Discovery of novel Nodosilinea species (Cyanobacteria, Nodosilineales) isolated from terrestrial habitat in Ryukyus campus, Okinawa, Japan

  • Terrestrial cyanobacteria are extremely diverse. In urban areas, they can be found as black stains on the surface of building walls, stone monuments, or man-made structures. Many of the terrestrial cyanobacteria are still understudied. To expand knowledge of terrestrial cyanobacterial diversity, a polyphasic characterization was performed to identify 12 strains isolated from campus of University of the Ryukyus, Okinawa, Japan. Multigene phylogenetic analyses based on 16S rRNA gene and 16S-23S rRNA internal transcribed spacer (ITS) region showed that the isolated strains formed two independent subclades within Nodosilinea, and were distantly related to all described Nodosilinea species. The 16S-23S rRNA ITS secondary structures showed variations for D1-D1' and Box B domain, while V3 domain was almost identical among entire species of Nodosilinea, including the studied strains. In addition, a unique morphological character, i.e. forming nodule or spiral shape, was also observed in certain studied strains. According to polyphasic characterization, Nodosilinea coculeatus sp. nov. and Nodosilinea terrestrialis sp. nov., were proposed as two new species of terrestrial cyanobacteria from Okinawa.

Corrosion Behavior of Cu-Ni Alloy Film Fabricated by Wire-fed Additive Manufacturing in Oxic Groundwater

  • The growing significance of sustainable energy technologies underscores the need for safe and efficient management of spent nuclear fuels (SNFs), particularly via deep geological disposal (DGD). DGD involves the long-term isolation of SNFs from the biosphere to ensure public safety and environmental protection, necessitating materials with high corrosion resistance for DGD canisters. This study investigated the feasibility of a Cu-Ni film, fabricated via additive manufacturing (AM), as a corrosion-resistant layer for DGD canister applications. A wire-fed AM technique was used to deposit a millimeter-scale Cu-Ni film onto a carbon steel (CS) substrate. Electrochemical analyses were conducted using aerated groundwater from the KAERI underground research tunnel (KURT) as an electrolyte with an NaCl additive to characterize the oxic corrosion behavior of the Cu-Ni film. The results demonstrated that the AM-fabricated Cu-Ni film exhibited enhanced corrosion resistance (manifested as lower corrosion current density and formation of a dense passive layer) in an NaCl-supplemented groundwater solution. Extensive investigations are necessary to elucidate microstructural performance, mechanical properties, and corrosion resistance in the presence of various corroding agents to simplify the implementation of this technology for DGD canisters.

Effects of chlorine and chlorine compounds on morphology and function of Caulobacter cells (Caulobacter의 세포(細胞)의 형태(形態) 및 기능(機能)의 분화(分化)에 대한 염소 및 염소화합물의 영향(影響))

  • Caulubacter is distinctive in the morphology and replication and ubiquitous in the biosphere, especially in every type of aquatic environment. In water and waste-water treatment processes, chlorine and chlorine compounds have been used as a main disinfectant throughout the world. Therefore, Caulobacter in the waters should be affected by chlorination of the waters. The objective of this study is to determine the effects of the disinfectants on Caulobacter cells and on the developmental processes of the cells. The Caulobacter swarmer cells were disinfected by chlorine at pH 7.0 minutes of the reaction with 2.0 mg/l of infected at pH 10.0. The swarmer cells treated with 2.0 or 4.0 mg/l of chlorine for 15 minutes lost their flagella and were observed by electron microscopy to be damaged on their cell surfaces, discharging some cellular materials. When the chlorinated swarmers and untreated control samples were recultivated in fresh PYE broth medium, the control swarmers multiplicated exponentially after one-hour lag phase, whereas the chlorinated swarmers extended the lag phase to about four hours. During the extended lag phase, the cells were proved by electron microscopy to be grown and be in predivisional step, but no swarmer cell was found. When the stalked cells were chlorinated, almost all the cells were observed to have their stalks broken and some cellular materials discharged. In those samples recultivated, many cells differentiated to possess an abnormally elongated stalk with several crossbands on it. This suggests that the chlorine-shocked Caulobacter cells can develope to abnormal morphology in water environments which they can survive and regrow in.

A Comparison of the Land Cover Data Sets over Asian Region: USGS, IGBP, and UMd (아시아 지역 지면피복자료 비교 연구: USGS, IGBP, 그리고 UMd)

  • A comparison of the three land cover data sets (United States Geological Survey: USGS, International Geosphere Biosphere Programme: IGBP, and University of Maryland: UMd), derived from 1992-1993 Advanced Very High Resolution Radiometer(AVHRR) data sets, was performed over the Asian continent. Preprocesses such as the unification of map projection and land cover definition, were applied for the comparison of the three different land cover data sets. Overall, the agreement among the three land cover data sets was relatively high for the land covers which have a distinct phenology, such as urban, open shrubland, mixed forest, and bare ground (>45%). The ratios of triple agreement (TA), couple agreement (CA) and total disagreement (TD) among the three land cover data sets are 30.99%, 57.89% and 8.91%, respectively. The agreement ratio between USGS and IGBP is much greater (about 80%) than that (about 32%) between USGS and UMd (or IGBP and UMd). The main reasons for the relatively low agreement among the three land cover data sets are differences in 1) the number of land cover categories, 2) the basic input data sets used for the classification, 3) classification (or clustering) methodologies, and 4) level of preprocessing. The number of categories for the USGS, IGBP and UMd are 24, 17 and 14, respectively. USGS and IGBP used only the 12 monthly normalized difference vegetation index (NDVI), whereas UMd used the 12 monthly NDVI and other 29 auxiliary data derived from AVHRR 5 channels. USGS and IGBP used unsupervised clustering method, whereas UMd used the supervised technique, decision tree using the ground truth data derived from the high resolution Landsat data. The insufficient preprocessing in USGS and IGBP compared to the UMd resulted in the spatial discontinuity and misclassification.

On Surface Ozone Observed in the Seoul Metropolitan Area during 1989 and 1990 (서울 수도권 지역의 광화학오존에 관한 연구)

  • Atmospheric $O_3$ in the biosphere is formed under the favourable meteorological condition, when the primary pollutants, such as $NO_2, HC, CO, CH_4$, etc., react with over constituents. Observed annual average concentrations for 1989 and 1990 were 11.8 and 10.4 ppb, respectively. THe number of days measured ozone over 80 ppb in Seoul were 36 in 1989 and 39 in 1990. In general, monthly maximum values occurred in May and August. In comparison with 1 $\sim$ 2 maxima of $O_3$ distribution in large cities in other countries, it was found that there were 3 $\sim$ 4 maxima in Seoul and its suburbs. Topographic effects, resulted by wind channelling in the Han River valley and by the blocking of air pollutants in the mountain, appeared to produce multiple centres of $O_3$ maxima in Seoul. Surface $O_3$ values were low with decreasing solar radiation, when the cloudiness increased and precipitation occurred. According to 12 cases examined, 2 cases shown here, $O_3$ values exceeding 80 ppb were occurred when the Korean peninsular was under the influence of the backside airflows with high intensity of solar radiation. Occasionally, sea breezes were observed to occur in warm seasons, and the chanelling effect of the Han River valley appeared to increase the general wind (speed) to the east side of Seoul. In this meso-scale situation $O_3$ in downwind is highly correlated with precursors. The sea breeze of 2 $\sim$ 4 m $s^{-1}$ will take 3 $\sim$ 5 hours to transport photochemical precursors for 20 $\sim$ 50 km. In turn the areas of maximum $O_3$ occurrence in Seoul are in the range of meso-scale transport of air pollutants.

  • A comparison study is performed on dose assessment programs including DOSE, LIMCAL, and PABLM, DOSE is a program developed for preliminary safety assessments of the low- and intermediate- level radioactive waste disposal, and the others are existing programs applicable to similar calculations. The results show acceptable agreement within an order of magnitude(mrem/yr) except for C-14 and Pu-239. PABLM results higher dose for C-14 while lower value for Pu-239 in comparison with those from DOSE or LIMCAL. It is found that the discrepancy in C-14 is due to difference in transport model introduced and that in Pu-239 is from the different value of dose conversion factor to each program.

  • This study was aimed to establish the method and process of the Biotope Mapping System.(B.M.S.) The main aspects of research for biotope were divided by 4 schemes such as biosphere, geosphere, antroposphere and evaluation informations. The cartographic function of GIS(geographic information system) was also used to mapping them. The B.M.S. had been progressed with 5 steps : (1) making research goals, (2) constructing the graphic and attribute databases, (3) classifying 3 types of biotope such as tree/shrub biotop, grass biotop and wetland biotop. (4) cross-analyzing biotope types against land use, habitat characters, relief characters and danger/disturbance elements, (5) evaluating the biotope's potentialities. Only 3 steps of them were done in this study. The results of applicating the B.M.S on the research site as followings : 1. The interdisciplinary researches were needed to develop the B.M.S in Korea since ecological habitat conditions of Germany, England are differed from them of Korea. 2. The distributions of the land uses in Maejuri, Seunghwan eup in Chonan city were mainly recorded by forest(29.82%), orchard(14.11%) and landscaping around building site(9.05%). 3. The percentages of the Tree/Shrub Biotope type were ranged by the orchard type(23.8%), natural and artificial deciduous type with natural coniferous(23.6%) and etc. 4. The Grass Biotope type was mainly recorded by the wild grassland type(27.99%), garden type(14.21%) and peddy field with wild grassland(13.24%). 5. The distributions of the wetland were pointed high at the wetland type with reed marsh and edge vegetation around wetland(54.79%) and etc.

Land Cover Classification over East Asian Region Using Recent MODIS NDVI Data (2006-2008) (최근 MODIS 식생지수 자료(2006-2008)를 이용한 동아시아 지역 지면피복 분류)

  • A Land cover map over East Asian region (Kongju national university Land Cover map: KLC) is classified by using support vector machine (SVM) and evaluated with ground truth data. The basic input data are the recent three years (2006-2008) of MODIS (MODerate Imaging Spectriradiometer) NDVI (normalized difference vegetation index) data. The spatial resolution and temporal frequency of MODIS NDVI are 1km and 16 days, respectively. To minimize the number of cloud contaminated pixels in the MODIS NDVI data, the maximum value composite is applied to the 16 days data. And correction of cloud contaminated pixels based on the spatiotemporal continuity assumption are applied to the monthly NDVI data. To reduce the dataset and improve the classification quality, 9 phenological data, such as, NDVI maximum, amplitude, average, and others, derived from the corrected monthly NDVI data. The 3 types of land cover maps (International Geosphere Biosphere Programme: IGBP, University of Maryland: UMd, and MODIS) were used to build up a "quasi" ground truth data set, which were composed of pixels where the three land cover maps classified as the same land cover type. The classification results show that the fractions of broadleaf trees and grasslands are greater, but those of the croplands and needleleaf trees are smaller compared to those of the IGBP or UMd. The validation results using in-situ observation database show that the percentages of pixels in agreement with the observations are 80%, 77%, 63%, 57% in MODIS, KLC, IGBP, UMd land cover data, respectively. The significant differences in land cover types among the MODIS, IGBP, UMd and KLC are mainly occurred at the southern China and Manchuria, where most of pixels are contaminated by cloud and snow during summer and winter, respectively. It shows that the quality of raw data is one of the most important factors in land cover classification.

Impacts of Land Surface Boundary Conditions on the Short-range weather Forecast of UM During Summer Season Over East-Asia (지면경계조건이 UM을 이용한 동아시아 여름철 단기예보에 미치는 영향)

  • In this study, the impacts of land surface conditions, land cover (LC) map and leaf area index (LAI), on the short-range weather forecast over the East-Asian region were examined using Unified Model (UM) coupled with the MOSES 2.2 (Met-Office Surface Exchange Scheme). Four types of experiments were performed at 12-km horizontal resolution with 38 vertical layers for two months, July and August 2009 through consecutive reruns of 72-hour every 12 hours, 00 and 12 UTC. The control experiment (CTRL) uses the original IGBP (International Geosphere-Biosphere Programme) LC map and old MODIS (MODerate resolution Imaging Spectroradiometer) LAI, the new LAI experiment (NLAI) uses improved monthly MODIS LAI. The new LC experiment (NLCE) uses KLC_v2 (Kongju National Univ. land cover), and the new land surface experiment (NLSE) uses KLC_v2 and new LAI. The reduced albedo and increased roughness length over southern part of China caused by the increased broadleaf fraction resulted in increase of land surface temperature (LST), air temperature, and sensible heat flux (SHF). Whereas, the LST and SHF over south-eastern part of Russia is decreased by the decreased needleleaf fraction and increased albedo. The changed wind speed induced by the LC and LAI changes also contribute the LST distribution through the change of vertical mixing and advection. The improvement of LC and LAI data clearly reduced the systematic underestimation of air temperature over South Korea. Whereas, the impacts of LC and LAI conditions on the simulation skills of precipitation are not systematic. In general, the impacts of LC changes on the short range forecast are more significant than that of LAI changes.

Anti-MRSA action of Papenfussiella kuromo

  • Papenfussiella kuromo (PK) is a marine plant and an abundant ecological resource for the future; it is found in almost 80% of the terrestrial biosphere. The aim of this study was to investigate the antibacterial activity of PK against methicillin-resistant Staphylococcus aureus (MRSA), multidrug-resistant pathogen. The minimum inhibitory concentrations (MICs) of PK hexane fraction (PKH) against 7 strains of MRSA ranged from 1.0 to 2.0 mg/mL. In the checkerboard dilution method, a synergistic effect of the PKH and the antibiotics (oxacillin and norfloxacin) was seen. PKH markedly reduced the MIC of each of the 4 antibiotics against MRSA. The time-kill assay showed that the synergistic activity of PKH and an antibiotic reduced the bacterial counts below the lowest detectable limit after 24 h. These findings suggest that PKH has antibacterial activity, and may be important baseline data in future extensive studies of living marine resources as a source of compounds active against MRSA.
