Immobilization and properties of Streptomyces sp. S56 endoinulase (Streptomyces sp. S56 endoinulase의 고정화 및 성질)

  • Endoinulase from Streptomyces sp. S56 was immobilized by adsorption on DEAE-cellulose in 0.01 M citrate-sodium phosphate buffer, pH 6.0 and the properties of immobilized and free enzymes were investigated. The immobilized enzyme preparation, having 40 inulase activity units per dried matrix, revealed the maximal activity at $pH\;4.5{\sim}5.5$ and $55{\sim}60^{\circ}C$ and were most stable at pH 6 and 45^{\circ}C$. The immobilization caused a drop in optimum pH and affinity toward inulin, a slight increase in optimum temperature, an important increase in thermal stability and maximum reaction velocity. The immobilized endoinulase hydrolyzed the tuber extract of jerusalem artichoke and inulin, mainly into fructose and inulobise, degrading 63 and 78% of the total sugar respectively, within 48 hrs in batch reactor.

  • Biomass has been concerned as one of the alternative energy resources because it is renewable, abundant worldwide, eco-friendly, and carbon neutral. Quercus variabilis has been studied to understand pyrolysis reaction characteristics, and to evaluate the efficiency of bio-energy production from fast pyrolysis. Quercus variabilis were fast pyrolyzed in a bubbling fluidized bed reactor at various reaction conditions. The effects of pyrolysis temperature between $400^{\circ}C$ and $550^{\circ}C$ on product yields were investigated. The yield of bio-oil was changed between 36.98 wt% and 39.14 wt%, and those of gas yield was 33.40 and 36.96 wt% with increasing reaction temperature. The higher heating value (HHV) of bio-oil at $500^{\circ}C$ ($3.0{\times}U_{mf}$) was 20.18 MJ/kg. The gas compositions were similar for all reaction conditions such as CO, $CO_2$ and $CH_4$, and $CO_2$ selectivity was the highest (37.16~50.94 mol%). The bio-oil has high selectivities for furfural, phenol and their derivatives such as 1-hydroxy-2-propanone, 2-methoxy-phenol, 1,2-benzendiol, 2,6-dimethoxy-phenol.

Effects of Organic Fertilizers on Growth and Yield in Liriope platyphylla WANG et TANG (유기질 비료의 사용에 따른 맥문동 생육 및 수량)

  • This experiment was carried out to investigate the effect of several organic fertilizers on the growth and root yield of Liriope platyphylla was planted 30 cm ${\time}$ 10 cm in field from 1998 to 1999. Six treatments of organic fertilizers applied : N.P.K, NPKC (N,P,K+Compost), NPKCO (N.P.K+Compost+Oilcake),NPKCOF (N.P.K+Compost+Oilcake+Fowldropping), NPKCOFP (N.P.K+Compost+ Oilcake+Fowldropping+plant ash), CO (Compost+Oilcake), COF (Compost+Oilcake+fowldropping). Organic matter, available phosphate, and MgO are NaO of soil increased in all of plots with the addition of organic fertilizers. The leaf length, number of leaves and of tillers were increased by the use of organic fertilizers compared to N.P.K fertilizers compared with N.P.K fertilization. The number of tubers were increased by using the organic fertilization N.P.K fertilization and showed higher in the soil depth of 0${\sim}$20 cm. The dry root yield was showed higher in organic fertilizers than in N.P.K fertilizer and highest dry root yield resulted from NPKOF 39% and NPKCOFP 49%.

Development of the Automatic Watering System for the Soybean Sprouting Equipment by Siphon and Well Bucket (싸이펀과 두레박을 이용한 콩나물 재배기용 자동 수주 시스템 개발)

  • The new watering system for the cabinet type of soybean sprouting equipment was developed. It was made of one siphon $({\Phi}35 mm)$, one well bucket $(13{\times}10{\times}10\;cm)$ that its weight-center is changed by water level, and bottle-neck water tank (bottom: $18.5^W{\times}32^L{\times}29^H\;cm$, top: $14^W{\times}14^L{\times}25^H\;cm)$. The watering system can be automatically watered more than 750 mL/sec without electric supply. The soybean sprouting equipment is consisted of the watering system and nine sprouting buckets. In addition siphon phenomena brought about easily by narrowing same as bottle-neck shape top portion of water tank that inserted a siphon. The equipment with the watering system may be produced more than three box a day in the condition of $25{\pm}1^{\circ}C$, and $6{\sim}7$ watering times a day.

Optimization of Enzyme Treatment Condition for Clarification of Pomegranate Extract (석류추출액의 청징화를 위한 효소처리조건 최적화)

  • Response surface methodology was used to investigate clarification characteristics (turbidity, brown color, soluble solid, total sugar and reducing sugar) of enzyme in pomegranate extract. Enzyme was treated at 16 conditions including independent variables of temperature ($35{\sim}55^{\circ}C$), time ($30{\sim}70\;min$) and concentration ($0.02{\sim}0.10%$) based on central composition design. Turbidity was decreased with increase of enzyme concentration, and the minimum value of turbidity was 0.04 (OD) when 0.08% enzyme was treated at $37.99^{\circ}C$ for 60.90 min. Total sugar was affected by all treatment conditions and the maximum value was 8.37% when 0.03% enzyme was treated at $39.28^{\circ}C$ for 42.04 min. Reducing sugar and soluble solid were largely affected by enzyme concentration, and the maximum value of reducing sugar was 7.22% when 0.02% enzyme was treated at $42.96^{\circ}C$ for 46.21 min. The maximum value of soluble solid was 8.13% when 0.02% enzyme was treated at $46.91^{\circ}C$ for 42.13 min.

Studies on the Post-hatching Development of the Testis in Korean Native Chickens (한국 재래 닭 부화 후 고환 발달에 관한 연구)

  • Changes in the chicken testis from hatching to adulthood were studied in Korean native chickens of 1, 2, 4, 6, 8, 10, 12, 14, 16, 18, 21, 24, 28, 32, 44, 52 and 64 weeks (n=13 chickens per group) of age. The present study was to investigate in more detail the post-hatching development of testis in Korean native chickens. Testes of chickens were fixed by whole body perfusion using a fixative containing 2.5% glutaraldehyde in cacodylate buffer, processed and embedded in Epon-araldite. Using $1{\mu}m$ sections stained with methylene blue-azure II, qualitative and quantitative(stereological) morphological studies were performed. Sperm production was measured by routine technique. The average volume of a testis of 1 week old Korean native chickens was determined as 0.015 g and the parameter increased linearly from 1 week to 21 weeks days (28.9 g), and did not change from 21 weeks to 64 weeks. The volume density of the seminiferous tubules increased with age from 32.6% at week 1 to 92.89% at week 64. The volume density of the interstitium represents 67.4% of the testicular parenchyma at week 1. This proportion progressively diminished during development to reach a value of 7.11% at week 64. Total sperm production per testis increased significantly from 18 weeks to 28 weeks and remained unchanged. Sperm production per 1 g testis increased significantly from 18 weeks to 28 weeks, did not change significantly from 28 weeks to 52 weeks, and declined significantly at 64 weeks of age. The average diameter of the seminiferous tubules gradually increased with age from 1 week $(42.4{\mu}m)$ to 21 weeks $(412.8{\mu}m)$. The length of the seminiferous tubules was 0.34 m at 1 week, increased significantly in subsequent age groups and reached 72.2 m by weeks 64. The stage of germ cell development in seminiferous tubules was classified as 1) spermatogonia $(1\sim8\;weeks)$, 2) spermatogonia and spermatocytes $(10\sim12\;weeks)$, 3) spermatogonia, spermatocytes and round spermatids $(14\sim16\;weeks)$, and 4) speramatogonia, spermatocytes, spermatids and spermatozoa $(18\sim64\;weeks)$. These results clarified the pattern of changes in the testicular development in Korean native chickens from hatching to adulthood as 1) neonatal-prepubertal $(1\sim12\;weeks)$, 2) puberty$(14\sim18\;weeks)$, and adult$(21\sim64\;weeks)$.

Material Budgets in the Youngsan River Estuary with Simple Box Model (영산강 하구해역에서의 단순 박스모델에 의한 물질수지)

  • Budgets of fresh water, salt, DIP and DIN in the Youngsan river estuary were estimated seasonally in order to clarify the characteristics of material cycling and flux of nutrients with a simple box model. Inflow volumes of freshwater into system was approximately $36.481{\times}10^6{\sim}663.634{\times}10^6m^3/month$ and existing water mass of freshwater in system calculated by salt budget was approximately $2.515{\times}10^6{\sim}5.812{\times}10^6m^3$. Mean residence time of freshwater was calculated to be about 0.26~2.03 day. water exchange $1,248{\times}10^6{\sim}9,489{\times}10^6m^3/month$ assumed with salinity between estuary and adjacent ocean. Inflow mass of DIN and DIN were approximately 76.63~1,149.91 ton/month and 2.91~61.22 ton/month, respectively. Residence times of DIP and DIN were calculated to be 0.45~1.10 day and 0.28~1.92 day, respectively. The ratio of water residence time versus DIP, DIN residence time was calculated that freshwater residence time was longer than DIP, DIN residence time except for summer season. Thus, We assume that circulation of Nutrients in the system will happen rapidly except for summer season. Specially DIP in Winter could assume to outer input source existence because of seawater inflow in system and high DIP concentration in open sea.

Development of Analytical Method of Polycyclic Aromatic Hydrocarbons Deposited on Tree Leaves by GC/MS (GC-MS 에 의한 나뭇잎에 침착된 다환방향족 탄화수소의 분석)

  • A new effective and economic method was developed, removing interferences such as chlorophyll and lipid from leaves with small amounts of reagents and solvents in order to analyse PAHs(Polycyclic Aromatic Hydrocarbons). The extract from a soxhlet containing $4{\sim}5g$ of leaves and 100ml of dichlormethane and refluxed for 20 hrs was concentrated and eluted with 60ml of a hexane:dichloromethane (1:1) mixture through a column of 9mm wide inner diameter and 130mm long, packed from the bottom with 2.5g of $Al_2O_3$, 1.5g of $SiO_2$and 2g of anhydrous $Na_2SO_4$. The eluent was concentrated and loaded on a GPC column of 20mm wide inner diameter and 280mm long, packed with 12g of Bio-beads. The column was washed with 37ml of the hexane:dichloromethane(1:1) mixture. Another 43ml of the mixture was eluted as a PAH fraction and collected. This eluent was concentrated under gentle nitrogen to $50{\mu}l$ and analysed using GC-MS. The recoveries, obtained by comparing with the amounts of the internal standards of deuterated PAHs were $43.3{\sim}107.5%$(RSD $2.2{\sim}9.5%$).

Comparison of Cardinal Temperatures of Lettuce Using Bilinear, Parabolic, and Beta Distribution Functions (선형, 쌍곡선과 Beta 함수를 이용한 상추의 주요 온도 비교)

  • The objective of this study was to estimate cardinal temperatures for germination of lettuce (Lactuca sativar L.) using bilinear, parabolic, and beta distribution functions. Seeds of lettuce were germinated in a growth chamber at 7 constant temperatures: 10, 14, 16, 20, 24, 28, and $32^{\circ}C$. Four replicates of 100 seeds were placed on two layers of filter paper in a 9 cm petri-dish. Radicle emergence of 1 mm was scored as germination. The time course of germination was modeled using a logistic function. These minimum, optimum, and maximum temperatures were estimated by regression of the inverse of time to 50% germination rate against the temperature gradient. In bilinear function, minimum, optimum, and maximum temperatures were $7.9^{\circ}C$, $23.3^{\circ}C$, and $28.0^{\circ}C$, respectively. In parabolic function, minimum, optimum, and maximum temperatures were $9.7^{\circ}C$, $19.5^{\circ}C$, and $29.4^{\circ}C$, respectively. In beta distribution function, minimum, optimum, and maximum temperatures were $3.7^{\circ}C$, $20.7^{\circ}C$ and $32.0^{\circ}C$, respectively. Minimum, optimum, and maximum ranges of temperatures were $3.7{\sim}9.7^{\circ}C$, $19.5{\sim}23.3^{\circ}C$, and $28.0{\sim}32.0^{\circ}C$, respectively.

Optimization of Ethanol Extraction Conditions for Effective Components from Gastrodia elata Blume (천마 유효성분의 에탄올 추출조건 최적화)

  • Gastrodia elata Blume (GEB) is a traditional herbal plant that has been used in Asian countries for centuries as an anticonvulsant analgesic, and a sedative for treating general paralysis, epilepsy, vertigo, and tetanus. This study was designed to optimize conditions for ethanol extracts or GEB by analyzing and monitoring the extraction characteristics with response surface methodology. The extract was used for analysis of the effective components of GEB. The estimated optimal conditions were 63.62% in ethanol of 5.06 mL/g in solvent per sample, and 6.25 hr in extract time. The optimal extraction conditions for $ \gamma-aminobutyric$ acid, were 45.52%, 5.67 mL/g, and 6.04 hr, while those for $\rho-hydroxybenzyl$ alcohol were 62.73%, 5.02 mL/g, and 5.95 hr. Regression equation was generated for each variables and then superimposed them, such as soluble solid, $ \gamma-aminobutyric$ acid and $\rho-hydroxybenzyl$ alcohol content thereby resulting in superimposed values of extinction conditions like $45\sim65%,\;5\sim7mL/g$ and $5\sim7$ hr, respectively.
