국내 전통주 주박의 이용과 유용생리활성 (Application of the Lees of Domestic Traditional Wine and its Useful Biological Activity)

  • 발효 폐기물의 대부분을 차지하는 주박은 발효 후 알코올성 발효액을 필터, 원심분리 또는 증류 후에 얻어지는 찌꺼기 및 1차 제조된 제품의 숙성과정 중에 생성되는 찌꺼기를 말한다. 국내 전통주 주박은 식용 원재료와 다양한 약용작물을 첨가하고 이를 누룩으로 발효시켜 제조하여, 안전성이 확보되면서 생리활성이 우수한 장점을 가지고 있다. 최근의 탁주 위주의 전통주 시장 성장으로 주박 생산량은 빠르게 증가되고 있으며, 이의 폐기물 처리는 더욱 엄격해지고 있으나, 국내에서는 아직 전통주 주박의 효율적인 재이용에 대한 연구는 제한적이다. 본 총설에서는 전통주 주박의 산업동향, 연구동향 및 특허동향을 분석하고, 전통주 주박의 유용생리활성물질들을 제시하였다. 전통주 주박은 단순 발효폐기물이라는 인식에서 벗어나, 식량, 식품첨가물, 비료, 사료, 미생물 배양원, 기능성 식품소재, 향장 및 미용소재의 고부가가치 발효소재 공급원이라는 인식의 전환이 필요하며, 향후 재처리 기술, 유용물질 회수기술 개발을 위한 체계적인 산학관 연계와 연구가 필요하다.

  • An important aspect of indigenous medicine is the ability of traditional societis to manipulate the environment for their own benefit, for example for their health care. In Indonesia, this indigenous medicine is called Jamu. Jamu mostly consists of a mixture of herbals of the genus Curcuma, Zingiberaceae. There are 19 species of curcuma grown in Indonesia. Eleven of them are popular in the jamu preparations. Ethnopharma-cological surveys have shown that 50% of these species are used for post partum protection, dismenorrhea, 30% are used for the treatment of stomache and as cosmetics, 20% for the treatment of various diseases such as fever, worms, asthma, etc. Chemical studies show that they contain curcuminoids, volatile oils, flavonoids, starch, and resinous substance. Pharmacological studies of extracts and isolated bioactive compounds have shown that they have a broad pharmacological activity such as antibacterial, antifungal, anti-inflammatory, choleretic, antihepatotoxic, antitumor, antioxidant, and antihyperlipidemic activity.

Commonly used herbal medicines in the treatment for female reproductive dysfunction

  • Herbal medicines are widely used for centuries to treat illness and improve health in Asia and the use of these remedies has become a worldwide form of alternative therapy. However, there is very limited evidence from randomized controlled trials to support the efficacy of the vast majority of herbal products. Of the commonly used herbal medicines, systemic clinical experiences and promising experiments in animals have found potential evidence of efficacy for six herbal medicines (Unkei-to, Hachimijio-gan, Keishi-bukuryo-gan, Toki-shakuyaku-san, Shakuyaku-kanzo-to, Sairei-to) in the treatment for female reproductive dysfunction. Because herbs may contain potent bioactive substances, the physician should have an opportunity to outline more stringent regulation, similar to over-the counter drug.

Licochalcone A, a Major Phenolic Constituent of Glycyrrhiza inflata, Suppresses Angiogenin Expression in Colon Cancer Cells

  • Tumor angiogenesis, which is essential for tumor growth and tumor metastasis, depends on angiogenic factors produced by tumor cells and/or infiltrating cells such as endothelial cells and immune cells in tumor tissue. Previously, we reported that licochalcone A (LicA), an important bioactive compound of Glycyrrhiza inflate, suppresses angiogenesis, tumor growth and metastasis. In this study, we evaluated the effect of LicA on angiogenin production in colon cancer cells because angiogenin is an essential factor to regulate angiogenesis and tumor progression. When we examined the angiogenin levels in three human colon cancer cells, HT-29, SW480 and Caco-2, LicA treatment significantly reduced the amounts of angiogenin among three cancer cell lines. In an in vivo study in which mice were implanted with HT-29 cells, oral administration of LicA reduced angiogenin in tumor tissues when compared with vehicle-administered mice. These results suggest that reduced angiogenin in response to LicA treatment may play essential role to inhibit tumor growth, angiogenesis as well as metastasis.

원적외선 조사에 따른 가래나무 잎의 항산화 활성 변화 (Changes of Antioxidant Activity in Juglans mandshrica Maxim. Leaves by Far Infrared Ray Irradiation)

  • 가래나무 잎을 원적외선 30, 40 및 50 v에서 각각 10, 20 및 30분을 처리하였다. 원적외선을 조사한 잎의 총 폴리페놀 함량, 총 플라보노이드 함량, 잎 추출물의 DPPH radical 소거 활성 등을 분석한 결과, 원적외선 30 v에서 20분간 조사한 처리구에서 총 폴리페놀 함량은 25.43 mg/g로 , 총 플라보노이드 함량은 39.38 mg/g으로 가장 높게 나타났다. 그 반면에, 원적외선 50 v에서 30분간 조사한 처리구에서 총폴리페놀 함량은 16.93 mg/g으로, 총플라보노이드 함량은 23.78 mg/g으로 가장 낮게 나타났다. 전체적으로 원적외선 처리에 의해 잎에 함유한 항산화 물질의 함량의 증가는 물론, 그 추출물의 항산화 활성도 증가하는 것을 알 수 있었다. 하지만 원적외선 처리가 가래나무에 함유한 어떠한 특정적인 물질들의 증가에 관하여서는 추후에 더욱 많은 연구가 필요할 것으로 사료된다.

시금치에서 생장 및 생리활성물질에 대한 UV-A LED의 보광 효과 (Effects of Supplemental UV-A LED Radiation on Growth and Bioactive Compounds in Spinach)

  • 본 연구에서는 단기간의 UV-A 조사가 시금치(Spinacia oleracea L.)의 생장과 생리활성물질에 미치는 영향을 평가하였다. 시금치 묘는 200µmol·m-2·s-1 PPFD, white LED, 광주기 12시간, 온도 20℃, 상대습도 70%, 이산화탄소 농도 500µmol·mol-1의 수직농장 모듈에서 재배되었다. 파종 후 5주된 묘는 7일 동안 20W·m-2와 40W·m-2의 두 가지 에너지 수준에서 연속적으로 UV-A(피크파장: 385nm) 조사한 후 생육 특성, 광합성 파라미터, 이미지 형광, 총 페놀 함량, 항산화도, 그리고 총 플라보노이드 함량을 분석하였다. 결과적으로, UV-A20W 처리는 시금치의 생체중과 건물중을 증가시켰다. 하지만, UV-A 처리구와 대조구 사이의 광합성 파라미터에는 유의한 차이가 나타나지 않았다. 광계II의 최대양자수율(Fv/Fm)은 모든UV-A 처리에서 7일동안 지속적으로 감소했다. 또한, UV-A20W 처리에서 식물체당 총 페놀 함량과 항산화도는 처리 7일째 증대되었으며, 총 플라보노이드 함량은 처리 5일째부터 유의적으로 증가하였다. 이러한 결과는 UV-A LED 보광이 수직농장과 같은 폐쇄형 식물 생산 시스템에서 재배되는 시금치의 생장과 품질을 향상시킬 수 있음을 시사한다.

가락지나물의 생장과 생리활성물질 함량에 미치는 차광, 광질 및 화학적 엘리시테이션 (Effect of Shading, Light Quality, and Chemical Elicitation on Growth and Bioactive Compound Content of Potentilla kleiniana Wight et Arnott)

  • 가락지나물은 장미과에 속하는 다년생 초본으로 potentillin, agrimoniin 등 생리활성물질은 한약재와 화장품 원재료로 이용되고 있다. 본 연구는 가락지나물의 재배 시 차광, 광질 및 화학적 elicitation에 따른 생육, 바이오매스, 엽록소함량, 생리활성물질 함량 축적효과를 조사하여 생산조건을 구명하고자 하였다. 온실에서 재배 중인 가락지나물을 각각 다른 차광조건[0% (200 μmol·m−2·s−1), 35% (95 μmol·m−2·s−1), 55% (65 μmol·m−2·s−1), 75% (40 μmol·m−2·s−1)]에서 60일간 재배한 결과, 엽록소 함량과 카로티노이드 함량은 35% 처리구에서 가장 높았으나 개체당 초장, 엽수와 바이오매스는 무차광구에서 가장 높은 것으로 조사되었다. 식물생장상 내(25 ± 2℃, 185 ± 3 μmol·m−2·s−1)에서 형광등과 3가지 혼합광원 [red:white:blue(RWB) = 8:1:1, red:blue(RB) = 8:2, red:green:blue(RGB) = 8:1:1] 처리구에서 재배한 결과, RWB 처리구에서 생장, 바이오매스, 엽록소함량, 총 페놀화합물과 플라보노이드 함량이 높은 것으로 나타났다. DPPH 라디칼 소거능은 모든 처리구에서 높았으며, 특히 형광등과 RGB 처리구에서 가장 높았다. 화학적 elicitor (SA; salicylic acid, MeJA; methyl jasmonate)를 0, 50, 100, 200 μM 농도별로 처리한 결과, 무처리구와 MeJA 50 μM 처리구에서 초장, 엽병직경, 바이오매스가 높게 조사되었다. 또한 MeJA 50 μM 처리구의 생체중과 건체중이 높은 것으로 나타났다. SA 50 μM 처리구에서의 총 페놀화합물(33.20 ㎎·g DW−1)과 플라보노이드 함량(7.18 ㎎·g DW−1)은 높게 분석되었으나 DPPH 라디칼 소거능은 MeJA 200 μM 처리구(88.65%)에서 유의적으로 높게 나타났다. 본 연구를 통해 가락지나물의 높은 생장, 바이오매스, 기능성물질 함량을 얻기 위한 온실과 식물생장상 내 최적의 차광 및 광질조건을 구명하였으며, 화학적 elicitation 농도에 따른 생리활성물질 함량과 항산화도를 확인하였다.

Selection of KYC 3270, a Cellulolytic Myxobacteria of Sorangium cellulosum, against Several Phytopathogens and a Potential Biocontrol Agent against Gray Mold in Stored Fruit

  • During 2002-2008 in Korea, 455 extracts from myxobacteria consisting of 318 cellulolytic and 137 bacteriolytic myxobacteria were isolated, which were then screened for antifungal activity against the phytopathogens Botrytis cinerea, Colletotrichum acutatum, Penicillium sp., Pyricularia grisea, and Phytophthora capsici. 204 isolates had antifungal activity, causing both a clear zone due to blocked spore germination and inhibition of mycelial growth; most (199) were from cellulolytic (Sorangium cellulosum) and only five were from bacteriolytic myxobacteria. B. cinerea, the best controlled among the five tested pathogens, had a unique group of antifungal isolates of myxobacterial extracts compared to the other pathogens' groups. Among seventy-nine bioactive myxobacteria, four isolates, KYC 3130, KYC 3247, KYC 3248 and KYC 3270, were selected and all were cellulolytic. Liquid culture filtrates of these four myxobacteria were applied to tomato, cherry tomato, strawberry, and kiwi fruits 5 h before inoculation with gray mold conidia; then the treated fruits were placed in an airtight container and the experiment was repeated six to eight times. Incidence (%) of gray mold on fruit of the infected control treatment was 84-98%, whereas it was only 5-21% after the KYC 3270 treatment. After KYC 3270 treatment of the four fruits, mold control was 79-95%, which was highest among the filtrates and statistically the same as treatment with fludioxonil, a registered chemical against gray mold of stored fruits.

Effect of chlorella culture solution using animal liquid manure on improving seed germination in perennial ryegrass

  • This experiments were conducted to evaluate the influence of Chlorella culture solution on seed germination of perennial ryegrass seeds. Chlorella are known to contain different bioactive compounds. In present research work, Chlorella culture solution using liquid manure as medium have been used to study their effects on germination and root length. The study conducted a germination experiment in petri-dishes. Four treatments were compared: non-treated control treated with distilled water, Chlorella culture solution and Chlorella culture filtrate, and liquid manure. The germination percentage of perennial ryegrass seeds was highest in the Chlorella culture solution treatment. Days required for 50, 70% seed germination was the fast in Chlorella culture solution and the Chlorella culture filtrate treatment. Root length of perennial ryegrass seeds was long by 1~2cm in the Chlorella culture solution compared with no treated control. The germination index of perennial ryegrass seeds was high by 180~202% in the Chlorella culture solution treatment compared to no treatment. Chlorella culture solution and the Chlorella culture filtrate have shown stimulatory effects in germination and development of root. Use of Chlorella culture solution and the Chlorella culture filtrate can be recommended to farmers as a ecofriendly practice for better germination and growth. Present research work reveals that Chlorella contain certain growth promoting substances which enhances seed germination.

Enzymatic transformation of ginsenosides in Korean Red Ginseng (Panax ginseng Meyer) extract prepared by Spezyme and Optidex

  • Background: In this study, we examined the effects of various enzymes on chemical conversions of ginsenosides in ginseng extract prepared by amylases. Methods: Rapidase, Econase CE, Viscozyme, Ultraflo L, and Cytolase PCL5 were used for secondary enzymatic hydrolysis after amylase treatment of ginseng extract, and ginsenoside contents, skin permeability, and chemical compositions including total sugar, acidic polysaccharide, and polyphenols were determined on the hydrolyzed ginseng extract. Results: Rapidase treatment significantly elevated total ginsenoside contents compared with the control (p < 0.05). In particular, deglycosylated ginsenosides including Rg3, which are known as bioactive compounds, were significantly increased after Rapidase treatment (p < 0.05). The Rapidase-treated group also increased the skin permeability of polyphenols compared with the control, showing the highest level of total sugar content among the enzyme treatment groups. Conclusion: This result showed that Rapidase induced the conversion of ginsenoside glycosides to aglycones. Meanwhile, Cytolase PCL5 and Econase treatments led to a significant increase of uronic acid (acidic polysaccharide) level. Taken together, our data showed that the treatments of enzymes including Rapidase are useful for the conversion and increase of ginsenosides in ginseng extracts or products.
