Nearinfra-red(NIR) reflectance NIR a spectra (400 to 2,100 nm) were collected on 32 beef samples to find feasibility of predicting beef tenderness. The study to predict beef tenderness was accomplished with the stepwise second differential data of the collected NIR spectra. Beef tenderness was measured by Warner-Bratzler(WB) shear force using a Universal Testing Machine(UTM). After modeling the relation between Warner-Bratzler shear force and NIR spectrum of 19 samples among the 32 beef samples, the verification was carried out through predicting the other 13 samples. The SEC and R$^2$ values in the prediction equation were 9.07(N) and 0.6463, respectively. The SEP and R$^2$ were 14.8(N) and 0.7082 (wave length 552 nm, 1988 nm) respectively. The result implied that it was possible to predict the beef tenderness using NIR spectrum and that the tenderness could be predicted non-destructively in real time.
Polysaccharides have attracted great attention for their wide range of applications in biological and medical fields. In this paper, the interaction of polysaccharides with human serum albumin (HSA) was systematically investigated by fluorescence (FL) spectroscopy and circular dichroism (CD) spectra under different conditions. The Stern-Volmer quenching constants ($K_a$) at different ionic strength and pH were calculated, and information of the structural features of HSA was discussed. FL and CD results indicate that both hydrophobic and electrostatic interactions play important roles during the binding process. The quenching of the fluorescence resulting the binding of polysaccharides and HSA is static.
In this study, 52 ureolytic bacterial strains were newly isolated from various environments. From these, 2 strains (TB-15 and TB-22) were selected based on their high urease activity. XRD spectra clearly showed presence of various sequestration products such as calcite and strontianite in samples. TB-22 showed 20~30% higher survivability upon Sr concentration (20 mM) than Sporosarcina pasteurii KCTC 3558. TB-15 and TB-22 showed 80~90% higher survivability at pH 6 than S. pasteurii. The results demonstrated that the 2 isolates colud be good candidates for the bioremediation of Sr contaminated sites.
To apply TMAH-based Py-MS to a field biological detection system for real-time classification of cell-type, reproducible patterns of the TMAH-based Py-MS spectra was known as a critical factor for classification but was seriously disturbed by quantity of cells injected into pyro-tube. This factor is an exterior variable that could not be complemented by improving the performance of the TMAH-based Py-MS instrument. One of idea to solve the knotty problem has been flashed from "Top-down proteomics for identification of intact microoganisms". That is, biomarker peaks are selected from complicate Py-MS spectra for intact microoganisms by tracing out their origins, based on Py-MS spectra for the featured components of different cell-types, in Top-down approach. This idea has been tested in classification of different Gram-type microoganisms. Through the analyses of spectra for the featured components - peptidoglycan and lipoteichoic acid for Gram-positive cells and lipopolysaccharide and lipid A for Gram-negative cells - with comparing to the spectra the corresponding Gram-type cells in the Top-down approach, biomarker peaks were selected to carry out PCA(Principal Component Analysis) in order to see classification of different Gram-types, resulting in significant improvement of their classification. Furthermore, weighting biomarker peaks on intact cell's spectra, based on the data for the featured components of the Gram-types, contributed to elevate classification performance.
The spectra of whole blood EDTA samples from two people were generated using a CARY 5E (UV-VIS-NIR) spectrophotometer from 400 to 1000nm which contain visible and NIR region. Only the data between 400 and 800nm were used to analyze the components of blood. Using the same spectrophotometer, the spectra of Water, normal saline, plasma were generated These spectra were subtracted from each blood sample, and then the first derivative of each of the subtracted data was taken to minimize baseline variations and indicated the wavelength-shift of peak and valley. Normalization and division between two blood samples were used to correlate the quantity ratio of specific components with feature of spectra. Samples were controlled at $30^{\circ}C,\;37^{\circ}C$, ambient temperature.
Pyrolysis-mass spectrometry, incorporating an in situ thermal hydrolysis and methylation(THM) step, has been used to study biological materials for bacteria, toxin and virus. Newly developed pyrolyzer was used to decompose biological materials, and tetramethylammonium hydroxide(TMAH) was used as a methylation reagent. Chemical ionization(CI) using ethanol and ion trap mass spectrometer(ITMS) were used to ionize and analyze of pyrolysis components, respectively. Analytical characteristics of bacteria (including spore), virus and toxin were analyzed. Also acquisition and interpretation of mass spectra as biomarkers for classification/identification of biological material s were explained.
Five secondary metabolites, including a new isopimarane derivative xylaroisopimaranin A (1), were isolated from the endophytic fungus Xylaralyce sp. (HM-1), and their structures were elucidated by 1D, 2D NMR, MS and CD spectra. Their bioactivities were performed to antibacterial, Hep G2 cells cytotoxicity and brine shrimp inhibition. The biological evaluation results showed that the xylaroisopimaranin A (1), xylabisboein B (2), griseofulvin (3), 5-methylmellein (4) and mellein-5-carboxlic acid (5) displayed no significant Hep G2 cells cytotoxicity and antibacterial acitivity, but they inhibited the brine shrimp with $IC_{50}$ from 0.5 to $25{\mu}mol/mL$.
Genipin, aglycone of geniposide isolated from fruits of Gardenia jasminoides, was transformed into blue pigments through reaction with glycine and methylamine. The blue pigments formed from glycine-reacted genipin were passed through Bio-Gel P-2 resin yielding fractions GG1 and GG2, and those from methylamine-reacted genipin were separated into fractions GM1-GM4. The first eluted higher molecular-weight fractions, GG1 and GM1, had higher tinctorial strength than the later eluted lower molecular-weight fractions, GG2 and GM2-GM4, respectively. $^1H-NMR$ spectra of GG1 and GM1 showed very broad peaks indicating that structures of the pigments were highly polymeric. $^1H-NMR$ spectra of GG2, GM3, and GM4 showed several sharp peaks at aliphatic and aromatic regions with accompanying broad peaks, although the spectrum of GM2 was rather simple. Determination of the structural and physical nature of the isolated pigments is in progress.
Objectives : Though a proportion of Alzheimer's disease(AD) patients treated with donepezil have shown positive response on cognition, but the responders' characteristics are still uncertain. This study attempts to identify whether a single oral dose of donepezil(5mg) can change cognition and the relationship between single dose responder items and long-term responder are examined. Methods : Twenty-three AD patients for single donepezil challenge study group and eleven AD patients for controls were participated in the study. Seven days after baseline study for neuropsychological test and EEG, same studies were rechecked after donepezil medication in study group. In donepezil study groups, 12 weeks after donepezil medication, neuropsychological test and EEG were rechecked. Results : After single donepezil challenge, forward digit span, Rey-Osterrieth Complex Figure Test copy, SVLT delayed recall were significantly improved, and beta spectra power in anterior, theta spectra power in posterior field were significantly decreased. According to linear regression analysis, forward digit span after single donepezil challenge was significantly positive correlated with long-term responders. Conclusions : This study suggests that single donepezil medication can significantly change cognitive functions and EEG in AD patients. Among these responsive items, forward digit span was significantly correlated with long-term responder.
우리나라 재배종(栽培種) Santa rosa자두 중(中)에 함유(含有)된 두가지 antocyanin 색소(色素)를 분리확인(分離確認)하였다. 자두로부터 0.5% HCl methanol로 색소(色素)를 추출(抽出)하여 Amberlite IRC-50 이온 교환수지(交換樹脂) column 으로 정제(精製)하고, paper chromatography에 의하여 개별색소(個別色素)를 분리(分離)하였다. 두 색소(色素)는 다음과같은 실험(實驗)을 거쳐 얻은 결과(結果)들에 기초(基礎)하여 cyanidin-3-xyloglucoside 및 cyaniden-3-monoglucoside 임을 확인(確認)하였다. 여러 용매계(溶媒系)에 의한 paper chromatography의 Rf치(値), 부분산가수분해(部分酸加水分解), 결합당(結合糖)의 paper chromatography에 의한 확인(確認), 결합당(結合糖)의 단계가수분해(段階加水分解), aglycone의 alkali분해(分解), 및 가시부(可視部) 흡수(吸收) spectra. 자두중(中)에 함유(含有)된 주색소(主色素)는 cyanidin-3-monoglucoside였다.
