Gas chromatography를 이용한 profiling과 신속한 동정을 허브류에 존재하는 유기산의 분석에 적용하여 보았다. 흡착제로 Chromosorb P와 용출 용매로 diethyl ether를 사용하는 고체상 추출법을 이용하여 허브추출물로부터 유기산을 분리한 후, tert-butyldimethylsilyl 유도체로 만들어 극성이 다른 DB-5와 DB-17 컬럼으로 동시분석하였다. 두 컬럼에서 분리된 유기산 피크들을 dual capillary column system의 retention index(RI) library 탐색과 RI specta 비교로 신속하고 정확하게 동정할 수 있었으며 이들을 GC-MS로 확인하였다. 허브추출물들의 유기산 분석 결과 fatty acid, dioic acid, hydroxy acid, aromatic acid등 24가지의 유기산이 동정되었고 각 허브의 종류에 따라 특징적인 유기산 GC-profile을 나타내었다. 각 허브추출물의 GC profile의 유기산 조성만을 단순화시킨 RI spectra로 표현한 결과, 각 허브의 유기산 패턴의 분별이 매우 용이하였다.

Metabolomics is the systematic identification and quantification of all metabolites in an organism or biological sample. NMR has been used as a major application tool in plant metabolomics such as quality control, chemotaxonomy, and analysis of genetically modified plants. Herbal medicines are the important therapeutics and are used to manage common diseases such as cold, inflammation, pain, heart diseases, liver cirrhosis, diabetes and central nerve system diseases. Herb plants include various kinds of species such as geranium, mint, and thyme and so on and contain different kinds of metabolites. We performed NMR-based metabolomics study on the seven different species of herb plants using $^1H$ NMR experiments and OPLS-DA to understand the correlation between the classification of herb plants and their metabolite contents. This study showed clear metabolic discrimination among various herb plants. This metabolmics study found several diagnostic NMR signals which are able to be used as bio-markers for identification of the specific herb plants among various species. Clear metabolic discrimination of herb plants suggests three chemotaxonomic groups of herb species.
Typical radiation cross-linked hydrogels has the characteristic that high water content, but low emission efficiency of active ingredients. Therefore, the hydrogel was prepared by the addition to collagen, which is closely related to the formation of skin wrinkles in biocompatibility and highly water-soluble carboxymethyl cellulose sodium salt (CMC) in order to preparation of hydrogels has excellent emission efficiency of active ingredients. Hydrogels were prepared by dissolving CMC and collagen each of 0.5%, 10% concentration in deionized water. Then, prepared hydrogels are performed by gamma-radiation at 1, 3, 5 kGy irradiation dose. The results showed that the gel fraction of after irradiated 3 kGy hydrogel was higher than before irradiated gelation as long as the 55.3%. The swelling rate of irradiated 3 kGy hydrogel was lower than the non-irradiated sample. The compressive strength of 3 kGy irradiated hydrogel was the highest. The visco-elastic did not show any significant differences, even after irradiation. The CMC hydrogel in this study suggested a potential use as a material for the mask pack for improved emission efficiency of the active ingredient and anti-wrinkles.
To study on antioxidant effects in the liver of 40-week-old mouse, the sample were orally pretreated 5mg/kg/day for 5 days with red ginseng saponin components(total saponin, protopanaxadiol saponin, protopanaxatriol saponin, ginsenoside-Rd, ginsenoside-Re, compound-K) for 5 days. The ability of saponin to protect the mouse liver from oxidative damage was examined by determining the activity of superoxide dismutase(SOD), glutathione peroxidase(GPx) and the contents of glutathione, the level of malondialdehyde, The only protopanaxadiol among the ginseng saponin fractions was significantly increased the hepatic SOD activity(p<0.01). The red ginseng saponin induced a slight increase of GPx activity, especially ginsenoside Rd, compound K and protopanaxatriol treatments significantly increased its activity. The content of glutathione was significantly increased by total saponin, protopanaxadiol and ginsenoside Rd(p<0.01), but the oxidized glutathione level was lowered in all the red ginseng saponin. Finally, the level of malondialdehyde was significantly decreased by ginsenoside Rd and protopanaxadiol. In conclusion, protopanaxadiol and ginsenoside Rd among the saponin fraction were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation. This comprehensive antioxidant effects of red ginseng saponin seems to be by a certain action of saponin other than a direct antioxidant action.
암 조직에는 다양한 형태의 세포가 존재하지만, 이들의 조성을 실험적으로 확인하기는 매우 어렵다. 본 연구에서는 유전자 발현 데이터에 통계적 기계학습 모델을 적용하여 각 샘플의 세포 조성을 추론하고, 이러한 세포 조성이 암조직과 정상 조직간에 차이가 있는지를 확인하였다. 두 가지 서로 다른 회귀 모델을 이용하여 세포 조성을 예측한 결과 CD8 T cell과 Neutrophil이 구강암 조직에서 정상 조직에 비해 증가함을 확인할 수 있었다. 또한 비지도학습 중 하나인 t-SNE를 적용하여, 유추된 세포 조성에 의해 정상 조직과 구강암 조직이 서로 군집을 이루고 있음을 확인하였고, 지도 학습 기반의 다양한 분류 알고리즘들을 이용하여 세포 조성 정보를 이용하여 구강암과 정상 조직을 예측하는 것이 가능함을 보였다. 이 연구는 구강암의 면역 세포 침투에 대한 이해도를 증진하는데에 도움을 줄 수 있을 것이다.
In this study, the dyeing properties of supercritical fluid dyed nylon fabrics were investigated which use two types of dyes for dyeing nylon. For other dyeing conditions were referred to related literature, and dyeing was performed with different dyeing concentrations. Dyeability was confirmed through measurement of washing fastness and color coordinate, and a calibration curve of each dye was drawn up and the absorbance of the residual dye was measured to confirm the amount of residual dye and the dye exhaustion rate at the corresponding concentration. As a result of color difference measurement, the color intensity increased as the concentration increased, but the increase was insignificant at high concentration. This tendency was more obvious in C.I. Disperse Orange 155 than in C.I. Disperse Yellow 42. The dye absorption rate also decreases as the concentration increases, but at 0.85% o.w.f concentration, C.I. Disperse Yellow 42 was 97.29% and C.I. Disperse Orange 155 was 93.77%. For both dyes, the wash fastness dropped by 0.5 to 1 class from the sample that was dyed at a concentration of 0.5% o.w.f in the wash fastness test.
본 연구에서는 브로콜리 잎 헥산 분획물의 항염 효과를 평가하여 기능성 식품 및 화장품 소재로의 적용 가능성을 확인하였다. LPS-자극된 RAW264.7 세포에서 전염증성 사이토카인의 생성, iNOS와 COX-2의 발현, MAPK (ERK, JNK, p38) 및 브로콜리 잎 헥산 분획을 사용한 NF-κB의 인산화를 분석하였다. 브로콜리 잎 헥산 분획은 TNF-α, IL-4, IL-6, IL-1β 등의 전염증성 사이토카인의 분비와 iNOS와 COX-2의 발현을 억제했습니다. 또한, 브로콜리 잎 헥산 분획물은 MAPK와 NFκB의 인산화를 감소시켰다. 따라서 브로콜리 잎 헥산 분획물은 식품 및 화장품에서 천연 항염증 소재로 적용 가능성이 있는 것으로 판단된다. 향후 항염증 기전 및 주요 생리활성 물질의 규명에 대한 연구가 필요할 것으로 생각된다.
Background: Age-related macular degeneration (AMD) is a significant visual disease that induces impaired vision and irreversible blindness in the elderly. However, the effects of ginseng berry extract (GBE) on the retina have not been studied. Therefore, this study aimed to investigate the protective effects of GBE on blue light (BL)-induced retinal damage and elucidate its underlying mechanisms in human retinal pigment epithelial cells (ARPE-19 cells) and Balb/c retina. Methods: To investigate the effects and underlying mechanisms of GBE on retinal damage in vitro, we performed cell viability assay, pre-and post-treatment of sample, reactive oxygen species (ROS) assay, quantitative real-time PCR (qRT-PCR), and western immunoblotting using A2E-laden ARPE-19 cells with BL exposure. In addition, Balb/c mice were irradiated with BL to induce retinal degeneration and orally administrated with GBE (50, 100, 200 mg/kg). Using the harvested retina, we performed histological analysis (thickness of retinal layers), qRT-PCR, and western immunoblotting to elucidate the effects and mechanisms of GBE against retinal damage in vivo. Results: GBE significantly inhibited BL-induced cell damage in ARPE-19 cells by activating the SIRT1/PGC-1α pathway, regulating NF-kB translocation, caspase 3 activation, PARP cleavage, expressions of apoptosis-related factors (BAX/BCL-2, LC3-II, and p62), and ROS production. Furthermore, GBE prevented BL-induced retinal degeneration by restoring the thickness of retinal layers and suppressed inflammation and apoptosis via regulation of NF-kB and SIRT1/PGC-1α pathway, cleavage of caspase 3 and PARP, and expressions of apoptosis-related factors in vivo. Conclusions: GBE could be a potential agent to prevent dry AMD and progression to wet AMD.
Objectives: The aim of this study was to compare the total phenolic content of Korean Acanthopanax senticosus stems harvested in different seasons and provide basic data for developing functional reinforcement products based on the optimal harvesting time. Methods: Each sample harvested in different seasons was extracted and concentrated twice for 2 hours using 70% ethyl alcohol. Phenolic compounds were analyzed using high-performance liquid chromatography for simultaneous multi-component analysis of 14 compounds, including syringaresinol and so on. Results: The results showed that the stem of Korean Acanthopanax senticosus harvested in winter (November 29th) (EAS-5) had the highest phenolic content of 1.038%. The stem of Korean Acanthopanax senticosus harvested in autumn (October 1st) (EAS-4) showed the second-highest phenolic content of 0.764%, followed by the stem of Korean Acanthopanax senticosus harvested in spring (February 2nd) (EAS-1) with a content of 0.390%. On the other hand, the stem of Korean Acanthopanax senticosus harvested in the summer (June 2nd) (EAS-3) showed the lowest content at 0.342%. In conclusion, the stem of Korean Acanthopanax senticosus harvested in winter (EAS-5) showed the highest phenolic compound content. Conclusions: Considering the extraction yield and the total phenolic content, as well as the concentrations of key functional components such as eleutheroside B, chlorogenic acid, and syringaresinol in the 70% ethyl alcohol extract of Korean Acanthopanax senticosus, it is suggested that the stems of Korean Acanthopanax senticosus harvested during the winter season are suitable for the development of novel materials with enhanced anti-obesity functionality.

Objective: This study aimed to identify the single-nucleotide polymorphisms (SNPs) in the dual-specificity phosphatase 8 (DUSP8) and insulin-like growth factor 2 (IGF2) genes and to explore their effects on inosine-5'-monophosphate (IMP), inosine, and hypoxanthine contents in Korean native chicken -red-brown line (KNC-R Line). Methods: A total sample of 284 (males, n = 127; females n = 157) and 230 (males, n = 106; females, n = 124) aged of 10 weeks old KNC-R line was used for genotyping of DUSP8 and IGF2 genes, respectively. One SNP (rs313443014 C>T) in DUSP8 gene and two SNPs (rs315806609A/G and rs313810945T/C) in IGF2 gene were used for genotyping by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and KASP methods, respectively. The Two-way analysis of variance of the R program was used to associate DUSP8 and IGF2 genotypes with nucleotide contents in KNC-R chickens. Results: The DUSP8 (rs313443014 C>T) was polymorphic in KNC-R line and showed three genotypes: CC, CT, and TT. The IGF2 gene (rs315806609A/G and rs313810945T/C) was also polymorphic and had three genotypes per SNP, including GG, AG, and AA for the SNP rs315806609A/G and genotypes: CC, CT, and TT for the SNP rs313810945T/C. Association resulted into a strong significant association (p<0.01) with IMP, inosine, and hypoxanthine. Moreover, the significant effect of sex (p<0.05) on nucleotide content was also observed. Conclusion: The SNPs in the DUSP8 and IGF2 genes might be used as genetic markers in the selection and production of chickens with highly flavored meat.
