Chemistry and Biology of Ras Farnesyltransferase

  • Mutated forms of ras are found in many human tumors and the rate of incidence is significantly higher in colon and pancreatic cancers. The protein product from the ras oncogene is a small G-protein, $p21^{ras}{\;}(Ras)$ that is known to playa key role in the signal transduction cascade and cell differentiation and proliferation. Mutated Ras is unable to regulate itself and remains constantly activated, leading to uncontrolled cell growth. The function of Ras in signal transduction requires its location near the growth factor receptor at the cell membrane. However, Ras does not have a transmembrane domain. Ras requires farnesylation to increase its hydrophobicity and subsequent plasma membrane association for its transforming activity. This key post-translational modification is catalyzed by the enzyme Ras farnesyltransferase (FTase), which transfers a farnesyl group from farnesylpyrophosphate to the C-terminal cysteine of the Ras protein. The requirement has focused attention on FTase as a target for therapeutic intervention. Selective inhibition of FTase will prevent Ras protein from association with the plasma membrane, leading to a disruption of oncogenic Ras function.

  • Monoclonal antibodies (Mabs) have been used as diagnostic and analytical reagents since hybridoma technology was invented in 1975. In recent years, antibodies have become increasingly accepted as therapeutics for human diseases, particularly for cancer, viral infection and autoimmune disorders. An indication of the emerging significance of antibody-based therapeutics is that over a third of the proteins currently undergoing clinical trials in the United States are antibodies. Until the late 1980's, antibody technology relied primarily on animal immunization and the expression of engineered antibodies. However, the development of methods for the expression of antibody fragments in bacteria and powerful techniques for screening combinatorial libraries, together with the accumulating structure-function data base of antibodies, have opened unlimited opportunities for the engineering of antibodies with tailor-made properties for specific applications. Antibodies of low immunogenicity, suitable for human therapy and in vivo diagnosis, can now be developed with relative ease. Here, antibody structure-function and antibody engineering technologies are described.

The Factors influencing Customer Satisfaction with and Revisiting Coffee Shops in Korea: The Moderating Roles of Psychological Value

  • This study aims to find what attributes of consumer satisfaction are more important when consumers choose coffee shops. Factors when customers choose coffee shops that are considered to be more crucial, such as taste, price, brand, and atmosphere, were tested and also relations between satisfaction and revisit were studied. As a result, factors as 'taste', 'price', 'brand', and 'atmosphere' were found to significantly affect satisfaction; in addition, the path that satisfaction leads to revisit was found to be significant. However, consumers' coffee shop selection attributes differed depending on their psychological consumption value. The path-coefficients from taste and price to satisfaction were more significant in the function-oriented group, meanwhile the path-coefficient from brand to satisfaction was significant in the emotion-oriented group (+) and the function-oriented group (-). The results of this study suggest attributes of selecting coffee shops and provide meaningful implications of consumer value when they choose the attributes.

A BIO-ECONOMIC MODEL OF TWO-PREY ONE-PREDATOR SYSTEM

  • We propose a model based on Lotka-Volterra dynamics with two competing spices which are affected not only by harvesting but also by the presence of a predator, the third species. Hyperbolic and linear response functions are considered. We derive the conditions for global stability of the system using Lyapunov function. The optimal harvest policy is studied and the solution is derived in the interior equilibrium case using Pontryagin's maximal principle. Finally, some numerical examples are discussed. The nature of variations in the two prey species and one predator species is studied extensively through graphical illustrations.

Assembly and Function of Seed Endophytes in Response to Environmental Stress

  • Seeds are colonized by diverse microorganisms that can improve the growth and stress resistance of host plants. Although understanding the mechanisms of plant endophyte-host plant interactions is increasing, much of this knowledge does not come from seed endophytes, particularly under environmental stress that the plant host grows to face, including biotic (e.g., pathogens, herbivores and insects) and abiotic factors (e.g., drought, heavy metals and salt). In this article, we first provided a framework for the assembly and function of seed endophytes and discussed the sources and assembly process of seed endophytes. Following that, we reviewed the impact of environmental factors on the assembly of seed endophytes. Lastly, we explored recent advances in the growth promotion and stress resistance enhancement of plants, functioning by seed endophytes under various biotic and abiotic stressors.

$Bio-pin^{(R)}$을 이용한 고정성 보철물의 탈락저항강도의 비교연구 (A COMPARATIVE STUDY OF THE RESISTANCE TO DISLODGEMENT OF FIXED PROSTHESES USING $BIO-PIN^{(R)}$)

  • Statement of problem. The current trend in prosthodontics is the adoption of a conservative approach to preparing dental prostheses by minimizing the amount of sound tooth structure removal during preparation. Purpose. The major disadvantage of the adhesion bridge is the inherently poor resistance to dislodgement that its use in areas subjected to high occlusal load is limited. The purpose of this study was to compare the dislodgement resistance of $Bio-pin^{(R)}$, conventional 3-unit and adhesion bridges. Material and methods. The experimental groups were classified as follows : Group I : 3-unit bridge cemented using $Super-Bond^{(R)}$ C&B Group II : Adhesion bridge cemented using $Super-Bond^{(R)}$ C&B Group III : $Bio-pin^{(R)}$ design adhesion bridge without incorporation of $Bio-pin^{(R)}$ (cemented using $Super-Bond^{(R)}$ C&B) Group IV-1 : $Bio-pin^{(R)}$ retained adhesion bridge incorporating a single $Bio-pin^{(R)}$ (cemented using $Super-Bond^{(R)}$ C&B) Group IV-2 : $Bio-pin^{(R)}$ retained adhesion bridge incorporating a single $Bio-pin^{(R)}$ (cemented using $Panavia^{(R)}$ F) Group V : $Bio-pin^{(R)}$ retained adhesion bridge incorporating two $Bio-pins^{(R)}$ (cemented using $Super-Bond^{(R)}$ C&B) Results. The results of this study were as follows : 1. Significant differences in dislodgement resistance of the restorations were found between Group I, Group II and Group III (p<0.05). No significant differences in dislodgement resistance of the restorations were observed between Group I Group IV-1 and Group V. However, there were significant differences in dislodgement resistance between Group II and the other groups (p<0.05). 2. No significant differences in dislodgement resistance of the restorations were observed between GroupIV-1 and GroupIV-2, both of which utilized a single $Bio-pin^{(R)}$. However, significant differences were observed when Group III was compared to either GroupIV-1 or Group V (p<0.05). 3. No significant differences in dislodgement resistance relative to the type of dental cements used were found. Conclusion. From the above results, it is concluded that the dislodgement resistance of $Bio-pin^{(R)}$ bridge restorations utilizing a single $Bio-pin^{(R)}$ is similar to that of a conventional 3-unit bridge. The results also suggest that $Bio-pin^{(R)}$ bridge restorations using a single $Bio-pin^{(R)}$ are a viable alternative to the conventional 3-unit bridge when minimal removal of sound tooth structure and fulfillment of both function and esthetic aspects are considered.

Biotransformation of Flavonoids with O-Methyltransferase from Bacillus cereus

  • O-Methylation is a common modification reaction found in nature, and is mediated by an O-methyltransferase (OMT). OMTs have been mainly studied in plants, whereas only a few OMTs have been studied in microbes. When searching the Bacillus cereus genome, four putative small molecular OMTs were identified, among which BcOMT-1 was cloned and expressed in E. coli as a his-tag fusion protein. The whole cell expressing BcOMT-1 was used to methylate several flavonoids. Eriodictyol, luteolin, quercetin, and taxifolin, all of which contain 3' and 4' hydroxyl groups, served as methyl group acceptors for BcOMT-1, whereas naringenin, apigenin, 3,3'-dihydroxyflavone, and 3,4'-dihydroxyflavone did not function as substrates. Analysis of the reaction products using HPLC showed two different peaks, and NMR revealed that the methylation position was at the hydroxyl group of either carbon 3' or 4'. Therefore, this showed that BcOMT-1 used flavonoids containing ortho hydroxyl groups and transferred a methyl group to either of two hydroxyl groups.

Rapid Separation of Cellular Cyclosophoraoses Produced by Rhizobium Species

  • A very rapid and efficient separation technique for cellular rhizobial cyclosophoraoses was developed based on fractional precipitation and partition chromatography. Cyclosophoraoses are known to function in the osmotic regulation and root nodule formation of legumes during the nitrogen fixation process. Cyclosophoraoses are produced as unbranched cyclic (1longrightarrow12)-${\beta}$-D-glucans in Agrobacterium or Rhizobium species. Recent research has shown that cyclosophoraoses can form inclusion complexation with various unstable or insoluble guest chemicals, thereby implying great potential for industrial application. Typical separation of pure cellular cyclosophoraoses has been so far carried out by several time-consuming steps, including size exclusion, anion exchange, and desalting liquid chromatographies, with a relatively poor recovery. However, the proposed method demonstrated that the successive application of fractional ethanol precipitation and one step of silica gel-based flash column chromatography was enough to simultaneously purify neutral or anionic forms of cyclosophoraoses. This novel technique is very rapid and provides a high recovery.

Role of ginseng in the neurovascular unit of neuroinflammatory diseases focused on the blood-brain barrier

  • Ginseng has long been considered as an herbal medicine. Recent data suggest that ginseng has antiinflammatory properties and can improve learning- and memory-related function in the central nervous system (CNS) following the development of CNS neuroinflammatory diseases such as Alzheimer's disease, cerebral ischemia, and other neurological disorders. In this review, we discuss the role of ginseng in the neurovascular unit, which is composed of endothelial cells surrounded by astrocytes, pericytes, microglia, neural stem cells, oligodendrocytes, and neurons, especially their blood-brain barrier maintenance, anti-inflammatory effects and regenerative functions. In addition, cell-cell communication enhanced by ginseng may be attributed to regeneration via induction of neurogenesis and angiogenesis in CNS diseases. Thus, ginseng may have therapeutic potential to exert cognitive improvement in neuroinflammatory diseases such as stroke, traumatic brain injury, multiple sclerosis, Parkinson's disease, and Alzheimer's disease.

Ameliorative Effect of Chitosan Complex on Miniature Pig Sperm Cryopreservation

  • Cryopreservation is mainly used for preservation of boar sperm. However, this method stresses the sperm by reactive oxygen species (ROS), and the conception rate and the litter size are not more efficient than the liquid preservation of spermatozoa. Therefore, we use chitosan which is a natural product derived antioxidant compound. We used GnHA (chitosan+hyaluronic acid) and GnHG (chitosan hydrogel) as chitosan complexes to cryopreserve boar sperm for improve sperm metabolism and function. Sperm parameter (sperm motility, progressive motility, path velocity, straight-line velocity, curvilinear velocity) is measured by computer-assisted sperm analysis (CASA) using frozen sperm with GnHA or GnHG (0, 0.25, 0.5, 1 mg/mL), respectively. Also, lipid peroxidation analysis using malondialdehyde (MDA) is performed to confirm the antioxidative effect of chitosan in frozen spermatozoa. CASA analysis showed GnHA and GnHG are effective against cryopreserved boar sperm. And antioxidant effect is measured by lipid peroxidation analysis. GnHA and GnHG, which is chitosan complex are effective for boar sperm cryopreservation by antioxidant effect.
