Establishment of Microwave-Assisted Extraction Conditions for Antioxidative Extracts from Pleurotus eryngii (새송이버섯의 항산화성 추출물 제조를 위한 마이크로웨이브 추출조건 설정)

  • Through the application of a microwave-assisted extraction, the aim of study was to investigate the extraction conditions for the total yield, electron donating ability, total polyphenol contents and nitrite scavenging effect from Pleurotus eryngii. This experiment was conducted under the following extraction conditions: the microwave power (60, 90, 120 W), sample to solvent ratio (1:50, 1:20, 1:10), extraction time (1, 5, 10, 15 mins), and extractant like water, $50\%$ EtOH and $99\%$ EtOH. During the increase of microwave power ranging from 60 to 120 W, the maximized effective components were obtained when the microwave power was set at 60 W. By decreasing the ratio down to 1:10 (g/mL), the extraction of effective components but not total yield was the highest at 1:10 (g/mL). As going up to 15 min, the maximum extraction of effective components was achieved at 1 min for the extraction time and was little affected at over 5 min. In conclusion, the most appropriate conditions for the extraction of effective compounds from Pleurotus eryngii were as follows: the microwave power at 60 W, 1:10 of the sample to solvent ratio, extraction time at 1 min and water as the extractant.

Optimizing the Hot-water Extraction Conditions for Acanthopanacis cortex Using Response Surface Methodology (반응표면분석법을 이용한 오가피의 열수 추출조건 최적화)

  • In order to examine the effective components and antioxidative characteristics of Acanthopanacis cortex, response surface methodology was used to optimize the hot water extraction process by analyzing and monitoring the extraction condition characteristics. The total extraction yield was optimized using the extraction temperature of $84.28^{\circ}C$, extraction time of 3.59 hr and 18.37 mL/g as the amount of solvent per sample. For the effective component of eleutheroside E, the optimal extraction temperature, time and amount of solvent per sample were $81.16^{\circ}C$, 3.56 hr, and 19.06 mL/g, respectively; also, for total phenolics, the optimal conditions were $88.45^{\circ}C$, 4.33 hr, and 21.12 mL/g, respectively. Regression equations were deduced for each variable and then eleutheroside E, chlorogenic acid, and antioxidative characteristics were superimposed with the optimal conditions to obtain values for each extraction process factor. The predicted results were $81{\sim}88^{\circ}C$, $3.5{\sim}4.3hr$, and $18{\sim}21mL/g$, respectively. The estimated values for the optimal extraction conditions of Acanthopanacis cortex's effective components and antioxidative characteristics were similar to the experimental results.

Rheological properties of flour dough containing roasted rice bran (볶은 쌀겨를 첨가한 밀가루 반죽의 rheology 특성)

  • This study was carried out to investigate the effect of roasted rice bran (RRB) on the rheological properties of bread dough. According to farinograph analysis, the consistency of the control sample was greatest. There were no significant differences in water absorption (p<0.05). Lower values of development time, stability, and time to breakdown, which were affected, by the addition of RRB, were observed for RRB-containing dough samples, compared to the control dough sample. Addition of RRB significantly increased the mixing tolerance index (MTI). According to rheofermentometer analysis, the values of H'm, $T^{\prime}_1$, and retention volume decreased with increase in the amount of RRB added. According to the rapid visco analyzer (RVA) analysis, peak viscosity, holding strength, and setback values were greater in the control than in the RRB-containing samples. The addition of RRB to the flour influenced rheological properties like fermentation volume and acidity. The total acidity increased with the increase in the amount of RRB added. The present study has indicated that there was no significant difference between the rheological properties of the control and 5% RRB-containing dough samples. Therefore, the addition of 5% RRB could be an effective way to produce functional flour bread without affecting its desirable physical properties.

Anti-inflammatory effect in macrophages according to the mixing ratio of acemannan and aloesin (Acemannan과 aloesin의 혼합 비율에 따른 대식세포에서의 항염증 효과)

  • This study aims to confirm the anti-inflammatory activities of acemannan and aloesin, which have been studied for various efficacies at various mixed sample ratios. The mixed samples were mixed at a ratio of 1:1 (AA-1), 1:2 (AA-2), 1:3 (AA-3), 2:1 (AA-4), and 3:1 (AA-5). Seven samples were evaluated for their cytotoxic ability on macrophages, and the results showed that all cell viability was over 90% at a concentration of 100 ㎍/mL. First, due to the NO production inhibitory activity, a better inhibitory effect was achieved when using a mixed sample rather than a single material. Afterward, the activity of inhibiting the production of PGE2, TNF-α, and IL-6 was confirmed using a mixed sample. It was confirmed that AA-2 had the best inhibitory activity on producing PGE2, TNF-α, and IL-6 rather than AA-1, AA-3, AA-4, and AA-5. For this reason, experiments were conducted using AA-2 to determine the protein expression levels of iNOS and COX-2, which are inflammation-related proteins. It was confirmed that AA-2 inhibited iNOS and COX-2 protein expression by 25.01% and 27.27%, respectively, compared to the LPS-alone treatment group. In conclusion, the mixed sample of acemannan and aloesin is judged to have anti-inflammatory activity and can potentially to be used as a functional material.

Preliminary Study on Human Papillomavirus Frequency and Specific Type-distribution in Vulva Cancer from Thai Women

  • Vulva cancer is rare among all gynecological cancer worldwide, including Thailand, and mainly affects older women. Persistent high risk type infection of human papillomavirus (HPV) is the one important factor for developing cancer. In this study, we focused on HPV DNA investigation and type-specific distribution of HPV in 25 formalin-fixed paraffin-embedded (FFPE) samples collected from Thai women with vulva cancer histologically confirmed by the National Cancer Institute, Thailand, during 2003-2011. HPV DNA detection and genotyping were undertaken with polymerase-chain reaction and enzyme-immunoassay using GP5+/bio6+ consensus specific primers and digoxigenin-labeled specific oligoprobes, respectively. Human ${\beta}$-globin genes was used as the internal control. Our results showed that 44% (11/25) of all vulva cancer samples were HPV-positive. All of them are high risk HPV type infection, detected as single (63.64%, 7/11) and/or double infections (4/11, 36.36%). HPV 16 was the most common type identified in vulva cancer, followed by HPV 35, 33, 18 and 58. In conclusion, this study presented that HPV-16 is observed at the highest frequency in this cancer, similar to cervical cancer, with HPV 18 being less frequent. Although the sample size was small and could not represent overall incidence and prevalence in Thai women, these preliminary data for vulva cancer are of interest since they reinforce the necessity for HPV screening or vaccination in Thailand.

Prognostic Factors of Idiopathic Facial Palsy: A Retrospective Study

  • Objectives : The purpose of this study was to evaluate the clinical prognostic factors affecting facial palsy in 98 idiopathic facial palsy patients who were hospitalized and treated in 2015, using retrospective statistical analysis. Methods : We investigated patients with idiopathic facial nerve palsy, admitted to a Korean medical hospital in 2015, and examined patients' variables and therapeutic variables. For analysis of clinical data, an independent sample t-test, analysis of variance (ANOVA), and simple regression analysis were performed using IBM SPSS version 24.0. Results : 1. The initial degree of facial palsy showed statistical significance with age. The older the age, the more severe the initial palsy. 2. Following treatment degree of facial palsy was statistically significant with age, hypertension, and fasting blood sugar (FBS). The higher the value, the slower the recovery from facial palsy. There was a statistical significance with the number of treatments in a Korean medical hospital. The more frequent the treatment, the faster the facial palsy recovery. 3. Degree of facial palsy after 12 months was statistically significant with age, hypertension, diabetes, FBS, and the initial severity of facial palsy. The higher the value, the slower the facial palsy recovery. 4. Sex, left or right sided palsy, alcohol consumption, smoking, history of facial palsy, season of onset, total number of treatments and bio chemistry (BC), complete blood cell count (CBC), urinalysis (UA) factors had no statistical significance with prognosis of facial palsy. Conclusion : Age, season of onset, hypertension, diabetes, FBS, initial severity of facial palsy, and the number of treatments at a Korean medical hospital showed statistical significance. The number of treatments at the Korean medical hospital positively correlated with facial palsy prognosis, and the others variables showed a negative correlation with facial palsy prognosis.

Comparison of Cellular Features Diagnostic of Papillary Thyroid Carcinoma in Liquid-Based (Cell Scan 1500TM) Preparations and Conventional Smears

  • The study compared the cytological features of papillary thyroid carcinoma (PTC) in liquid-based preparations (LBPs) and conventional Pap (CP) smears from fine needle aspiration (FNA), and assessed the feasibility of LBP using the Cell Scan $1500^{TM}$ processor on thyroid FNA samples. Thyroid FNA samples were obtained from 883 consecutive patients. Each sample was divided into two and used for LBPs and CP smears. All were screened independently in a double-blind manner. From the 883 cases, 95 cases were diagnosed as PTC in one or both types of preparation (10.8%). PTC was diagnosed via CP smears in 83 cases (87.4%) and via LBPs in 70 cases (73.7%). However, there were differences in categorization between the paired preparations: Twelve (12) PTCs were misinterpreted in CP smears and 25 PTCs in LBPs. There was a significant discrepancy in the rate of detection of the diagnostic features, with LBPs having a lower detection rate. One (1) case (1.2%) of CP smears and 16 cases (22.9%) of LBPs were categorized as unsatisfactory/nondiagnostic in a total of the 95 PTCs. To conclude, the detection rate of the diagnostic features of PTC is lower in Cell Scan 1500TM samples than in CP smears. However, there are some cases in which a diagnosis of PTC is made in LBPs, but not in CP smears. Therefore, definitive cancer diagnosis in thyroid FNA preparations is likely to result from agreement between direct smears and Cell Scan 1500TM preparations.

  • In this study, ozone oxidation experiment was carried out for the removal of fluorescent whitening agent which is widely used in textile dyeing and paper industry. The stilbene fluorescent whitening agent has been industrialized since the earliest, and the amount of current production is the highest. Due to the characteristics of the fluorescent whitening agent that can not be removed by conventional wastewater treatment methods, the fluorescent whitening agent in wastewater treatment has difficulty in using as recycled water in the process. Pre-treatment ozone oxidation experiment was conducted prior to the introduction of Membrane Bio Reactor(MBR) treatment process by converting biodegradable materials into biodegradable materials. The removal efficiencies of fluorescent whitening agents, a diaminostilbenedisulfonic acid derivative by ozone oxidation were evaluated by $UV_{254}$ Scan, $COD_{Mn}$, T-N and color using a synthetic wastewater sample ($COD_{Mn}=433.0mg/{\ell}$) and paper and paper mill wastewater ($COD_{Mn}=157.2mg/{\ell}$).

Prediction of Nutrient Composition and In-Vitro Dry Matter Digestibility of Corn Kernel Using Near Infrared Reflectance Spectroscopy

  • Nutritive value analysis of feed is very important for the growth of livestock, and ensures the efficiency of feeds as well as economic status. However, general laboratory analyses require considerable time and high cost. Near-infrared reflectance spectroscopy (NIRS) is a spectroscopic technique used to analyze the nutritive values of seeds. It is very effective and less costly than the conventional method. The sample used in this study was a corn kernel and the partial least square regression method was used for evaluating nutrient composition, digestibility, and energy value based on the calibration equation. The evaluation methods employed were the coefficient of determination ($R^2$) and the root mean squared error of prediction (RMSEP). The results showed the moisture content ($R^2_{val}=0.97$, RMSEP=0.109), crude protein content ($R^2_{val}=0.94$, RMSEP=0.212), neutral detergent fiber content ($R^2_{val}=0.96$, RMSEP=0.763), acid detergent fiber content ($R^2_{val}=0.96$, RMSEP=0.142), gross energy ($R^2_{val}=0.82$, RMSEP=23.249), in vitro dry matter digestibility ($R^2_{val}=0.68$, RMSEP=1.69), and metabolizable energy (approximately $R^2_{val}$ >0.80). This study confirmed that the nutritive components of corn kernels can be predicted using near-infrared reflectance spectroscopy.

  • Polybrominated diphenyl ethers (PBDEs) are a class of brominated flame retardants (BFRs) which have taken much interest recently due to their potential hazardous effects to human body and ecosystem. Many countries and European community prohibits the usage of certain BFRs in electronics and electronic devices (e.g. RoHS). In this perspectives, Korea Research Institute of Standards and Science (KRISS) has designed and practiced proficiency testing programs based on the ISO/IEC 17043 in order to assist laboratory accreditation activities. The programs for interlaboratory comparisons include congeners of PBDE (PBDE-154, 183, 206, 209) in high impact polystyrene (HIPS). A sample bottle that contains 10 g granular HIPS was distributed to 35 participating laboratories and the test results were calculated by the statistical procedure using z-scores to evaluate performance of each laboratory. The results and the laboratory's performance were discussed.
