유제품의 기능성 펩타이드 (Health Functional Peptides from Milk Products)

  • Various peptides derived from food are among the most potent physiologically active agents known, and include anticancer peptides, angiotensin converting enzyme(ACE) inhibitor exhibiting antihypertension action, opioid peptides, antithrombotic peptides, hypocholesterolemic peptides, immunomodulators, calcium absorption enhancers, and other peptides. Hydrophobic peptides extracted from a Cheddar-type cheese slurry were fractionated by gel chromatography and repeated HPLC. A peptide fraction from HPLC showed high cytotoxicity on the tumor cell lines such as a human colon carcinoma, and comprised of Tyr, Ser, Leu, Gly, and others. Hypocholesterolemic peptides were isolated from peptic hydrolyzates of casein and soy proteins. Macropeptides of 1,000${\sim}$5,000 dalton were effective on reducing the cholesterol level of mouse serum. Peptides showing high Krigbaum hydrophobicity and ANS surface hydrophobicity resulted in high hypocholesterolemic effect and fecal steroid concentrations. Caseinomacropeptides(CMP) were isolated from whey powder and treated with soluble and immobilized trypsin to obtain antithrombotic peptides. One fraction from the CMP hydrolyzed with immobilized trypsin for 24h exhibited high antithrombotic activity with 52.5% inhibition of platelet aggregation. These result suggested that peptides from various milk products could be utilized as a good bioactive agents for developing health functional foods.

  • The microdilution assay recommended by NCCLS (National Committee for Clinical Laboratory Standards) is one of the standardized methods of antibiotic susceptibility test. This method has been widely used clinically to obtain MIC values of antibiotics on pathogenic microorganisms. It is more convenient, rapid and simple to test many samples than other test methods such as agar diffusion assay and broth macrodilution assay. The screening of antimicrobial agents from microbial extracts is too laborious in its process. Therefore, a number of screening methods having more simple procedure have been developed. In our laboratory, we applied microdilution assay for screening the antimicrobial agents. This assay showed dose-response results and was more sensitive than disc diffusion assay in our system. We tested 200 samples of microbial extracts originated from 100 microbial strains and selected several samples as potential candidates. In this report, we show that the microdilution assay is more convenient method in screeing of antibiotic susceptibility than those previously reported.

Antibacterial Activity of Sophoraflavanone G Isolated from the Roots of Sophora flavescens

  • This study investigated the antibacterial activities of sophoraflavanone G from Sophora flavescens in combination with two antimicrobial agents against oral bacteria. The combined effect of sophoraflavanone G and the antimicrobial agents was evaluated using the checkerboard method to obtain a fractional inhibitory concentration(FIC) index. The sophoraflavanone G+ampicillin(AM) combination was found to have a synergistic effect against S. mutans, S. sanguinis, S. sobrinus, S. gordonii, A. actinomycetemcomitans, F nucleatum, P. intermedia, and P. gingivalis, whereas the sophoraflavanone G+gentamicin(GM) combination had a synergistic effect against S. sanguinis, S. criceti, S. anginosus, A. actinomycetemcomitans, F nucleatum, P. intermedia, and P. gingivalis. Neither combination exhibited any antagonistic interactions(FIC index>4). In particular, the MICs/MBCs for all the bacteria were reduced to one-half$\sim$one-sixteenth as a result of the drug combinations. A synergistic interaction was also confirmed by time-kill studies for nine bacteria where the checkerboard suggested synergy. Thus, a strong bactericidal effect was exerted through the drug combinations, plus in vitro data suggested that sophoraflavanone G combined with other antibiotics may be microbiologically beneficial rather than antagonistic.

  • 본 연구는 상업용 Ortho MTA에 생체활성 유리 나노입자(bioactive glass nano particles)에 구리(Cu) 0.5, 1.0, 2.0, 4.0 wt%를 첨가하여 새로운 Cu-BGn MTA를 조성하고 세균부착실험을 통한 항균효과와 물성을 평가하였다. 경화시간과 압축강도는 ISO 6876(2012) 규격에 맞추어 직경 4 mm, 두께 6 mm 시편을 제작하여 산출하였으며, 항균효과는 S. mutans, E. faecalis 2개의 균주를 이용하여 평가하였다. 실험 결과 경화시간과 압축강도는 Cu-BGn 첨가와는 통계적으로 유의한 차이를 보이지 않았다(p>0.05). 항균실험 결과 대조군 Ortho MTA와 비교하여 Cu-BGn을 4.0 wt% 첨가한 S. mutans 실험군에서 낮은 부착 양상을 보였으며 통계적으로 유의한 차이가 나타났다(p<0.05). E. faecalis 실험군에서도 4.0 wt% 첨가한 군에서 통계적으로 유의한 차이를 보였다(p<0.05). 이는 Cu-BGn의 세균부착 억제 효과가 있음을 입증한 것이라 사료된다. 향후 구강환경을 재현시킬 수 있는 다양한 환경에서의 심도 있는 연구가 필요할 것으로 생각된다.

생체활성 유리 나노입자 첨가량에 따른 치면열구전색제의 물성평가와 세균부착 억제 효과 (Effect of Physical Properties and Bacterial Adherence Inhibition of Pit and Fissure Sealant Containing Bioactive Glass Nano Particles(BGn))

  • 본 연구는 불소 미방출 치면열구전색제 $Concise^{TM}$에 제조한 생체활성 유리 나노입자(bioactive glass nano particles: BGn)를 0.5, 1.0, 2.0 wt%를 첨가하여 새로운 치면열구전색제를 조성하고 세균부착실험을 통한 세균부착 억제 효과와 물성을 평가 하였다. 물흡수도와 용해도는 ISO 4049(2009) 규격에 맞추어 직경 10 mm, 두께 2 mm 시편을 제작하여 무게를 측정하여 산출하였으며 세균부착효과는 S. mutans, S. aureus, E. coli 3개의 균주를 이용하여 평가하였다. 실험 결과 물흡수도는 BGn 첨가가 증가할수록 높은 값을 보였으며 용해도는 첨가될수록 낮은 용해도를 보였다(p<0.05). 세균부착실험 결과 대조군 $Concise^{TM}$과 비교하여 BGn을 첨가한 S. mutans 실험군에서 다소 낮은 부착 양상을 보였으나 통계적 유의한 차이는 나타나지 않았으나, S. aureus 실험군과 E.coli 실험군에서는 통계적 유의한 차이를 보였다(p<0.05). 이는 BGn의 세균부착 억제 효과가 있음을 입증한 것이라 사료된다. 향후 BGn 첨가양에 따른 효율성과 폭 넓은 물성 연구가 필요할 것으로 사료된다.

Kitasatospora sp. MJM383 Strain Producing Two Antitumor Agents, Streptonigrin and Oxopropaline G

  • MJM383, a rare actinomycete sp. strain originated from Chinese soils, was isolated through an antimicrobial screening system. The analysis of 16S rDNA sequences and biochemical characterization determined the strain to belong to genus Kitasatospora. Both NMR and ESI mass data of its purified bioactive compounds revealed Kitasatospora sp. MJM383 to produce two antitumor agents, streptonigrin and oxopropaline G, which have been known to be produced from Streptomyces species. This is the first report to demonstrate the presence of antitumor agents produced by genus Kitasatospora.

Isolation and Structure Determination of a Proteasome Inhibitory Metabolite from a Culture of Scytonema hofmanni

  • Cyanobacteria, blue-green algae, are a rich source of bioactive secondary metabolites with many potential applications. The ubiquitin-proteasome proteolytic system plays an important role in selective protein degradation and regulates cellular events including apoptosis. Cancer cells are more sensitive to the proapoptotic effects of proteasome inhibition than normal cells. Thus, proteasome inhibitors can be potential anticancer agents. Cyanobacteria have been shown to be a rich source of highly effective inhibitors of proteases. A proteasome inhibitor was screened from an extract of the culture of Scytonema hofmanni on the basis of its inhibitory activity, which led to the isolation of nostodione A with an $IC_{50}$ value of 50${\mu}M$. Its structure was determined by spectroscopic methods such as $^{1}H$-NMR and ESI-MS spectral analyses.

유색작물의 Cyanidin-3-Glucoside 최적 추출분석 (Quantitative Analysis of Cyanidin-3-Glucoside from Purple-Colored Crops)

  • Anthocyanins are water-soluble glycosides and acylglycosides of anthocyanidins, having different color variations due to its substitution patterns. Anthocyanins, present in various fruits, vegetables and crops as natural colorant, have been well characterized for its bioactive properties, anti-oxidant, anti-cancer, anti-proliferative and anti-inflammatory properties. During extraction and purification, the factors, such as pH, temperature, oxygen, light, enzymes, nucleophilic agents, sugar derivatives and co-pigments, have affected on anthocyanin stability. For this reason, the extraction method should be thoroughly checked for the qualitative/quantitative analysis of anthocyanin in particular plant material. To identify the optimum extraction method of cyanidin-3-glucoside, major anthocyanin of dark purple-colored grains, Oryza sativa cv. Heugjinjubyeo, Phaselous vulgaris, Phynchosia gngularis, Sesamum indium, Rhynchosia nulubilis and Lablab purpureus, reversed-phase HPLC analysis using solvent system of acetonitrile, methanol and water were accomplished.

Recent Discovery of Bioactive Natural Products from Taiwanese Marine Invertebrates

  • The secondary metabolites from Taiwanese marine soft corals and sponges have attracted much attention because they possess considerable potential biological activities. To explore the origin of bioactivity, many cytotoxic natural products were isolated and characterized in the past few years. For examples, The lipophilic extracts from marine sponges Petrosia elastica and Ircinia formosana were found active against several human tumor cells. The investigation of the gorgonian Junceela has also resulted in the discovery of a series of new juncenolides. Bioassay-directed fractionation of Clavularia viridis yielded seven new prostanoids. These compounds have been tested and evaluated as potential antitumor agents. The soft corals of the genus Cespitularia produced novel secondary metabolites with diverse chemical structures and interesting biological activities. Four new norditerpenoids, designated cespitulactones and cespihypotins were isolated from Cespitularia hypotentaculata. Cespitulactones are novel structures having a bond cleavage between C-10 and C-11. In addition, three novel diterpenes were isolated from C. taeniata and designated cespitulactams A, B and C having a phenylethyl amino side chain.
