산림소유역 유출수의 비강우일 비점오염물질 농도 변화 및 유출 특성 분석 (Changes in temporal and spatial stream water concentrations and analysis on nonpoint source runoff in forested watersheds on non rainfall days)

  • This study was carried out to analyze the monthly runoff concentration on non rainfall days in order to prepare basic data to compare the runoff concentration on rainfall days in 7 forest watersheds in the Republic of Korea. Forest stream water has been collected through 15 times of sampling in each watershed and analyzed based on the changes in concentration of Biochemical Oxygen Demand(BOD), Chemical Oxygen Demand(COD), Total Organic Carbon(TOC), Total Nitrogen(TN), and Total Phosphorus(TP). The average concentration was 0.8 mg/L for BOD, 1.4 mg/L for COD, 0.8 mg/L for TOC, 1.85 mg/L for TN and 0.002 mg/L for TP during non rainfall days. Coniferous forested watersheds showed higher value of TN and TP concentration. Concentrations of BOD and TP in early March (p<0.01) were affected by melt water flow input in spring season. Significant differences (p<0.01) in concentrations were observed in BOD and TOC, indicating seasonal rainfall and vegetation growth impacts on forest stream quality. Concentration of TN and TP showed significant positive correlation, and weak negative correlation was found in the concentration of BOD and TOC. It is expected that result of forest stream water on non rainfall days could be basic information in managing non-point source from forest watersheds.

Residual Level, Histology, and Blood Biochemistry of Tebuconazole: A Repeated Dose 28-Day Oral Toxicity Study in Pigs

  • In this study, we investigated the residual properties of tebuconazole-treated pigs. Twenty pigs were treated with different concentrations (0.25, 1.25, 2.5, 12.5, and 25 mg/kg bw/d) of tebuconazole for 28 d. Blood biochemistry, histology, and residual levels were analyzed using the VetTest analyzer, Masson's trichrome staining kit, and liquid chromatography-mass spectrometry, respectively. The final body weights were not significantly different between the control and treatment groups. Alkaline phosphatase, blood urea nitrogen, cholesterol, and gamma-glutamyl transpeptidase levels were significantly different from those of the control after exposure for 14 d. However, alanine aminotransferase levels showed changes only after exposure to pesticides for 28 d. The biochemical parameters were separated during the experimental period (14 d versus 28 d) by principal component analysis. Based on variable importance plots, blood urea nitrogen/creatinine ratio, blood urea nitrogen, glucose, and gamma-glutamyl transpeptidase are candidate biomarkers for tebuconazole exposure. The residual levels were observed at T4 (12.5 mg/kg bw/d) and T5 (25 mg/kg bw/d) in the liver and fat tissues, respectively. Fibrosis increased in the liver, kidney, and fat tissues, depending on the tebuconazole concentration. In conclusion, the residue limits of tebuconazole and the physiological changes caused by dietary tebuconazole in pigs provide important information for establishing maximum residue limits of pork and pork products.

Identification, Characterization, and Efficacy Evaluation of Bacillus velezensis for Shot-Hole Disease Biocontrol in Flowering Cherry

  • Though information exists regarding the pathogenesis of the shot-hole disease (SH) in flowering cherry (FC), there has been a lack of research focusing on SH management. Therefore, here, we investigated the inhibitory activities of antagonistic bacteria against SH pathogens both in vitro and in vivo as well as their biochemical characteristics and bioactive compounds. Two biosurfactant-producing bacterial antagonists, identified as Bacillus velezensis strains JCK-1618 and JCK-1696, exhibited the best effects against the growth of both bacterial and fungal SH pathogens in vitro through their cell-free culture filtrates (CFCFs). These two strains also strongly inhibited the growth of the pathogens via the action of their antimicrobial diffusible compounds and antimicrobial volatile organic compounds (VOCs). Crude enzymes, solvent extracts, and biosurfactants of the two strains exhibited antimicrobial activities. Liquid chromatography/electrospray ionization time-of-flight mass spectrometric analysis of the partially purified active fractions revealed that the two antagonists produced three cyclic lipopeptides, including iturin A, fengycin A, and surfactin, and a polyketide, oxydifficidin. In a detached leaf assay, pre-treatment and co-treatment of FC leaves with the CFCFs led to a large reduction in the severity of the leaf spots caused by Epicoccum tobaicum and Bukholderia contaminans, respectively. In addition, the two antagonists produced indole-3-acetic acid, siderophore, and a series of hydrolytic enzymes, along with the formation of a substantial biofilm. To our knowledge, this is the first report of the antimicrobial activities of the diffusible compounds and VOCs of B. velezensis against the SH pathogens and their efficiency in the biocontrol of SH.

Characterization of Pseudomonas sp. NIBR-H-19, an Antimicrobial Secondary Metabolite Producer Isolated from the Gut of Korean Native Sea Roach, Ligia exotica

  • The need to discover new types of antimicrobial agents has grown since the emergence of antibiotic-resistant pathogens that threaten human health. The world's oceans, comprising complex niches of biodiversity, are a promising environment from which to extract new antibiotics-like compounds. In this study, we newly isolated Pseudomonas sp. NIBR-H-19 from the gut of the sea roach Ligia exotica and present both phenotypes and genomic information consisting of 6,184,379 bp in a single chromosome possessing a total of 5,644 protein-coding genes. Genomic analysis of the isolated species revealed that numerous genes involved in antimicrobial secondary metabolites are predicted throughout the whole genome. Moreover, our analysis showed that among twenty-five pathogenic bacteria, the growth of three pathogens, including Staphylococcus aureus, Streptococcus hominis and Rhodococcus equi, was significantly inhibited by the culture of Pseudomonas sp. NIBR-H-19. The characterization of marine microorganisms with biochemical assays and genomics tools will help uncover the biosynthesis and action mechanism of antimicrobial metabolites for development as antagonistic probiotics against fish pathogens in an aquatic culture system.

Ruminal impaction with an indigestible foreign body in long-tailed goral (Naemorhedus caudatus)

  • A 6-year-old female long-tailed goral (Naemorhedus caudatus, goral) was admitted to the Gangwon wildlife medical rescue center due to severe starvation and hypothermia. The goral displayed clinical signs of severe weight loss, dehydration, lethargy, regurgitation, hypothermia, and bloating, with blood tests showing a decrease in albumin, calcium, glucose, and sodium. Supportive care was given through heating, oxygen supply, and fluid therapy, but the goral's condition worsened, and the goral died three days after admission. Radiographic and computed tomography scans revealed the presence of an indigestible foreign body (IFB) in the rumen, which was confirmed during the necropsy. The cause of goral's death was identified as chronic rumen impaction caused by the IFB (a ball of string), with other pathological findings including bloating, the presence of serous ascites, and congestion of digestive organs. Blood biochemical changes, clinical signs, and veterinary medical imaging diagnosis can be helpful in diagnosing ruminal impaction. This case report can provide valuable information for the diagnosis and treatment of ruminal impaction caused by IFB in endangered gorals, which may include rumenotomy or other surgical procedures.

Copper Tolerance of Novel Rhodotorula sp. Yeast Isolated from Gold Mining Ore in Gia Lai, Vietnam

  • In this study, twenty-five yeast strains were isolated from soil samples collected in the gold mining ore in Gia Lai, Vietnam. Among them, one isolate named GL1T could highly tolerate Cu2+ up to 10 mM, and the isolates could also grow in a wide range of pH (3-7), and temperature (10-40 ℃). Dried biomass of GL1 was able to remove Cu2+ effectively up to 90.49% with a maximal biosorption capacity of 18.1 mg/g at pH 6, temperature 30 ℃, and incubation time 60 min. Sequence analysis of rDNA indicated this strain was closely related to Rhodotorula mucilaginosa but with 1.53 and 3.46% nucleotide differences in the D1/D2 domain of the 28S rRNA gene and the ITS1-5.8S rRNA gene-ITS2 region sequence, respectively. Based on phylogenetic tree analysis and the biochemical characteristics, the strain appears to be a novel Rhodotorula species, and the name Rhodotorula aurum sp. nov. is proposed. This study provides us with more information about heavy metal-tolerant yeasts and it may produce a new tool for environmental control and metal recovery operations.

The Hazardous Components and Prevailing Rate of Sarcopenic Obesity in Younger Women : Based on 2008-2011 Korean National Health and Nutrition Examination Surveys

  • Purpose : Sarcopenic obesity is associated with adverse health consequences in females. Nevertheless, there has been limited research on the hazardous components and prevailing rates of sarcopenic obesity among younger women. This study aimed to identify the hazardous components and prevailing rates of sarcopenic obesity in younger females. Methods : This study utilized data based on 2008~2011 from the Korea National Health and Nutrition Examination Surveys by the centers for disease control and prevention. The analysis was concentrated on a subset of 1,520 women aged between 30 and 39 years of age. The participants answered a questionnaire to gather demographic information. They then underwent a physical examination to measure the human detention variables, which was conducted utilizing bioelectrical impedance analysis. The blood pressure and blood laboratory tests were determined using established laboratory protocols for evaluating blood parameters. Results : This study included 1,520 patients aged 30~39 years old. The mean age of the participants was 34.97 (years)±2.74 and the overall prevailing rate was 1.84 %. The hazard components in human dimensions were the height, weight, body mass index (BMI), waist circumference (WC) and skeletal muscle mass index (SMI). The hazard components in biochemical and blood pressure were high fasting glucose, increased triglyceride, elevated total cholesterol, high systolic blood pressure, and increased diastolic blood pressure with p-values <.05. Conclusion : This study examined the hazardous components and prevailing rates of sarcopenic obesity in younger women living in the community. The results contribute to the current body of knowledge on sarcopenic obesity and shed light on possible hazardous components in a younger female population. Based on these findings, there should be increased health and medical attention towards the prevention, management, and health promotion related to reducing risk factors for sarcopenic obesity in younger women.

Effect of water temperature on protein requirement of Heteropneustes fossilis (Bloch) fry as determined by nutrient deposition, hemato-biochemical parameters and stress resistance response

  • Background: Dietary protein requirements are dependent on a variety of factors and water temperature is one of the most important abiotic factors affecting protein requirement of fish. This study was, therefore, conducted to investigate effects of water temperature on dietary protein requirement of fry Heteropneustes fossilis which has high demand in most of the Asian markets. Methods: Quadruplicate groups of 30 fish per treatment (2.97 ± 0.65 cm; 5.11 ± 0.34 g) were fed seven isoenergetic diets (17.9 kJ g-1 gross energy; 14.99 kJ g-1 digestible energy) containing dietary protein levels ranging from 28 to 52% at two water temperatures (18 and 26 ℃). Experimental diets were fed to apparent satiation as semi-moist cakes thrice daily at 17:00, 12:00, and 17:30 h for 12 weeks. For precise information, various growth parameters, protein deposition, hematological parameters, metabolic enzymes, and stress response were analyzed, and effects of water temperature on dietary protein requirement was recommended on the basis of response from above parameters. Results: Groups held at 26 ℃ attained best growth, feed conversion, and protein deposition at 44% dietary protein indicating that temperature affected dietary protein requirement for optimum growth of H. fossilis fry and protein requirement seems to be satisfied with 44% dietary protein. Interestingly, interactive effects of both dietary protein levels and temperature were not found (P > 0.05). Fish reared at 18 ℃ had comparatively higher values for aspartate and alanine transferases than those reared at 26 ℃ water temperature which exhibited normal physiological value for these enzymes indicating that body metabolism was normal at this temperature. Hematological parameters also followed same pattern. Furthermore, fish reared at 26 ℃ water temperature exhibited more resistant to thermal stress (P < 0.05). The 95% maximum plateau of protein deposition data using second-degree polynomial regression analyses exhibited dietary protein requirement of fry H. fossilis between 40.8 and 41.8% of diet at 26 ℃ water temperature. The recommended range of dietary protein level and protein/digestible energy ratio for fry H. fossilis is 40.8-41.8% and 27.21-27.88 mg protein kJ-1 digestible energy, respectively. Conclusions: Information developed is of high significance for optimizing growth potential by making better utilization of nutrient at 26 ℃ and, to develop effective management strategies for mass culture of this highly preferred fish species.

토양 및 퇴적토 환경 시료로부터 DNA 추출하는 방법에 대한 고찰 (A Review on the Current Methods for Extracting DNA from Soil and Sediment Environmental Samples)

  • 토양미생물은 토양 및 퇴적토 환경에서 생물학적 물질순환의 중요한 역할을 맡고 있다. 이러한 중요성으로 인해 토양미생물 생태 다양성과 군집구성이 토양 및 퇴적토 생태환경과 자연저감능력을 평가하는 지표로 유용하게 쓰일 수 있다. 보다 정확한 다양성과 군집구조 분석을 위해서는 다양한 토양 및 퇴적토 시료로부터 분석에 필요한 DNA수율과 순도를 확보해야 한다. 지금까지 토양 및 퇴적토에서 DNA추출하는 방법들에 대한 다양한 기초연구가 이루어졌지만, 실제 환경생태영향평가와 분해기능평가에 사용시 DNA추출방법 선정에 대한 지침 및 정보는 매우 부족하다. 이에 본 연구에서는 문헌조사를 통해서 다양한 방법들의 토양 및 퇴적토 내 미생물 DNA 추출 특성을 비교 분석하고, 현재 주로 사용되고 있는 DNA추출방법을 토양 및 퇴적토 환경평가나 오염지하수토양 자연저감능평가에 활용 할 경우 고려해야 할 기술적 사항에 대해 고찰하였다. 본 연구를 통해 하나의 특정 추출 방법으로는 미생물다양성, 군집구성 및 개체간 상호작용에 대한 정보를 얻기에는DNA양과 순도 차원에서 충분하지 않으며, 토양 및 퇴적토 미생물 군집분석의 목적에 따라 적합한 방법의 선택이 매우 중요함을 알 수 있었다.
