상급종합병원 병동 간호사의 환자안전 문화인식, 잡 크래프팅, 간호순회인식이 환자안전 관리활동에 미치는 영향 (Associations of perceptions of patient safety culture, job crafting, and perceptions of patient rounding with patient safety management activities among tertiary hospital nurses)

  • Purpose: This study investigated patient safety management activities among general hospital nurses and aimed to identify the factors influencing patient safety management activities. Methods: A cross-sectional descriptive design included 159 ward nurses working at two tertiary hospitals located in Busan city. Data were collected with self-reported questionnaires from April 8 to April 20, 2024 and were analyzed using descriptive statistics, the t-test, analysis of variance, Pearson correlation coefficients, and multiple regression in SPSS 29.0. Results: Factors significantly influencing patient safety management activities in nurses were perceptions of patient safety culture (β = .47, p < .001) and perceptions of patient rounding (β = .31, p < .001). The explanatory power of these variables was 43%. Conclusion: This study found that perceptions of patient safety culture and perceptions of patient rounding had significant effects on nurses' patient safety management activities. Thus, it is necessary to promote patient safety management activities among nurses through the development of intervention programs to improve perceptions of patient safety culture and perceptions of patient rounding.

Chloronicotinyl 유도체의 합성 및 생물활성 검정 (Synthesis and biological activities of Chloronicotinyl derivatives)

  • Chloronicotinyl 유도체의 기본 골격인 3-pyridylmethylamine의 amine기를 phosphite group으로 치환하여 chloronicotinyl 유도체를 합성하였고, 이들의 살충활성과 살균활성을 확인하였다. 살충활성 검정 결과, 500 ppm 에서 phosphonate에 methyl기와 butyl기가 치환된 compound 4와 2개의 butyl, 2,2,2-trifluorotehtyl, 2-ethylhexyl, phenyl기가 치환된 compound 5, 6, 7, 8이 벼멸구에 대해서 90%의 살충활성을 나타내었다. 그러나 이들 화합물들은 배추좀나방과 두점박이응애에 대해서는 낮은 살충활성을 나타내었으며, phosphonate에 halogen이나 heterocycle이 치환된 compound $10{\sim}21$에서도 벼멸구, 배추좀나방, 담배거세미나방, 두점박이응애, 복숭아혹진딧물에 대하여 거의 살충활성을 나타내지 않았다. Chloronicotinyl 유도체의 벼도열병균, 벼문고병균, 오이잿빛 곰팡이병균, 토마토역병균, 밀녹병균, 보리흰가루병균에 대한 살균활성을 검정한 결과, 10 ppm에서 phosphonate에 butyl기와 4-nitrophenyl기가 치환된 compound 10이 벼도열병균에 대해 86%의 살균활성을 보였다. 인 함유 chloronicotinyl 유도체들 중 일부가 높은 살균활성을 나타낸다는 것은 이 유도체들이 신규 살균제로 개발될 가능성이 있다는 것을 시사하고 있다.

Evaluation of Luminescent P450 Analysis for Directed Evolution of Human CYP4A11

  • Cytochrome P450 4A11 (CYP4A11) is a fatty acid hydroxylase enzyme expressed in human liver. It catalyzes not only the hydroxylation of saturated and unsaturated fatty acids, but the conversion of arachidonic acid to 20-hydroxyeicosatetraenoic acid (20-HETE), a regulator of blood pressure. In this study, we performed a directed evolution analysis of CYP4A11 using the luminogenic assay system. A random mutant library of CYP4A11, in which mutations were made throughout the entire coding region, was screened with luciferase activity to detect the demethylation of luciferin-4A (2-[6-methoxyquinolin-2-yl]-4,5-dihydrothiazole-4-carboxylic acid) of CYP4A11 mutants in Escherichia coli. Consecutive rounds of random mutagenesis and screening yielded three improved CYP4A11 mutants, CP2600 (A24T/T263A), CP2601 (T263A), and CP2616 (A24T/T263A/V430E) with ~3-fold increase in whole cells and >10-fold increase in purified proteins on the luminescence assay. However, the steady state kinetic analysis for lauric acid hydroxylation showed the significant reductions in enzymatic activities in all three mutants. A mutant, CP2600, showed a 51% decrease in catalytic efficiency ($k_{cat}/K_m$) for lauric acid hydroxylation mainly due to an increase in $K_m$. CP2601 and CP2616 showed much greater reductions (>75%) in the catalytic efficiency due to both a decrease in $k_{cat}$ and an increase in Km. These decreased catalytic activities of CP2601 and CP2616 can be partially attributed to the changes in substrate affinities. These results suggest that the enzymatic activities of CYP4A11 mutants selected from directed evolution using a luminogenic P450 substrate may not demonstrate a direct correlation with the hydroxylation activities of lauric acid.

아로니아, 블랙커런트, 마키베리의 기능성 성분 및 생리활성 비교 (Comparison of Bioactive Constituents and Biological Activities of Aronia, Blackcurrant, and Maquiberry)

  • 본 연구에서는 아로니아, 블랙커런트, 마키베리의 생리활성을 비교하기 위하여 총폴리페놀, 총플라보노이드, 총안토시아닌 함량, DPPH radical 소거능, ABTS radical 소거능, FRAP 활성, 환원력, tyrosinase 저해 효과, 아질산염 소거능 등을 측정하였다. 총폴리페놀 함량은 마키베리가 73.66 mg GAE/g으로 가장 높게 나타났고 그다음으로 아로니아 59.26 mg GAE/g, 블랙커런트 43.70 mg GAE/g 순이었다. 총플라보노이드 함량도 같은 경향을 나타내어 마키베리 28.08 mg/g, 아로니아 24.26 mg/g, 블랙커런트 16.82 mg/g 순으로 나타났으며, 총안토시아닌 함량은 마키베리가 12.82 mg/g으로 가장 높았고, 아로니아와 블랙커런트가 각각 9.52 mg/g과 8.95 mg/g으로 유의적인 차이를 보이지 않았다. 마키베리는 DPPH 및 ABTS radical 소거능, FRAP, 환원력에서 유의적으로 높은 활성을 나타내었고, 시료 농도 5 mg/mL에서는 양성대조군으로 사용한 ascorbic acid보다 우수한 효과를 보여주었다. 그다음으로는 아로니아가 우수한 활성을 보였으며 블랙커런트는 대부분의 기능성 실험 결과 가장 낮은 활성을 나타내었다. 이상의 결과를 종합하여 볼 때 마키베리는 천연 항산화제 및 건강 기능성 식품재료로서의 활용 가능성이 매우 높을 것으로 생각된다.

Discovery, semisynthesis, biological activities, and metabolism of ocotillol-type saponins

  • Ocotillol-type saponins are one kind of tetracyclic triterpenoids, sharing a tetrahydrofuran ring. Natural ocotillol-type saponins have been discovered in Panax quinquefolius L., Panax japonicus, Hana mina, and Vietnamese ginseng. In recent years, the semisynthesis of 20(S/R)-ocotillol-type saponins has been reported. The biological activities of ocotillol-type saponins include neuroprotective effect, antimyocardial ischemia, antiinflammatory, antibacterial, and antitumor activities. Owing to their chemical structure, pharmacological actions, and the stereoselective activity on antimyocardial ischemia, ocotillol-type saponins are subjected to extensive consideration. In this review, we sum up the discovery, semisynthesis, biological activities, and metabolism of ocotillol-type saponins.

녹비식물 혼합재배에 휴믹물질 투입이 정화처리토양의 생물학적 토양 건강성 지표에 미치는 영향 (Effects of Mixed Planting of Green Manure Crops Supplemented with Humic Substance on the Biological Soil Health Indicators of Reclaimed Soils)

  • The effects of green manure crops, hairy vetch and sesban, supplemented with HS (humic substance) on biological soil health indicators was studied in a pot containing two kinds of reclaimed soil previously contaminated with petroleum hydrocarbons; a soil remediated by land-farming (DDC) and another soil by low-temperature thermal desorption (YJ). Treatments include no plant (C), plants only (H), and plants+2% HS (PH), which were evaluated in a pot containing respective soil. Biological indicators include microbial community analysis as well as soil enzyme activities of dehydrogenase, 𝛽-glucosidase, N-acetyl-𝛽-D-glucosaminidase (NAG), acid/alkaline phosphatase, arylsulfatase, and urease. Results showed an increase of enzyme activities in pot soils with plants and even greater in soils with plants+HS. The enzyme activities of DDC soil with plants (DDC_P) and with plants+HS (DDC_PH) increased 1.6 and 3.9 times on average, respectively than those in the control. The enzyme activities YJ soil with plants (YJ_P) and with plant+HS (YJ_PH) increased 1.8 and 3.8 times on average, respectively than those in the control. According to microbial community analysis, the relative abundance of nitrogen-fixing bacteria in DDC and YJ soil was increased from 1.5% to 7% and from 0 to 5%, respectively, after planting hairy vetch and sesban. This study showed that mixed planting of green manure crops with a supplement of humic substance is highly effective for the restoration of biological health indicators of reclaimed soils.

손바닥 선인장 (Opuntia humifusa) 줄기 추출물의 생리활성 (A Study on Biological Activities of Opuntia humifusa Cladode Extracts)

  • 손바닥 선인장(Opuntia humifusa) 줄기 열수와 75% 에탄올 추출물의 일부 생리활성을 조사한 결과는 다음과 같다. DPPH 전자공여능은 열수 추출물에서 79.07%, 에탄올 추출물에서 82.54%를 보였다. 각 세포주에 대한 세포독성은 열수 추출물이 에탄올 추출물을 처리(100 ${\mu}g/well$)한 경우에서 보다 전반적으로 높게 나타났다. 열수 추출물은 HeLa와 AGS 세포주에서 50% 이상 세포 독성을 나타내었다. 항균 효과를 조사한 결과 그람 음성세균인 S. aureus KCCM 11593에 대해서는 물과 75% 에탄올 추출물에 있어서 항균효과를 나타내었으나 나머지 세균에 있어서는 항균 효과가 없었으며, 오히려 세포 생장효과가 있는 것으로 나타났다. 또한 NO 생성 저해 효과에 있어서는 물 추출물(34.31%)이 에탄올 추출물(25.59%)에서 보다 더 항염증효과가 높은 것으로 나타났다. 전구지방세포(3T3-L-1)의 분화에 미치는 영향에서는 처리 농도가 증가할수록 지방세포의 분화가 증가되어 물과 에탄올 추출물 모두 80과 100 ${\mu}g/mL$처리구 에서는 분화촉진제인 IBMX + dexamethasone의 처리구보다 다소 높은 분화정도를 보였다. 이와 같은 결과로 볼 때 천년초 줄기는 항염, 항균, 항산화, 항암효과에서 우수한 활성을 나타내어 앞으로 여러 가지 유용한 기능성 식품과 화장품 소재로의 개발을 기대할 수 있을 것으로 보인다.

Various Biological Activities of Ramie (Boehmeria nivea)

  • The purpose of this study was to evaluate the biological activities of extracts of ramie (Boehmeria nivea (L.) Gaud.), hereafter referred to as Bn. Bn extracts from various collecting area were extracted with methanol. Two extracts from our study, Bn-13 and -82, showed significant antioxidant properties, likely due to their ability to scavenge free radicals. In addition, Bn extracts showed stronger anti-bacterial activity against Escherichia coli (Bn-40), Stapylococcus aureus (Bn-33), and Helicobacter pylori (Bn-05). In addition, this study was conducted to evaluate the anti-inflammatory effects of Bn extracts in lipopolyssacharide (LPS)- and interferon-${\gamma}$ (IFN-${\gamma}$)-stimulated RAW 264.7 macrophages cells. Bn-37 significantly inhibited the production LPS/IFN-${\gamma}$-induced nitric oxide. The most noteworthy anti-cancer effect was found in Bn-23. Bn-08 showed inhibition of aldose reductase. This study provides basic information for the development of functional foods.

소청룡탕 효능에 관한 기초 실험 연구 문헌 분석 (Analysis of Biological Experiment on Socheongryong-tang (Xiaoqinglong-tang))

  • Objectives: To investigate the scientific evidence of Korean Medicine (KM), papers on Socheongryong-tang (Xiaoqinglong-tang), which is frequently used in medical clinics or hospitals of Korean medicine, were collected and analyzed. Methods: Papers were classified by the registration of domestic or international journals, the year of publication, experimental models and the subjects of biological activities. The mechanisms of biological activity in accordance with therapeutic effects of Socheongryong-tang (Xiaoqinglong-tang) were noted. Results: Among 98 papers included, 21 were published in domestic journals whereas 35 were in Chinese journals and 43 in Japanese journals. Most reported biological activities were amelioration of asthma. Socheongryong-tang (Xiaoqinglong-tang) regulated interleukin and interferon and immunoglobulin, inhibited the production of nerve growth factor, endotheliln-1, nitric oxide, toll-like receptor-4, p-Akt and increased extracellular signal regulated kinase and cyclin D1, which led to decreased bronchi inflammation and bronchoconstriction, and inhibited the proliferation of airway smooth muscle cells, mucus secretion and airway hyperresponsiveness. In addition, Socheongryong-tang (Xiaoqinglong-tang) also restored tissues injured by asthma so that respiratory function recovered. Conclusions: Amelioration of asthma by Socheongryong-tang (Xiaoqinglong-tang) is supported by objective and scientific evidence.
