A Comprehensive Review of Diffusing Alpha-Emitters Radiation Therapy (DaRT): From Dosimetry to Its Biological Effectiveness

  • Diffusing alpha-emitters radiation therapy (DaRT) represents a groundbreaking development in cancer therapy, offering a solution to the limitations of conventional radiation therapy. By deploying 224Ra embedded seeds, DaRT achieves targeted delivery of high-dose alpha particles directly to tumor sites, showing considerable efficacy in tumor control and minimal damage to adjacent healthy tissues. This comprehensive review analyzes the published literature regarding mechanisms, seed production, dose calculation, measurement, and biological experiments related to DaRT. It includes in-depth discussions on mathematical models, Monte Carlo simulations for dose distribution, real-time in vivo dosimetry developments, and biological experiments both in vitro and in vivo. Clinical trial outcomes are also examined to evaluate the therapy's effectiveness in various cancer types. DaRT utilizes 224Ra-labeled seeds, using the decay chain of 224Ra to deliver alpha particles effectively within a tumor. Several asymptotic diffusion-leakage models were developed to calculate the alpha dose distribution of DaRT. In vivo dosimetry techniques have been developed for real-time monitoring. Biological experiments demonstrated the cytotoxic effects of DaRT across various cancer cells, with varying radiosensitivity. Additionally, the enhanced effects of combined therapy with chemotherapy and immunotherapy were suggested by many in vivo studies. Clinical trials have shown high complete response rate in squamous cell carcinoma, with minimal side effects, suggesting DaRT's feasibility and safety. DaRT emerges as a highly localized cancer treatment method with minimal side effects compared to traditional radiation therapy. It directly ablates tumors and potentially enhances immune responses, indicating a significant advance in cancer therapy. Future research and ongoing clinical trials will further elucidate its efficacy across different cancer types and in combination with other treatments.

Vortex behavior in the inertial flow of viscoelastic fluids past a confined cylinder

  • The effect of molecular parameters on the steady vortex behaviors in the inertial viscoelastic flow past a cylinder has been investigated. FENE-CR model was considered as a constitutive equation. A recently developed iterative solution method (Kim et al., (in press)) was found to be successfully applicable to the computation of inertial viscoelastic flows. The high-resolution computations were carried out to understand the detailed flow behaviors based on the efficient iterative solution method armed with ILU(0) type pre-conditioner and BiCGSTAB method. The discrete elastic viscous split stress-G/streamline upwind Petrov Galerkin (DEVSS-G/SUPG) formulation was adopted as a stabilization method. The vortex size decreased as elasticity increases. However, the vortex enhancement was also observed in the case of large extensibility, which means that the vortex behavior is strongly dependent upon the material parameters. The longitudinal gradient of normal stress was found to retard the formation of vortex, whereas the extensional viscosity played a role in the vortex enhancement. The present results are expected to be helpful for understanding the inertial vortex dynamics of viscoelastic fluids in the flow past a confined cylinder.

  • Excessive amounts of volatile organic compounds (VOCs) and odorants discharged into the environment are highly dangerous to human health as well as to ecological systems. Biological treatments of waste gas streams, called biofiltration, containing VOCs and odorous compounds has gained much attention because biofilters are more cost effective and environmentally friendly than conventional air pollution control technologies. This review provides an overview of biotrickling filtration, which is a type of biofiltration including continuous trickled-water flow inside filter media, for VOC and odor abatement. The configuration, design, cost effectiveness, removal capacity and environmental impact of this techniques and the future research and development needs in this area are all considered.

Production of 1,2-Propanediol from Glycerol in Saccharomyces cerevisiae

  • Glycerol has become an attractive carbon source in the biotechnology industry owing to its low price and reduced state. However, glycerol is rarely used as a carbon source in Saccharomyces cerevisiae because of its low utilization rate. In this study, we used glycerol as a main carbon source in S. cerevisiae to produce 1,2-propanediol. Metabolically engineered S. cerevisiae strains with overexpression of glycerol dissimilation pathway genes, including glycerol kinase (GUT1), glycerol 3-phosphate dehydrogenase (GUT2), glycerol dehydrogenase (gdh), and a glycerol transporter gene (GUP1), showed increased glycerol utilization and growth rate. More significant improvement of glycerol utilization and growth rate was accomplished by introducing 1,2-propanediol pathway genes, mgs (methylglyoxal synthase) and gldA (glycerol dehydrogenase) from Escherichia coli. By engineering both glycerol dissimilation and 1,2-propanediol pathways, the glycerol utilization and growth rate were improved 141% and 77%, respectively, and a 2.19 g 1,2- propanediol/l titer was achieved in 1% (v/v) glycerolcontaining YEPD medium in engineered S. cerevisiae.

  • Multivariate analysis and batch monitoring on a pilot-scale sequencing batch reactor (SBR) are described for integrated wastewater treatment management system, where a batchwise multiway independent component analysis method (MICA) are used to extract meaningful hidden information from non-Gaussian wastewater treatment data. Three-way batch data of SBR are unfolded batch-wisely, and then a non-Gaussian multivariate monitoring method is used to capture the non-Gaussian characteristics of normal batches in biological wastewater treatment plant. It is successfully applied to an 80L SBR for biological wastewater treatment, which is characterized by a variety of error sources with non-Gaussian characteristics. The batchwise multivariate monitoring results of a pilot-scale SBR for integrated wastewater treatment management system showed more powerful monitoring performance on a WWTP application than the conventional method since it can extract non-Gaussian source signals which are independent and cross-correlation of variables.

Biocontrol Traits and Antagonistic Potential of Bacillus amyloliquefaciens Strain NJZJSB3 Against Sclerotinia sclerotiorum, a Causal Agent of Canola Stem Rot

  • Bacillus amyloliquefaciens strain NJZJSB3 has shown antagonism of several phytopathogens in vitro, especially Sclerotinia sclerotiorum. Both the broth culture and cell suspension of strain NJZJSB3 could completely protect the detached leaves of canola (Brassica napus) from S. sclerotiorum infection. In pot experiments, the application of strain NJZJSB3 cell suspension ($10^8CFU/ml$) decreased the disease incidence by 83.3%, a result similar to commercially available fungicide (Dimetachlone). In order to investigate the potential biocontrol mechanisms of strain NJZJSB3, the nonvolatile antifungal compounds it produces were identified as iturin homologs using HPLC-ESI-MS. Antifungal volatile organic compounds were identified by gas chromatography-mass spectrometry. The detected volatiles toluene, phenol, and benzothiazole showed antifungal effects against S. sclerotiorum in chemical control experiments. Strain NJZJSB3 also produced biofilm, siderophores and cell-wall-degrading enzymes (protease and ${\beta}$-1,3-glucanase). These results suggest that strain NJZJSB3 can be a tremendous potential agent for the biological control of sclerotinia stem rot.

  • Activated sludge processes are broadly used in the biological wastewater treatment processes. The activated sludge processes are complex systems because of the many factors such as the variation of influent flowrate and ingredients, the complexity of biological reactions, and the various operation conditions. The main motivation o this research is to develop an intelligent control strategy for activated sludge process (ASP). ASP is a complex and nonlinear dynamic system owing to the characteristic of wastewater, the change in influent flowrate, weather conditions, and so on. The mathematical model of ASP also includes the uncertainty which is a ignored or unconsidered factor from process designers. The ASP model based on Matlabⓡ/Simulinkⓡ is developed in this paper. And the model performance is examined by IWA (International Water Association) and COST (European Cooperation in the filed of Scientific and Technical Research) data. The model tests derive steady-state results of 14 days. In this paper, fuzzy logic control approach is applied to handle DO concentrations. The fuzzy logic controller includes two inputs and one output to adjust air flowrate. The objective function for the optimization, in the implemented evolutionary strategy, is formed with focusing on improving the effluent quality and reducing the operating cost.

  • The oxygen saturation of blood can be measured by sensing the difference absorption in optical spectra of Hb and $HbO_2$, as the well known previous study. [1] In this study we developed the noninvasive tissue reflectance oximeter(TRO) using three kinds of LEDs which produce a peak spectral emission at a wavelength of 565, 660 and 940nm. And we tested the unction of the TR oximeter by comparing the output signals measured on normal tissue to measured on low oxygenated tissue. The results showed that oxygen saturation of blood and biological tissue can be monitored from the separation arrangement light source and detector.

  • The effect of unattached valve leaflet on flow field downstream of a floating and flapping polyurethane heart valve prosthesis was investigated. With a triggering system and a time-delay circuit the instantaneous velocity field downstream of the valve was measured by particle image velocimetry (PIV) in conjunction with the opening posture of a flexible valve leaflet during a cardiac cycle. Reynolds shear stress distribution was calculated from the velocity fields and wall shear stress was directly measured by hot-film anemometry (HFA). The floating motion of the valve leaflet resulted in the reduction of pressure drop and recirculating flow region downstream of the valve.

  • Studies on some biodegradable polymers and other materials such as hydrogels have shown the promising potential for a variety of surgical applications. Postoperative adhesion caused by the natural consequence of surgical wound healing results in problems of the repeated surgery. Recently, scientists have developed absorbable anti-adhesion barriers that can protect a tissue from adhesion in case they are in use; however, they are dissolved when no longer needed. Although these approaches have been attempted to fulfill the criteria for adhesion prevention, none can perfectly prevent adhesions in all situations. Overall of this work, a new method to fabricate an anti-adhesion membrane using biodegradable polymer and hydrogel has been developed. The ideal barrier for preventing postoperative adhesion would have the following properties; it should be (i) resorbable (ii) non-reactive (iii) easy to apply (iv) capable of being fixed in position. In order to fulfill these properties, we adopted solid freeform fabrication method combined with surface modification which includes the hydrogel coating, therefore, inner or outer structure can be controlled and the property of anti adhesion can be improved.
