Storage Stability of Anthocyanin Pigment Isolated from a Wasted Grape Peels (거봉(Black Olympia) 포도과피로부터 분리된 Anthocyanin색소지 저장안정성)

  • To develop the use of natural pigment for food, anthocyanins were isolated from grape peels which were wasted much in korea, and their characteristics were as fellows. pH has a marked influences on the color of the grape peels anthocyanin solution(GPAS). At low pH the color of GPAS was more stable than high pH. With increasing pH the color gradually fades as colorless pseudobases are formed. It showed characteristic bathnochromic shift as the solution increased. Among the sugars tested, glucose showed the most protective effect on the color of GPAS to raise the color stability, while fructose showed an adverse effect. Orgarnic acid such as citric acid prevented the degradation of anthocyanin, but ascorbic acid lowered stability of color considerably. The effect of light on GPAS was found to be very deleterious. The pigment degradation can be minimized by shielding the light from the pigment solution.

  • The optimum cultural conditions for production of Monascus natural pigment by Monascus purpureus MK2 were investigated in submerged culture. This strain was showed the maximum production of monascus natural pigment in the optimal medium of 3.0% wheat flour, 0.15% $NaNO_3$, 0.2% $K_{2}HPO_4$ and 0.05% $MgSO_4{\cdot}7H_{2}O$, at pH 7.0. The maximum production of this pigment was achieved at $30^{\circ}C$ for 7 day cultivation under 130 rpm shaking. At optimal condition, Monascus purpureus MK2 produced 29.10, 36.84 and 48.92 units of yellow, orange and red pigment, respectively.

Embryonic Development of Siberian Sturgeon Acipenser baerii under Hatchery Conditions: An Image Guide with Embryological Descriptions

  • Normal embryonic development at a constant temperature ($18^{\circ}C$) has been described for the Siberian sturgeon Acipenser baerii (Acipenseriformes). Hormone-induced spawning and artificial insemination were performed to prepare embryonic batches for embryologic examination. After insemination, early cleavages of the Siberian sturgeon embryos continued for 7 h post-fertilization (HPF), showing the typical pattern of uneven holoblastic cleavage. Blastulation and gastrulation began at 9 HPF and 19 HPF, respectively. Epiboly formation (2/3 covered) was observed at 25 HPF during gastrulation. Neurulation was initiated with the formation of a slit-like neural groove from the blastopore at 33 HPF. During neurulation, the primary embryonic kidney (pronephros) and s-shaped heart developed. The embryos underwent progressive differentiation, which is typical of Acipenseriform species. A mass hatching was observed at 130 HPF, and the average total length of the hatched prolarvae was 10.5 mm. The hatched prolarvae possessed a typical pigment plug (yolk plug). The results of this study are valuable not only as a reference guide for the artificial propagation of Siberian sturgeon in hatcheries but also as the basis for the derivation of developmental gene expression assays for this species.

  • 본 연구에서는 미세조류에 광스트레스를 가해서 특정 색소의 증가를 유도하였다. 광스트레스를 미세조류인 N.oculata에 각 각 가했을 때, 안세라크산틴 6.6 배 제아크산틴 8.6 배, 증가함을 HPLC로 분석하였다. 이러한 광스트레스에 노출되면 크산토필 회로에서 비올라산틴이 안세라크산틴과 제아크산틴으로 역위되어 광보호를 하기 때문이다.

Lycopene-Induced Hydroxyl Radical Causes Oxidative DNA Damage in Escherichia coli

  • Lycopene, which is a well-known red carotenoid pigment, has been drawing scientific interest because of its potential biological functions. The current study reports that lycopene acts as a bactericidal agent by inducing reactive oxygen species (ROS)-mediated DNA damage in Escherichia coli. Lycopene treatment elevated the level of ROS-in particular, hydroxyl radicals ($^*OH$)-which can damage DNA in E. coli. Lycopene-induced DNA damage in bacteria was confirmed and we also observed cell filamentation caused by cell division arrest, an indirect marker of the DNA damage repair system, in lycopene-treated E. coli. Increased RecA expression was observed, indicating activation of the DNA repair system (SOS response). To summarize, lycopene exerts its antibacterial effects by inducing $^*OH$-mediated DNA damage that cannot be ameliorated by the SOS response. Lycopene may be a clinically useful adjuvant for current antimicrobial therapies.

Evaluation of Antioxidant Potential and UV Protective Properties of Four Bacterial Pigments

  • In the present study, four distinctly colored bacterial isolates that show intense pigmentation upon brief ultraviolet (UV) light exposure are chosen. The strains are identified as Micrococcus luteus (Milky yellow), Cryseobacterium pallidum (Yellow), Cryseobacterium spp. (Golden yellow), and Kocuria turfanensis (Pink) based on their morphological and 16S rDNA analysis. Moderate salinity (1.25%), 25-37℃ temperature, and pH of 7.2 are found to be the most favorable conditions of growth and pigment production for all the selected isolates. The pigments are extracted using methanol: chloroform (1:1) and the purity of the pigments are confirmed by high-performance liquid chromatography (HPLC) and thin-layer chromatography (TLC). Further, Fourier transform infrared (FTIR) and UV-Visible spectroscopy indicate their resemblance with carotenoids and flexirubin family. The antioxidant activities of the pigments are estimated, and, all the pigments have shown significant antioxidant efficacy in 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picryl-hydrazyl (DPPH), and ferric reducing antioxidant power (FRAP) assays. The UV protective property of the pigments is determined by cling-film assay, wherein, at least 25% of UV sensitive Escherichia coli survive with bio-pigments even after 90 seconds of UV exposure compared to control. The pigments also hold a good sun protective factor (SPF) value (1.5-4.9) which is calculated with the Mansur equation. Based on these results, it can be predicted that these bacterial pigments can be further developed into a promising antioxidant and UV-protectant for several biomedical applications.

Statistical Optimization of Medium Components for the Production of Prodigiosin by Hahella chejuensis KCTC 2396

  • Prodigiosin is a natural red pigment with algicidal activity against Cochlodinium polykrikoides, a major harmful red-tide microalga. To increase the yield of prodigiosin production by Hahella chejuensis KCTC 2396, significant medium components were determined using a two-level Plackett-Burman statistical design technique. Among 12 components included in basal medium, $NaHCO_3$, ${Na}_{2}{SiO}_{3}$, ${NH_4}{NO_3}$, ${Na}_{2}{SO}_{4}$ and $CaCl_2$ were determined to be important for prodigiosin production. The medium formulation was finally optimized using a Box-Behnken design as follows: 1% sucrose; 0.4% peptone; 0.1 % yeast extract; and (g/l): NaCl, 20.0; ${Na}_{2}{SO}_{4}$, 9.0; $CaCl_2$, 1.71; KCl, 0.4; and (mg/l): ${H_3}{BO_3}$, 10.0; KBr, 50.0; NaF, 2.0; $NaHCO_3$, 45.0; ${Na}_{2}{SiO}_{3}$, 4.5; ${NH_4}{NO_3}$, 4.5. The predicted maximum yield of prodigiosin in the optimized medium was 1.198 g/l by the Box-Behnken design, whereas the practical production was 1.495 g/l, which was three times higher than the basal medium (0.492 g/l).

  • The purpose of this study was to understand the current trend with regard to the material of jeans and the making method of jeans pattern and the characteristics of washing processing through the overall production status of jeans in casual brands and to offer the basic data for producing the high efficient jeans. The results were as follows. According to the survey of production status of jeans in casual brands, the fabric mixture was highest in the order of non-stretch denim 100% cotton, stretch denim cotton/spandex mix and denim with $1{\sim}2%$ weft direction spandex mix. The most frequently used processing method for denim was in the order of normal finishing, mercerization finishing, soft finishing and pigment finishing. The most frequently used method of washing finishing for jeans was in the order of forming by embossing washing, bio stone washing, normal washing, bio washing, and bio stone bleach washing. The average shrinkage was higher on waist circumference and pants length of warp direction rather than hips circumference, thigh circumference, hem circumference of weft direction.

A Novel Rapid Fungal Promoter Analysis System Using the Phosphopantetheinyl Transferase Gene, npgA, in Aspergillus nidulans

  • To develop a convenient promoter analysis system for fungi, a null-pigment mutant (NPG) of Aspergillus nidulans was used with the 4'-phosphopantetheinyl transferase (PPTase) gene, npgA, which restores the normal pigmentation in A. nidulans, as a new reporter gene. The functional organization of serially deleted promoter regions of the A. nidulans trpC gene and the Cryphonectria parasitica crp gene in filamentous fungi was representatively investigated to establish a novel fungal promoter assay system that depends on color complementation of the NPG mutant with the PPTase npgA gene. Several promoter regions of the trpC and crp genes were fused to the npgA gene containing the 1,034-bp open reading frame and the 966-bp 3' downstream region from the TAA, and the constructed fusions were introduced into the NPG mutant in A. nidulans to evaluate color recovery due to the transcriptional activity of the sequence elements. Serial deletion of the trpC and crp promoter regions in this PPTase reporter assay system reaffirmed results in previous reports by using the fungal transformation step without a laborious verification process. This approach suggests a more rapid and convenient system than conventional analyses for fungal gene expression studies.

Analytical Method for Determination of Laccaic Acids in Foods with HPLC-PDA and Monitoring (식품 중 락카인산 성분 분리정제를 통한 분석법 확립 및 실태조사)

  • Major components of lac coloring include laccaic acids A, B, C, and E. The Korean Food Additive Code regulates the use of lac coloring and prohibits its use in ten types of food products including natural food products. Since no commercial standards are available for laccaic acids A, B, C, and E, a standard for lac pigment itself was used to separate laccaic acids from the lac pigment molecule. A standard for each laccaic acid was then obtained by fractionation. To obtain pure lac pigment for use in food by High performance Liquid Chromatography Photo Diode Array (PDA), a C8 column yielded the best resolution among various tested columns and mobile phases. A qualitative analytical method using High Performance Liquid Chromatography (HPLC) Tandem Mass(LC-MS/MS) was developed. The conditions for fast and precise sample preparation begin with extraction using methanol and 0.3% ammonium phosphate, followed by concentration. The degree of precision observed for the analyses of ham, tomato juice and Red pepper paste was 0.3-13.1% (Relative Standard Deviation (RSD%)), degree of accuracy was 90.3-122.2% with r2=0.999 or above, and recovery rate was 91.6-114.9%. The limit of detection was 0.01-0.15 ㎍/mL, and the limits of quantitation ranged from 0.02 to 0.47 ㎍/mL. Lac pigment was not detected in 117 food products in the 10 food categories for which the use of lac pigment is banned. Multiple laccaic acids were detected in 105 food products in 6 food categories that are allowed to use lac color. Lac pigment concentrations range from 0.08 to 16.67 ㎍/mL.
