Modulation of the aqueous extract of Bupleuri radix on glycine-induced current in the acutely dissociated rat periaqueductal gray neurons

  • Bupleuri radix (Umbelliferae), the dried root of Bupleurum Chinense DC, has been clinically used to mitigate pain sensation. The descending pain control system consists of three major components, and modulation of pain in the periaqueductal gray is the most extensively studied descending pain control system. However, the relation of Bupleuri radix on the descending pain control system has not been clarified. In the present study, modulation of the aqueous extract of Bupleuri radix on glycine-induced ion current in the acutely dissociated periaqueductal gray neurons was investigated by using nystatin-perforated patch-clamp technique under voltage-clamp condition. In the present results, the glycine-induced ion current was significantly suppressed by 0.1 mg/ml Bupleuri radix, while treatment with $10^{-5}\;M$ naltrexone, opioid receptor antagonist, alleviated Bupleuri radix-induced inhibition on glycine-induced ion current. The present study showed that the aqueous extract of Bupleuri radix may activate descending pain control system through inhibition on glycine-induced ion current in the periaqueductal gray neurons and this effect is mediated by opioid receptors.

Effect of Microalgal Extracts of Tetraselmis suecica against UVB-Induced Photoaging in Human Skin Fibroblasts

  • Exposure of cells to ultraviolet B (UVB) radiation can induce production of free radicals and reactive oxygen species (ROS), which damage cellular components. In addition, these agents can stimulate the expression of matrix metalloproteinase (MMP) and decrease collagen synthesis in human skin cells. In this study, we examined the anti-photoaging effects of extracts of Tetraselmis suecica (W-TS). W-TS showed the strongest scavenging activity against 2,2-difenyl-1-picrylhydrazyl (DPPH) and peroxyl radicals, followed by superoxide anions from the xanthine/xanthine oxidase system. We observed that the levels of both intracellular ROS and lipid peroxidation significantly increased in UVB-irradiated human skin fibroblast cells. Furthermore, the activities of enzymatic antioxidants (e.g., superoxide dismutase) and the levels of non-enzymatic antioxidants (e.g., glutathione) significantly decreased in cells. However, W-TS pretreatment, at the maximum tested concentration, significantly decreased intracellular ROS and malondialdehyde (MDA) levels, and increased superoxide dismutase and glutathione levels in the cells. At this same concentration, W-TS did not show cytotoxicity. Type 1 procollagen and MMP-1 released were quantified using RT-PCR techniques. The results showed that W-TS protected type 1 procollagen against UVB-induced depletion in fibroblast cells in a dose-dependent manner via inhibition of UVB-induced MMP-1. Taken together, the results of the study suggest that W-TS effectively inhibits UVB-induced photoaging in skin fibroblasts by its strong anti-oxidant ability.

동일한 속 원핵생물들의 보존 유전자와 대사경로 (Conserved Genes and Metabolic Pathways in Prokaryotes of the Same Genus)

  • 원핵생물 분류의 기본단위인 종(species)의 동정에 16S rDNA가 사용되지만 한계가 있고 원핵생물의 속(genus)에 대한 연구가 많지 않다. 본 연구에서는 보존 유전자를 확보한 COG database와 대사경로를 확보한 MetaCyc database에 공통적인 원핵생물 중 속이 같고 종이 다른 13개 속 28개의 원핵생물을 대상으로 속 수준에서 연구하였다. 전체 유전자에서 core-genome인 속 보존 유전자의 비율은 최저 27.62%(Nostoc 속)에서 71.76%(Spiribacter 속)의 범위로 평균 46.72%였다. 각 원핵생물에서 core-genome의 비율이 낮으면 특이한 생명현상을 보이거나 서식지가 다양할 수 있을 것이다. 속 수준의 공통 대사경로의 비율은 최저 58.79%(Clostridium 속)에서 최대 96.31%(Mycoplasma 속), 평균 75.86%로 core-genome의 비율보다 높았다. 비교대상을 확장하면 속 특이 보존 유전자와 대사경로는 확인할 수 없었다. 보존 유전자와 대사경로 보유 계통수에서는 대체로 같은 속의 구성원들이 가장 인접하였으며, Bacillus속과 Clostridium 속이 그룹을 형성하였고, 고세균끼리 그룹을 형성하였다. 보존 유전자 보유계통수에서는 Acidobacteria, Cyanobacteria, Proteobacteria 문(phylum)의 Granulicella, Nostoc, Bradyrhizobium의 3개 속이 하나의 그룹을 형성하였다. 본 연구 결과는 (i) 각 계통 단계에서 보존유전자와 대사경로의 확인, (ii) 수평적 유전자 전달 또는 부위 지정 돌연변이를 통한 균주의 개선 등의 분야에 기초자료로 활용될 수 있을 것이다.

생체신호를 통한 헬스케어 디바이스 기술 동향 연구 (A Study on the Trend of Healthcare Device Technology by Biometric Signal)

  • 최근 개인의 생체 데이터를 수집·활용하여 적시에 효과적인 예방과 치료를 제공하는 맞춤형 의료 및 서비스가 가능하며, 사용자의 헬스케어에 활용되고 있다. 실시간 건강관리 서비스로 개발하여 스마트폰, PC 태블릿 등을 활용하여 각 개인에게 정보를 제공·수집된 데이터를 통해 경보하고 관리자에게 분석 데이터 및 결과를 알려서 양방향 커뮤니케이션을 지원하며, 이에 따른 건강관리를 구성할 수 있는 시점에 직면하고 있다. 본 연구에서는 생체신호를 통한 헬스케어 디바이스와 실시간 건강관리 시스템을 포함하는 스마트 웨어러블 헬스케어 기술 동향을 분석하여 현재 시점에서의 기술개발 특허 출원에 대비하고자 한다. 스마트 웨어러블 헬스케어 기술 관련 특허는 연구개발 집중도가 비교적 높은 것으로 조사되었다. 분석 구간 전반에 걸쳐 한국은 실시간 헬스케어 시스템에 대한 특허 활동에 집중하고 있으며, 미국 및 유럽은 생체기술을 통한 헬스케어 디바이스에 대한 특허 활동을, 일본은 생체기술을 통한 헬스케어 디바이스 및 시스템 전반에 걸쳐 특허 활동을 활발하게 하는 것으로 나타났다. 생체기술을 통한 헬스케어 디바이스와 실시간 건강관리 시스템에 대한 특허는 최근 한국에서 시장을 주도하고 있으며, 해당 기술에 대한 특허 및 시장을 확보하고 있는 것으로 나타남에 따라 스마트 웨어러블 헬스케어 기술은 시장진입 장벽이 높은 분야로 판단된다. 향후 기술을 상업화시킬 계획이 있다면 제품의 형상, 제조방법 및 기타 관련 기술 시스템에 대한 특허권을 획득하여 특허 포트폴리오를 구축하는 것이 중요하다.

금강 하구 자연수계 생물체의 군집 분석 및 질병 원인체 검사 (Community Analysis and Pathogen Monitoring in Wild Cyprinid Fish and Crustaceans in the Geum River Estuary)

  • Freshwater farms are primarily located adjacent to rivers and lakes, facilitating the introduction and spread of pathogens into natural systems. Therefore, it is necessary to continuously monitor natural aquatic organisms, the breeding environment, and infection rates by pathogenic organisms. Fish and crustaceans were sampled 4 times in the Geum River estuary in 2016. The samples were analyzed for the presence of pathogens for reportable communicable diseases, including KHVD (koi herpesvirus disease), SVC (spring viraemia of carp), EUS (epizootic ulcerative syndrome) and WSD (white spot disease); parasite abundance was also examined. The dominant fish species were deep body bitterling Acanthorhodes macropterus (21.4%), followed by skygager Erythroculter erythropterus (12.7%). For crustaceans, Palaemon paucidens and Chinese mitten crab Eriocheir sinensis were dominant. Sixty fish and 36 crustacean species were examined for reportable communicable diseases. When using a specific primer set for each disease, PCR analysis did not detect any reportable communicable diseases in the samples. Some instances of Dactylogyrus, copepods, nematodes and metacercaria were detected. However, the PCR results indicated that the metacercaria were not Clonorchis sinensis.

Effects of White Radish (Raphanus sativus) Enzyme Extract on Hepatotoxicity

  • Raphanus sativus (Cruciferaceae), commonly known as radish is widely available throughout the world. From antiquity it has been used in folk medicine as a natural drug against many toxicants. The present study was designed to evaluate the hepatoprotective activity of radish (Raphanus sativus) enzyme extract (REE) in vitro and in vivo test. The $IC_{50}$ values of REE in human liver derived HepG2 cells was over 5,000 ${\mu}g/ml$ in tested maximum concentration. The effect of REE to protect tacrine-induced cytotoxicity in HepG2 cells was evaluated by MTT assay. REE showed their hepatoprotective activities on tacrine-induced cytotoxicity and the $EC_{50}$ value was 1,250 ${\mu}g/ml$. Silymarin, an antihepatotoxic agent used as a positive control exhibited 59.7% hepatoprotective activity at 100 ${\mu}g/ml$. Moreover, we tested the effect of REE on carbon tetrachloride ($CCl_4$)-induced liver toxicity in rats. REE at dose of 50 and 100 mg/kg and silymarin at dose of 50 mg/kg were orally administered to $CCl_4$-treated rats. The results showed that REE and silymarin significantly reduced the elevated levels of serum enzyme markers induced by $CCl_4$. The biochemical data were supported by evaluation with liver histopathology. These findings suggest that REE, can significantly diminish hepatic damage by toxic agent such as tacrine or $CCl_4$.

밀리타리스 동충하초 열수추출물의 멜라닌 분비 억제능 효과 (Inhibitory Effects of Melanin Secretion on B16 Melanoma cell of Cordyceps militaris Water Extract)

  • The present study aims to evaluate Cordyceps militaris water extract (CMWE) with a view to develop melanogenesis inhibitors. Inhibitory activities of CMWE against tyrosinase, L-DOPA(L-3,4-dihydroxyphenylalanine) oxidation, and melanin biosynthesis in B16 mouse melanoma cells were investigated. CMWE, at $5000\;{\mu}g/ml$, inhibited tyrosinase activity of 71% and DOPA oxidation of 40% as reacting with L-DOPA. Furthermore, B16 mouse melanoma cell survived over 50% from low to high dose on MTT assay, and CMWE markedly inhibited (> 50%) melanin synthesis at $5000\;{\mu}g/ml$. The inhibitory effect of CMWE on melanogenesis was attributed to enhancement of tyrosinase degradation. Key enzyme of melanin biosynthesis is tyrosinase which catalyses a beginning step from tyrosine to DOPA quinine and melanin formation step, respectively. These results indicated that CMWE may be a potential source of novel whitening agents for cosmetic or therapeutic application.

Mono Sodium Glutamate 발효로 얻은 GABA 코팅 쌀에 의한 수면유도 효과 (Sleep-Induction Effects of GABA Coated Rice from Fermentation of Mono Sodium Glutamate)

  • GABA는 포유류의 신경전달물질의 분비를 억제하며, 신경안정 작용, 스트레스 해소, 우울증 완화, 불면 등에 효과가 있는 것으로 알려져 있다. 곡류식품에 존재하는 GABA 함량은 일반미 1~4 mg/100 g, 현미 4~8 mg/100 g으로 알려져 있지만, 그 함량이 낮아 자연 식품으로부터 섭취하는 양으로는 생리활성을 기대하기 어렵다. 본 연구에서는 mono sodium glutamate (MSG)발효로 얻은 GABA 코팅 쌀이 신경안정제 뿐만 아니라 수면유도제로 쓰이는 melatonin과 serotonin의 생성 촉진 효과를 알아보았다. 그 결과 melatonin의 경우 GABA (25 mg/mL) 투여군은 $3.515{\pm}0.149pg/mL$, 일반미 공급군의 경우 $2.632{\pm}0.188pg/mL$이며, GABA 코팅 쌀 공급군은 $3.578{\pm}0.158pg/mL$로 나타났다. Serotonin 함량을 측정한 결과 GABA (25 mg/mL), 일반미 공급군은 각각 $5.183{\pm}0.142ng/mL$, $4.784{\pm}0.108ng/mL$가 측정되었으며 GABA 코팅 쌀을 공급한 실험군은 $5.918{\pm}0.169ng/mL$의 serotonin이 생성되었다. GABA 코팅 쌀의 지속적인 섭취로 인해 melatonin과 serotonin은 유의적인 증가는 있었지만, 누적효과는 없었다. 기능성 식품으로 개발된 GABA 코팅 쌀을 지속적으로 섭취하면 일반미에서 섭취하여 얻을 수 있는 melatonin과 serotonin의 유도효과를 얻을 수 있고, GABA 25 mg/mL를 섭취하는 것과 비슷한 효과를 나타낼 수 있다고 사료된다.

Cryoprotective Properties of Exopolysaccharide (P-21653) Produced by the Antarctic Bacterium, Pseudoalteromonas arctica KOPRI 21653

  • Twenty-five bacterial strains that secrete mucous materials were isolated from sediment obtained from King George Island, Antarctica. Seven of these strains proved capable of producing cryoprotective exopolysaccharides. The strain KOPRI 21653 was selected for the further study of an anti-ice-nucleating polysaccharide (ANP), which originated from a polar region. KOPRI 21653 was identified as Pseudoalteromonas arctica as the result of 16S rRNA analysis. The exopolysaccharide, P-21653, was purified completely from the KOPRI 21653 cell culture via column chromatography and protease treatment. The principal sugar components of P-21653 were determined to be galactose and glucose, at a ratio of 1:1.5, via GC-MS analysis. The cryoprotective activity of P-21653 was characterized via an E. coli viability test. In the presence of 0.1% (w/v) P-21653, the survival ratio of E. coli cells was as high as 82.6% over three repeated freeze-thaw cycles. The survival ratio decreased drastically to 71.5 and 48.1 %, respectively, in five and seven repeated cycle conditions; however, the survival ratios were greater over three (96.6-92.1%) to seven (100.5-91.6%) freeze-thaw cycles in the presence of 0.5 and 1.0% (w/v) P-21653. In addition, at much lower concentrations (0.1-1.0%), P-21653 resulted in survival ratios (83.1-98.4%) similar to those of two commercially available cryoprotectants ($V_{EG}$ plus X-1000, 92.9% and $V_{M3}$, 95.3%), which were utilized at the recommended concentrations (90%). The biochemical characteristics of exopolysaccharide P-21653 reflect that this compound may be developed as a useful cryoprotectant for use in medical applications and in the food industry.

Fructus Amomi Cardamomi Extract Inhibits Coxsackievirus-B3 Induced Myocarditis in a Murine Myocarditis Model

  • Coxsackievirus B3 (CVB3) is the main cause of acute myocarditis and dilated cardiomyopathy. Plant extracts are considered as useful materials to develop new antiviral drugs. We had previously selected candidate plant extracts, which showed anti-inflammatory effects. We examined the antiviral effects by using a HeLa cell survival assay. Among these extracts, we chose the Amomi Cardamomi (Amomi) extract, which showed strong antiviral effect and preserved cell survival in CVB3 infection. We investigated the mechanisms underlying the ability of Amomi extract to inhibit CVB3 infection and replication. HeLa cells were infected by CVB3 with or without Amomi extract. Erk and Akt activities, and their correlation with virus replication were observed. Live virus titers in cell supernatants and viral positive- and negative-strand RNA amplification were measured. Amomi extract significantly increased HeLa cell survival in different concentrations ($100-10{\mu}g/ml$). CVB3 capsid protein VP1 expression (76%) and viral protease 2A-induced eIF4G1 cleavage (70%) were significantly decreased in Amomi extract ($100{\mu}g/ml$) treated cells. The levels of positive- (20%) and negative-strand (80%) RNA were dramatically decreased compared with the control, as revealed by reverse transcription-PCR. In addition, Amomi extract improved mice survival (51% vs 26%) and dramatically reduced heart inflammation in a CVB3-induced myocarditis mouse model. These results suggested that Amomi extract significantly inhibited Enterovirus replication and myocarditis damage. Amomi may be developed as a therapeutic drug for Enterovirus.
