Anthelmintic Efficiency of Nitroxynil against Fasciola hepatica, Eurythrema Pancreaticum and Paramphistomum sp. in Korean Native Goat (재래흑산양(在來黑山羊)의 흡충류(吸蟲類)(간질(肝蛭), 췌질(膵蛭), 쌍구흡충(雙口吸蟲))에 대(對)한 Nitroxynil(Trodax)의 구충효과(驅蟲效果))

  • The anthelmintic efficiency of nitroxynil(Trodax) at a dose rate of 10mg/kg was tested in naturally infected Korean native goat (Black goat) against Fasciola hepatica, Eurythrema pancreaticum, and Paramphistomum sp. The drug was administered with injection subcutaneouslly and the efficiency was measured by faecal examination microscopically. The results obtained were summarized as follows: 1. The anthelmintic efficiency of nitroxynil against Fasciola hepatica was shown 33.3% on day 7, 88.9% on day 14 and 94.4% on day 28 after the administration of the drug, respectively. The reduction rates of egg per gram of faeces (E. P. G.) against Fasciola hepatica in faecal examination were shown 86.8% on day 7, 97.7% on day 14, and 99.7% on day 28 postmedication. 2. The anthelmintic efficiency of nitroxynil against Paramphistomum sp. was shown 0% on day 7, 10.5% on day 14, and 36.8% on day 28 after the administration of drug. The reduction rates of E. P. G. against the parasite were shown 13.2% on day 14, and 39.4% on day 28 postmedication. 3. The anthelmintic efficiency of nitroxynil against Eurythrema pancreaticum was shown 0% on day 7, 16.6% on day 14, and 25% on day 28 after the administration of the drug.

Morphological Characteristics and Growth Performance of Korean Native Black Goats (재래흑염소의 형태적 특징 및 생육특성)

  • This study aims to provide basic data for investigating the phenotypic traits and growth characteristics of Korean native black goats. The Jangsu and Tongyoung strains were shown to have black hair, while the Dangjin strain was shown to have black or dark brown long hair. Only the Tongyeong strain was found to have no wattle and one goat each from the females and males of the Dangjin strain was found to have no horns. The lengths of the ears and tails were similar for the female and male goats, while the lengths of horns and beards were longer in the male goats. The birth weight was 2 kg on average, with that of male goats tending to be slightly heavier than that of the females. Weights at the weaning period were found to be slightly heavier for the males. Also, at 12 months, the weights of the male goats by strain were shown to be heavier than that of the females. The body length and depth by strain were found to be higher for the male goats, while no differences in the chest width and girth between the strains or gender were found. These findings are expected to be used as a basis for differentiating between native and crossbred Korean black goat strains and for improving Korean native black goats.

  • In vitro maturation and activation of oocytes are primary steps towards biotechnological manipulation in embryology. The objectives of the present study were to determine the oocyte recovery rate per ovary, in vitro maturation rates of oocytes and rates of parthenogenetically activation of matured oocytes in Black Bengal goats. All visible follicles were aspirated to recover follicular fluid from individual ovaries (number of ovaries = 456). The immature cumulus oocyte complexes (COCs; n = 1289) were cultured in tissue culture medium (TCM)-199 supplemented with 10% (v/v) fetal bovine serum (FBS) for 27 hours at $39^{\circ}C$ with 5% $CO_2$ in humidified air. The matured oocytes (n = 248) were activated with 5 ${\mu}M$ ionomycin for 5 minutes followed by treatment with 2 mM 6-dimethylaminopurine (6-DMAP) for 4 hours. After activation, oocytes were cultured for another 14 hours in TCM-199 supplemented with bovine serum albumin (BSA) at $39^{\circ}C$ with 5% $CO_2$ in humidified air. The pronucleus formation in activated oocytes was determined by staining with 1% orcein (whole mount technique). Matured oocytes (n = 176) without activation stimuli were used as control. The mean number of oocytes recovered per ovary was $3.5{\pm}0.5$. The proportion of oocytes matured in vitro, confirmed by the presence of first polar body, was $42.1{\pm}4.7%$. Parthenogenetic activation, evidenced by formation of pronucleus, occurred in $37.2{\pm}15.8%$ of matured oocytes. No pronucleus formation was observed in control oocytes. In conclusion, a combination of ionomycin and 6-DMAP induces activation in one third of Black Bengal goats' oocytes.

Effect of Feeding Alfalfa and Concentrate on Meat Quality and Bioactive Compounds in Korean Native Black Goat Loin during Storage at 4℃

  • The primary aim of this study was to evaluate the effect of feeding alfalfa: Concentrate at different ratios (8:2 or 2:8) to Korean native black goats (KNBG) for 90 days on meat quality and bioactive compound content. Feeding KNBG alfalfa and concentrate at different ratios did not impact meat pH, color, microorganism composition, volatile basic nitrogen levels, or lipid oxidation. The low alfalfa (KLA) group exhibited increased oleic acid and monosaturated fatty acid levels, both of which impact the palatability traits of meat. The abundance of bioactive compounds increased in the loin meat of the KLA group, leading to an increase in antioxidant activities. Our results suggest that feeding alfalfa and concentrate at a 2:8 ratio to KNBG can increase taste-related fatty acids and bioactive compounds in loin meat, relative to that achieved by feeding at an 8:2 ratio. Further investigation is required to evaluate the quality and the metabolites of bioactive compounds in KNBG meat and the effect of the different dietary ratios of forage and concentrate.

Attitudes toward Complementary and Alternative Medicine in Suwon City (수원시 주민의 대체의학 수용실태 조사)

  • Objectives: The aim of this study was to investigate the attitudes toward complementary and alternative medicine among 1,490 residents(339 households) in Suwon city. Methods: All respondents were asked about types, frequency, effects, side-effects, views, and cost of complementary or alternative medicine through a questionnaire from July 24th to 27th. Six therapies were investigated: diet; acupuncture/ massage/ chiropractic etc., mind control such as Ki/ Yoga/ spiritual therapy/ relaxation therapy etc.; nutritional supplements, cultural remedies; and Herb medications Results: The results of this survey were as follows: 35.6% of respondents had experiences with at least one or more types of complementary and alternative medicine. The average number of different types of therapies used was 3.4. More experience with various types of therapies were found among those respondents of higher education, older age group, higher income, married group, religious group than among the opposite groups of respondents. Herb medications were used most frequently(39.8%), followed by minor grains(37.9%), Ginseng(23.8%), Boshintang(21.5%), acupuncture(20.3%), Gaesojou(15.3%) Gingko nut(12.0%), mushroom(11.5%), Cupping therapy(10.2%), and black goat(0.0%). Acupuncture and Herb medications were used for treatment of hypertension the most frequently; minor grains or silkworm for treatment of diabetic mellitus; vegetables for treatment of obesity; acupuncture, Cupping Therapy, Herb medications for treatment of rheumatism; and acupuncture, Herb medications, or exercises for treatment of Cerebro Vascular Accident(CVA). The average costs of treatment were 108,000 Won for hypertension, 87,200 Won for diabetic mellitus, 16,800 Won for obesity, 68,800 Won for rheumatism, and 87,500 Won for CVA. Among 10.9% of respondents, there were 13 cases of side-effects with acupuncture, Herb medications, and Gaesojou. Among the cases of side-effects, majority was due to Herb medications. Respondents reported that Cupping Therapy was the most effective, followed by acupuncture, Ginseng, Gingko nut, Boshintang, black goat, minor grains, Gaesojou, Herb medications, vegetables, and mushroom. In response to the views of complementary and alternative medicine which they had used, they recommended minor grains first, followed by Ginseng, acupuncture, Gingko nut, Cupping Therapy, vegetables, Boshintang, black goat, mushroom and Herb medications. In contrast, they did not recommend Herb medications, acupuncture, nor Gaesojou. Conclusions: These findings indicate that many people use various complementary and alternative medicine without any guidelines for treatment of serious chronic diseases not even to invigorate themselves. It is, therefore, suggested that medical doctors or scientists verify the true effects or side-effects from the most common complementary or alternative therapies through experiments. Also medical doctors should provide a comfortable atmosphere for discussion among doctors and patients who would like to try these therapies.

Effects of Triladyl-egg Yolk Diluents on the Viability of Frozen Korean Black-goat Spermatozoa from Cauda Epididymis and Electro-ejaculated Semen (Triladyl-난황 희석제가 한국 재래 흑염소의 정소상체 및 전기자극 유래 정자의 융해 후 생존성에 미치는 영향)

  • To preserve genetic materials, cryopreservation of the semen from live animals is the main technique to establish cryo-banking system which could be used for artificial insemination and embryo transfer. However, the population of Korean black goat (KBG) becomes to dwindle in number and is now faced genetic erosion by crossbreeding with non-native breeds in small KBG farms. In this study, simple freezing method was used to preserve frozen semen from KBG using spermatozoa of cauda epididymis (CE) and electro-stimulated semen (ES). The negative effects of seminal plasma on fresh sperm was confirmed using precipitation test of Triladyl egg yolk diluent and sperm viability after thawing was compared between CE and ES spermatozoa. When seminal plasma of fresh ES semen was washed with semen washing media (SWM), the rates of live sperm shown no significant difference between CE and ES spermatozoa before freezing. However, the survival rate of frozen/thawed CE sperm was higher than ES ($74.6{\pm}10.6%$ vs $53.8{\pm}5.2%$) with significant difference (p < 0.05). The results of longevity test on frozen/thawed sperm from CE showed healthier sperm than ES. Therefore, spermatozoa from CE could be used for cryo-banking system in KBG lines. The more studies are needed to increase survival rate of ES semen.

Serological and virological investigation of pestiviruses in Korean black goats

  • Blood samples were collected from 672 goats in 60 farms from five provinces of Korea between November 2009 and August 2011. The prevalence of antibodies to pestiviruses was investigated. The examination for antibodies was performed using an enzyme-linked immunosorbent assay (ELISA) detecting antibodies to the bovine viral diarrhea virus (BVDV) and border disease virus (BDV). All blood samples were screened using reverse transcription-polymerase chain reaction (RT-PCR) with primer pairs specific to common pestivirus genome regions. The observed individual seroprevalence was 1.49% and herd seroprevalence was 11.67%. Also, the specific genomes to pestiviruses were detected in 3 out of the 915 clinical samples (0.45%). Based on the nucleotide sequence data, detected pestiviruses were belonged to two BVDV type-1 and one BVDV type-2. The pestivirus infection has been occurred among Korean black goats. However, our results indicate that the prevalence of pestiviruses in black goats was not significantly higher on farms with cattle.

Occurrence, clinical signs, postmortem lesions and etiology of enterotoxaemia in Black Bengal goats

  • A year round study was carried out to investigate the etiology, clinical signs, postmortem lesions and occurrence of naturally occurring enterotoxaemia in Black Bengal goats. Sixteen goats of different age and sex died in different seasons with sigh associated with enterotoxaemia made the materials of this study. Accidental access to large amount of concentrate was noted as one of the predisposing factors although few cases were reported to occur without known diet change. Younger animals(50%) and males(62.50%) were found more prone to the disease and it was likely to be more prevalent during winter(50%) followed by at rainy season(31.25%) and summer(18.75%). Diarrhoea(81.25%), dullness(56.25%), drooping of the ears(50%), anorexia(43.75%) were recorded as major clinical signs whereas enterocolitis(100%), lung edema(87.50%), fluid filled intestines(87.50%), enlarged mesenteric lymph nodes(56.52%) etc. were most common post mortem lesions found. A few cases showed lesions on heart(31.25%), brain(25%) and/or liver/spleen(18.75%) but no lesion was found on kidney. Thus the so called 'pulpy kidney' lesion was absent. Intestinal contents were subjected to conventional bacteriological culture based methods to identify the causal agents. Based on the morphological, cultural and biochemical properties the causal agent was identified as Clostridium perfringens. Despite the study was carried out at certain area it showed a clear picture of goat enterotoxaemia in terms of etiology, clinical signs, postmortem lesions and occurrence of goat enterotoxaemia in Bangladesh.

Changes in Feed Value, Forage Productivity, and Grazing Intensity at Native Pasture Grazed by Growing Korean Native Female Goat(Capra hircus coreanae) (육성 암컷 흑염소 방목 야초지에서 사료가치, 사초 건물 수량 및 방목 강도의 계절별 변화에 관한 연구)

  • The study was conducted to determine effects on forage productivity, feed value, grazing intensity and livestock productivity in growing Korean native female goat grazing in native pasture. Its with average initial body weight of $14.10{\pm}3.6kg$ and an average age of 4 months were used in this study. Dry matter content of native pasture was the highest at $33.48{\pm}2.56%$ in June, and the content was significantly increased from spring to autumn (p<0.05). Crude protein was maintained between 11% and 12% on average. Nutrient content was maintained at a certain level in native pasture, but there were differences due to the different types of wild grasses produced in each season. The productivity of forage crops increased from June, but decreased after August and showed a characteristic of grassland where productivity decreased rapidly in spring and autumn. The average grazing intensity are 39 head/ha. Black goat average daily gain was 80.2g/d. The stable weight gain in grazing is that the nutrient requirement of the black goat was met by supplementing the concentrated feed during grazing. study, can be expected that the productivity of livestock can be increased through the proper feeding of supplementary feed and maintenance of grazing intensity.

Comparative Histological Study on the Parafollicular Cells of Mammals (각종 포유동물 갑상선내의 소포방세포에 관한 비교조직학적 연구)

  • Comparative studies on the parafollicular cells of the some mammalian species from five different orders were carried out; i.e., man from Primates, cattle, pig, and black goat from Artiodactyla, dog from Carnivora, rabbit from Lagomorpha, rat, mouse, and squirrel from Rodentia. For this study, various special techniques for the parafollicular cells, including Grimelius' silver impregnation method (Sawicki and Bajko, 1974), Singh's argentaffin method (Singh, 1964), HCl-toluidine blue stain (Sawicki, 1971), and HCl-lead hematoxylin stain (Solcia et al., 1969), were applied. Authors obtained the following results: 1. Number of parafollicular cells in the same area of thyroid tissues are significantly different from species to species. Number of cells were largest in dog and less cells were found in the following orders; rat, squirrel, mouse, rabbit, cattle, pig, black goat and finally the smallest number in man. 2. Distribution of parafollicular cells within thyroid gland are significantly different from portion to portion in case of cattle, rabbit, squirrel and mouse, but it is not significant in dog, man, pig, black goat and rat (see Table 1-1 and 1-2). 3. In dog, clustered parafollicular cells are located usually in the interfollicular space, and groups of parafollicular cells are located in the para-and/or inter-follicular positions in rabbit. But in the other animals parafollicular cells are found solitarily in the intra-and/or para-follicular positions. 4. The shape of parafollicular cells shows oval to round contour in dog, but it is polymorphic, for example, spindle, conical, oval, round or elongated with cytoplasmic processes, in the other animals. 5. Size of parafollicular cells is larger in cattle, dog and pig, smaller in rat, mouse and squirrel, and medium-size in rabbit, man and black goat. 6. Parafollicular cells of pig, cattle, dog and squirrel are observed to contain densely packed granules, whereas those of mouse, rat and man contain relatively scanty granules. 7. Parafollicular cells of all the mammals show more or less positive reaction to Grimelius' argyrophile silver impregnation method, HCl-toluidine blue stain and HCl-lead hematoxylin stain, whereas they show negative reaction to argentaffin method (see Table 2). 8. Considering the above finding, it is concluded that there are species differences in the distribution, location and shape of parafollicular cells, and infer that preferable staining method should be selected for reliable detection of parafollicular cells, beacuse staining methods applied on the cells in this study show variable reactions according to species.
