우유와 노인건강 (Milk and Health of Elderly People)

  • As the development of medical technology and the elevation of the standard of living, the population rate of elderly people in Korea is increasing gradually. To keep a good lift quality of the elderly, both appropriate exercise and nutrients intake are necessary for them. Dairy products are known for the good source of variable nutrients including functional components and bioactive peptides such as Ig, lactoferrin, MFGM, OPP, CPP, GMP, sialic acid etc that are required especially for elderly people. However, they are classified as the low dairy products consumption group recently. For the promotion of dairy product consumption of elderly people, variable and specialized dairy products for the elderly should be researched and developed with the strengthened publicity activities.

  • A new health paradigm may be evolving that would place emphasis on the positive aspects of diet, identifying components that are physiologically active and that contribute to the prevention of disease onset. It has been increasingly difficult to evaluate the impact of new bioactive materials and food products on the well being of society. Thus, testing systems for both health function and toxicity have become very elaborate, complex, and interrelated, making their interpretation difficult and open to controversy. To select the proper starting materials, to screen the appropriate health functionality and to determine the efficacy of product, a reliable, reproducible, sensitive and predictive assay is required. Particularly, in uitro assay is the first stage in preclinical test on physiologically active materials and have many advantages in terms of time, cost and convenience. However, several factors as well as some limitations should be considered in this assay system. This presentation, therefore, will address the use of in uitro assays for evaluating physiological functionality of foods coupled with general consideration.

Antitumor constituents from some Korean medicinal plants

  • For the screening of bioactive natural products, the benzene or methanol extracts from 93 medicinal plants of Korea were prepared, and tested for the cytotoxicity against L1210 cells and for the antitumor action (Bae et al., 1992 and 1996). Of 93 extracts tested, 6 samples showed a cytotoxicity in both benzene and methanol extract, 39 samples in benzene and 13 samples in methanol extract. The benzene extract of the root of Scutellaria indica L., Sophora fIavescens Solander ex Aiton, Carpesium abrotanoides L., Gymnaster koraiensis (Nakai) Kitamura, Pyrola japonica Klenze, and Forsythiae Fructus showed a potent cytotoxic activity. This observation led to isolate active cytotoxic components, some of which demonstrated some antitumor action. In addition, the structure-activity relationship was discussed.

  • Marine ecosystems are often characterized by a high biological diversity, and it corresponds to a high chemical diversity. Up to present, more than 20,000 new bioactive substances have been isolated from marine organisms, where considerable numbers of these naturally occurring derivatives are developed as potential candidates for pharmaceutical application. In this process, screening of natural products from marine organisms that could potentially inhibit the expression of metalloproteinases has gained a huge popularity. Cancer is considered as one of the deadliest diseases in the medical field. Matrix metalloproteinase (MMPs) can degrade extracellular matrix (ECM) components and play important roles in a variety of biological and pathological processes. Matrix metalloproteinase inhibitors (MMPIs) have been identified as potential therapeutic candidates for metastasis, arthritis, chronic inflammation and wrinkle formation.

Biocompatible Nanoparticles with Well-defined Surface Chemistry for Smart Drug Delivery

  • Many nanomaterials are being harnessed as critical components in various systems for biomedical applications including diagnosis, imaging, and drug delivery. Those systems necessitate biocompatibility and low toxcity within effective dose range while achieving enough efficacy. Even though many nanomaterials enjoy successful demonstrations in bioapplications, lack of biocompatibility and high cytotoxicity often become hurdles for practical bioapplications. On the other hand, it is important to achieve enough efficiency based on chemically well-defined systems with efforts to understand mechanism at molecular level. Here, we developvarious biocompatible nanomaterials based on simple procedure using dextran as both reducing agent and surface coating. Dextran is one of the popular biocompatible polymers that have been used for drug delivery and biosensors. Dextran coated nanomaterials showed excellent colloidal stability, flexible surface chemistry for conjugation of bioactive molecules and low cytotoxicity with successful demonstrations in various bioapplications.

  • I classified the bone graft material according to the component. Most bone graft material is composed of inorganic and organic constituent. Organic component such DBM is associated with osteoinduction. Inorganic components such as hydroxyapatite, $\beta$-TCP, calcium sulfate, bioactive glass, polymer are associated with osteoconduction. Autogenous bone graft is ideal material. We can select any biocompatible material for the restoration of small filling defect with intact bony wall. However, we should select first osteogenetic and osteoinductive material to regenerate the viable bone tissue.

복어간 폐기물로부터 Tetrodotoxin, DHA 및 EPA의 분리 (Separation of Tetrodotoxin, DHA and EPA from Pufferfish Liver Waste)

  • The present study was undertaken to separate the available components effectively, such as tetrodotoxin(TTX), docosahexaenoic acid(DHA, C22:6,ω-3) and eicosapentaenoic acid (EPA, C20:5,ω -3) from pufferfish liver waste, which are known to have high values as bioactive materials. By using ultrafiltration, it was possible to separate high contents of 68mg TTX from pufferfish liver waste. In contrast, by activated charcoal column, it was to obtain about 54mg TTX. The recovering ratios were 65.3% and 45.0% in the two different methods of ultrafiltration and activated charcoal column, respectively. From the results of HPLC and gas chromatography-mass spectrometry(GC-MS), the obtained toxins were identified to be TTX and its derivatives. In addition, it was also possible to obtain 72.3g DHA and 11.4g EPA from 1kg of pufferfish liver by high performance liquid chromatography (HPLC). These amounts of DHA and EPA were also 17.70% and 1.04% in the total lipid of pufferfish liver oil from analysis of gas chromatography(GC), respectively.

Inhibition of Homodimerization of Toll-like Receptor 4 by 6-Shogaol

  • Toll-like receptors (TLRs) play a critical role in sensing microbial components and inducing innate immune and inflammatory responses by recognizing invading microbial pathogens. Lipopolysaccharide-induced dimerization of TLR4 is required for the activation of downstream signaling pathways including nuclear factor-kappa B ($NF-{\kappa}B$). Therefore, TLR4 dimerization may be an early regulatory event in activating ligand-induced signaling pathways and induction of subsequent immune responses. Here, we report biochemical evidence that 6-shogaol, the most bioactive component of ginger, inhibits lipopolysaccharide-induced dimerization of TLR4 resulting in the inhibition of $NF-{\kappa}B$ activation and the expression of cyclooxygenase-2. Furthermore, we demonstrate that 6-shogaol can directly inhibit TLR-mediated signaling pathways at the receptor level. These results suggest that 6-shogaol can modulate TLR-mediated inflammatory responses, which may influence the risk of chronic inflammatory diseases.

Functionality of Dairy Foods on Osteoporosis

  • Osteoporosis is a systemic skeletal disease characterized by decreased bone mass, which results in a markedly increased risk of traumatic fractures. This disease is a worldwide health problem with a high prevalence. Recently, various dietary components have been found to minimize the risk of developing osteoporosis through their ability to stimulate bone formation and optimize bone health. Among them, probiotics and fermented milk can have beneficial effects to human health. Bioactive compounds derived from probiotics in fermented milk can especially modulate physiological functions related to bone health. Here, we review the evidence to support these insights into newly found functionality of dairy foods for osteoporosis prevention.

Lignans from the Stem Barks of Kalopanax septemlobus

  • As a part of an ongoing collaborative program to discover novel bioactive components of plant origin, the stem barks of Kalopanax septemlobus were extracted with MeOH, and successively partitioned with CH$_2$Cl$_2$, EtOAc, BuOH and water. Repeated column chromatographic separation of the CH$_2$Cl$_2$ fraction resulted in the isolation of four compounds. Their structures were identified as vladinol E (1), (-)-simulanol{4-[3-hydroxymethyl-5-((E)-3-hydroxypropenyl)-7-methoxy-2,3-dihydrobenzofuran-2-yl]-2,6-dimethoxy-phenol} (2), vladinol F (3), and (${\pm}$)-secoisolariciresinol (4). This is the first report on the isolation of these compounds from Kalopanax species.
