A Theoretical Study of the Formation of Benzene Excimer: Effects of Geometry Relaxation and Spin-state Dependence

  • Geometry relaxation effects on the formation of benzene excimer were investigated by means of ab initio calculation at SOS-CIS($D_0$)/aug-cc-pVDZ level. In the case of T-shaped dimer configuration, intermolecular interactions in the excited states are found to be nearly the same as those in the ground state and structural deformations are limited within a single molecule; the geometry relaxation effects are then negligible and singlet-triplet energy gap remains constant. As for face-to-face eclipsed dimer, on the other hand, both molecules undergo structural change. As a result, intermolecular interactions in the excited states are significantly different than those in the ground state. Although the intermolecular distances obtained from potential energy curve calculation with frozen molecular structures are in qualitative agreement, the excited-state binding energies are notably overestimated with respect to those at optimized structures. In particular, the effects are calculated to be larger in $T_1$ state and hence singlet-triplet energy gap, which reduces markedly in this configuration, is underestimated without relaxation.

Association of Insulin-like growth factor binding protein 2 genotypes with growth, carcass and meat quality traits in pigs

  • Background: This study was conducted to investigate the potential association of variation in the insulin-like growth factor binding protein 2 (IGFBP2) gene with growth, carcass and meat quality traits in pigs. IGFBP2 is a member of the insulin-like growth factor binding protein family that is involved in regulating growth, and it maps to a region of pig chromosome 15 containing significant quantitative trait loci that affect economically important trait phenotypes. Results: An IGFBP2 polymorphism was identified in the Michigan State University (MSU) Duroc ${\times}$ Pietrain $F_2$ resource population (n = 408), and pigs were genotyped by MspI PCR-RFLP. Subsequently, a Duroc pig population from the National Swine Registry, USA, (n = 326) was genotyped using an Illumina Golden Gate assay. The IGFBP2 genotypic frequencies among the MSU resource population pigs were 3.43, 47.06 and 49.51 % for the AA, AB and BB genotypes, respectively. The genotypic frequencies for the Duroc pigs were 9.82, 47.85, and 42.33 % for the AA, AB and BB genotypes, respectively. Genotype effects (P < 0.05) were found in the MSU resource population for backfat thickness at $10^{th}$ rib and last rib as determined by ultrasound at 10, 13, 16 and 19 weeks of age, ADG from 10 to 22 weeks of age, and age to reach 105 kg. A genotype effect (P < 0.05) was also found for off test Longissimus muscle area in the Duroc population. Significant effects of IGFBP2 genotype (P < 0.05) were found for drip loss, 24 h postmortem pH, pH decline from 45 min to 24 h postmortem, subjective color score, CIE $L^*$ and $b^*$, Warner-Bratzler shear force, and sensory panel scores for juiciness, tenderness, connective tissue and overall tenderness in MSU resource population pigs. Genotype effects (P < 0.05) were found for 45-min pH, CIE $L^*$ and color score in the Duroc population. Conclusions: Results of this study revealed associations of the IGFBP2 genotypes with growth, carcass and meat quality traits in pigs. The results indicate IGFBP2 as a potential candidate gene for growth rate, backfat thickness, loin muscle area and some pork quality traits.

dicA promoter DNA에 붙는 H-NS 단백질에 의한 dicA 유전자의 발현 조절 (H-NS binding on dicA promoter DNA inhibits dicA gene expression)

  • H-NS는 대장균에서 DNA 결합 단백질로 수많은 유전자의 발현에 영향을 주는 것으로 잘 알려져 있다. DicA 단백질은 dicF, dicB의 발현을 억제하여 대장균의 분열을 조절한다. dicA의 발현에 Cnu, H-NS의 관여 여부는 CnuK9E 돌연변이가 $37^{\circ}C$에서 dicA의 발현을 억제하여 대장균이 길게 자라는 현상을 일으키며 처음 알려졌다. 하지만 Cnu와 H-NS 두 단백질이 어떻게 dicA의 발현을 조절하는지에 대한 분자적인 기작 연구는 잘 되어있지 않다. 본 연구에서 H-NS가 dicA와 dicC 유전자의 프로모터 부근에 염기서열 특이적으로 결합하며, $37^{\circ}C$ 보다 $25^{\circ}C$에서 DNA 더 잘 결합하는 것을 확인하였다. 그리고 EMSA를 통해 Cnu는 H-NS의 DNA 결합의 oligomeric state를 변화시키는 방식으로 작용하는 것을 보여주었다. In vivo transcription assay와 real time PCR을 통해 H-NS가 제거된 대장균에서 dicA 프로모터 활성이 높아지고, 분열 초기 dicA의 발현이 조절 받지 못하고 증가하는 것으로 보아, H-NS는 dicA의 발현에 억제자로서 기능한다.

Characterization of a novel posttranslational modification in polypyrimidine tract-binding proteins by SUMO1

  • Polypyrimidine tract-binding protein 1 (PTBP1) and its brain-specific homologue, PTBP2, are associated with pre-mRNAs and influence pre-mRNA processing, as well as mRNA metabolism and transport. They play important roles in neural differentiation and glioma development. In our study, we detected the expression of the two proteins in glioma cells and predicted that they may be sumoylated using SUMOplot analyses. We confirmed that PTBP1 and PTBP2 can be modified by SUMO1 with co-immunoprecipitation experiments using 293ET cells transiently co-expressing SUMO1 and either PTBP1 or PTBP2. We also found that SUMO1 modification of PTBP2 was enhanced by Ubc9 (E2). The mutation of the sumoylation site (Lys137) of PTBP2 markedly inhibited its modification by SUMO1. Interestingly, in T98G glioma cells, the level of sumoylated PTBP2 was reduced compared to that of normal brain cells. Overall, this study shows that PTBP2 is posttranslationally modified by SUMO1.

흰쥐의 뇌의$A_1$ 아데노신 수용체에 작용하는 역효현제에 관한 연구 (Inverse Agonists at $A_1$ Adenosine Receptors in Rat Cerebral Cortex)

  • 전통적인 수용체 이론에 따르면 상경적 길항제는 효현제와 수용체의 같은 부위에 작용하지만, 효능(efficacy)이 없기 때문에 생물학적 반응을 일으키지는 않는다. 그러나 최근에 발표되는 자료들에 따르면 모든 길항체의 효능(efficacy)이 0가 아니라 음수도 될 수가 있다고 생각된다. 이러한 음수의 효능을 갖는 약물을 역효현제라 부른다. 본 연구에서는 쥐의 cerebral cortex에서 얻은 membranes을 사용하여, $A_1$ 아데노신 수용체에 작용하는 역효현제를 인구하였다. 8개의 길항제로 알려진 약물들이 G단백에 대한 $[^{35}S]GTP_{\gamma}S$ 결합을 감소시키는 정도를 측정함으로써 역효현제의 특성을 검색하였다. 효현제에 의한 $[^{35}S]GTP_{\gamma}S$ 결합의 증가는 이틀 길항제틀에 의해 완전히 억제되었지만, 검색한 8개의 길항제는 두 군으로 구분되었다. DPCPX를 포함한 7개 길항제는 효현제 부재시의 basal $[^{35}S]GTP_{\gamma}S$ binding을 통계적으로 의의있게 감소시켜 역효현제의 특성을 나타내는 반면, CCS-15943은 basal $[^{35}S]GTP_{\gamma}S$ binding에 아무런 영향을 주지 않았다. NEM을 membranes에 처치하면 PIA에 의한 $[^{35}S]GTP_{\gamma}S$ binding이나 basal binding 둘다 감소하는데 이는 $[^{35}S]GTP_{\gamma}S$ binding의 상당부분이 G단백의 activated state를 나타내는 것을 알 수 있다. 또한 $[^3H]DPCPX$를 이용한 competitive binding assay에서 0.1 mM GTP는 효현제인 PIA의 apparent affinity를 감소시켰으며, DPCPX의 apparent affinity는 증가시키고, CGS-15943에는 아무런 영향을 미치지 않았다. 이것은 상기의 $[^{35}S]GTP_{\gamma}S$ binding의 결과를 뒤받침해 주는 결과라고 생각된다.

산화환원에 따른 hHSF1의 DNA binding domain의 역할 (The Role of DNA Binding Domain in hHSF1 through Redox State)

  • 다양한 종류의 박테리아에서부터 사람의 세포에 이르기까지 환경적인 스트레스나 병에 의한 스트레스 혹은 스트레스가 없는 상황에서도 열충격반응(heat shock response) 유도되어진다. 열충격반응에 노출된 세포에서는 모든 단백질의 발현이 정지되는 반면, 열충격단백질(heat shock proteins: HSPs)은 발현되어 스트레스로부터 세포를 보호한다. HSF1(heat shock factor 1)이라는 HSPs 유도단백질은 열충격반응시 단량체형태에서 삼중체의 형태로 구조변화를 일으켜 heat shock element(HSE)라고 불리우는 HSP gene의 발현 promoter에 특이적으로 결합하게 되어 HSPs를 발현시킨다. Human HSF1(hHSF1)은 다섯 개의 시스테인 잔기를 가지고 있는데 이 시스테인의 thiol(-SH)기는 강한 친전자성을 띔으로 급격히 산화되거나 질산화된다. 이러한 고찰은 시스테인 잔기가 산화 환원 의존적인 황산기/이황화결합 전환을 통해 구조적인 변화를 가져온다는 사실을 의미하고 있다. 따라서 본 연구에서는 여러 가지 산화환원제를 이용하여 HSF1에 존재하는 다섯 개의 시스테인 잔기의 역할과 삼량체 형성에 관여하는 잔기에 대하여 알아보고자 하였다. 또한 이황화결합을 통한 삼량체형성의 구조적변화의 관점에서 HSF1의 구조 변화와 DNA 결합력과의 상관관계에 관하여도 알아보고자 하였다. 본 연구결과로 HSF1의 DNA binding domain은 삼량체를 형성하는 구조적인 변화를 통해서 DNA에 대한 결합력이 증가되는 것을 알 수 있었는데 이것은 삼량체가 됨으로서 HSF1의 내부에 위치해 있던 DNA binding domain이 외부로 노출 되어져 DNA에 쉽게 결합할 수 있게 된다는 사실을 시사한다.

산소 분압에 따른 산화주석 박막의 전계효과 이동도 변화 분석 (Analysis on the Field Effect Mobility Variation of Tin Oxide Thin Films with Oxygen Partial Pressure)

  • Bottom-gate tin oxide ($SnO_2$) thin film transistors (TFTs) were fabricated on $N^+$ Si wafers used as gate electrodes. 60-nm-thick $SnO_2$ thin films acting as active layers were sputtered on $SiO_2/Al_2O_3$ films. The $SiO_2/Al_2O_3$ films deposited on the Si wafers were employed for gate dielectrics. In order to increase the resistivity of the $SnO_2$ thin films, oxygen mixed with argon was introduced into the chamber during the sputtering. The mobility of $SnO_2$ TFTs was measured as a function of the flow ratio of oxygen to argon ($O_2/Ar$). The mobility variation with $O_2/Ar$ was analyzed through studies on crystallinity, oxygen binding state, optical properties. X-ray diffraction (XRD) and XPS (X-ray photoelectron spectroscopy) were carried out to observe the crystallinity and oxygen binding state of $SnO_2$ films. The mobility decreased with increasing $O_2/Ar$. It was found that the decrease of the mobility is mainly due to the decrease in the polarizability of $SnO_2$ films.

Mutational Analysis of Two Conserved Active Site Tyrosine Residues in Matrilysin

  • The ionization of tyrosine residue is known to be involved in the stabilization of transition-state in catalysis of astacin based upon the astacin-transition state analog structure. Two tyrosine residues, Tyr-216 and Tyr-219, are conserved in all MMPs related with astacin family, We replaced Tyr-216 and Tyr-219 into phenylalanine, respectively and the zinc binding properties, kinetic parameters, and pH dependence of each mutant are determined in order to examine the role of tyrosine residue in matrilysin catalysis. Both mutants contain two zinc atoms per mol of enzyme, indicating that either tyrosime does not affect the zinc binding property of the enzyme. Y216F and Y219F mutants are highly active and the kcat/Km values are only decreased 1.1-1.5-fold compared to the wild-type enzyme. The decrease in the activity of the mutants is essentially due to the increase in Km value. The pH dependencies of the kcat/Km values for both mutants are similar to the corresponding dependencies obtained with the wild type enzyme. The pKa values at the alkaline side of both mutants are not changed. These kinetic and pH dependence results indicate that the ionization of active site tyrosine residue of matrilysin is not reflected in the kinetics of peptide hydrolysin as catalyzed by astacin.

수축중인 근육막에서 actin과 myosin 결합의 구조변화에 관한 연구 (A Study On Structure Change of Binding Actin and Moysin On The Contracting Muscle Membrane)

  • The effects of the applied stretch and MgADP binding on the structure of the actin and myosin cross-bridges in rabbit fibers in the rigor state have been investigatedwith improved resolution by x-ray diffraction using synchrotron radiation. To clarify the structure of the ATP hydrolysis intermediates formed by actin and myosin cross-bridges,the effects of various phosphate analogs in the of MgADP on the structure of the thin and thick filaments in glycerinated rabbit muscle fibers in the rigor state investigated by x-ray diffraction with a short exposure time using synchrotron radiation. These results strongly suggest that when MgADP and phosphate analogs such as metallofluorides(BeF3 and AlF4)and vanadate(VO4(Vi)) were added the rigor fibers in the presence of the ATP-depletion backup system, the intensities of the actin-based layer lines were markedly weakened. We found that the intensity of the 14.5 nm-based meridional reflections increase by 20-50% when phosphate analogs such as metallofluorides(BeF3 and AlF4) and vanadate(VO4(Vi)) was added to the rigor muscle.

$Cr^{6+}$가 흰쥐 신장세포내 미토콘드리아의 호흡에 미치는 영향과 저분자 크롬결합물질의 작용 (Effects of Hexavalent Chromium on Mitochondrial Respiration of Rat Kidney and Function of Low-Molecular-Weight, Chromium-Binding Substances)

  • To investigate the effects of hexavalent chromium on mitochondrial respiration of rat kidney, various hexavalent chromium concentrations were treated, then respiration and electron transfer enzyme activities were measured. Ultrastructural changes at state IV respiration of mitochondria were also observed. Then, to investigate protective role against hexavalent chromium in the body, low-molecular-weight, chromium-binding substances (LMCr) were purified from livers of rabbits 2hr after intravenously administrated with sodium dichromate at a dose of 74mg per kg body weight. And then, respiration rates of mitochondria treated with LMCr, hexavalent chromium containing 0.7mM chromium were measured. Hexavalent chromium decreased state IV respiration rates and electron transfer enzyme activities of mitochondria, and increased labile membrane and swelling. And partial inhibitions of condensed to orthodox conformational change were observed. Respiration rates of mitochondria treated with LMCr containing 0.7mM chromium did not differ from that of the non-treated mitochondria. But respiration rates of 0.7mM hexavalent chromium-treated mitochondria decreased by 42%, compared to non-treated mitochondria. These results suggest that LMCr may play an important role in detoxification of toxic hexavalent chromium.
