Effect of Toluene Application to Skin on the Enhancement of Liver Injury in $CCl_4$-Pretreated Rats

  • To evaluate an effect of toluene application to skin on the enhancement of liver damage in $CCl_4$-pretreated rats, toluene (35 mg/$cm^2$) was sequentially applied for 5 days to the skin of liver damaged rats with $CCl_4$ (6 times every other day: 0.1 ml/100 g body weight-50% $CCl_4$ in olive oil) On the basis of the functional and morphological findings in rat liver, appling toluene to the skin in liver damaged animals led to the more enhanced liver damage. In addition, by applying toluene to skin of liver damaged rats, the hepatic cytochrome P450 content was somewhat more increased, but the hepatic benzylalcohol dehydrogenase activity was significantly decreased (P<0.001), whereas benzaldehyde dehydrogenase activity was not statistically changed. In conclusion, the toluene application to skin in liver-damaged rat led to enhancement of liver injury that may be due to the accumulation of toluene metabolite in liver.

Estimation of Death Time by Changes of Postmortem Xanthine Oxidase Activity in Rats

  • To evaluate the postmortem changes in activities of oxygen free radical metabolizing enzymes, the rats were sacrificed with cervical dislocation and were kept in an incubator at $25^{\circ}C$, 70% of humidity for 12 hours. The activities of aniline hydroxylase, catalase, glutathione-S-transferase and superoxlde dismutase were decreased with the time. On the other hand, the activity and type conversion ratio (type D ${\to}$type O) of hepatic xanthine oxidase (XO) were gradually increased. From these changes of XO, the estimation of death time (mathematical equation) could be determined with the least square method. To clarify the cause of increasing XO activity, enzyme kinetics were examined. The Km values of XO were decreased with the time. In conclusion, the determination of liver XO activity might be used for the estimation of death time in the early postmortem period.

The Effect of Vision Training on Exophoria and Intermittent Extropia Using MYSTERY CIRCLE System (MYSTERY CIRCLE 시스템을 이용한 폭주부족형 사위 및 간헐성 사시 환자의 시기능 훈련 효과 연구)

  • Purpose: The purpose of this study was to investigate the effect of vision training using MYSTERY CIRCLE system on patients with exophoria and intermittent extropia. Mehtods: The vision training observation group consists of 26 clients who had exophoria (n=18) and intermittent extropia (n=8) without opthalmic diseases or vertical phoria. Our clients took each vision training once a week for eight weeks and were investigated for the symptoms changes of binocular vision. Results: According to the results, MISTERY CIRCLE vision training methods improved the binocular vision function and sensation symptoms. For Worth 4 Dot test, Titmus test and red lenses showed fusion rate changes. Conclusions: The study proved that MISTERY CIRCLE vision training system for patients with binocular vision was effective for the improvement of the binocular vision symptoms of these patients.

Super-resolution Microscopy with Adaptive Optics for Volumetric Imaging

  • Optical microscopy is a useful tool for study in the biological sciences. With an optical microscope, we can observe the micro world of life such as tissues, cells, and proteins. A fluorescent dye or a fluorescent protein provides an opportunity to mark a specific target in the crowd of biological samples, so that an image of a specific target can be observed by an optical microscope. The optical microscope, however, is constrained in resolution due to diffraction limit. Super-resolution microscopy made a breakthrough with this diffraction limit. Using a super-resolution microscope, many biomolecules are observed beyond the diffraction limit in cells. In the case of volumetric imaging, the super-resolution techniques are only applied to a limited area due to long imaging time, multiple scattering of photons, and sample-induced aberration in deep tissue. In this article, we review recent advances in super-resolution microscopy for volumetric imaging. The super-resolution techniques have been integrated with various modalities, such as a line-scan confocal microscope, a spinning disk confocal microscope, a light sheet microscope, and point spread function engineering. Super-resolution microscopy combined with adaptive optics by compensating for wave distortions is a promising method for deep tissue imaging and biomedical applications.

The Impact of Refractive Errors on Quality of Life in Elementary School Children (초등학교 학생의 근시도가 삶의 질에 미치는 영향)

  • Purpose: This study investigated the impact of ametropia and myopia on health-related quality of life (QoL) measures in elementary schoolers. Methods: Elementary school children of 92 aged 12 to 13 were divided into emmetropia and myopia groups by spherical equivalent. Then myopia was classified into the low, moderate and high myopia groups. Vision-related QoL scores were determined using PedsQL 4.0 (Pediatric Quality of Life Inventory) with physical health (8 items), emotional functioning (4 items), social functioning (5 items) and school functioning (5 items). Results: The total QoL score in the myopia group appeared lower than that in the emmetropia group, however the difference was not statistically significant (p>0.05). When it comes to physical health (running or exercising) and social functioning (getting along with friends or being teased)-related questions, the QoL score in myopia was low compared with the emmetropia group (p<0.05). High myopia showed a low score in physical health items but there was no significant difference in overall QoL scores in comparison with other groups (p<0.05). Conclusions: Refractive errors does not have a great impact on the total health-related QoL in elementary school children but it causes discomfort in physical health and social functioning.

Research on Visual Function Before and After Prescription of Vertical Prism (수직사위 교정 전·후 시기능 변화에 대한 연구)

  • Purpose: This study was undertaken to study on visual function before and after prescription of vertical prism. Methods: 80 subjects (10-30ages) who had been precribed vertical prism and 1 year after they were represcribed vertical prism. Results: Phoria (p=0.000), supra vergence (p=0.000), NPC (p=0.003), NPA (right eye) (p=0.005), NPA(left eye) (p=0.000) showed a significant change. Conclusion: Vertical prism prescription was reduced during revisit. Supra vergence, NPC, NPA(right eye), NPA(left eye) showed a significant change. Binocular Accommodative lag was not associated with vertical prism prescription and binocular vision findings.

  • In this paper the conceptual design and development of a compact vertical type soft x-ray microscope is described. This x-ray microscope operates in the water window wavelength region(2.3 ${\sim}$ 4.4nm), where natural contrast between carbon(protein) and oxygen(water) allows imaging of unstained biological material their natural, hydrated environment. Until now, operational x-ray microscopes are based on synchrotron radiation sources, which limit their accessibility. Many biologists would benefit from having the x-ray microscope as a tool among other tools in their own laboratory, For this purpose we introduced the compact vertical type soft X-ray microscope with 50 nm resolution for biomedical application. The compact vertical type soft x-ray microscope is based on a laser plasma x-ray source, doubled ellipsoidal condenser reflective optics, diffractive zone plate optics and MCP coupled with CCD to record an x-ray image.

  • A Fresnel lens is an optical component which can be used as a cost-effective, lightweight alternative to conventional continuous surface optics. Fresnel lens solar concentrators continue to fulfill a market requirement as a system component in high volume cost effective Concentrating Photovoltaic (CPV) electricity generation. The basic principles of the fresnel lens are reviewed and some practical examples are described. To investigate the performance space of the Fresnel lens, a fast simulation method which is a hybrid between raytracing and analytical computation is employed to generate a cache of simulation data. Injection molders are warming up to the idea of cycling their tool surface temperature during the molding cycle rather than keeping it constant. Heat and cool process are now also finding that raising the mold wall temperature above the resin's glass-transition or crystalline melting temperature during the filling stage and product performance in applications from automotive to packaging to optics. This paper deals with the suitability of Fresnel lenses of imaging and non-imaging designs for solar energy concentration. The concentration fresnel lens confirmed machinability and optical transmittance and roughness measure through manufactured the prototype.

Demonstration of the Usefulness of Optical Coherence Tomography in Imaging a Mouse Tail Model of Lymphedema

  • To investigate the usefulness of optical coherence tomography (OCT) for imaging lymphedema, we directly compared it to other histological methods in a mouse model of lymphedema. We performed detailed imaging of the lymphedema lesion on a mouse tail. We imaged the mouse tail in vivo with OCT and created histopathological samples. We constructed a spectrometer-based OCT system using a fiber-optic Michelson interferometer. The light was directed to 50:50 couplers that split the light into reference and sample arms. Backscattered light from a reference mirror and the sample produced an interference fringe. An OCT image of the lymphedema model revealed an inflammatory reaction of the skin that was accompanied by edema, leading to an increase in the light attenuation in the dermal and subcutaneous layers. Similar to OCT image findings, histological biopsy showed an inflammatory response that involved edema, increased neutrophils in epidermis and subdermis, and lymphatic microvascular dilatation. Furthermore, the lymphedema model showed an increase in thickness of the dermis in both diagnostic studies. In the mouse tail model of lymphedema, OCT imaging showed very similar results to other histological examinations. OCT provides a quick and useful diagnostic imaging technique for lymphedema and is a valuable addition or complement to other noninvasive imaging tools.

In Vivo Enhanced Indocyanine Green-Photothermal Therapy for a Subconjunctival Tumor

  • Indocyanine green (ICG) is a dye approved for use in clinical diagnostics. ICG remains in the intravascular space following intravenous administration, due to its ability to rapidly bind to the plasma proteins, and its therapeutic potential has been studied in well-vascularized cutaneous tumors. Here we have evaluated the clinical response of a subconjunctival tumor to photothermal therapy (PTT) using an ICG-enhanced near-infrared diode laser and its adverse effects, in a rabbit. 22 male New Zealand white rabbits with subconjunctival tumors were enrolled (control group 6, laser-only group 8, laser-with-ICG group 8). Rabbits in the laser-with-ICG group received ICG (twice, 2 mg/kg each time, intravenously) directly followed by irradiation with a diode laser (λ = 810 nm). Rabbits in the laser-only group were irradiated with the diode laser. ICG angiography, ultrasonography, and pathologic examination were performed to evaluate PTT response at specific time points (0, 2, and 4 weeks after PTT). Two weeks after initial treatment, the eight rabbits treated by laser with ICG showed a 100% response rate. There was no clinical response in both laser-only and control groups. ICG-PTT is a potential and effective palliative therapeutic modality for subconjunctival tumors.
