A Technique for the Prevention of Greenhouse Whitefly (Trialeurodes vaporariorum) Using the Entomopathogenic Fungus Beauveria bassiana M130

  • The possibility of using hyphomycete fungi as suitable biocontrol agents against greenhouse whitefly has led to the isolation of various insect pathogenic fungi. Among them is Beauveria bassiana, one of the most studied entomopathogenic fungi. The objective of this study was to use B. bassiana M130 as an insecticidal agent against the greenhouse whitefly. M130 isolated from infected insects is known to be a biocontrol agent against greenhouse whitefly. Phylogenetic classification of M130 was determined according to its morphological features and 18S rRNA sequence analysis. M130 was identified as B. bassiana M130 and showed chitinase (342.28 units/ml) and protease (461.70 units/ml) activities, which were involved in the invasion of the host through the outer cuticle layer, thus killing them. The insecticidal activity was 55.2% in petri-dish test, 84.6% in pot test, and 45.3% in field test. The results of this study indicate that B. bassiana has potential as a biological agent for the control of greenhouse whitefly to replace chemical pesticides.

  • Emodin (3-methyl-1,6,8-trihydroxyanthraquinone) is a natural chemotherapeutic compound with diverse biological properties including an antitumor activity. Emodin, a specific inhibitor of the protein tyrosine kinase, has a number of cellular targets in related to it. Its inhibition activity affects the mammalian cell cycle regulation in specific oncogene. Practically, it has been proven to inhibit HER-2/neu tyrosine kinase expressing breast cancer cells as an anticancer agent. 2-[123I]iodoemodin has been synthesized and evaluated human breast cancer cells (MDA-MB-231, MCF-7, fibroblast as a control) which express basal levels of HER-2/neu tyrosine kinase to investigate its suitability as a breast cancer imaging agent and 2-iodoemodin has been synthesized as a standard compound. The radiochemical yield of the 2-[123I]iodoemodin was about 72% and its radiochemical purity was over 97% after purification. The radioactivity of the 2-[123I]iodoemodin was increased in a time dependent manner in both cell lines and the ratio of MDA-MB-231 and MCF7 to fibroblast was 2.9 and 1.7, respectively.

  • A genus, Eocanthecona Bergroth (Hemiptera: Heteroptera: Pentatomidae: Asopinae), is reported from the Korean Peninsula for the first time based on the species of Eocanthecona japanicola (Esaki & Ishihara, 1950). The morphological information such as description and diagnosis, and biological information are provided.

Investigations on Bacteria as a Potential Biological Control Agent of Summer Chafer, Amphimallon solstitiale L. (Coleoptera: Scarabaeidae)

  • Studying the bacteria of hazardous insects allows the opportunity to find potentially better biological control agents. Therefore, in this study, bacteria from summer chafer (Amphimallon solstitiale L., Coleoptera: Scarabaeidae) we isolated and identified the insecticidal effects of bacteria isolated from A. solstitiale and Melolontha melolontha L. (common cockchafer, Coleoptera: Scarabaeidae) and the mixtures of these bacterial isolates were investigated on A. solstitiale larvae. Crystals from Bacillus sp. isolated from M. melolontha were also purified, and tested against the second and third-stage larvae of A. solstitiale. The bacterial isolates of A. solstitiale were identified as Pseudomonas sp., Pseudomonas sp., Bacillus cereus and Micrococcus luteus, based on their morphology, spore formation, nutritional features, and physiological and biochemical characteristics. The insecticidal effects of the bacterial isolates determined on the larvae of A. solstitiale were $90\%$ with B. cereus isolated from A. solstitiale, and $75\%$ with B. cereus, B. sphaericus and B. thuringiensis isolated from M. melolontha within ten days. The highest insecticidal effects of the mixed infections on the larvae of A. solstitiale were $100\%$ both with B. cereus+B. sphaericus and with B. cereus+B. thuringiensis. In the crystal protein bioassays, the highest insecticidal effect was $65\%$ with crystals of B. thuringiensis and B. sphaericus isolated from M. melolontha within seven days. Finally, our results showed that the mixed infections could be utilized as microbial control agents, as they have a $100\%$ insecticidal effect on the larvae of A. solstitiale.

A Study on the Fermentation Characteristics of Garbages by the C/N Ratio Control using Kudzu Creeper and Sawdust (칡넝쿨 및 톱밥을 이용한 C/N비 조절에 따른 음식물찌꺼기의 발효특성에 관한 연구)

  • In this study, to determine the optimum fermentation process for the feed production of food wastes and estimate the practical value of fermented feed using kudzu creeper and sawdust as bulking agent. This study considered initial C/N ratio control as the fermentation process variables. The results are summarized as follows. Minimum water contents of byproducts in the fermentation feed production showed 3994(kudzu), 37%( sawdust) at the C/N ratio 25 and 45%(kudzu, sawdust) at the C/N ratio 35. Temperature variations in the fermentation feed production at the C/n ratio 25 indicated 68'C(kudzu), 70'C (sawdust).Optimum condition of consists of fermentation process of water content, C/N ratio and permeability (porous structure of the mixture). For optimum fermentation of gravitationally, dewatered garbage, the proper mixing ratios of kudzu(moisture contents : 17.3%) and sawdust(moisture contents : 13.2%) were 41% and 39%, respectively Major biological reaction in the aerobic fermentation feed production occurred during 12~14hrs

  • The biological control efficacy of a greenhouse soil bacterial mixture of Lactobacillus farraginis, Bacillus cereus, and Bacillus thuringiensis strains with antinematode activity was evaluated against the root-knot nematode Meloidogyne incognita. Two control groups planted in soil drenched with sterile distilled water or treated with the broad-spectrum carbamate pesticide carbofuran were used for comparison. The results suggest that the bacterial mixture is effective as a biocontrol agent against the root-knot nematode.

Fungichromin Production by Streptomyces padanus PMS-702 for Controlling Cucumber Downy Mildew

  • Streptomyces padanus PMS-702 strain produces a polyene macrolide antibiotic fungichromin and displays antagonistic activities against many phytopathogenic fungi. In the present study, experimental formulations were assessed to improve the production of fungichromin, the efficacy of PMS-702 on the suppression of sporangial germination, and the reduction of cucumber downy mildew caused by Pseudoperonospora cubensis. PMS-702 strain cultured in a soybean meal-glucose (SMG) medium led to low levels of fungichromin accumulation and sporangial germination suppression. Increasing medium compositions and adding plant oils (noticeably coconut oil) in SMG significantly increased fungichromin production from 68 to $1,999.6{\mu}g/ml$. Microscopic examination reveals that the resultant suspensions significantly reduced sporangial germination and caused cytoplasmic aggregation. Greenhouse trials reveal that the application of PMS-702 cultural suspensions reduced downy mildew severity considerably. The addition of Tween 80 into the synthetic medium while culturing PMS-702 further increased the suppressive efficacy of downy mildew severity, particularly when applied at 24 h before inoculation or co-applied with inoculum. Fungichromin at $50{\mu}g/ml$ induced phytotoxicity showing minor necrosis surrounded with light yellowish halos on cucumber leaves. The concentration that leads to 90% inhibition (IC90) of sporangial germination was estimated to be around $10{\mu}g/ml$. The results provide a strong possibility of using the S. padanus PMS-702 strain as a biocontrol agent to control other plant pathogens.

Development Feasibility of Water-Floating Fungicide Formulation for the Control of Sheath Blight(Caused by Rhizotonia solani) in Rice (수도(水稻) 문고병방제용(紋枯病防除用) 수면부유제(水面浮遊劑)의 개발(開發) 가능성(可能性)에 관(關)한 연구(硏究))

  • Some physico-chemical properties and biological activities of new fungicide formulations were tested to investigate a feasibility of water-floating formulation development for sheath blight control in rice. Pencycuron[1-(4-chlorobenzyl)-1-cyclopentyl-3-phenylurea] and flutolanil$({\alpha},{\alpha},{\alpha}$-trifluoro-3'-isopropoxy-O-toluanilide) were chosen as toxicants for the formulations. Vegetable oil and surface active agents were used as a floating agent and spreader, respectively. All the formulations tested showed an excellent spreadability on the water having over $35cm^2/mg$ and were chemically stable, which the degradation rates of active ingredients were less than 10% after 12 weeks of storage at $50^{\circ}C$. Most of the applied test formulation of pencycuron was retained within 0.5cm of the surface paddy water, while that of flutolanil was vertically dispersed in the water. Inhibition activity of the tested pencycuron formulation on the sclerotia germination of the pathogen in paddy water was maintained over 30 days after the formulation treatment. Control effect of 4% pencycuron water-floating formulated with surface active agent of hydro-lipophyllic balance 4.3 on the disease in rice was equal to the reference fungicide(pencycuron 25% WP) when the former was treated one day before the transplantation of rice seedlings. Overflowing the submerged paddy water after the formulation treatment resulted in a deterious effect on the disease control.

  • The black cutworm, Agrotis ipsilon (Hufnagel) is a major insect pest of economic crops including turfgrasses on golf courses. The entomopathogenic nematode, Steinernema carpocapsae GSN1 strain (ScG), a Korean isolate, is an effective biological control agent for soil dwelling and greenhouse insect pests in Korea. In addition, ScG is commercially produced in Korea. We conducted laboratory, greenhouse, and field trials to evaluate efficacy of ScG against black cutworms in turfgrasses. A rate of 63 infective juveniles (Ijs) per larva killed >90% of $3^{rd}$ instars feeding in cups of artificial diet within 3 days. In greenhouse trials against cutworms feeding in pots of turfgrass, efficacy of ScG was higher against $4^{th}$ instars than against $2^{nd}$ instars (90.0 vs 81.2% mortality, respectively, at $2,000Ijs\;pot^{-1}$) in perennial ryegrass, and higher against $3^{rd}$ instars in creeping bentgrass, Agrostis palustris than in zoysiagrass, Zoysia japonica (96.7 vs 52.5% mortality at $100,000Ijs\;m^{-2}$) in pot. The corrected mortality of $4^{th}$ instar was 79.9% at the rate of $100,000Ijs\;m^{-2}$ in the creeping bentgrass in the field. So ScG could be used as biological control agent against black cutworm in turfgrass of golf courses.

  • Mollugin has been reported to have various biological activities including antineoplastic, antitumor, antiviral against the hepatitis B virus, anti-aging and antimutagenic activities. An effective and concise synthesis of mollugin in two steps including kinetic control from the cheap starting material 1,4-naphthoquinone has been introduced, and mollugin derivatives thus prepared are screened for their inhibition ability against the hepatitis C virus (HCV) and the dihydrobenzochromene structure might be an additional anti-HCV agent as a new leading compound.
