Anti-Selective Dihydroxylation Reactions of Monosubstituted and (E)-Ester Conjugated Allylic Amines by Bulky Alkyl Groups

  • The $O_sO_4$-catalyzed dihydroxylations of a monosubstituted allylic amine and $\gamma-amino-\alpha,\;\beta-unsaturated$ (E)-esters with bulky alkyl groups showed a high anti-selectivity. Since the acyclic conformation of N-acyloxy protected allylic amines was efficiently controlled by a bulky t-Bu or OBO ester group, the anti diastereoselectivity of >12.5:1 was obtained without applying a chiral reagent. The synthetic utility of the present method was demonstrated by a stereoselective and efficient synthesis of an $\alpha$-glucosidase inhibitor 15 from commercially available N-Cbz-L-serine 6 in 11 steps and 31% yield.

솔비탄 메타크릴레이트의 효소적 합성 - 반응온도와 아실 공여체의 영향 - (Enzymatic Synthesis of Sorbitan Methacrylate Effect of Reaction Temoerature and Acyl Donor)

  • In this research, the chemo-enzymatic synthesis of sorbitan methacrylate was investigated to optimize reaction conditions. Firstly, sorbitan was manufactured by sorbitol cyclic reaction in the presence of p-toluenesulfonic acid (p-TSA) as catalyst material. Secondly, sorbitan methacrylate was synthesized by immobilized lipase Novozyme 435 with acyl donors in t-butanol. As a result of enzymatic synthesis of sorbitan methacrylate, the conversion yield reached about $65\%$ in the condition of initial sorbitan conc. 50 g/L, enzyme content $3\%$ (w/v) , molar ratio 1:3, reaction temperature 50^{circ}C and reaction time 42 hrs using methyl methacrylate as acyl donor. Comparing with acyl donors and reaction temperature, the conversion yield reached about 18, 65 and $80\%$ with methacrylic acid, methyl methacrylate and vinyl methacrylate as acyl donor, respectively. And optimum reaction temperature was 60, 50, and 50^{circ}C, respectively

  • Human blood consists of 55% of plasma and 45% of blood cells such as white blood cell (WBC) and red blood cell (RBC). In plasma, there are many kinds of promising biomarkers, which can be used for the diagnosis of various diseases and biological analysis. For diagnostic tools such as a lab-on-a-chip (LOC), blood plasma separation is a fundamental step for accomplishing a high performance in the detection of a disease. Highly efficient separators can increase the sensitivity and selectivity of biosensors and reduce diagnostic time. In order to achieve a higher yield in blood plasma separation, we propose a novel fluid wing structure that is optimized by COMSOL simulations by varying the fluidic channel width and the angle of the bifurcation. The fluid wing structure is inspired by the inertial particle separator system in helicopters where sand particles are prevented from following the air flow to an engine. The structure is ameliorated in order to satisfy biological and fluidic requirements at the micro scale to achieve high plasma yield and separation efficiency. In this study, we fabricated the fluid wing structure for the efficient microfluidic blood plasma separation. The high plasma yield of 67% is achieved with a channel width of $20{\mu}m$ in the fabricated fluidic chip and the result was not affected by the angle of the bifurcation.

The Use of Cassava Chips as an Energy Source for Lactating Dairy Cows Fed with Rice Straw

  • Thirty-six crossbred (70% Holstein Friesian) cows in mid-lactation were assigned to one of four treatments. The dietary treatments were concentrate based, containing 13.5, 27.0, 40.5 and 54.0% of cassava in concentrate replacing ground maize (16.0% CP). There were curvilinear responses to intake of organic matter, non-structural carbohydrate and metabolisable energy. Cassava and corn fed in a ratio of 50:50 maximised organic matter, metabolisable energy intake; milk yield, milk protein and lactose yield. Milk fat yield was not affected by levels of inclusion. Dietary treatment did not influence ruminal pH, ammonia and volatile fatty acid concentrations or plasma glucose. The low market price for cassava resulted in a lower concentrate feed cost. The optimal level of cassava in a dairy cow diet is suggested as being between 20.0 and 30.0% of cassava in dry matter intake when fed with rice straw.

홍삼 추출물의 제조에서 단백질 분해효소의 활용 (Potential of proteolytic enzyme treatment for production of Korean red ginseng extract)

  • 본 연구에서는 홍삼 추출물 제조과정에서 추출 수율을 향상시킬 수 있는 효소를 선별하고 최적의 반응조건을 조사하였다. 상업용 단백질 분해효소 중 Alcalase가 단백질과 탄수화물 수율 향상에 효과적이었으며, 효소 사용량은 홍삼 중량의 2%, 반응 시간은 1.5시간이 적당하였다. 최적의 반응조건으로 홍삼을 Alcalase로 처리한 결과 대조구보다 고형분 수율은 45.1%에서 71.1%로 50% 이상, 총페놀 함량은 0.44%에서 0.80%로 80% 이상 증가하였으며, 항산화 활성은 대조군과 매우 유사하였다. 또한 진세노사이드 함량은 대조구의 1.48 mg/g에서 1.98 mg/g으로 30% 이상 증가하였다.

Biological Control of Alternaria Fruit Rot of Chili by Trichoderma Species under Field Conditions

  • Trichoderma strains were evaluated under field conditions to assay their efficacy in suppressing Alternaria fruit rot disease and promoting chili plant growth. The experiment was conducted at the Botanical Garden, Rajshahi University, Bangladesh from July 2006 to March 2007. Application of Trichoderma harzianum IMI 392432 significantly (p = 0.05) suppressed the disease compared to Alternaria tenuis (T2) treatment and improved both growth and yield. The treatment T4 (T. harzianum IMI-392432 + A. tenuis) was most effective in reducing disease percentage (72.27%) compared to A. tenuis (T1) treatment. The highest seed germination rate (85.56%) and the highest growth and yield (12.5 g/plant) was also recorded in the same treatment (T4), followed by T5 (T. harzianum IMI-392433 + A. tenuis), T6 (T. harzianum IMI-392434 + A. tenuis), T2 (T. virens IMI-392430 + A. tenuis), and T3 (T. pseudokoningii IMI-392431 + A. tenuis) treatment, while single treatment with A. tenuis significantly decreased these values.

  • 수도재배(水稻栽培)에 있어서의 퇴비(堆肥) 및 인산(燐酸)의 시용방법(施用方法)을 확립(確立)하기 위(爲)해서 포장시험(圃場試驗)과 시험전후(試驗前後)의 각구표토(各區表土)의 유기물함량(有機物含量)을 조사(調査)한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 퇴비(堆肥) 및 인산(燐酸)은 전량기비(全量基肥)로 하는것보다는 기비(基肥)와 유수형성기(幼穗形成期) 2주전(週前) 2회(回)에 분시(分施)하는 것이 정조수량(精粗收量)을 증대(增大)하는 효과(?果)가 컸으며 유기물(有機物)이 토양중(土壤中)에서 소모(消耗)되는 양(量)이 적을 수록 수량(收量)이 많아지는 경향(傾向)이 뚜렷했다. 2. 퇴비(堆肥)와 인산(燐酸)을 별도(別途)로 시용(施用)하는것 보다는 혼합시용(混合施用)하는 것이 정립수량증대면(精粒收量增大面)에서 좋은 경향(傾向)을 보였다. 3. 유기물(有機物)이 분시회수(分施回數)를 증대(增大)시키면 더욱 증수(增收)할수 있을 것이라고 추정(推定)되었다.

Effect of Nitrogen and Sulphur Application on Nitrate Reductase and ATP-sulphurylase Activities in Soybean

  • A number of field experiments were conducted to assess the role of combined application of nitrogen and sulfur to increase the seed and oil yield of nonnodulating soybean (Glycine max (L) Merr.) cv. PK-416 $(V_1)$ and cv. PK-1024 $(V_2)$. Six combinations of N and S in three replicates each were used for this purpose i.e. $0\;S+23.5kg\;N\;ha^{-1}(T_1);\;0\;S+23.5+20kg\;N \;ha^{-1}(T_2);\;40\;S+23.5kg\;N\;ha^{-1}(T_3);\;40\;S+23.5+20kg\;N\;ha^{-1}(T_4);\;20+20\;S+23.5kg\;N\;ha^{-1}(T_5);\;20+20\;S+23.5+20kg\;N\;ha^l(T_6)$. Nitrate reductase (NR) and ATP-sulphurylase activities in the leaves were measured at various growth stages as the two enzymes catalyze the rate limiting steps of the assimilatory pathways of nitrate and sulphate, respectively. The activities of these enzymes were strongly correlated with seed yield. The higher seed, oil and protein yields were achieved with the treatment $T_6$ in both the cultivars due to optimization of NR activity and ATP-sulphurylase activity, as these parameters were influenced by N and S assimilation. Any variation from this combination was observed to decrease the activity of these enzymes resulting in reductions in the seed, oil and protein yield of soybean.

Improvement of Biocontrol of Damping-off and Root Rot/Wilt of Faba Bean by Salicylic Acid and Hydrogen Peroxide

  • Rhizoctonia solani, Fusarium solani, F. oxysporum, and Macrophomina phaseolina were found to be associated with root rott and wilt symptoms of faba bean plants collected from different fieldes in New Valley governorate, Egypt. All the obtained isolates were able to attack faba bean plants (cv. Giza 40) causing damping-off and root rot/wilt diseases. R. solani isolates 2 and 5, F. solani isolate 8, F. oxysporum isolate 12 and M. phaseolina isolate 14 were the more virulent ones in the pathogenicity tests. Biocontrol agents (Trichoderma viride and Bacillus megaterium) and chemical inducers (salicylic acid [SA] and hydrogen peroxide) individually or in combination were examined for biological control of damping-off and root rot/wilt and growth promoting of faba bean plants in vitro and in vivo. Both antagonistic biocontrol agents and chemical inducers either individually or in combination inhibited growth of the tested pathogenic fungi. Biocontrol agents combined with chemical inducers recorded the highest inhibited growth especially in case SA + T. viride and SA + B. megaterium. Under green house and field conditions, all treatments significantly reduced damping-off and root rot/wilt severity and increased of survival plants. Also, these treatments increased fresh and weights of the survival plants in pots compared with control. The combination between biocontrol agents and chemical inducers were more effective than used of them individually and SA + T. viride was the best treatment in this respect. Also, under field conditions, all these treatments significantly increased growth parameters (plant height and number of branches per plant) and yield components (number of pods per plant and number of seeds per plant, weight of 100 seeds and total yield per feddan) and protein content in both seasons (2010~2011 and 2011~2012). Faba bean seeds soaked in SA + T. viride and SA + B. megaterium were recorded the highest growth parameters and yield components. Generally, the combination between biocontrol agents and chemical inducers recorded the best results for controlling damping-off and root rot/wilt diseases in greenhouse and field with addition improved plant growth and increased yield components in field.

Disrupting Escherichia coli: A Comparison of Methods

  • The often-encountered problem of disrupting bacteria for the purpose of extracting soluble protein has generated various methods. Many require specialized equipment. Very often, especially during preliminary studies, investigators need a simple, fast, and inexpensive method for cell disruption that preserves biological activity. This paper compares some simple and inexpensive methods for cell disruption, such as bead-vortexing, freesing-thawing, French pressing, and sonication. It also provides some tips to increase protein yield and preserve biological activity. If performed under optimal conditions, bead-vortexing gives protein yields that are comparable to French pressing and sonication. It also preserves the activities of labile enzymes and releases periplasmic enzymes. Vortexing with glass beads appears to be the simplest method for cell disruption.
