Effect of Mode of Binding Linkage on Monolayer Assembly of Zeolite

  • During the monolayer assembly of zeolite microcrystals using sonication with stacking (SS) method, the factors that govern the degree of close packing (DCP) between the microcrystals, the rate of attachment (rA) of microcrystals onto the substrate, the degree of coverage (DOC), and the binding strength (BS) between each crystal and the substrate were investigated for each mode of binding linkage (MBL). The tested MBLs were covalent linkage (CL), ionic linkage (IL), and polyelectrolyte-mediated ionic linkage (p-IL). Unlike the monolayers of zeolite crystals assembled on glass with a covalent linkage, the strong BS, very high DOC, and very high DCP do not decrease during monolayer assembly on glass through ionic linkages. This results indicate that the surface migration of crystals undergo linkage-nondestructively when crystals were attached to the substrates through ionic linkages.

Conformation and Linkage Studies of Specific Oligosaccharides Related to H1N1, H5N1, and Human Flu for Developing the Second Tamiflu

  • The interaction between viral HA (hemagglutinin) and oligosaccharide of the host plays an important role in the infection and transmission of avian and human flu viruses. Until now, this interaction has been classified by sialyl(${\alpha}2-3$) or sialyl(${\alpha}2-6$) linkage specificity of oligosaccharide moieties for avian or human virus, respectively. In the case of H5N1 and newly mutated flu viruses, classification based on the linkage type does not correlate with human infection and human-to-human transmission of these viruses. It is newly suggested that flu infection and transmission to humans require high affinity binding to the extended conformation with long length sialyl(${\alpha}2-6$)galactose containing oligosaccharides. On the other hand, the avian flu virus requires folded conformation with sialyl(${\alpha}2-3$) or short length sialyl(${\alpha}2-6$) containing trisaccharides. This suggests a potential future direction for the development of new species-specific antiviral drugs to prevent and treat pandemic flu.

Aspergillus niger 유래의 Transglucosidase의 고정화 (Immobilization of Transglucosidase from Aspergillus niger)

  • 이소말토올리고당의 생산에 사용되는 Aspergillus niger 유래의 TG의 고정화를 위해 여러가지 방법을 비교하여 고정화방법을 선정하였다. 이온결합과 흡착, entrapment, 공유결합과 금속이온을 이용한 결합 등을 이용하여 TG의 고정화를 시도하였다. TG의 고정화를 위해서는 수율면에서 우수하고(61.3%) 간편성도 있는 CNBr-activated sepharose 4B를 이용한 고정화 방법을 선정하였다. DEAE-sephadex를 이용한 이온결합에 의해서는 33.1%, Diaion HP-20를 이용한 흡착법에 의해서는 22.5%의 수율을 보였으나 CNBr-activated sepharose 4B를 이용한 고정화 방법에 비해 수율과 안정성이 낮아 제외하였다. Entrapment에 의한 고정화는 bead내의 확산저항으로 인해 최종산물의 생성에 불리한 것으로 판단되었고 magnesium silicate, silica gel, glass bead 등의 담체를 이용한 고정화방법은 낮은 수율로 인해, 금속이온을 이용한 경우 역시 낮은 수율과 장시간의 준비로 인해 고정화방법으로 부적당하다고 판단되었다.

  • This paper describes the real-time data linkage of the field control system for distributed control in nuclear power plant environment. The most important keys of digital control system in nuclear power plant are the reliability and stability of system, and real-time control ability. This Paper brought up the hardware construction using a new method about the design of each station located upon control transmission network to improve real-time ability of field control system, and measured the station binding time between devices connected to field control module. And it was confirmed performance improvement of overall system for real-time data linkage between control devices.

Identification of Novel SNPs in Bovine Insulin-like Growth Factor Binding Protein-3 (IGFBP3) Gene

  • The insulin-like growth factors (IGFs), their receptors, and their binding proteins play key roles in regulating cell proliferation and apoptosis. Insulin-like growth factor binding protein-3 (IGFBP3, OMIM #146732) is one of the proteins that bind to the IGFs. IGFBP3 is a modulator of IGF bioactivity, and direct growth inhibitor in the extravascular tissue compartment. We identified twenty-two novel single nucleotide polymorphisms (SNPs) in IGFBP3 gene in Korean cattle (Hanwoo, Bos taurus coreanae) by direct sequencing of full gene including -1,500 bp promoter region. Among the identified SNPs, five common SNPs were screened in 650 Korean cattle; one SNP in promoter (IGFBP3 G-854C), one in 5'UTR region (IGFBP3 G-100A), two in intron 1 (IGFBP3 G+421T, IGFBP3 T+1636A), and one in intron 2 (IGFBP3 C+3863A). The frequencies of each SNP were 0.357 (IGFBP3 G-854C), 0.472 (IGFBP3 G-100A), 0.418 (IGFBP3 G+421T), 0.363 (IGFBP3 T+1636A) and 0.226 (IGFBP3 C+3863A), respectively. Haplotypes and their frequencies were estimated by EM algorithm. Six haplotypes were constructed with five SNPs and linkage disequilibrium coefficients (|D'|) between SNP pairs were also calculated. The information on SNPs and haplotypes in IGFBP3 gene could be useful for genetic studies of this gene.

  • The synthesis and ion binding properties of calix[4]pyrrole bearing pyrene moieties appended to one side of the calix[4]pyrrole are reported. The key feature is the presence of flexible fluorescence arms attached to the calix[4]pyrrole ring in a cis-fashion. The preliminary solution phase anion and cation binding studies revealed that the systems can be in fact as viable sensors for anionic guest.

Marker Production by PCR Amplification with Primer Pairs from Conserved Sequences of WRKY Genes in Chili Pepper

  • Despite increasing awareness of the importance of WRKY genes in plant defense signaling, the locations of these genes in the Capsicum genome have not been established. To develop WRKY-based markers, primer sequences were deduced from the conserved sequences of the DNA binding motif within the WRKY domains of tomato and pepper genes. These primers were derived from upstream and downstream parts of the conserved sequences of the three WRKY groups. Six primer combinations of each WRKY group were tested for polymorphisms between the mapping parents, C. annuum 'CM334' and C. annuum 'Chilsung-cho'. DNA fragments amplified by primer pairs deduced from WRKY Group II genes revealed high levels of polymorphism. Using 32 primer pairs to amplify upstream and downstream parts of the WRKY domain of WRKY group II genes, 60 polymorphic bands were detected. Polymorphisms were not detected with primer pairs from downstream parts of WRKY group II genes. Half of these primers were subjected to $F_2$ genotyping to construct a linkage map. Thirty of 41 markers were located evenly spaced on 20 of the 28 linkage groups, without clustering. This linkage map also consisted of 199 AFLP and 26 SSR markers. This WRKY-based marker system is a rapid and simple method for generating sequence-specific markers for plant gene families.

Certification of Gibroblase Cell Adhesion and Spreading Mediated by Arg-Gly-Asp (RGD) Sequence on Thermo-Reversible Hydrogel

  • In an effort to regulate the mammalian cell behavior in entrapment with a gel, we have functionalized hydrogels with the putative cell-binding (-Arg-Gly-Asp-)(RGD) domain. An adhesion molecule of Gly-Arg-Gly-Asp-Ser (GRGDS) peptides, a cell recognition ligand, was induced into thermo-reversible hydrogels, composed of N-isopropylacrylamide with small amounts of acrylic acid (typically 2-5 $mol\%$ in feed), as a biomimetic extracellular matrix (ECM). The GRGDS containing a p(NiPAAm-co-AAc) copolymer gel was studied in vitro for its ability to promote the spreading and viability of cells by introducing a GRGDS sequence. Hydrogel with no adhesion molecule was a poor ECM for adhesion, permiting spreading of only $3\%$ of the seeded cells for 36h. By immobilizing the peptide linkage into the hydrogel, the conjugation of RGD promoted $50\%$ of proliferation for 36h. However, the GREDS sequence, nonadhesive peptide linkage, conjugated hydrogel showed only $5\%$ of the seeded cell for the same time period. In addition, with the serum-free medium, only GRGDS peptides conjugated to hydrogel was able to promotecell spreading, while there was no cell proliferation in the hydrogel without GRGDS. Thus, the GRGDS peptide-conjugated thermo-reversible hydrogel specifically mediated the cell spreading. This result suggests that utilization of peptide sequences conjugating with the cell-adhesive motifs can enhance the degree of cell surface interaction and influence the long-term formation of ECM in vitro.

Positively Charged Silver Nanoparticles Threaded on Carbon Nanotube for the Efficient Delivery of Negatively Charged Biomolecules

  • Silver nanoparticle (Ag-NPs)-immobilized and amine-functionalized carbon nanotubes (MWCNTs), MWCNT-Ag-$NH_2$, were easily prepared in order to develop an efficient delivery system of biomolecules without complicated processes of manufacture. For this, Ag-NPs-immobilized MWCNTs, MWCNT-Ag, were initially prepared in order to create large surface area to enable more efficient linkage with guest-molecules using pristine MWCNTs. The Ag-NPs on MWCNTs were further positively functionalized with 2-aminoethanthiol to allow ionic linkage with biomolecules. Ultimately, the positively charged delivery system proved to be highly effective for the binding capacity of bovine serum albumin (BSA) as a negatively charged model protein, when compared to that of lysozyme used as a positively charged model protein. The releasing profile of BSA was observed in almost linear pattern for about two weeks in a saline solution. This study demonstrated the potential usefulness of the pristine MWCNTs in conjunction with Ag-NPs for the selective delivery of many (negatively or positively) charged biomolecules including proteins and genes.
