The diet composition of Leiognathus nuchalis were studied using 350 specimens collected from October 2010 to September 2011 in the coastal waters of Gadeok-do, Korea. L. nuchalis was carnivorous feeder that consumed mainly copepods, crab larvae, and shrimp larvae. Its diet also included small quantities of polychaetes, amphipods, euphausiids, bivalve larvae, and gastropod larvae. The size of L. nuchalis ranged from 3.0 to 9.8 cm in body length (BL). L. nuchalis underwent an ontogenetic dietary shift. 3~5 cm size individuals fed mainly on copepods. 5~7 cm size individuals fed mainly crab larvae and shrimp larvae. 8~10 cm size individuals fed mainly polychaetes. The size increases, the weight of food organisms are constantly increased.
Five species of intertidal clams including Ruditapes philippinarum, Tegillarca granosa, Solen strictus, Heteromacoma irus, and Coecella chinensis were tested for the presence of the protozoan parasite, Perkinsus sp. using fluid thioglycollate medium (FTM) fortified with antibiotics and histological techniques. Each individual clam was placed in a test tube filled with 10ml FTM, placed in totally dark place, and incubated over a week. After incubation the clam tissues were stained with Lugol's iodine solution and examined under a light microscope to find out any hypnospores of Perkensus sp. in the tissues. Cross-sections of the clams were also embedded in paraffin, sliced to 3um, and stained with Harry's hematoxylene and Picro eosine to observe the presence of tomont or trophozoites. Perkinsus sp. were found in the presence of tomont or trophozoites. Perkinsus sp. were found in the tissues of R. philippinarum collected from Kangjin and Wando, along the south coast of Korea. However, Perkinsus sp. was not found in four other species of clams nor R. philippinaurm collected from Kimnyong and Waido in Cheju. A size-dependent Perkinsus sp. infection was found in R. philippinarum collected rom Kangjin and Wando the clams smaller than 15mm in shell width do not exhibit and Perkinsus sp. while other clams greater than 20mm in shell width exhibit almost 100% infection. To determine the number of Perkinsus sp. in the clams, FTM cultured clam tissues were digested with 2M NaOH solution and the number of hypnospores in the tube were counted. The number of hypnospores counted from the tissues indicated that each Manila clam contains 100,000 to 3,500,000 Perkinsus cells or 20,000 to 1,000,000 cells per gram tissue wet weight. The results of cell counts also suggests that such a high occurrence of Perkinsus sp. in the clam may cause mortality, as already reported from other studies of Perkinsus spp.

Oysters Crassostrea gigas are a globally popular shellfish for human consumption. As filter-feeding bivalve mollusks, oysters may harbor many microorganisms and chemicals that could pose potential human health risks. The objective of this study was to investigate the suitability of cultured oysters for raw consumption or use in seafood products by measuring concentrations of harmful microorganisms and chemicals in their flesh. Microbial concentrations in cultured oysters were found to be: $1.0{\times}10^2-6.0{\times}10^4CFU/g$ (viable cell counts), not detected $(ND)-5.4{\times}10^3CFU/g$ (coliform bacteria), $ND-1.3{\times}10^2CFU/g$ (E. coli), and $ND-4.6{\times}10^3CFU/g$ (Vibrio parahaemolyticus). Other pathogenic bacteria, including Enterohemorrhagic E. coli (EHEC), Listeria monocytogenes, Staphylococcus aureus, and Salmonella spp., were not detected in any samples. Heavy metal concentrations of cultured oysters were ND-0.239 mg/kg (total mercury), ND-1.091 mg/kg (lead), ND-0.968 mg/kg (cadmium). The concentrations of benzo(a)pyrene ranged from $0.280-0.880{\mu}g/kg$. Paralytic shellfish poison ranged from ND-0.58 mg/kg, while diarrhetic shellfish poison was not detected. No radioactivity was detected. These results suggest that oysters intended for raw consumption or use in seafood products should be subjected to chemical and biological controls.
In order to investigate the live food value of microalgae for efficacious rearing of larvae and spats of bivalve, we studied growth rates of four microalgal species (Isochrysis galbana, Pavlova lutheri, Chaetoceros simplex, Tetraselmis tetrathele) cultured in different environmental conditions. These include changes in temperatures (20, 25, 30 and $35^{\circ}C$), salinities (20, 25, 30 and 33 psu) and light intensities (60, 100 and $140{\mu}mol\;m^{-2}s^{-1}$). The growth rate of I. galbana was faster at $25^{\circ}C$ than that of $20^{\circ}C$. At $25^{\circ}C$ the highest growth rate of I. galbana was observed at 33 psu (0.413) and the lowest at 20 psu (0.368) in 10 days of culture (P < 0.05). The growth rate of I. galbana was lower at 25 psu (0.383) than that of 30 psu and higher than that of 20 psu (P < 0.05). Similar temperature and salinity-dependent changes were also found in P. lutheri and T. tetrathele. C. simplex showed faster growth rate at $30^{\circ}C$ than that of $25^{\circ}C$. The highest growth rate of C. simplex was observed at 33 psu (0.428) and the lowest at 20 psu (0.389) in 10 days of culture (P < 0.05). Upon exposure to the light with different intensities, all four microalgal species showed a significantly faster growth rate at $140{\mu}mol\;m^{-2}s^{-1}$ than at $100{\mu}mol\;m^{-2}s^{-1}$ (P < 0.05).
This study was conducted to find out the changes of survivorship, respiration and organ structure of Mactra veneriformis exposed to copper (Cu). Experimental period was four weeks. Experimental groups were composed of one control condition and three copper exposure conditions (0.025, 0.050 and 0.100 mg/L). The results of the study confirmed that copper induces reduction of survival rate and respiration rate and histopathology of organ structure of the bivalve. In the copper concentration of 0.100 mg/L, mortality was 100% after Cu exposure of 3 weeks. Respiration rate was observed exposure groups lower than control decline by 75%. Histological analysis of organ system illustrated expansion of hemolymph sinus, disappearance of epidermal layer and degeneration of connective tissue layer of the mantle. Also, histological degenerations as epithelial necrosis and hyperplasia of mucous cells are recognized in the gill and it was observed expansion of hemolymph sinus, disruption of epithelial layer, decrease of mucous cell and degeneration of connective tissue layer in the foot. In the digestive diverticulum, it was showed atrophy of basophilic cell and degeneration of epithelial cell in the digestive tubules, and as the concentration of copper increased the accumulation of lipofuscin increased.
In a series of feeding experiments using the newly established the first laboratory strain of a temperate photosynthetic ciliate, Mesodinium rubrum MR-MAL01, direct evidence was obtained for ingestion of the cultured M. rubrum cells by the bivalves like the Korean scallop, Chlamys farreri and Manila clam. Ruditapes phil-ippinarum. Each experimental Korean scallop and Manila clam removed up to 9,590 and 23,200 cells min-1, respectively. Small particles almost identical to the ruptured cells of MR-MAL01 culture were observed in the gut of the experimental bivalves, although no intact M. rubrum cell was found. In a 28 days rearing experiment, Isoshrysis galbana (KMCC H-2), a microalga, supported better growth of small Manila clam spat (0.46 mm in shell length) than MR-MAL01 strain. For the large Manila clam spat (0.84 mm in shell length), however. MR-MAL01 strain was a better prey item. Mass culture methods for this photosynthetic marine ciliate may be developed for use as live feed in aquaculture of the spat and broodstock of bivalve and small-mouthed fish larvae.

VHSV is a major viral agent that affects freshwater and marine fish, causing serious economic losses in aquaculture in the world. Due to their filter-feeding activity, bivalve mollusks may act as viral transmitters after accumulation of the fish viruses released into seawater from infected fish. Amplification by RT-PCR was carried out to investigate the presence of VHSV in pacific oysters (Crassotrea gigas) and blue mussels (Mytilus edulis), inhabiting regions around aquatic farms in Korea. Primers designed from conserved regions of VHSVs allowed us to detect four different types of VHSV in a single PCR. Twenty two of the eighty four samples showed positive results of VHSV in a 2-step RT-PCR. Using six positive samples from three different regions in Korea, we cloned and sequenced the glycoprotein (G) gene (467-bp long) of VHSVs. Genetic analysis of the VHSVs detected in shellfish in various geographical areas of Korea showed highly restricted results to VHSV type Iva. This was in agreement with the reports showing only a single genotype of VHSV (Iva genotype) in outbreaks in cultured or wild fish in Korea. Consequently, we investigated VHSVs carried by bivalve mollusks inhabiting the vicinity of aquatic farms, and revealed correlationship between the type of viral accumulated in shellfish by filter-feeding, and those detected in disease outbreaks in fish.
Exposure to air and increased temperature induced successful spawning in Mytilus edulis, M. coruscus Crassostrea gigas and Pinctata fucata martensii. Developmental durations required for an egg to attain D-shaped larva and the D-shaped larva to reach pediveliger stage were estimated in these bivalves. Size of fertilized eggs was the largest (70.3 ${\mu}m$) in M. coruscus and the smallest (45.3 ${\mu}m$) in P. fucata martensii. At 17$^{\circ}C$, M. edulis and M. coruscus attained D-shaped larval stage within 48 hours after fertilization but those of C. gigas and B. fucata martensiii within 24 and 22 hours at 21 and 26$^{\circ}C$, respectively. The development duration required for a D-shaped larva to attain pediveliger stage was the longest (27 days) in M. coruscus and ranged between 20 and 22 days for the others. The shell length of the pediveliger was the longest (274.9 ${\mu}m$) in C. gigas and smallest (190.9 ${\mu}m$) in P. fucata martensii Length and height of larval shell was highly correlated with each other in all the 4 species. The shell height of C. gigas was more than the shell length beyond the size of 100 ${\mu}m$ shell length. However, shell length of the others was always longer than shell height at the larval stage.
This study attempted to verify the possibility of using germ cell aspiration (GCA) method as a non-fatal technique in studying the life-history of equilateral venus, Gomphina veneriformis (Veneridae) and granular ark, Tegillarca granosa (Arcidae). Using twenty-six gauge 1/2" (12.7mm) needle, GCA was carried out in equilateral venus through external ligament. In granular ark, GCA was performed by preventing closure of the shells by inserting a tongue depressor between the shells while still open. The success rate of sex identification using the GCA method was 95.6% for the equilateral venus (n=650/680) and 94.3% for the granular ark (n=707/750). Mortality of equilateral venus, which spent 33 days under wild conditions, was 13.8% (n=90/650) while the mortality of granular ark, which spent 390 days under wild conditions, was 2.4% (n=17/707). Although we believe that GCA does not appear to cause death in equilateral venus or granular ark, the success rate in employing of this methodology may differ depending on the level of proficiency of the researcher and reproductive stage of the bivalve. This study concludes that GCA is a convenient non-fatal methodology, which can be employed to identify sex and investigate gonadal maturity in Gomphina veneriformis and Tegillarca granosa.

Manila clam, Ruditapes philippinarum, is commercially and ecologically important marine bivalve in Korea and japan. However, clam landings in the two countries have dramatically declined since the 1980-1990's. In the present study, the protozoan parasite, Perkinsus sp., lectin (host's defense-related glycoprotein) and histopathological features were investigated in Manila clams collected from Gomso Bay in Korea and Ariake Bay in japan (one of the largest clam beds in each country) during summer and fall, 2002-2003. DNA sequences of non-transcribe spacer (NTS), internal transcribed space. (ITS) and 5.85 rRNA of Perkinsus sp. were identical to those of P. atlanticus that was reported in Europe and Korea. For diagnosis of Perkinsus, the fluid thioglycollate medium (FTM) and the 2 M NaOH lysis methods were used. Prevalence of the parasite varied from 92.5-98.7% in Gomso Bay and 35.5-37.9% in Ariake Bay. Infection intensity, in terms of the number of Perkinsuscells per gram tissue wet weight, in the clams of Gomso Bay in fall 2002 averaged 1,010,077-470,937 recording approximately100 times higher than that of Ariake Bay, and these were twice higher than those of summer samples in each location. Mean hemagglutination titer of the clams from Gomso Bay was approximately 60-folds higher than that of clams from Ariake Bay in 2002. In histological preparation of the clams from Gomso Bay in 2002, trophozoites of P. atlanticus were in groups and resulted in severe inflammatory response of host clam. Prevalence of the trematod, Cercaria tapes-like in the clams of Gomso Bay and Ariake Bay were 8.8 % and 10.5% respectively. In conclusion, the clams from Gomso Bay showed more severe pathologic symptoms and higher immune response than those of the clams from Ariake Bay.
