  • We are designed the delivery constitution technique that is to be measure the safe RFID statusof thewireless delivery system level (WDSL) on the delivery system tracking system. The delivery system level condition by the delivery system tracking system is organized with the RFID system. As to inspection a wireless network of the wireless network, we are found of the delivery value with wireless network by the upper take form. The concept of delivery system level is organized the reference of wireless level for delivery signal by the delivery RFID tracking system. Further, symbolizing a safe deliveryof the WDSL of the medium-minimum interval of the RFID tracking system, and the delivery wireless network RFID that was the delivery value of the far delivery of the DSTS-FA-φMED-MIN with 5.80±1.20 units, that was the delivery value of the convenient delivery of the DSTS-CO-φMED-MIN with 4.06±(-0.04) units, that was the delivery value of the flank delivery of the DSTS-φMED-MIN with 0.91±0.07 units, that was the delivery value of the vicinage delivery of the DSTS-VI-φMED-MIN with 0.18±(-0.03) units. The RFID will be to look into at the safe of the RFID tracking systemwith wireless network bythe delivery system level on the WDSL that is supply the wireless tracking system by the delivery system level system. We will be possible make to curb of a tracking system that to put the wireless signal and to use of the delivery data of RFID level by the delivery system.

Performance of Quantitative Real-Time PCR for Detection of Tuberculosis in Granulomatous Lymphadenitis Using Formalin-Fixed Paraffin-Embedded Tissue

  • Although culture is the gold standard method to identify mycobacteria, its use in tuberculous lymphadenitis (TBL) is limited due to formalin fixation of the submitted specimens. We evaluated the performance of quantitative real-time PCR (q-PCR) for Mycobacterium Tuberculosis (MTB) in granulomatous lymphadenitis using formalin-fixed paraffin-embedded (FFPE) tissues. From 2000 to 2010, a total number of 117 cases of lymph node samples with granulomatous inflammation which were surgically removed and fixed in formalin were studied. Hematoxylin & Eosin (H&E) and Ziehl-Neelsen-stained (ZN) slides were reviewed. qPCR using Real TB-Taq$^{(R)}$ was performed for all cases to identify Mycobacterium tuberculosis. Thirteen non-tuberculous lymphadenopathy cases were used as negative control. Cervical lymph nodes were more frequently affected (60%, 70/117) than other sites. ZN stain for acid fast bacilli was positive in 19 (16.24%) cases. qPCR for tuberculosis was positive in 92 (78.63%) cases. Caseous necrosis was found in 103 (88.03%) cases. While the ZN stain and qPCR were both negative in all control cases, the qPCR showed a significantly higher positive rate (78.63% vs. 16.24%) compared to ZN stain in histologically diagnosed TBL. Quantitative real-time PCR proves to be more sensitive than ZN stain for diagnosis of tuberculous lymphadenitis.

  • Breast cancer is one of the most common cancers affecting women worldwide. Although the survival rate of breast cancer has increased, breast cancer still results in a high mortality rate. Breast cancer deaths are caused by metastasis that occurs in organ dysfunction. Recently, there have been many studies on circulating tumor cells (CTCs), which are related to breast cancer metastasis in the blood. Recent studies have demonstrated that some CTCs do not express epithelial markers. Therefore, in this study, total RNA was extracted from blood without separating out the CTCs, and the characteristics of the CTCs were analyzed by RT-qPCR. Cyclin D1 and twist-related protein 1 (TWIST1) are well-known markers for predicting the prognosis of patients with breast cancer. However, few studies have demonstrated the use of CCND1 and TWIST1 in blood as diagnostic and prognostic markers of breast cancer. In this study, patients with late-stage breast cancer had overexpressed CCND1 and TWIST1 than patients with different stages of breast cancer (P < 0.001 and P < 0.01, respectively). The relative expression level of CCND1 in survivors was higher than in patients who died (P = 0.06). The relative expression level of TWIST1 in survivors was lower than in patients who died (P = 0.08). Overall CCND1 and TWIST1 were not useful as markers for the diagnosis of breast cancer through blood. However, we showed the possibility of using CCND1 and TWIST1 as prognostic markers, and a large-scale study is needed to confirm the usefulness of these prognostic markers.

  • In this study, the effects of $Ca^{2+}$ and cyclic adenosine monophosphate (cAMP) on microsomal triglyceride (TG) transfer protein (MTP) activity were investigated in rat liver. MTP activity was high when liver contained low levels of cAMP, which was induced by administration of glucose, or high levels of total $Ca^{2+}$ and TG. However, MTP activity increased by high levels of $Ca^{2+}$ and TG was reduced in a dose-dependent manner by treatment with dibutyryl-cAMP (db-cAMP), a cAMP analogue. Conversely, when homogenates of liver from normal rats, with low levels of total $Ca^{2+}$ and high levels of cAMP, were incubated with thapsigargin, a $Ca^{2+}$-inducer, MTP activity was increased in a dose-dependent manner compared to control. Therefore, our results suggest that high levels of $Ca^{2+}$ cause hypertriglyceridemia through the elevation of MTP activity, as opposed to high levels of cAMP, which suppress MTP activity and inhibit hypertriglyceridemia.

Water Extract from Rice Bran Fermented with Lactobacillus plantarum Hong Inhibits Thromboxane A2 Production Associated Microsomal Enzyme Activity in Human Platelets

  • In this study, we investigated the effect of rice bran water extract fermented with Lactobacillus plantarum Hong (RBLw), on activities of cyclooxygenase-1 (COX-1) and thromboxane $A_2$ synthase (TXAS), thromboxane $A_2$ ($TXA_2$) production associated microsomal enzymes and evaluated its the antiplatelet effect. RBLw, containing 13.5 mg of ferulic acid, dose-dependently inhibited ADP-induced platelet aggregation, and inhibited the production of $TXA_2$, an aggregation molecule. In addition, RBLw directly inhibited COX-1 activity in a dose-dependent manner, but not TXAS activity in platelet microsomal fraction having cytochrome c reductase (an endoplasmic reticulum marker enzyme) activity and expressing COX-1 (72 kDa) and TXAS (60.5 kDa) proteins. These results suggest that RBLw selectively inhibited the activity of COX-1 rather than TXAS to attenuate $TXA_2$ production in ADP-activated platelets. Thus, we demonstrate that RBLw might have direct COX-1 antagonistic function for platelet aggregation-mediated diseases, such as thrombosis, myocardial infarction, atherosclerosis, and ischemic cerebrovascular disease.

  • Blood pressure is possible to diagnose a disease associated with blood pressure and judgment the current health of patients. Automatic blood pressure monitor capable of measuring a blood pressure easily in hospital and at home have become spread. In this study, we developed the blood pressure simulator (BPS) that can test the arm-type automatic blood pressure monitor that is commonly used in hospital. BPS is to produce a pressure similar to the pressure wave generated in the human blood using a servo disk motor. Then, using the silicon tube, it implements the situations such as human blood vessels, and to output the generated pressure waveform. Simply the BPS's phantom put on the cuff and it is able to simulate blood pressure. So anyone can quickly test the blood pressure monitor within one minute and it is possible to shorten the test time required for the automatic blood pressure monitor. In Performance test, the trends and the standard deviation of the values measured in the BPS is similar to the value of the measured pressure from people with normal blood pressure. Thus, the development BPS showed a possibility of taking into account the actual blood pressure measurement environment simulator.

  • The development of technology has led to ubiquitous health care service, which enables many patients to receive medical services anytime and anywhere. For the ubiquitous health care environment, real-time measurement of biomedical signals is very important, and the medical instruments must be small and portable or wearable. So, such devices have been developed to measure biomedical signals. In this study, we develop the biomedical monitoring device which is sensing the PPG signal, one of the useful signal in the field of ubiquitous healthcare. We design a watch-like biomedical signal monitoring system without a finger probe to prevent the user's inconvenience. This system obtains the PPG from the radial artery using a sensor in the wrist band. But, new device developed in this paper is easy to get the motion artifacts. So, we proposed new algorithm removing the motion artifacts from the PPG signal. The method detects motion artifacts by changing the degree of brightness of the light source. If the brightness of the light source is reduced, the PPG pulses will disappear. When the PPG pulses have disappeared completely, the remaining signal is not the signal that results from the changing blood flow. We believe that this signal is the motion artifact and call it the noise reference signal. The motion artifacts are removed by subtracting the noise reference signal from the input signal. We apply this algorithm to the system, so we can stabilize the biomedical monitoring system we designed.

Estimation of the Blood Pressure Using Point Variation Aspect of Dicrotic Notch on Pulsating Waveform at Each Cardiac Periods (주기별 맥동파형의 절흔점 위치변화 특성을 이용한 혈압 추정)

  • In the study, novel blood pressure estimation method was proposed to improve the accuracy of oscillometric method. The proposed algorithm estimated the blood pressure by comparing and analyzing the point variation aspect of dicrotic notch on pulsating waveform during each cardiac cycle. The waveforms of each cardiac cycle were extracted by maximum points. The extracted pulsating waveforms were applied by re-sampling, end-matching, and normalization. The systolic and diastolic blood pressures were estimated by point variation aspect of dicrotic notch. The blood pressures, which were estimated from proposed algorithm, were compared and analyzed by blood pressures from oscillometric methods and auscultation. The systolic blood pressure from oscillometric methods were +0.88 mmHg more than proposed algorithm, and 1.875 less than the diastolic blood pressures from proposed algorithm. The systolic and diastolic blood pressures from auscultation were 2.89 mmHg and 3.44 mmHg less than the blood pressures from proposed algorithm. As the errors between blood pressures from proposed algorithm, oscillometric method and auscultation were less than 5 mmHg, the proposed algorithm was effective.

Performance of HPV E6/E7 mRNA Genotyping Test on Paired Cervical Cancer Exfoliated Cells and Formalin Fixed Paraffin Embedded Tissues

  • Investigation of human papillomavirus (HPV) in archival formalin-fixed paraffin-embedded (FFPE) material is important for understanding cervical carcinogenesis. The objective of the present study was to identify the high risk HPVs (HR-HPVs) using HPV E6/E7 mRNA testing from archival tissues in cervical cancer and the relation to HR-HPVs genotypes in paired cervical exfoliated cells. HPV E6/E7 mRNA testing and DNA chip testing were performed in 79 paired cervical FFPE tissues and exfoliated cells from women with histologically confirmed squamous cell carcinoma and adenocarcinoma. Overall agreement in HR-HPVs detection from FFPE samples and cytology samples were 98.5% in HPV 16, 100% in HPV 18, HPV 31, HPV 33, HPV 58, HPV 66, and HPV 68. Type-specific agreement between FFPE samples and cytology samples was 89.1% in HPV positive, 93.5% in HPV 16 and more than 70% in the other HR-HPVs. In conclusion, HR-HPVs were reliably detected in paired FFPE and cytology samples with some variation in type-specific detection.

Dual Electrospinning to Manufacture Hybrid Nanofibrous Scaffold using Polyurethane and Poly(Ethylene Oxide) (Polyurethane과 Poly(Ethylene Oxide)를 이용한 hybrid 나노섬유 지지체의 제작)

  • The object of this study is to investigate the potential of dual-electrospun polymer based structure for vascular tissue engineering, especially for the medium or small sue blood vessels. Polyurethane(PU), which is known to be biocompatible in this area, was electrospun with poly(ethylene oxide) (PEO). Concentration of PU was fixed at 20wt%, while that of PEO was set from 15 to 35wt%. Morphological features were observed by SEM image and measurement of porosity and cellular responses were tested before and after extracting PEO from the hybrid scaffolds by immersing the scaffolds into distilled water. The diameter of PEO fibers were ranged from 200nm to 500nm. The lower concentration of PEO tended to show beads. The porosity of the scaffolds after extracting PEO was highly increased with higher concentration of PEO as expected. Also, higher proliferation rate of smooth muscle cells was observed at higher concentration of PEO than at the lower concentration and without PEO. As conclusions, this dual electrospinning technique combined with PU and PEO is expected to overcome the current barrier of cell penetration by providing more space for cells to proliferation.
