  • To yield high concentrations of protein expressed by genetically modified Escherichia coli, it is important that the bacterial strains are cultivated to high cell density in industrial bioprocesses. Since the expressed target protein is mostly accumulated inside the E. coli cells, the cellular product formation can be directly correlated to the bacterial biomass concentration. The typical way to determine this concentration is to sample offline. Such manual sampling, however, wastes time and is not efficient for acquiring direct feedback to control a fedbatch fermentation. An E. coli K12-derived strain was cultivated to high cell density in a pressurized stirred bioreactor on a pilot scale, by detecting biomass concentration online using a capacitance probe. This E. coli strain was grown in pure minimal medium using two carbon sources (glucose and glycerol). By applying exponential feeding profiles corresponding to a constant specific growth rate, the E. coli culture grew under carbon-limited conditions to minimize overflow metabolites. A high linearity was found between capacitance and biomass concentration, whereby up to 85 g/L dry cell weight was measured. To validate the viability of the culture, the oxygen transfer rate (OTR) was determined online, yielding maximum values of 0.69 mol/l/h and 0.98mol/l/h by using glucose and glycerol as carbon sources, respectively. Consequently, online monitoring of biomass using a capacitance probe provides direct and fast information about the viable E. coli biomass generated under aerobic fermentation conditions at elevated headspace pressures.

  • The present paper resents information on the dietary intake and biochemical status of elderly who are living in different types of residence. On hundred and sixty five women(age 65-80 years old) were divided into three groups ; those who are living alone (LA) , living with family (LF) , and living in institutions (LI) within the same general community. Nutrient intake, body fat content, and biochemical measurements in blood and urine were examined statistically and potential environment-related influence are discussed. Elderly women living alone had significantly lower intake of nutrients than elderly women were not living alone LF group showed significantly higher fat consumption than LA and LI groups. Body fat content, plasma albumin and calcium levels in elderly women who lived with other people were significantly higher than those in elderly women living alone, but fat weights and lean body mass between groups were not different. None of the plasma lipid fractions were significantly different among the three groups except LDL-cholesterol (LDL-C) concentration. In the LI group, LDL-C was lower than that of LF and LA groups. From the above results, serious nutritional deficiency has been shown in elderly women that live alone. Therefore, nutritional education and social help should be carried out to improve these situations for elderly people.

  • This purpose of this study was to investigate the nutrient intake, the health status as determined self-assessment checklist, biochemical indicators of elderly Korean women. We interviewed and 55 female subjects living alone in the Urban aged over 65 years. Information on their dietary intake was collected by 24-hour Recall method. Their health status was determined by a NSI checklist. Biochemical indicators were performed in whole blood and plasma of subjects. Except for protein, Fe, all of the elderly subjects belonged to over moderate nutritional risk. The average daily nutrient intake of the elderly was below the level of the recommended dietary allowances (RDA) for Koreans. A relationship between their health risk score and nutrient intake was observed negatively (not significantly). They had a risk of anemia as hemoglobin and hematocrit of subjects were under the normal value. Therefore, the reason that health risk score and health status badly was thought for lower nutrient intake.

  • Somatic embryogenesis has become a major tool in the study of plant embryology, as it is possible in culture to manipulate cells of many plant species to produce somatic embryos in a process that is remarkably similar to zygotic embryogenesis. Traditionally, the process has been studied by an examination of the ex vitro factors which influence embryo formation. Later structural, physiological and biochemical approaches have been applied. Host recently, molecular tools are being used. Together, these various approaches are giving valuable information on the process. This article gives an overview of somatic embryogenesis by reviewing information on the morphogenesis, physiology, biochemistry and molecular biology of the process. Topics covered include a brief description of the factors involved in the production of embryogenic cells. Carrot cell suspension is most commonly used, and the development of a high frequency and synchronous system is outlined. At the physiological and biochemical lev-els various topics, including the reactivation of the cell cycle, changes in endogenous growth regulators, amino acid, polyamine, DNA, RNA and protein metabolism, and embryogenic factors in conditioned medium are all discussed. Lastly, recent information on genes and molecular markers of the embryogenic process are outlined. Somatic embryogenesis, the best example of totipotency in plant cells, is not only an important tool in studies in basic biology, but is potentially of equal significance in the micropropagation of economically important plants.

  • Spiropyran-doped organic-inorganic hybrid coatings were prepared starting from glycidoxypropyl triethoxysilane and vinyltriethoxysilane by a sol-gel method. They were applied as a thin layer to polycarbonate sheets and their photochromic properties were investigated. The effect of polarity of solvents dissolving the spiropyran was investigated on the photochromic properties. The decoloration rate of the spiropyran decreased with increasing the polarity of solvents dissolving the spiropyran because the open form of the spiropyran was easily stabilized in the polar gel matrix.

  • This study was carried out to collect information to establish a framework for meal management and nutritional service for prevention of chronic degenerative disease in the industrial workers. We investigated the health concerned life-style, nutrient intakes, anthropometry and biochemical parameters in the male workers in Korea by work condition. Anthropometric parameters of height, weight, waist, hip and triceps skinfold thickness were measured and biochemical parameters including Urine pH, hemoglobin, blood glucose, total cholesterol, GOT, GPT, $\gamma$-GPT and blood pressure were determined for 101 subjects(50 office workers : 33.9yr, 51 laborers : 34.4yr). To assess the nutrient intakes and diet quality of workers, dietary intake was measured by one day 24-hour recall method. Result of anthropometric parameters of height(p<0.01), weight(p<0.05), hip(p<0.05), TSF(p<0.01) were significantly higher in office workers than in laborers. Biochemical parameters were not significantly difference in both of them. Average daily nutrient intakes of both groups was higher than the Korean RDA and report on 2001 National Health & Nutrition Survey. The calcium(p<0.01), iron(p<0.05), vitamin A(p<0.01), vitamin B1(p<0.01) and niacin(p<0.05) intakes in laborers were significantly higher than in office workers. Nutrition adequacy ratio(NAR), mean adequacy ratio(MAR) and index of nutritional quality(INQ) of laborers were higher than those of office workers. This results indicated that the onset possibility of chronic degenerative disease appeared higher in office workers. Therefore, meal menagement and nutritional service for prevention of chronic degenerative disease of industrial works needs to be more variant guidelines.

  • Biochemical characteristics of 24 Pongamia pinnata genotypes (candidate plus trees) from three agroclimatic zones were estimated and molecular characterization through RAPD markers was done. Various biochemical characters viz. seed oil, total carbohydrates, protein, acid value and Iodine number recorded significant variation among different genotypes. The highest seed oil content was 41.87% while seeds of 14 genotypes recorded above average (32.11%) for the trait. Seed oil and protein content exhibited a significant positive correlation and moderate heritability. Out of the initially selected twenty-five random primers, twenty-two RAPD primers were found to be highly reproducible and produced a total of 183 loci of which 147 (80.32%) loci were polymorphic. Percentage of polymorphism varied from 44% to 100% with an average of 80.62%. High level of genetic variation was found among different genotypes of P. pinnata. Both molecular and oil content (biochemical) markers appeared useful in analyzing the extent of genetic diversity in Pongamia and the result of these analyses will help to better understand the genetic diversity and relationship among populations. Overall, the Pongamia genotypes included in the study showed a correlation with their geographical origins such that genotypes from the same region tend to have higher genetic similarity as compared to those from different regions. However, in UPGMA based Nei's analysis, some genotypes were found not to be grouped based on geographical origins possibly due to the exchange of germplasm over time between farmers across the regions. The results from oil content analyses showed that several genotypes in 'Central and Western Plateau' agroclimatic zone of Jharkhand displayed a good potential for high oil content. The study provides insight about P. pinnata populations in Jharkhand (India) and constitutes a set of useful background information that can be used as a basis for future breeding strategy and improvement of the species.
