  • The scaling relationship of the longest relaxation time of a single chain of semiflexible biological polyelectrolyte has been investigated by performing well-established coarse-grained Brownian dynamics simulations. Two kinds of longest relaxation times were estimated from time-sequences of chain trajectories, and their behaviors were interpreted by applying the scaling law for different molecular weights of polyelectrolyte and Debye lengths. The scaling exponents for longest stress relaxation and rotational relaxation are found in the ranges of 1.67-1.79 and 1.65-1.81, respectively, depending on the physicochemical interaction of electrostatic Debye screening. The scaling exponent increases with decreasing screening effect, which is a special feature of polyelectrolytes differing from neutral polymers. It revealed that the weak screening allows a polyelectrolyte chain to follow the behavior in good solvent due to the strong electrostatic repulsion between beads.

  • Continuous production of lactic acid from glucose by Lactobacillus rhamnosus with cell recycling using an acoustic cell settler was carried out. The performance of the system, such as the concentration of cell and product were compared with the control experiment without recycling. The acoustic settler showed cell separation efficiency of 67% during the continuous operation and the cell concentration in the fermentor with recycle exceeded that of the control by 29%. Com-pared with the control, tactic acid production was increased by 40%, while glucose consumption was only increased by 8%. The higher value of lactic acid production to substrate consumption (Yp/s, product yield coefficient) achieved by cell recycling is interpreted to indicate that the recycled cell mass consumes less substrate to produce the same amount of product than the control Within system environmental changes due to the longer mean cell residence time induced the cells maintaining the metabolic pathways to produce Less by-Product but more product, lactic acid.

  • In this study, the enzymatic process for biodiesel production was optimized using a mixture of immobilized Rhizopus oryzae and Candida rugosa lipases. The optimal temperature and agitation speed for biodiesel production were $45^{\circ}C$ and 300 rpm, respectively. The optimal ratio of R. oryzae and C. rugosa lipases in the mixture was 3:1 (w:w). When 3 mmol of methanol was the initial reaction medium and 3 mmol of methanol was added every 1.5 h during biodiesel production, biodiesel conversion was over 98% at 4 h. In addition, when the immobilized lipase mixture was reused, biodiesel conversion exceeded 80% after 5 reuses.

  • Public perceptions of nanotechnology and its potential risk can be an important measure for the sustainable advances of the technology. We conducted a survey for public (N =599) and experts (N = 165). They answered the same questions and results were analyzed. 74% of the public have not heard about the potential risk of nanotechnology and 77% expected that nanotechnology/materials are not harmful to the human and nature. 74% of experts realized the potential hazards by nanomaterials. The results represented that large perception gab between public and experts has existed in Korea. Interestingly, Korean public thought that nanotechnology is more profitable and less risky than that of American. We suggest that 'now is the best time to give appropriate information on the potential risk of nanotechnology to Korean public without preconception or exaggeration.'

  • Heavy metal contamination and biological toxicity of mine drainages and sediments in abandoned mines were studied. Mine drainages had pH at a range of 2.94~7.86, and contained heavy metals at a toxic level. For coal mines, toxicity of mine drainage to Daphnia magna was attributable to acidic pH. In addition to the low pH, suspended heavy metals such as Zn and Cu contributed to toxicity of mine drainages at abandoned metalliferous mines. All mine sediments studied in this work showed biological toxicity to Chironomus riparius, having mortality at a range of 15~60%. However, its relationship with physicochemical properties including heavy metal content of the sediments was not statistically explained. Exceptionally clay ($< 2{\mu}m$ particle) content was negatively correlated with the biological toxicity for sediment samples collected at the same abandoned mines.

  • In the previous paper (Bull. Korean Chem. Soc., 2011, 32, 2997), the hybrid molecules between ${\alpha}$-lipoic acid (ALA) and polyphenols (PPs) connected with neutral 2-(2-aminoethoxy)ethanol linker (linker-1) showed new biological activity such as butyrylcholinesterase (BuChE) inhibition. In order to increase the binding affinity of the hybrid compounds to cholinesterase (ChE), the neutral 2-(2-aminoethoxy)ethanol (linker 1) was switched to the cationic 2-(piperazin-1-yl)ethanol linker (linker 2). The $IC_{50}$ values of the linker-2 hybrid molecules for BuChE inhibition were lower than those of linker-1 hybrid molecules (except 9-2) and they also had the same great selectivity for BuChE over AChE (> 800 fold) as linker-1 hybrid molecules. ALA-acetyl caffeic acid (10-2, ALA-AcCA) was shown as an effective inhibitor of BuChE ($IC_{50}=0.44{\pm}0.24{\mu}M$). A kinetic study using 7-2 showed that it is the same mixed type inhibition as 7-1. Its inhibition constant (Ki) to BuChE is $4.3{\pm}0.09{\mu}M$.

  • The efficacy of integrated ozone oxidation-biodegradation treatment was examined in the treatment of petrochemical wastewater with a special focus on the overall treatment time. When raw wastewater with chemical oxygen demand(COD) of 70-80 mg/L was oxidized by ozone, approximately 20% of initial COD was removed in less than 1.5 min at a dosing rate of 400 mg $O_3/L{\cdot}h $. No further decrease in COD was observed for the extended ozone treatment up to 30 min. Biological treatment alone showed a rapid reduction of COD to 40-50 mg/L, subsequently resulting in the decreased rate of COD removal. Pre-treatment by ozone before biological treatment did not significantly affect the specific rate of COD removal in a biological treatment. When ozone oxidation followed biological treatment, the extent of COD removal by ozone oxidation was greater compared to that of biologically-treated wastewater for a shorter time. Taken together, it was decided that the biological treatment time could be reduced if the treatment processes of concern will be properly arranged.

  • The Herpes simplex virus type 1 (HSV-1) strain F glycoprotein H (gH) gene in the pHLB-4 plasmid was recombinated into a baculovirus expression vector (lacZ-HcNPV) to construct a recombinant virus GH-HcNPV expressing gH. The sequences of gH and its expression were analyzed. The gH gene was located in the 6.41 kb BglII fragment. The open reading frame (ORF) of the gH gene was 2,517 bp and codes 838 amino acid residues. Insect cells infected with this recombinant virus synthesized a high level of the matured and gX-gH fusion protein with approximately 112 kDa. The fusion gH protein was localized on the membrane of the insect cells as seen by using immunofluorescence assay and accumulated in the cultured media by the SDS-PAGE and immunoprecipitation assays. The amino acid sequence presents additional characteristics compatible with the structure of a viral glycoprotein: signal peptide, putative glycosylation sites and a long C-terminal transmembrane sequence. Antibodies raised in mice to this recombinant protein recognized viral gH and neutralized the infectivity of HSV-1 in vitro. These results demonstrate that it is possible to produce a mature protein by gene transfer in eukaryotic cells, and indicate the utility of the HcNPV-insect cell system for producing and characterizing eukaryotic proteins. Furthermore, the neutralizing antibodies would appear to protect mice against HSV; accordingly, this particular recombinant protein may be useful in the development of a subunit vaccine.

  • A variety of wireless devices are commercially available now. Most of studies, however, have been directed to the biological effects of mobile-phone EMF. In this study, dosimetric analysis for wireless devices of head-mounted display type and a wristwatch type were made to investigate possible biological effects of these devices. SAR (Specific Absorption Rate) distributions were calculated using FDTD (Finite Difference Time Domain) method, for adult human models such as standard Korean human model and VHP(Visible Human Project) model, as well as scaled models. Measurements were also performed for SAM phantom wearing a simplified prototype for a wireless device for validation of the simulation results. It has been found that children are more vulnerable to such exposure, and these devices could cause some biological effects for relatively lower power compared to conventional mobile pones.

  • The effect of silkworm hemolymph on the expression of recombinant protein in Escherichia coli was investigated. The addition of silkworm hemolymph to the culture medium increased the production of recombinant $\beta$-galactosidase in E. coli. The production was dependent on the concentration of the added silkworm hemolymph, which increased 2-, 5-, and 8-fold in media supplemented with 1, 3, and $5\%$ silkworm hemolymph, respectively. To identify the effective component, the silkworm hemolymph was fractionated by gel filtration column chromatography. A fraction, with a molecular weight of about 30 K was identified as the effective component.
