Investigation of hematological analyses in Gyeongju DongGyeong dog populations (경주지방의 경주개 동경이(무미 또는 단미 형태의 개)에서 혈액상 분석에 대한 실태조사)

  • Investigations for hematologic and biochemical values for Gyeongju DongGyeong dogs were performed. This study were conducted to determine normal blood and serum chemical values on DongGyeong dogs. Normal blood and serum chemical values were studied in 47 healthy dogs (male 28, female 19). The hematologic and serum chemistry results were confirmed by age, sex, shape of tail. The hematological values showed no difference from Korea native dogs. The hematocrit values ($49.0{\pm}8.1$(%)) of DongGyeong dogs (2-3 years group) were significantly higher than other dogs. The mean levels of MCV ($61.6{\pm}3.8({\mu}m^3)$) in DongGyeong dogs were lower than general dogs. The levels of cholesterol($220{\pm}80.5$(mg/dl)) and inorganic phosphorus ($3.6{\pm}1.0$ (mg/dl)) of DongGyeong dogs (2~3 years group) were significantly higher than Korea native dogs. Totally concentration of triglyceride ($66.2{\pm}17.0$ (mg/dl)) of DongGyeong dogs were significantly lower than Korea native dogs. The mean values of BUN were increased with age but no significantly. In conclusion, data obtained from this study may be valuable as a standard for interpretation of the results in hematologic and biochemical analysis of Gyeongju DongGyeong dog populations.

Comparison of hematological and serum biochemical parameters among small breed dogs (국내 반려견의 품종에 따른 혈구 및 생화학 수치 비교 연구)

  • Hematological and serum biochemical analyses are routinely used to screen dogs for disease and monitor their clinical progression. These values in dogs may be affected by internal factors, such as breed and age, and external factors like environment, diet, and lifestyle. Apparently, subclinical breed-related variations in hematologic and serum biochemical results have been characterized for Bernese Mountain dogs, Alaskan Malamutes, English Setters, and Golden Retrievers. It is possible that some breed-related differences in clinical pathology analyses may reflect evidence of underlying disease and impact clinical decision plan. In this study, we estimated hematologic and serum biochemical parameters in small breed dogs and compared among four different breed dogs. Blood samples were collected from 38 domestic dogs that were 10 Beagles, 9 Malteses, 10 Poodles, and 9 Miniature Schnauzers. In terms of RBC count, Beagle ($7.2{\pm}0.24{\times}10^3cells/{\mu}L$) showed significantly (P<0.05) higher count than other breeds (Maltese, $6.51{\pm}0.25{\times}10^3cells/{\mu}L$; Poodle, $6.35{\pm}0.24{\times}10^3cells/{\mu}L$; and Miniature Schnauzer, $6.35{\pm}0.25{\times}10^3cells/{\mu}L$). For MCV, Miniature Schnauzer ($67.05{\pm}0.84fl$) results were significantly (P<0.05) higher than Beagle with $64.33{\pm}0.8fl$. For cholesterol value, Beagle ($163.3{\pm}9.15mg/dl$) had significantly (P<0.05) higher values than Miniature Schnauzer with $119.12{\pm}9.64mg/dl$. There was no statistically difference among breeds in terms of enzyme markers of liver diseases. In conclusion, data obtained from this study may be valuable as breed-related variability for interpretation of the results in hematologic and serum biochemical analysis among four small breed dogs.

Biochemical Characters of Polygalacturonase Produced by Botryosphaeria dothidea (사과 겹무늬썩음병균(Botryosphaeria dothidea)이 생산하는 Polygalacturonase의 생화학적 특성)

  • The polygalacturonase (PG) production in rotten apples by Botryosphaeria dothidea was purified by using gel filtration and ion exchange column chromatography, and the biochemical characters of PG were investigated. The purified PG appeared as a single band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) with approximate molecular weight of 49 kilodalton (kDa). The molecular weight was equal to the native molecular weight estimated by gel filtration. The Km and Vmax values of PG were 0.51 mg/ml and 90.9 $\mu$M/min/ml, respectively. Optimum pH was 4.0~5.0, and the PG activity was stable from pH 5.0~10.0. Optimum temperature of the enzyme activity was 4$0^{\circ}C$. The PG activity was relatively stable at 2$0^{\circ}C$, but it was reduced 45% at 4$0^{\circ}C$ and completely inactivated at 8$0^{\circ}C$. The PG activity was considerably inhibited by Cu2+, Zn2+, SDS and EDTA, whereas it was not effected by Ca2+, K+, Mg2+ or Na+ ions.

Effects of age on intestinal phosphate transport and biochemical values of broiler chickens

  • Objective: The objective of this experiment was to characterize the mRNA expression profile of type IIb sodium-inorganic phosphate cotransporter (NaPi-IIb) and the biochemical values of serum alkaline phosphatase (AKP), calcium, inorganic phosphorus, tibial ash and minerals of broiler chickens with aging. Methods: A total of 56 one-day-old Arbor Acres male broiler chickens were used. Broiler chickens were weighed and samples were collected weekly from day 1. Results: The result showed that before the growth inflection point, ash, calcium, and phosphorus content in the tibia of broiler chickens increased with growth (before 3 weeks of age), although there were no significant differences in chicks at different ages in the later period of the experiment and weight gain rate was relatively slow at this stage (4 to 6 weeks). NaPi-IIb gene expression in the small intestine in the early growth stage was higher than that in the later growth stage. Expression of calbindin and the vitamin D receptor protein in the intestinal mucosa increased with age in the duodenum and jejunum. Serum AKP activity first increased and subsequently decreased after peaking at 1 week of age, but there was no significant difference after 3 weeks of age. Conclusion: These results show that compared with the early growth stage, the weight-gain rate of broiler chickens in the late growth stage gradually decreased with gradual tibia maturation, along with weaker positive transport of phosphorus in the intestine and reinforced re-absorption of phosphorus in the kidney, which might be the reason that phosphorus requirement in the late growth stage was decreased.

Clinical blood chemistry analysis in chickens infected with reticuloendotheliosis virus (세망내피증 바이러스 감염 닭에서의 혈액화학치 변화)

  • Body weights and blood biochemical values in chickens infected with reticuloendotheliosis virus (REV)-HI, a Korean isolate, were studied. REV-HI causes severe body weight depression in chickens inoculated but not in chicken contact-infected. Body weights of infected chickens in 3, 4, and 5 weeks after infection were 78%, 76% and 65% of those of control respectively. Blood glucose levels in REVinfected chickens were extremely high compared with those in control (226 $\geq$ 21 vs. 814 $\geq$91.3 mg/dl in week 2) during the experiment period. Triglyceride levels in REV-infected chickens were significantly higher in week 2 and 3, whereas in week 4, REV-infected chickens showed significantly lower levels than the control. Blood lipase, amylase and alkaline phosphatase levels of REV-infected chickens in week 2 were significantly higher, whereas cholesterol, magnesium and calcium values in week 4 were significantly lower than the control. Other blood biochemical values such as alkaline aminotransferase, aspartate aminotransferase, and $\gamma$-glutamyltransferase were nonsignificantly different from the control. These above results suggest that weight depression by REV may be related with increase of blood glucose, which indicated that REV-infected chickens could not use blood glucose as energy source.

External and Internal Glucose Mass Transfers in Succinic Acid Fermentation with Stirred Bed of Immobilized Actinobacillus succinogenes under Substrate and Product Inhibitions

  • This paper is dedicated to the study on the external and internal mass transfers of glucose for succinic acid fermentation under substrate and product inhibitions using a bioreactor with stirred bed of immobilized Actinobacillus succinogenes cells. By means of the substrate mass balance for a single particle of biocatalysts, considering the kinetic model adapted for both inhibitory effects, specific mathematical models were developed for describing the profiles of the substrate concentration in the outer and inner regions of biocatalysts and for estimating the substrate mass flows in the liquid boundary layer surrounding the particle and inside the particle. The values of the mass flows were significantly influenced by the internal diffusion velocity and rate of the biochemical reaction of substrate consumption. These cumulated influences led to the appearance of a biological inactive region near the particle center, its magnitude varying from 0 to 5.3% of the overall volume of particles.

Fabrication and Characterisation of a Novel Pellicular Adsorbent Customised for the Effectvie Fluidised Bed Adsorption of Protein Products

  • A dense pellicular solid matrix has been fabricated by coating 4% agarose gel on to dense zironia-silica(ZS) spheres by watr-in-oil emulsification . The agarose evenly laminated the ZS bead to a depth of 30㎛, and the resultin gpellicular assembly was characterised by densities up to 2.39g/mL and a mean particle dimeter of 136 ㎛. In comparative fluidisation tests, the pellicular solid phase exhibited a two-fold greater flow velocity than commercial benchmark ad-sorbents necessary to achieve common values of bed expansion. Furthermore, the perlicular parti-cles were characterised by improved qualities of chromatographic behaviour, particularly with re-spect to a three-fold increase in the apparent effective diffusivity of lysozyme within a pellicular assembly modified with Cibacron Blue 3GA. The properties of rapid protein adsorption/desorp-tion were attributed to the physical design and pellicular deployment of the reactive surface in the solid phase. When combined with enhanced feedstock throughput, such practical advantages recommend the pellicular assembly as a base matrix for the selective recovery of protein products from complex, particulate feedstocks(whole fermentation broths, cell disruptates and biological extracts).

A Novel Strategy for Thermostability Improvement of Trypsin Based on N-Glycosylation within the Ω-Loop Region

  • The Ω-loop is a nonregular and flexible structure that plays an important role in molecular recognition, protein folding, and thermostability. In the present study, molecular dynamics simulation was carried out to assess the molecular stability and flexibility profile of the porcine trypsin structures. Two Ω-Loops (fragment 57-67 and fragment 78-91) were confirmed to represent the flexible region. Subsequently, glycosylation site-directed mutations (A73S, N84S, and R104S) were introduced within the Ω-loop region and its wing chain based on its potential N-glycosylation sites (Asn-Xaa-Ser/Thr consensus sequences) and structure information to improve the thermostability of trypsin. The result demonstrated that the half-life of the N84S mutant at 50℃ increased by 177.89 min when compared with that of the wild-type enzyme. Furthermore, the significant increase in the thermal stability of the N84S mutant has also been proven by an increase in the Tm values determined by circular dichroism. Additionally, the optimum temperatures of the wild-type enzyme and the N84S mutant were 75℃ and 80℃, respectively. In conclusion, we obtained the thermostability-improved enzyme N84S mutant, and the strategy used to design this mutant based on its structural information and N-linked glycosylation modification could be applied to engineer other enzymes to meet the needs of the biotechnological industry.

Hematological analyses of Japanese macaque (Macaca fuscata) housed in Everland Zoological Gardens (국내 동물원에서 사육 중인 일본원숭이의 혈액상)

  • Hematological and serum biochemical values were assessed from 20 clinically healthy Japanese Macaques raised in Everland Zoological Gardens and compared to the International Species Information System (ISIS) reference range that is used internationally as standard for wildlife animals. Taking our standard on sexual maturation at age 4, tRBC values in Macaques under age 4 were significantly lower than those over age 4, but the Hb and PCV values were significantly higher. Compared to the ISIS standard, the tRBC values in Macaques under age 4 were significantly lower whereas the Hb and MCHC values were significantly higher. Moreover, in the samples of Macaques over age 4, the PCV and MCV values were significantly lower than the ISIS standard. On serum biochemistry values the creatinine and amylase values in the Macaques under age 4 were significantly lower than those over age 4. In comparison with the ISIS standard, the values of ALT, ALP, BUN, IP, $Ca^{2+}$ and $K^+$ in the Macaques under age 4 did have no significant difference. The values of TP, GGT, tBil, amylase, TG and UA were significantly higher than the ISIS standard, but the values of albumin, AST, glucose, creatinine, cholesterol, CPK, LDH, $Na^+$ and Clwere significantly lower. In contrast, the values of TP, albumin, ALT, ALP, creatinine, cholesterol, amylase, TG, IP and $Na^+$ in the Macaques over age 4 did have no significant difference, but the values of GGT, BUN, tBil, UA and $Ca^{2+}$ were significantly higher, while the values of AST, glucose, CPK, LDH, $K^+$ and $Cl^-$ were significantly lower. On the other hand, there was no significant difference in hematological and serum biochemical values between the groups of male and female.

Analysis of Biochemical Compositions and Nutritive Values of Six Species of Seaweeds (해조류 6종의 생화학적 조성 및 영양적 가치 분석)

  • The biochemical compositions and nutritive values of six species of seaweeds were analyzed to determine their applicability in functional foods or ingredients. The biochemical compositions (moisture, ash, protein, lipid, and dietary fiber) and fatty acid contents were determined for the following seaweed extracts: Phaeophyceae (Laminaria japonica, Hizikia fusiformis, and Undaria pinnatifida), Rhodophyceae (Porphyra tenera and Gracilaria verrucosa), and Chlorophyceae (Ulva lactuca). The moisture content (% dry weight) ranged from 11.47% to 13.94%, ash from 19.15% to 26.50%, protein from 5.08% to 15.44%, lipid from 2.75% to 4.43%, and dietary fiber from 36.84% to 52.98%. C14:0, C16:0, C18:0, C16:1, C18: 1n-3, C18:2n-6, C18:3n-6, C20:4n-6, and C20:5n-3 represented the predominant proportions of fatty acids. Interestingly, docosahexaenoic acid (C22:6n-3, DHA) was either not found or only detected in trace amounts in the analyzed seaweeds. The levels of n-3 fatty acid were higher than other polyunsaturated fatty acids, and the n-6/n-3 ratio was very low. These results indicate that seaweed inhabiting Korean coastal areas will be beneficial to human health.
