Characteristic of Aromatic Amino Acid Substitution at α96 of Hemoglobin

  • Replacement of valine by tryptophan or tyrosine at position $\alpha$96 of the $\alpha$ chain ($\alpha$96Val), located in the ${\alpha}_1{\beta}_2$ subunit interface of hemoglobin leads to low oxygen affinity hemoglobin, and has been suggested to be due to the extra stability introduced by an aromatic amino acid at the $\alpha$96 position. The characteristic of aromatic amino acid substitution at the $\alpha$96 of hemoglobin has been further investigated by producing double mutant r Hb ($\alpha$42Tyr$\rightarrow$ Phe, $\alpha$96Val$\rightarrow$Trp). r Hb ($\alpha$42Tyr$\rightarrow$Phe) is known to exhibit almost no cooperativity in binding oxygen, and possesses high oxygen affinity due to the disruption of the hydrogen bond between $\alpha$42Tyr and $\beta$99Asp in the ${\alpha}_1{\beta}_2$ subunit interface of deoxy Hb A. The second mutation, $\alpha$96Val$\rightarrow$Trp, may compensate the functional defects of r Hb ($\alpha$42Tyr$\rightarrow$Phe), if the stability due to the introduction of trypophan at the $\alpha$96 position is strong enough to overcome the defect of r Hb ($\alpha$42Tyr$\rightarrow$Phe). Double mutant r Hb ($\alpha$42Tyr$\rightarrow$Phe, $\alpha$96Val$\rightarrow$Trp) exhibited almost no cooperativity in binding oxygen and possessed high oxygen affinity, similarly to that of r Hb ($\alpha$42Tyr$\rightarrow$Phe). $^1$H NMR spectroscopic data of r Hb ($\alpha$42Tyr$\rightarrow$Phe, $\alpha$96Val$\rightarrow$Trp) also showed a very unstable deoxy-quaternary structure. The present investigation has demonstrated that the presence of the crucible hydrogen bond between $\alpha$42Tyr and $\beta$99Asp is essential for the novel oxygen binding properties of deoxy Hb ($\alpha$96Val$\rightarrow$Trp).

A Study on the Surface Acid Amount and Surface Charge Density of Acid Treated γ-Alumina (산처리한 γ-알루미나의 표면 산량과 표면 전하밀도)

  • This study was carried out to investigate the relation between surface acidity and interfacial electrical characteristics of surface-treated ${\gamma}-alumina$ whose surface activity was increased. The points of zero charge (P. Z. C.) of ${\gamma}-alumina$ whose surface is treated with the sulfuric, nitric and hydro-chloric acid of various concentration were measured from the site-binding theory and mass transport method. The surface active sites were measured by amine titration method and Hammett indicator method. The interfacial properties at alumina/KCl(aq) interface were measured by potentiometric titration. From the experimental results, the following results were obtained. Pure ${\gamma}-alumina$ surface acidity decreases with the increase of calcination temperature at strength $H_o{\leq}+9.3$ Surface-treated alumina acidic properties increase with the anion loading on alumina surface. The surface ionization constants decrease with anion loading on alumina surface, then P. Z. C. decreases with acid amount on alumina surface. Acid amount of surface treated alumina can be correlated with surface charge density at strength $H_o{\leq}+4.8$ as follows. $SO_4^2-/Al_2O_3:Q_A=-0.172ln(0.0418{\sigma}+1.448)$ $NO_3^-/Al_2O_3:Q_A=-0.024{\sigma}-0.0189$ $Cl^-/Al_2O_3:Q_A=-0.01{\sigma}-0.2006$.

Expression and regulation of prostaglandin transporters, ATP-binding cassette, subfamily C, member 1 and 9, and solute carrier organic anion transporter family, member 2A1 and 5A1 in the uterine endometrium during the estrous cycle and pregnancy in pigs

  • Objective: Prostaglandins (PGs) function in various reproductive processes, including luteolysis, maternal pregnancy recognition, conceptus development, and parturition. Our earlier study has shown that PG transporters ATP-binding cassette, subfamily C, member 4 (ABCC4) and solute carrier organic anion transporter family, member 2A1 (SLCO2A1) are expressed in the uterine endometrium in pigs. Since several other PG transporters such as ABCC1, ABCC9, SLCO4C1, and SLCO5A1 are known to be present in the uterine endometrium, this study investigated the expression of these PG transporters in the porcine uterine endometrium and placenta. Methods: Uterine endometrial tissues were obtained from gilts on day (D) 12 and D15 of the estrous cycle and days 12, 15, 30, 60, 90, and 114 of pregnancy. Results: ABCC1, ABCC9, SLCO4C1, and SLCO5A1 mRNAs were expressed in the uterine endometrium, and levels of expression changed during the estrous cycle and pregnancy. Expression of ABCC1 and ABCC9 mRNAs was localized mainly to luminal and glandular epithelial cells in the uterine endometrium, and chorionic epithelial cells during pregnancy. Conceptuses during early pregnancy and chorioallantoic tissues from mid to late pregnancy also expressed these PG transporters. $Estradiol-17{\beta}$ increased the expression of ABCC1 and SLCO5A1, but not ABCC9 and SLCO4C1 mRNAs and increasing doses of $interleukin-1{\beta}$ induced the expression of ABCC9, SLCO4C1, and SLCO5A1 mRNAs in endometrial explant tissues. Conclusion: These data showed that several PG transporters such as ABCC1, ABCC9, SLCO4C1, and SLCO5A1 were expressed at the maternal-conceptus interface, suggesting that these PG transporters may play an important role in the establishment and maintenance of pregnancy by regulating PG transport in the uterine endometrium and placenta in pigs.

  • This study was performed to investigate the effect of humic acid on the adsorption of arsenic onto hematite and its binding mechanism through the chemical speciation modeling in the binary system and the adsorption modeling in the ternary system. The complexation modeling of arsenic and humic acid was suitable for the binding model with the basis of the electrostatic repulsion and the effect of bridging metal. In comparison with the experimental adsorption data in the ternary system, the competitive adsorption model from the binary intrinsic equilibrium constants was consistent with the amount of arsenic adsorption. However, the additive rule showed the deviation of model in the opposite way of cationic heavy metals, because the reduced organic complexation of arsenic and the enhanced oxyanionic competition diminished the adsorption of arsenic. In terms of the reaction mechanism, the organic complex of arsenic, neutral As(III) and oxyanionic As(V) species were transported and adsorbed competitively to the hematite surface forming the inner-sphere complex in the presence of humic acid.

Regulation of SPIN90 by Cell Adhesion and ERK Activation

  • SPIN90 was identified to farm molecular complex with $\betaPIX$, WASP and Nck. This complex shows that SPIN90 interacts with Nck in a manner dependent upon cell adhesion to extracellular matrix, but $SPIN90{\cdot}{\beta}PIX{\cdot}WASP$ complex was stable even in suspended cells. This suggests that SPIN90 serves as an adaptor molecule to recruit other proteins to Nck at focal adhesions. SPIN90 was phosphorylated by ERK1, which was, itself, activated by cell adhesion and platelet-derived growth factor. Such phosphorylation of SPIN90 likely promotes the interaction of the $SPIN90{\cdot}{\beta}PIX{\cdot}WASP$ complex and Nck. It thus appears that the interaction of the $SPIN90{\cdot}{\beta}PIX{\cdot}WASP$ complex with Nck is crucial for stable cell adhesion and can be dynamically modulated by SPIN90 phosphorylation that is dependent on cell adhesion and ERX activation. SPIN90 directly binds syndapin I, syndapin isoform II-1 and II-s via its PRD region in vitro, in vivo and also associates with endocytosis core components such as clathrin and dynamin. In neuron and fibroblast, SPIN90 colocalizes with syndapins as puntate form, consistent with a role for SPIN90 in clathrin-mediated endocytosis pathway. Overexpression of SPIN90 N-term inhibits receptor-mediated endocytosis. Interestingly, SPIN90 PRD, binding interface of syndapin, significantly blocks internalization of transferrin, demonstrating SPIN90 involvement in endocytosis in vivo by interacting syndapin. Depletion of endogenous SPIN90 by introducing $\alpha-SPIN90$ also blocks receptor-mediated endocytosis. Actin polymerization could generate farce facilitating the pinch-out event in endocytosis, detach newly formed endocytic vesicle from the plasma membrane or push out them via the cytosol on actin tails. Here we found that SPIN90 localizes to high actin turn over cortical area, actin-membrane interface and membrane ruffle in PDGF treated cells. Overexpression of SPIN90 has an effect on cortical actin rearrangement as filopodia induction and it is mediated by the Arp2/3 complex at cell periphery. Consistent with a role in actin organization, CFP-SPIN90 present in actin comet tail generated by PIP5 $kinase\gamma$ overexpression. Therefore this study suggests that SPIN90 is functional linker between endocytosis and actin cytoskeleton.

  • ${\pi}$-A isotherms of the Palmitic acid(PA) with increasing pH shifted to the low area/molecule due to the dissociation of PA at the air-water interface. More condensation of PA monolayers occurred by the addition of Mg$^{2+}$, Ca$^{2+}$ and Ba$^{2+}$ ion in subphase. This condensing effect was increased with increasing the concentration of these ions. Due to the interaction with each ion, PA were formed Mg, Ca, Ba-Palmitate complex. The binding structure between alkaline earth ion and carboxylate ligand in PA has been identified by IR spectrometry. The order of condensing effect of alkaline earth ions at pH 8 was Ca$^{2+}$ > Ba$^{2+}$+ > Mg$^{2+}$. The condensing effect except for Mg$^{2+}$ decreased with increasing atomic number. Whereas, the condensing effect in pure water system decreased with decreasing atomic number in the sequence: Ba$^{2+}$ > Ca$^{2+}$ > Mg$^{2+}$. The miscibility of binary system of PA and hexadecanol in monolayer showed that the miscibility was good for the pure water system. But, in the buffered pH 8 system, bad miscibility was found.

Automotive Pre-primed Coatings with Automotive Structural Adhesive for Non-weldable Binding Process (자동차 구조용 접착제를 이용한 자동차용 Pre-primed 도료의 비용접식 접합공정 적용)

  • Currently, automotive pre-primed coatings has been developed to overcome environmental regulations and to reduce manufacturing cost in automotive industry. By these reasons, an automotive pre-primed system has been investigated to remove the wash and pre-treatment process using a roll coating application. It is required to develop non-weldable pre-primed system for automotive structural adhesives, because pre-primed sheet coated with organic compounds is hard to be assembled by welding process. Primer 1 (polyester type) and primer 2 (urethane type) were designed to satisfy flexibility and formability for non-weldable pre-primed system. According to the results of physical property test of the primers, adhesion test such as single-lap shear test and T-peel test, primer 1 (polyester type) had better physical properties such as pencil hardness, solvent resistance, flexibility and adhesion with automotive adhesive than that of primer 2 (polyurethane type). In addition, the possibility of the non-weldable pre-primed system was applicable to automotive assembly process in place of welding process.

A Study on the Kiosk UI Reflecting the Elderly's Characteristics (고령자의 특성을 반영한 패스트푸드점 키오스크 UI 연구)

  • This study was conducted to improve the use experience of elderly people in fast food kiosks which are actively utilizing 'un-tact' technology. First, the Kiosk design analysis criteria were discussed through literature research. Then, the UI screens of McDonald's, Lotteria and Burger King, which are currently serving kiosks in Korea, were divided into colors, layouts and buttons. Next, the usability evaluation and survey of the elderly revealed problems with the interface design of the current kiosk. As a result, older people do not recognize buttons without an outline as buttons, and errors occur when layout changes or when viewing is complicated. In addition, horizontal navigation and vertical layout menus were error-free, and the design of buttons was error-free with the same color for the same function. Research shows that the Kiosk UI design is preferred by older people. First, buttons in the form of outline or solid are recommended. Second, vertical binding of buttons combined with images and text is recommended. Third, the screen layout should be consistent with the direction of view flow and manipulation, and should avoid increasing the number of menus and information. Fourth, page navigation is recommended in horizontal form and menu arrangement is vertical form. Finally, buttons that perform the same function are efficient for buttons to use the same color, and for different functions, applying a complementary colors can reduce confusion.

The Effect of the Heat Treatment of the ZrO2 Buffer Layer and SBT Thin Film on Interfacial Conditions and Ferroelectric Properties of the SrBi2Ta2O9/ZrO2/Si Structure (ZrO2 완충층과 SBT 박막의 열처리 과정이 SrBi2Ta2O9/ZrO2/Si 구조의 계면 상태 및 강유전 특성에 미치는 영향)

  • To investigate the possibility of the $ZrO_2$ buffer layer as the insulator for the Metal-Ferroelectric-Insulator-semiconductor (MFIS) structure, $ZrO_2$ and $SrBi_2Ta_2O_9$ (SBT) thin films were deposited on the P-type Si(111) wafer by the R.F. magnetron-sputtering method. According to the process with and without the post-annealing of the $ZrO_2$ buffer layer and SBT thin film, the diffusion amount of Sr, Bi, Ta elements show slight difference through the Glow Discharge Spectrometer (GDS) analysis. From X-ray Photoelectron Spectroscopy (XPS) results, we could confirm that the post-annealing process affects the chemical binding condition of the interface between the $ZrO_2$ thin film and the Si substrate. Compared to the MFIS structure without the post-annealing of the $ZrO_2$ buffer layer, memory window value of MFlS structure with post-annealing of the $ZrO_2$ buffer layer were considerably improved. The window memory of the Pt/SBT (260 nm, $800^{\circ}C)/ZrO_2$ (20 nm) structure increases from 0.75 to 2.2 V under the applied voltage of 9 V after post-annealing.

High Temperature Properties of Fire Protection Materials Using Fly Ash and Meta-Kaolin (Fly Ash 및 Meta-Kaolin을 활용한 내화성 마감재의 고온특성)

  • The serious issue of tall building is to ensure the fire-resistance of high strength concrete. The fire resistant finishing method is necessarily essential in order to satisfy the fire resistance time of 3 h required by the law. The fire resistant finishing method is installed by applying a fire resistant material as a method of shotcrete or a fire resistant board to high strength concrete surface. This method can reduce the temperature increase of the reinforcement embedded in high strength concrete at high temperature due to the installation thickness control. This study is interested in identifying the effectiveness of inorganic alumino-silicate compounds including the inorganic admixture such as fly ash and meta-kaolin as the fire resistant finishing materials through the analysis of fire resistance and components properties at high temperature. The study results show that the fire resistant finishing material composed of fly ash and meta-kaolin has the thermal stability of the slight decrease of compressive strength at high temperature. These thermal stability is caused by the ceramic binding capacity induced by alkali activation reaction by the reason of the thermal analysis result not showing the decomposition of calcium hydrate. Inorganic compounds composed of fly ash and meta-kaolin is evaluated to be very effective as the fire resistance material for finishing to protect the concrete substrate by the reason of those simplicity in both application and manufacture. The additional study about the adhesion in the interface with concrete substrate is necessary for the purpose of the practical application.
