Anti-skin Aging Potential of Alcoholic Extract of Phragmites communis Rhizome

  • Chronological aging and photoaging affect appearance, causing wrinkles, pigmentation, texture changes, and loss of elasticity in the skin. Phragmites communis is a tall perennial herb used for its high nutritional value and for medicinal purposes, such as relief from fever and vomiting and facilitation of diuresis. In this study, we investigated the effects of ethanol extract of P. communis rhizome (PCE) on skin aging. The total flavonoid and total phenolic content in PCE were 2.92 ± 0.007 ㎍ of quercetin equivalents (QE) and 231.8 ± 0.001 ㎍ of gallic acid equivalents (GAE) per 100 mg of dried extract (n = 3). The half-maximal inhibitory concentration (IC50) values of PCE for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) and hydrogen peroxide scavenging activities were 0.96 and 0.97 mg/mL, respectively. PCE showed inhibitory effects on tyrosinase when L-tyrosine (IC50 = 1.25 mg/mL) and L-3,4-dihydroxyphenylalanine (IC50 = 0.92 mg/mL) were used as substrates. PCE treatment up to 200 ㎍/mL for 24 h did not cause any significant cytotoxicity in B16F10 melanocytes, human dermal fibroblasts (HDFs), and HaCaT keratinocytes. In B16F10 melanocytes, PCE (25 and 50 ㎍ /mL) inhibited melanin production and cellular tyrosinase activity after challenge with α-melanocyte-stimulating hormone (α-MSH; p < 0.05). In HDFs, PCE suppressed the mRNA expression of matrix metalloproteinase-1 (MMP-1) and reduced the activity of elastase (p < 0.05). In addition, ultraviolet B (UVB)-mediated downregulation of hyaluronic acid synthase-2 gene expression in HaCaT keratinocytes was also effectively suppressed by PCE treatment. Overall, our results showed that PCE has potential anti-skin aging activity associated with the suppression of hyperpigmentation, wrinkle formation, and reduction in dryness. PCE is a promising candidate for the development of an anti-skin aging cosmetic ingredient.

쥐 흑색종 세포에서 산화적 스트레스에 의한 MMPs의 발현과 활성에 대한 흑도라지 추출물의 억제 효과 (Inhibitory Effect of Aged Black Platycodi Radix Extract on Expression and Activation of Matrix Metalloproteinases in Oxidative-stressed Melanoma Cells)

  • 도라지는 민간에서 항궤양, 항진통, 항알러지, 혈관과 히스타민 억제 및 항산화 효과가 있는 것으로 알려져 있고, 그 뿌리인 길경은 아시아에서 식용으로 즐겨먹는 식품 소재로 사포닌이 과량 함유되어 있는 것으로 보고 되어 있으나, 그 작용기전은 과학적으로 해명되지 않았다. 본 연구에서 처음으로 흑도라지가 특정한 공정기술을 이용하여 개발되었으며, 산화적 스트레스 및 MMPs) 대한 ABPRE의 억제 효과를 조사하였다. ABPRE의 항산화효과를 조사하기 위해서 DPPH radical, hydroxyl radical, reducing power, hydrogen peroxide, lipid peroxidation 및 DNA 산화에 대한 연구를 수행하였다. 연구 결과 ABPRE는 DPPH radical 소거에는 효과를 나타내었으나 지질과산화에 의한 malondialdehyde의 생성은 억제하지 않았다. 그러나, ABPRE의 존재 하에서 산화적 DNA 손상이 억제되었다. 또한 gelatin zymography 및 western blot 분석에서 ABPRE는 PMS (phenazine methosulfate)에 의해서 자극된 MMP-2의 발현과 활성을 감소시켰으며, ABPRE는 항산화 효과 및 tyrosinase 발현 억제에 의한 L-DOPA로 유발된 melanin 생성을 저해 하였을 뿐만 아니라 Nrf2에 의해서 조절되는 항산화 효소인 SOD-1과 SOD-2의 발현을 증가시키는 것으로 나타났다. 그러므로 ABPRE는 항산화와 관련 있는 암전이의 예방을 위한 항암제의 후보 소재로 그 가능성이 기대된다.

Morroniside Protects C2C12 Myoblasts from Oxidative Damage Caused by ROS-Mediated Mitochondrial Damage and Induction of Endoplasmic Reticulum Stress

  • Oxidative stress contributes to the onset of chronic diseases in various organs, including muscles. Morroniside, a type of iridoid glycoside contained in Cornus officinalis, is reported to have advantages as a natural compound that prevents various diseases. However, the question of whether this phytochemical exerts any inhibitory effect against oxidative stress in muscle cells has not been well reported. Therefore, the current study aimed to evaluate whether morroniside can protect against oxidative damage induced by hydrogen peroxide (H2O2) in murine C2C12 myoblasts. Our results demonstrate that morroniside pretreatment was able to inhibit cytotoxicity while suppressing H2O2-induced DNA damage and apoptosis. Morroniside also significantly improved the antioxidant capacity in H2O2-challenged C2C12 cells by blocking the production of cellular reactive oxygen species and mitochondrial superoxide and increasing glutathione production. In addition, H2O2-induced mitochondrial damage and endoplasmic reticulum (ER) stress were effectively attenuated by morroniside pretreatment, inhibiting cytoplasmic leakage of cytochrome c and expression of ER stress-related proteins. Furthermore, morroniside neutralized H2O2-mediated calcium (Ca2+) overload in mitochondria and mitigated the expression of calpains, cytosolic Ca2+-dependent proteases. Collectively, these findings demonstrate that morroniside protected against mitochondrial impairment and Ca2+-mediated ER stress by minimizing oxidative stress, thereby inhibiting H2O2-induced cytotoxicity in C2C12 myoblasts.

Regulation of toll-like receptors expression in muscle cells by exercise-induced stress

  • Objective: This study investigates the expression patterns of toll-like receptors (TLRs) and intracellular mediators in horse muscle cells after exercise, and the relationship between TLRS expression in stressed horse muscle cells and immune cell migration toward them. Methods: The expression patterns of the TLRs (TLR2, TLR4, and TLR8) and downstream signaling pathway-related genes (myeloid differentiation primary response 88 [MYD88]; activating transcription factor 3 [ATF3]) are examined in horse tissues, and horse peripheral blood mononuclear cells (PBMCs), polymorphonuclear cells (PMNs) and muscles in response to exercise, using the quantitative reverse transcription-polymerase chain reaction (qPCR). Expressions of chemokine receptor genes, i.e., C-X-C motif chemokine receptor 2 (CXCR2) and C-C motif chemokine receptor 5 (CCR5), are studied in PBMCs and PMNs. A horse muscle cell line is developed by transfecting SV-T antigen into fetal muscle cells, followed by examination of muscle-specific genes. Horse muscle cells are treated with stressors, i.e., cortisol, hydrogen peroxide (H2O2), and heat, to mimic stress conditions in vitro, and the expression of TLR4 and TLR8 are examined in stressed muscle cells, in addition to migration activity of PBMCs toward stressed muscle cells. Results: The qPCR revealed that TLR4 message was expressed in cerebrum, cerebellum, thymus, lung, liver, kidney, and muscle, whereas TLR8 expressed in thymus, lung, and kidney, while TLR2 expressed in thymus, lung, and kidney. Expressions of TLRs, i.e., TLR4 and TLR8, and mediators, i.e., MYD88 and ATF3, were upregulated in muscle, PBMCs and PMNs in response to exercise. Expressions of CXCR2 and CCR5 were also upregulated in PBMCs and PMNs after exercise. In the muscle cell line, TLR4 and TLR8 expressions were upregulated when cells were treated with stressors such as cortisol, H2O2, and heat. Migration of PBMCs toward stressed muscle cells was increased by exercise and oxidative stresses, and combinations of these. Treatment with methylsulfonylmethane (MSM), an antioxidant on stressed muscle cells, reduced migration of PBMCs toward stressed muscle cells. Conclusion: In this study, we have successfully cultured horse skeletal muscle cells, isolated horse PBMCs, and established an in vitro system for studying stress-related gene expressions and function. Expression of TLR4, TLR8, CXCR2, and CCR5 in horse muscle cells was higher in response to stressors such as cortisol, H2O2, and heat, or combinations of these. In addition, migration of PBMCs toward muscle cells was increased when muscle cells were under stress, but inhibition of reactive oxygen species by MSM modulated migratory activity of PBMCs to stressed muscle cells. Further study is necessary to investigate the biological function(s) of the TLR gene family in horse muscle cells.

하이브리드 수열탄화기술을 이용한 캄보디아 망고 폐기물 고형연료화 실증플랜트 (2T/day) 제안 (Proposal of a Pilot Plant (2T/day) for Solid Fuel Conversion of Cambodian Mango Waste Using Hybrid Hydrothermal Carbonization Technology)

  • 하이브리드수열탄화 (Hybrid HTC) 기술은 2가지 이상의 유기성폐기물을 혼합한 특허 받은 열역학 공정으로 공정온도 180~250℃, 압력 20~40 bar에서 반응시간이 2시간 이내이며 에너지 소비가 적고, 폐기물의 부피감소 및 악취 저감효과가 크다. 폐기물 중 대부분의 탄소가 최종 생성물에 축적되므로 유기성 폐기물 고형연료화에 가장 적합한 기술로 평가받고 있다. 본 연구에서는 하이브리드 수열탄화기술을 활용하여 캄보디아 망고 폐기물을 대상으로 온도 및 반응시간의 변화에 따라 발열량 및 수율 등에 미치는 영향에 대하여 평가하였다. 본 연구를 통해 공정변수를 최적화하고, 전공정플랜트의 에너지 효율성을 향상시킬 수 있으며, 수연탄화기술에서 분해되어 가스가 생성되는데 이때 수소(H2) 및 메탄(CH4) 등 제조 및 생산기술개발을 할 수 있다. 본 연구 결과를 토대로 망고폐기물(2t/day)실증 물질수지 및 에너지 수지 도출과 함께 경제성도 평가하였다.
