경락 전위를 이용한 경락의 생체물리학적 연구 (Study on the Electric Potentials at Biophysical Meridian System)

  • Objectives : Assuming That the Characteristic of Meridian System Has Been Similar to This of Electric Potentials in Human Body and That Measurements of Electric Potential at Well and Sea Points in Branches of the Twelve Meridians Will Be Representative of Measurements of the Twelve Meridians, to Measure the Electric Potentials in Healthy Volunteers(HG), Patients Diagnosed As a Cerebral Infarction(CG), a Arrhythmia(AG), a Other Intervertebral Disc Disorder(IG), and a Joint Disorder(JG), and Then to Find Out the Characteristic of Biophysical Meridian System, Finally to Compare with the Results of the Electric Potential in Those Groups. Methods : We Selected Who HG Were Diagnosed by a Blood Test, Urine Examination and Differentiation of Syndromes by Five Viscera among Volunteers, CG by CT and Wind-Syndrome Caused by Hyperactivity of the Liver-Yang, AG by EKG, CT, and Deficiency or the Heart Blood, IG by X-ray or CT and Pain in Waist, and JG by X-ray and Knee Pain. Their Electric Potential of Well and Sea Points in the Meridians Were Measured by hysiograph. Results and Conclusions : Measurements Were Analyzed by Factor Analysis, Analysis of Variance, and Logistic Regression Model, We Obtained That Most of the Results, In Conclusion, Hold Good for the Classical Meridian Doctrine.

Interaction of ct-DNA with 2,4-Dihydroxy Salophen

  • In the present study, at first, 2,4-Dihydroxy Salophen (2,4-DHS), has been synthesized by combination of 1, 2-diaminobenzene and 2,4-dihydroxybenzaldehyde in a solvent system. This ligand containing meta-quinone functional groups were characterized using UV-Vis and IR spectroscopies. Subsequently, the interaction between native calf thymus deoxyribonucleic acid (ct-DNA) and 2,4-DHS, was investigated in 10 mM Tris/HCl buffer solution, pH 7.2, using UV-visible absorption and fluorescence spectroscopies, thermal denaturation technique and viscosity measurements. From spectrophotometric titration experiments, the binding constant of 2,4-DHS with ct-DNA was found to be (1.1 ${\pm}\;0.2)\;{\times}\;10^4\;M^{-1}.$ The fluorescence study represents the quenching effect of 2,4-DHS on bound ethidium bromide to DNA. The quenching process obeys linear Stern-Volmer equation in extended range of 2,4-DHS concentration. Thermal denaturation experiments represent the increasing of melting temperature of DNA (about 3.5 ${^{\circ}C}$) due to binding of 2,4-DHS. These results are consistent with a binding mode dominated by interactions with the groove of ct-DNA.

Estimation of Corn Growth by Radar Scatterometer Data

  • Ground-based polarimetric scatterometers have been effective tools to monitor the growth of crop with multi-polarization and frequencies and various incident angles. An important advantage of these systems that can be exploited is temporal observation of a specific crop target. Polarimetric backscatter data at L-, C- and X-bands were acquired every 10 minutes. We analyzed the relationships between L-, C- and X-band signatures, biophysical measurements over the whole corn growth period. The Vertical transmit and Vertical receive polarization (VV) backscattering coefficients for all bands were greater than those of the Horizontal transmit and Horizontal receive polarization (HH) until early-July, and then thereafter HH-polarization was greater than VV-polarization or Horizontal transmit and Vertical receive polarization (HV) until the harvesting stage (Day Of Year, DOY 240). The results of correlation analysis between the backscattering coefficients for all bands and corn growth data showed that L-band HH-polarization (L-HH) was the most suited for monitoring the fresh weight ($r=0.95^{***}$), dry weight ($r=0.95^{***}$), leaf area index ($r=0.86^{**}$), and vegetation water content ($r=0.93^{***}$). Retrieval equations were developed for estimating corn growth parameters using L-HH. The results indicated that L-HH could be used for estimating the vegetation biophysical parameters considered here with high accuracy. Those results can be useful in determining frequency and polarization of satellite Synthetic Aperture Radar stem and in designing a future ground-based microwave system for a long-term monitoring of corn.

NMR Study on the Internal Dynamics of Ketosteroid Isomerase

  • The backbone dynamics of ketosteroid isomerase, a homodimeric enzyme with 125 amino acid residues per subunit, has been studied in the presence/absence of a steroid ligand and 5% trifluoroethanol (TFE) by $^{15}$ M relaxation measurements. The relaxation data were analyzed using the model-free formalism to extract the model-free parameters (S$^2$, $\tau$$_{e}$, and R$_{ex}$ ). The results show that a large number of the residues, particularly those involved in the dimer interaction, exhibit reduced order parameters (S$^2$) in the steroid-bound enzyme, indicating the increased high-frequency (pico- to nanosecond) motions in the interface region upon ligand binding. The results also show that that the presence of 5 % TFE in free enzyme causes little change or slight increase in the order parameters for a number of residues in the dimer interface region. However, the majority of the residues in free enzyme exhibit reduced order parameters in the presence of 5 % TFE, indicating that the increase in entropy is partially responsible for the increased stability of KSI by 5% TFE.E.E.

Conformational Studies of Gaseous Proteins Using Mass Spectrometry

  • Conformations of the +5 to +13 charge state of ubiquitin ions have been studied in the gas phase by an Electron Capture Dissociation (ECD) mass spectrometry (MS) technique. This approach has showed that the conformations of the gaseous ions change from the compact to extended structures as the number of protons on the protein ions increases, consistent with previous collisional cross-section measurements by an ion-mobility MS. However, this observation is in contrast to that of the solution-phase where the unique native structure is usually found. The (un)folding stability and kinetics of these gaseous ions were further investigated experimentally using gradual blackbody-radiation or sudden laser-induced thermal heating, respectively. These studies have provided the evidence that the thermodynamics and kinetics of protein (un)folding in the gas phase are quite different from those of the native aqueous proteins.

  • Biodegredable and injectable nanocomposites based on polypropylene fumarate (PPF) as unsaturated polyester were prepared. The investigated polyester was crosslinked with three different monomers namely N-vinyl pyrrolidone (NVP), methyl methacrylate (MMA) and a mixture of NVP and MMA (1:1 weight ratio) and was filled with 45 wt% of hydroxyapatite (HA) incorporated with different concentrations of chemically treated natural bone powder (NBP) (5, 10 and 15 wt%) in order to be used in treatment of orthopedics bone diseases and fractures. The nanocomposites immersed in the simulated body fluid (SBF) for 30 days, after the period of immersion in-vitro bioactivity of the nanocomposites was studied through Fourier transform infrared (FTIR), scanning electron microscope (SEM), energy dispersive X-ray (EDX) in addition to dielectric measurements. The degradation time of immersed samples and the change in the pH of the SBF were studied during the period of immersion.

레이저에 의한 연골의 광학적 열적 특성변화 연구 (Optical and Thermal Characteristic Studies of Cartilage by Laser Irradiation)

  • Laser cartilage reshaping(LCR) is a promising method for reshaping cartilage by using laser irradiation to maintain permanently modifies its shape. However this method has not been fully understood due to the limited scientific researches. The purpose of this study is to analyze optical and thermal characteristics of cartilage during laser irradiation. After analyzing Monte Carlo simulation for the comparison of laser fluence distributions with different laser wavelengths the characterization of the spectral changes during Nd:YAG laser(${\lambda}$ = 1444 nm) irradiation was investigated in the ranges of 900-1700 nm with double integrating spheres. The surface temperature distribution changes during laser irradiation were investigated with an infrared camera. The quantitative measurements of optical and thermal characteristics in cartilage after laser irradiation were correlated with the transition of water flux(from bound to free water) and this study may be useful for better understanding of biophysical transformation phenomena in cartilage after laser heating.

  • Local probe techniques such as scanning probe microscopy (SPM) or atomic force microscopy (AFM) extended our perception into ultra small world. Specially, the sense of touching was extended by AFM into the micro- and nanoworld and has provided complementary new insights of the microscopic world. In addition, touching objects is an essential step before trying to manipulate things. SPM as a touch sensor not only measure the mechanical properties but also detect different properties such as magnetic, electrical, ionic, thermal, chemical and biophysical properties in nanoscale and even less. Obtaining biophysical measurements, monitoring dynamics and processes together with high-resolution imaging of the biomolecules and cells with rather simpler sample preparation than any other techniques give great attractions to the scientists experimenting with biological samples. Among the many AFM capabilities we will specifically introduce the force plot which is used to measure tip-sample interactions and its application this time.

Effective Validation Model and Use of Mobile-Health Applications for the Elderly

  • Objectives: Due to the uncontrolled increase of the mobile health applications and their scarce use by elderly for reason of absence credibility of measurements by lack scientific support, the aim of this study was to evaluate the differences between the biophysical measurements based on standard instrument against a mobile application using controlled experiments with elderly to propose an effective validation model of the developed apps. Methods: The subjects of the study (50 people) were elderly people who wanted to check their weight and cardiac status. For this purpose, two mobile applications were used to measure energy expenditure based on physical activity (Activ) and heart rate (SMCa) during controlled walking at specific speeds. Minute-by-minute measurements were recorded to evaluate the average error and the accuracy of the data acquired through confidence intervals by means of statistical analysis of the data. Results: The experimental results obtained by the Activ/SMCa apps showed a consistent statistical similarity with those obtained by specialized equipment with confidence intervals of 95%. All the subjects were advised and trained on the use of the applications, and the initial registration of data to characterize them served to significantly affect the perceived ease of use. Conclusions: This is the first model to validate a health-app with elderly people allowed to demonstrate the anthropometric and body movement differences of subjects with equal body mass index (BMI) but younger. Future studies should consider not only BMI data but also other variables, such as age and usability perception factors.

The Homeobox and Genetic Disease: Structure and Dynamics of Wild Type and Mutant Homeodomain Proteins

  • Structural and physical properties of type wild type and various selected mutants of the vnd/NK-2 homeodomain, the protein product of the homeobox, and the implication in genetic disease are reviewed. The structure, dynamics and thermodynamics have been Investigated by NMR and by calorimetry. The interactions responsible for the nucleotide sequence-specific binding of the homeodomain to its consensus DNA binding site have been identified. There is a strong correlation between significant structural alterations within the homeodomain or its DNA complex and the appearance of genetic disease. Mutations in positions known to be important in genetic disease have been examined carefully For example, mutation of position 52 of vnd/NK-2 results in a significant structural modification and mutation of position 54 alters the DNA binding specificity and amity The $^{15}N$ relaxation behavior and heteronuclear Overhauser effect data was used to characterize and describe the protein backbone dynamics. These studies were carried out on the wild type and the double mutant proteins both in the free and in the DNA bound states. Finally, the thermodynamic properties associated with DNA binding are described for the vnd/NK-2 homeodomain. These thermodynamic measurements reinforce the hypothesis that water structure around a protein and around DNA significantly contribute to the protein-DNA binding behavior. The results, taken together, demonstrate that structure and dynamic studies of proteins combined with thermodynamic measurements provide a significantly more complete picture of the solution behavior than the individual studies.
