  • Autoimmune regulator gene (Aire)는 흉선에서 발현되며 promiscuous genes으로 알려진 흉선에서 자가항원 발현을 조절한다. Aire 와 promiscuous genes은 흉선에서 T세포 tolerance와 자가면역에 관여한다. 말초 조직 즉 림프절에서 Aire의 역할을 알아보고자 림프절 구성 세포중 하나인 fibroblastic reticular cell (FRC)을 분리 확립하였다. 마우스 림프절로부터 분리된 FRC에서 Aire의 발현을 확인하였고 또한 promiscuous antigen인 insulin의 발현도 확인하였다. Aire 과발현 플라스미드로 형질전환 후 배양 FRC에서 Insulin의 발현이 증가하였다. 이것은 Aire가 FRC에서 promiscuous gene의 발현을 조절한다는 것을 보여주며 peripheral selection과 연관되어 있을 수 있다는 것을 제시한다.

Isolation and Characterization of the Colletotrichum acutatum ABC Transporter CaABC1

  • Fungi tolerate exposure to various abiotic stresses, including cytotoxic compounds and fungicides, via their ATP-driven efflux pumps belonging to ATP-binding cassette (ABC) transporters. To clarify the molecular basis of interaction between the fungus and various abiotic stresses including fungicides, we constructed a cDNA library from germinated conidia of Colletotrichum acutatum, a major anthracnose pathogen of pepper (Capsicum annum L.). Over 1,000 cDNA clones were sequenced, of which single clone exhibited significant nucleotide sequence homology to ABC transporter genes. We isolated three fosmid clones containing the C. acutatum ABC1 (CaABC1) gene in full-length from genomic DNA library screening. The CaABC1 gene consists of 4,059 bp transcript, predicting a 1,353-aa protein. The gene contains the typical ABC signature and Walker A and B motifs. The 5'-flanking region contains a CAAT motif, a TATA box, and a Kozak region. Phylogenetic and structural analysis suggested that the CaABC1 is a typical ABC transporter gene highly conserved in various fungal species, as well as in Chromista, Metazoans, and Viridiplantae. We also found that CaABC1 was up-regulated during conidiation and a minimal medium condition. Moreover, CaABC1 was induced in iprobenfos, kresoxim-methyl, thiophanate-methyl, and hygromycin B. These results demonstrate that CaABC1 is necessary for conidiation, abiotic stress, and various fungicide resistances. These results will provide the basis for further study on the function of ABC transporter genes in C. acutatum.

LED 광원의 다양한 광질이 배추 유묘의 유전자 발현에 미치는 영향 (Effects of Light Quality Using LEDs on Expression Patterns in Brassica rapa Seedlings)

  • 광은 모든 살아 있는 세포에 있어서 매우 중요한 신호이며 생물체에게 이익이 되면서 동시에 해가 되기도 하는데, 광 환경에 잘 적응한 유기체는 자연에서의 경쟁에서 더 잘 살아남을 수 있다. 광질과 식물 생육에 대한 정보는 LED와 같은 인공광을 효율적으로 사용하게 하여 경제적인 손실을 줄일 수 있다. 초기 1주 동안 단일 광파장에 노출된 배추 유묘의 생장을 보면 백색광 조건에서 생장한 묘와 비교했을 때 적색광과 청색광 조건에서는 유묘의 발달과 생육이 정상이었다. 암조건에서 자란 유묘는 하배축이 길고 황화된 반면, 초적외광 조건에서 자란 유묘는 황화되었으나 하배축이 거의 신장하지 않았다. 마이크로어레이를 이용하여 유전자 대량 발현분석 실험을 수행한 결과, 청색광 조건에서 유묘의 생육이 가장 왕성하였으며 적색광 조건과 비교하여 발현변화를 보인 유전자의 수가 2배 많았다. 각각의 광질 조건에서 발현 증가 된 유전자들을 기능별로 분류해 보면(GO 분석) 적색광과 청색광은 생물학적 과정(biological process)과 관련된 유전자의 발현에 영향을 많이 주었다. 특히 대사과정(metabolic process)과 발달과정(developmental process) 그리고 세포 요소(cellular component) 중 색소체(plastid)나 엽록체(chloroplast)관련 유전자들의 발현이 청색광 조건에서 더 많이 증가하였다. 또한 초적외광 조건에서도 다양한 유전자들이 발현 변화를 보였다. 본 논문은 다양한 배추 유묘의 생장에 대한 광질의 영향을 분자생물학적으로 증명하기 위해 수행되었으며 유묘의 배양에 LED 광원이 효율적으로 이용될 수 있음을 보여 주었다.

GroEL-GroES 샤페로닌에 의한 단백질 접힘에 있어서 온도와 변성조건의 영향 (Effect of temperature and denaturation conditions on protein folding assisted by GroEL-GroES chaperonin)

  • 이 연구의 목적은 대장균 분자 샤페론 GroEL의 시험관 내 단백질 접힘에 있어서 반응온도의 영향과 보조샤페론의 필요 여부를 자발적 재접힘이 가능한 온도와 그렇지 않은 온도조건에서 조사하는 것이다. 여러 조건하에서 GroEL에 의한 두 가지 기질 단백질의 재접힘을 반응속도론적으로 조사하기 위하여 GroEL에 의한 단백질 침전생성억제와 변성된 단백질의 재접힘을 광범위하게 조사하였다. 자발적 재접힘이 가능하지 않은 $37^{\circ}C$에서는 ATP와 완전한 GroEL 시스템이 변성된 폴리펩티드의 재접힘을 위하여 필요하다는 것을 확인하였다. 하지만, 자발적 재접힘이 가능한 낮은 온도에서는 자발적 재접힘과 샤페론 의존적 단백질 재접힘이 서로 경쟁하는 것을 알 수 있었다. 따라서 GroEL은 변성된 폴리펩티드의 자발적 접힘 경로를 더 효율적인 단백질 재접힘 경로인 샤페론 의존적 단백질 재접힘 경로로 유도하는 것으로 보인다.

High Pressure Inactivation Kinetics of Salmonella enterica and Listeria monocytogenes in Milk, Orange Juice, and Tomato Juice

  • Effects of pressure come-up and holding times on the inactivation of Salmonella enterica and Listeria monocytogenes were evaluated in deionized water, milk, orange juice, and tomato juice with pH 6.76, 6.85, 3.46, and 4.11, respectively. The inoculated samples were subjected to high pressure treatments at 300, 400, and 500 MPa for less than 10 min at $30^{\circ}C$. At 500 MPa, the numbers of S. enterica and L. monocytogenes in deionized water, orange juice, and tomato juice were reduced by more than 6 log CFU/mL during the come-up time. Compared to orange and tomato juices, milk showed a considerable baroprotective effect against S. enterica and L. monocytogenes. At 300 MPa, the D values for S. enterica in milk, orange juice, and tomato juice were 0.94, 0.41, and 0.45 min, while those for L. monocytogenes were 9.56, 1.11, and 0.94 min, respectively. Low pH resulted in a noticeable synergistic effect on the inactivation of S. enterica and L. monocytogenes in orange and tomato juices. Therefore, these results might provide more useful information for designing the entire high hydrostatic pressure (HHP) conditions, taking the come-up time reduction, and food system.

  • Silk proteins consist of two major proteins, fibroin and sericin. There is currently an enormous reawakening of interest in these silk proteins as a biomaterial due to their mechanical and biological properties based on the detailed findings. Novel method for determination of the crystalline structure of silk proteins in an atomic level using nuclear magnetic resonance (NMR) was reviewed. Recent application of silks to biomaterials and prospects for future were discussed.

Clinical presentation of a horse-derived biomaterial and its Biocompatibility: A Clinical Case Report

  • Purpose: The objective of this clinical presentation was to present a clinical case series report of socket preservation, sinus augmentation, and bone grafting using a horse-derived biomaterial. Methods: A horse-derived biomaterial was used in 8 patients for different indications including socket preservation following tooth extraction, osseous bone grafting, and sinus augementation procedures. Surgeries were performed by a well trained specialist and clinical radiographs were obtained at designated intervals. Biopsy cores of 2 ${\times}$ 8 mm prior to implant placement was obtained following a healing interval of 4 - 6 months. A clinical and histologic evaluation was performed to evaluate the clinical effectiveness and biocompatibility of the biomaterial. Results: All surgeries in 8 patients were successful with uneventful healing except for one case with membrane exposure that eventually resulted with a positive outcome. Radiographic display of the healing phase during different intervals showed increased radiopacity of granular nature as the healing time increased. No signs of adverse effect or infection was observed clinically and the tissues surrounding the biomaterial seemed well-tolerated with good intentional healing. The augmented sinuses healed uneventfully suggesting in part, good biocompatibility of the biomaterial. Dental implants placed following socket preservation were inserted with high initial torque suggesting good initial stability and bone quality. Conclusions: Our results show that at least on a tentative level, a horse-derived biomaterial may be used clinically in socket preservation, sinus augmentation, bone grafting techniques with good intentional healing and positive results.

Imporoved Method for the Preparation of Silk Fibroin Hyoysates

  • An improvement of methods in fibroin hydrolysates preparat significantly enlarges their applications to practical use. Acidic hydrolysis by hydrohloric acid is one of the methods for silk fibroin depolymeriza6tion. A low yield of final product and long time of the process are the demerits of this method. Possibility of preparation of water-soluble silk hydrolysates with more yield and less expenses is investigated in this study. Such possibility is occurred with the increasing tratment temperature and simultaneously decreasing treatment time, concentration of hydrochoric acid respectively, the concentration of sodium hydroxide used for neutraliza6tion of hydolysates after hydrolysis. Colour is decreased in this case and a small amount of activated, too. Protection of hydrolysates against precipitation after neutraliza6tion, and separation and during concentrating process is the other merit of this method. Creamy-coloured insoluble silk powder is the remainder of hydrolyzed fibroin. This is the only the immobiliza6tion of enzymes and other physiological active substances. Fine particles of this powder can be used as additives for artificial diets and cultural media, as well as raw materials for polymer membranes, etc.
