inhibited mycelial growth of B. cinerea causing tomato gray mold and zoospore germination and mycelial growth of P. infestans causing tomato late blight. Pseudomonas sp. SG3 producing rhamnolipid B could be used as a new biocontrol agent for the control of plant diseases occurring on tomato plants.

Prevotella intermedia is one of the most frequently implicated pathogens in human periodontal disease and has a requirement for hemin for growth. This study has identified a hemin-binding P. intermedia protein by expression of a P. intermedia genomic library in Escherichia coli, a bacterium which does not require or transport exogenous hemin. The genomic library of P. intermedia was constructed into plasmid pUC18, transformed into Escherichia coli strain $DH5{\alpha}$, and screened for recombinant clones using heminbinding activity by plating onto hemin-containing agar. Approximately 5,000 recombinant E. coli colonies were screened onto LB-amp-hemin agar, single clone(pHem1) was exhibited a clearly pigmented phonotype. The 2.5kb insert DNA of pHem1 was determined by restriction enzyme mapping. Southern blot analysis of BamHI, BglII, EcoRI, HindIII and PstI-digested P. intermedia DNA indicated that single copy of the gene was present in the genome. Northern blot analysis revealed that the size of transcript was approximately 1.8 kb. The cloned gene contained a single ORF, consisting of approximately 850-residue amino acids. A BLAST search of the Institute for Genomic Research genes with similar nucleotide sequence revealed no significant similarity It needs further investigation to clarify the mechanisms of heme uptake in P. intermedia.
[ ${\alpha},{\beta}$ ]-Unsaturated carboxamides 12 with trifluromethylated dihydro-1,4-dithiins were synthesized for the purpose of development of new agrochemical fungicide. Chlorination of trifluoromethylated ${\beta}$-ketoester 4 followed by tile reaction with 1,2-ethanedithiol gave intermediate 1,4-dithiane 9. Without purification of 9 substitution of hydroxy by chlorine followed by dehydrochlorination in the presence of triethylamine afforded trifluoromethylated dihydro-1,4-dithiin ethyl ester 7. Activation of the hydroxy of the carboxylic acid 10 obtained from the hydrolysis of 7 and then reacted with various amines gave the corresponding trifluoromethylated dihydro-1,4-dithiin carboxamides. Antifungal screening (in vivo) against typical plant diseases, Rice Blast, Rice Sheath Blight, Cucumber Gray Mold, Tomato Late Blight, Wheat Leaf Rust, and Barley Powdery Mildew of the synthesized compounds was carried out. As a result, most of the compounds shlowed weak antifungal activities and some compounds in which isopropyl group was substituted in meta of the phenyl showed antifungal activity (99%) at 250 ppm against the disease Wheat Leaf Rust.
In order to find out more effective countermeasures against growth damage of paddy rice due to the irrigation of livestock waste water, pot experiments were carried out in 1985 and 1986. Experiment I conducted in 1985 was to find out the effects of irrigation control and application of ameliorators to paddy soil with irrigation of animal waste water. Experiment II conducted in 1986 was to find out the amounts of wollastonite to be applied. The results obtained were as follows. Experiment I ; Rice yields were higher in the treatment of continuously submersion until harvest than in that of intermittent irrigation. The yield was also increased with the application of soil ameliorators and it showed higher increase in the treatment of zeolite and wollastonite. Nitrogen contents in plant were increased with the nitrogen application and more amounts of irrigation of waste water, but they were considerably decreased with the application of soil ameliorators. Experiment II ; Rice yields were increased with the application of wollastonite and the optimum level of wollastonite for maximum yield in condition of 3.0 g/pot of nitrogen application was belived to be 60g/pot. The increment of yield by the application of wollastinte was though to be due to increase of No. of grains per panicle and ripening ratio. Steriled neck occurred by rice disease such as rice blast was increased with increasing the amounts of nitrogen application and nitrogen contained in irrigated waste water but that was remarkably decreased with the application of wollastonite.
'Goamibyeo', a mid-late maturing ecotype with high amylose content in kernels, was developed by the rice breeding team of National Yeongnam Agricultural Experiment Station(NYAES) in 2000 and released in 2001. This variety was derived from the three way cross of Milyang 95//Kimcheonaengmi/$2^*Ilpumbyeo$ (in 1992 summer) through the pedigree breeding method and designated as 'Milyang 168' in 1997. The heading date of 'Goamibyeo' was Aug. 18 in ordinary season with culm length of 85 cm. However, 'Goamibyeo' showed susceptible to bacterial leaf blight, stripe virus and leaf blast disease. The amylose content of 'Goamibyeo' in milled rice kernels is about 26.7% with translucent and clear in chalkness. Thus, 'Goamibyeo' is expected to be used as a source grain for rice noodle industries. The milled rice yield potential of 'Goamibyeo' is about 5.38 MT/ha in local adaptability test of three years and it would be adaptable to Yeongnam plain of Korea.
Saemimyeon, a Tongil type, medium-late maturing rice variety, is especially used for preparing rice noodles. Its high amylose content was developed to fit market demands and to be affordable for rice processing industries. One of the high yielding lines, Milyang181 (Hanareum), was used in the final three-way cross of $IR50^*2$/YR18241-B-B-115-1-1 for yield improvement and cultivation stabilization, including disease resistance. YR24235-10-1-3, a high yielding and compact plant type, was selected and named Milyang278 after yield test at NICS (RDA, Miryang) in 2010. It was subjected to regional yield test at six sites in the middle and southern plain areas of South Korea. Saemimyeon heading occurs on August 12 and is a mid-late maturing cultivar, with resistance to leaf blast, rice stripe virus, and bacterial blight (K1-K3a), but it is susceptible to major diseases and insect pest infestation. Saemimyeon showed a high amylose content of 26.7%, with a relatively low KOH digestion value of 3.5, which are key factors in rice noodles and pasta processing. In the local adaptability tests, the yield of Saemimyeon was 7.08 MT/ha-an increase of approximately 106% compared to that of Dasan. Thus, Saemimyeon is suitable for cultivation in the southern and middle plain areas of South Korea.

In plant-pathogen interactions, Magnaporthe oryzae causes blast disease on more than 50 species of 14 monocot plants, including important crops such as rice, millet, and most 15 recently wheat. M. oryzae is a model fungus for studying plant-microbe interaction, and the main source for fungal pathogenesis in the field. Here we report that MoJMJD6 is required for conidium germination and appressorium formation in M. oryzae. We obtained MoJMJD6 mutants (ΔMojmjd6) using a target gene replacement strategy. The MoJMD6 deletion mutants were delayed for conidium germination, glycogen, and lipid droplets utilization and consequently had decreased virulence. In the ΔMojmjd6 null mutants, global histone methyltransferase modifications (H3K4me3, H3K9me3, H3K27me3, and H3K36me2/3) of the genome were unaffected. Taken together, our results indicated that MoJMJD6 function as a nuclear protein which plays an important role in conidium germination and appressorium formation in the M. oryzae. Our work provides insights into MoJMJD6-mediated regulation in the early stage of pathogenesis in plant fungi.
