Characterization of Yersinia ruckeri isolated from the farm-cultured eel Anguilla japonica in Korea

  • Yersinia (Y.) ruckeri has been recognized as a serious bacterial pathogen to several kinds of fish, including rainbow trout. However, there are no reports about the characteristics and pathogenicity of Y. ruckeri isolated from farm-cultured eels. In this study, we isolated and characterized Y. ruckeri from the farm-cultured eel Anguilla japonica in Korea. We investigated the phenotypic and genotypic characteristics of Y. ruckeri and tested the virulence of Y. ruckeri isolates on experimentally infected eels. Examination of the flagellar morphology of Y. ruckeri by electron microscopy showed peritrichous flagella in its cell body. Biochemical reaction studies showed overall identical profiles between the isolates and the reference strain of Y. ruckeri in API 20E and API ZYM tests. We sequenced the 16S rRNA of the Y. ruckeri (1,505 bp) for the genotypic characterization (National Center for Biotechnology Information accession number EU401667). Comparison of the 16S rRNA sequences with previously reported Y. ruckeri strains revealed similar phylogenetic relationships. In the virulence assay of the Y. ruckeri on eels, the eels exhibited listlessness, but Y. ruckeri was reisolated from those of the gills and kidneys.

Radionuclide Peritoneal Scintigraphy in Patients with Ascites and Pleural Effusion (방사성핵종 복막촬영술을 이용한 복수에 동반된 수흉의 감별 진단)

  • Simultaneous presence of ascites and pleural effusion has been documented in patients with cirrhosis of the liver, renal disease, Meigs' syndrome and in patients undergoing peritoneal dialysis. Mechanisms proposed in the formation of pleural effusion in most of the above diseases are lymphatic drainage and diaphragmatic defect. But sometimes, hepatic hydrothoraxes in the absence of clinical ascites and pleural effusion secondary to pulmonary or cardiac disease are noted. It is not always possible to differentiate between pleural effusion caused by transdiaphragmatic migration of ascites and by other causes based soly on biochemical analysis. Authors performed radionuclide scintigraphy after intraperitoneal administration of $^{99m}Tc-labeled$ colloid in 23 patients with both ascites and pleural effusion in order to discriminate causative mechanisms responsible for pleural effusion. Scintigraphy demonstrated the transdiaphragmatic flow of fluid from the peritoneum to pleural cavities in 13 patients correctly. In contrast, in 5 patients with pleural effusion secondary to pulmonary, pleural and cardiac diseases, radiotracers fail to traverse the diaphragm and localize in the pleural space. Ascites draining to mediastinal lymph nodes and blocked passage of lymphatic drainage were also clarified, additionaly. Conclusively, radionuclide peritoneal scintigraphy is an accurate, rapid and easy diagnostic tool in patients with both ascites and pleural effusion. It enables the causes of pleural effusion to be elucidated, as well as providing valuable information required when determining the appropriate therapy.

Effects of Intra-particle Composition on the Adhesive Properties and Water Dispersity of Water Dispersible Acrylic Emulsion Pressure Sensitive Adhesive (입자 내 조성 변화가 수분산성 아크릴 에멀젼 점착제의 점착 물성과 수분산성에 미치는 영향)

  • The effects of intra-particle composition on the adhesive properties and water dispersity of acrylic emulsion type pressure sensitive adhesives (PSAs) were investigated. In the case of PSA having uniform intra-particle composition, the higher holding strength made the water dispersity lower. By changing the intra-particle composition in hard core/soft shell type, however, it was possible to get PSAs showing both high holding strength and water dispersity. When the weight ratio of MAA/AA is 4/1, high holding strength, but low initial tack and very low water dispersity were observed in both cases of higher contents of (AA+MAA) in core and shell area. When the weight ratio of MAA/AA is 1/4, however, higher water dispersity and lower holding strength were indicated in the case of higher contents of (AA+MAA) in shell area.

  • By estimating probability distributions of the good solutions in the current population, some researchers try to find the optimal solution more efficiently. Particularly, finite mixtures of distributions have a very useful role in dealing with complex problems. However, it is difficult to choose the number of components in the mixture models and merge superior partial solutions represented by each component. In this paper, we propose a new continuous evolutionary optimization algorithm with distribution estimation by variational Bayesian mixtures of factor analyzers. This technique can estimate the number of mixtures automatically and combine good sub-solutions by sampling new individuals with the latent variables. In a comparison with two probabilistic model-based evolutionary algorithms, the proposed scheme achieves superior performance on the traditional benchmark function optimization. We also successfully estimate the parameters of S-system for the dynamic modeling of biochemical networks.

Review of Rice Quality under Various Growth and Storage Conditions and its Evaluation using Spectroscopic Technology

  • Purpose: Grain quality is a general concept that covers many characteristics, ranging from physical to biochemical and physiochemical properties. Rice aging during storage is currently a challenge in the rice industry, and is a complicated process involving changes in all of the above properties. Spectroscopic techniques can be used to obtain information on the quality of rice samples in a non-destructive manner. Methods: The objective of this review was to highlight the factors that contribute to rice quality and aging, and to describe various spectroscopic modalities, particularly vibrational and hyperspectral imaging, for the assessment of rice quality. Results: Starch and protein are the main components of the rice endosperm, and are therefore key factors contributing to eating and cooking quality. While the overall starch, protein, and lipid content in the rice grain remains essentially unchanged during storage, structural changes do occur. These changes affect pasting and gel properties, and ultimately the flavor of cooked rice. In addition, grain quality is significantly affected by growing and environmental conditions, such as water availability, temperature, fertilizer application, and salinity stress. These properties can be evaluated using spectroscopic techniques, and rice samples can be discriminated by using multivariate statistical analysis methods. Conclusion: Hyperspectral imaging and vibrational spectroscopy techniques have good potential for determining rice quality properties in a non-invasive manner, i.e., not requiring the introduction of instruments into the rice grain.

Using Reverse Dot Hybridization Method and 16S rRNA Gene (16S rDNA) for Identifying the Food Poisoning Microorganism in Foods (Reverse dot hybridization 방법과 16S rRNA gene(16S rDNA)을 이용한 식품에서 식중독균의 탐색)

  • DNA sequence information on small-subunit rRNA gene (16S rDNA) obtained from food-poisoning bacterial culture was used to investigate the presence of bacterial pathogens in food. By reverse dot blot detection method, presence of food-poisoning bacteria could be confirmed on hybridization of digoxigenin-labeled 16S rDNA Polymerase Chain Reaction (PCR) primer product and biotin-labeled specific oligonucleotide probe. Escherichia coli, Bacillus cereus. and Salmonella sp. were used as the representative food-poisoning bacterial microorganisms. An oligonucleotide probe, based on the variable region of 16S rRNA gene, was used as the specific probe. These tools may be more useful than classic biochemical method for rapid identification of contaminated food.

Distribution and Antimicrobial Susceptibility Patterns of Bacteria Isolated from Genital Tract in Thoroughbred Mares (더러브렛 씨암말의 생식기내 세균의 분포 및 항생제 감수성 양상)

  • This study carried out to investigate the genital tract bacterial flora of Thoroughbred mare in Jeju province during March and July, 2006. The specimens were collected from vaginal ucosa and clitorial fossa using a culture swab (BBL, USA) from 100 Thoroughbred mares. Colonies were selected blood and MacConkey agar plate, and identified as standard biochemical properties using Biolog system (Thermo, USA). In this study, 470 gram-negative strains were isolated more frequently than 249 gram-positive strains. We were Isolated Escherichia coli (19.8%), Proteus mirabillis (14.9%), Enterobacter nimipressuralis (7.4%), Enterobacter mobilis (4.7%), Aeromonas encheleia (4.3%), Pseudomonas aeroginosa (3.0%), Staphylococcus aureus (14.9%), Staphylococcus epidermidis (11.2%), Coagulasenegative Staphylococcus spp. (10.0%), Enterococcus faecalis (9.2%), Enterococcus faecium (8.0%), Actinomyces viscosus (7.2%), Micoroccus diversus(6.8%), Streptococcus dysgalactiae subsp. equisimilis(5.2%), Streptococcus equi subsp. zooepidemicus (3.2%), Other non-beta hemolytic Streptococcus spp. (2.0%) and many others from vaginal mucosa and clitorial fossa in Thoroughbred mares. No significant bacteria (Taylorella equigenitalis and Klebsiella pneumonia) were isolated from the mare genital tract. In antimicrobial agents susceptibility test, it shows a high sensibility in the antibiotics of the most which excepts the streptomycin and neomycinm, kanamycin, spectinomycin, compound sulfonamides. Especially, Staphylococcus epidermidis, Enterococcus spp. and Streptococcus spp. were visible a high sensibility in the all antibiotics. However, Staphylococcus aureus, Pseudomonas aeruginosa, Proteus spp. and E. coli were showed a high antibiotic resistance patterns. These results may provide the basic information to establish strategies for the treatment and prevention of reproductive disease in Thoroughbred mares in Korea.

Development of Simple Benthic Macroinvertebrates Index (SBMI) for Biological Assessment on Stream Environment (하천환경의 생물학적 평가를 위한 간이저서동물지수(SBMI)의 개발)

  • GPI (Group Pollution Index) using 29 indicator groups of Korean benthic macroinvertebrates was proposed in 1992, a higher category taxa-level index developed for rapid field assessment of organic water pollution. This study was performed to revise the assessment scheme of GPI based on taxonomic performance and ecological information accumulated since 1992. The original GPI was renamed SBMI (Simple Benthic Macroinvertebrates Index), and SBMI was based on saprobic valency of 26 indicator groups composed of higher category taxa (mainly family ~ phylum) excluding some genus or species-level taxa. SBMI revealed highly significant correlation with concentration of 5-day biochemical oxygen demand ($BOD_5$) (correlation coefficient r = 0.78, n = 569 sites), total suspended solids (r = 0.69), and total phosphorus (r = 0.77). Also, SBMI revealed strong correlation with Shannon-Weaver's species diversity (r = -0.85), Margalef's species richness (r = -0.85), and McNaughton's dominance (r = 0.83). Determination coefficient of SBMI to concentration of water quality items and values of community indices such as species diversity was 3 ~ 8 % and approximately 11 ~ 14 % higher than that of GPI, respectively. Correlation between SBMI and water quality factors or community indices such as species diversity did not reveal much difference compared to that of species-level indices, such as BMI (Benthic Macroinvertebrates Index) and ESB (Ecological Score of Benthic Macroinvertebrates). SMBI is a simple-qualitative index with higher category taxa easily identified, and is applicable for rapid field assessment of water environment impairment.

Effects of Au Nanoparticle Monolayer on or Under Graphene for Surface Enhanced Raman Scattering

  • Since first discovery of strong Raman spectrum of molecules adsorbed on rough noble metal, surface enhanced Raman scattering (SERS) has been widely used for detection of molecules with low concentration. Surface plasmons at noble metal can enhance Raman spectrum and using Au nanostructures as substrates of SERS has advantages due to it has chemical stability and biocompatibility. However, the photoluminescence (PL) background from Au remains a problem because of obtaining molecular vibration information. Recently, graphene, two-dimensional atomic layer of carbon atoms, is also well known as PL quenchers for electronic and vibrational excitation. In this study, we observed SERS of single layer graphene on or under monolayer of Au nanoparticles (NPs). Single layer graphene is grown by chemical vapor deposition and transferred onto or under the monolayer of Au NPs by using PMMA transfer method. Monolayer of Au NPs prepared using Langmuir-Blodgett method on or under graphene surface provides closed and well-packed monolayer of Au NPs. Scanning electron microscopy (SEM) and Raman spectroscopy (WItec, 532 nm) were performed in order to confirm effects of Au NPs on enhanced Raman spectrum. Highly enhanced Raman signal of graphene by Au NPs were observed due to many hot-spots at gap of closed well-packed Au NPs. The results showed that single layer graphene provides larger SERS effects compared to multilayer graphene and the enhancement of the G band was larger than that of 2D band. Moreover, we confirm the appearance of D band in this study that is not clear in normal Raman spectrum. In our study, D band appearance is ascribed to the SERS effect resulted from defects induced graphene on Au NPs. Monolayer film of Au NPs under the graphene provided more highly enhanced graphene Raman signal compared to that on the graphene. The Au NPs-graphene SERS substrate can be possibly applied to biochemical sensing applications requiring highly sensitive and selective assays.

Polyphasic Microbial Analysis of Traditional Korean Jeung-Pyun Sourdough Fermented with Makgeolli

  • Jeung-pyun, a fermented rice cake, is prepared by fermenting rice sourdough using makgeolli, a traditional Korean rice wine, in the presence of yeast and lactic acid bacteria (LAB). The goal of this study was to conduct biochemical and microbial analyses of five different rice sourdoughs, each fermented with a different commercial makgeolli, using culture-dependent and culture-independent approaches. All sourdough samples fermented with different makgeolli for 6.5 h showed different profiles in pH, total titratable acidity, organic acid concentration, and microbial growth. LAB belonging to different genera were identified based on colony morphology on modified MRS and sourdough bacteria agar medium. PCR-denaturing gradient gel electrophoresis analyses of the five sourdoughs showed different bands corresponding to LAB and yeast. 16S/26S rRNA gene sequence analyses of the samples confirmed that the predominant LAB in the five fermented rice doughs was Lactobacillus plantarum, Lb. pentosus, and Lb. brevis. Various other Lactobacillus spp. and Saccharomyces cerevisiae were common in all five fermented samples. This study provides comprehensive and comparative information on the microflora involved in fermentation of rice sourdough and signifies the need to develop effective starters to enrich the quality of jeung-pyun.
