The isoflavone glycosides are hydrolyzed by ${\beta}$-glucosidase from gut microbes to the bioactive aglycones. However, the specific bacteria from the human intestinal tract that are involved in the metabolism of these compounds are not known. This study was undertaken to develop a fermented soymilk which converts isoflavones to the more bioactive aglycones form using a Bifidobacterium strain. The ${\beta}$-glucosidase activity of 15 Bifidobacterium strains were measured during cell growth. Among them, Bifidobacterium sp. Int-57 was selected for this study, because it has the highest ${\beta}$-glucosidase activity. Growth, acid development, ${\beta}$-glucosidase activity, and the hydrolysis of daidzin and genistin were investigated in four soymilks inoculated with Bifidobacterium sp. Int-57. After 12 h of fermentation, the counts of viable Bifidobacterium sp. Int-57 in all the soymilks reached a level of more than $10^8$ cfu/ml, which was then maintained. The pH of soymilks started to decrease rapidly after 6 h of fermentation and leveled off after 18 h. The titratable acidity of BL# 1 soymilk, BL#2 soymilk, and JP#l soymilk increased from 0.18 to 1.21, 1.15, and $1.08\%$ over the fermentation period, respectively. After 24 h of fermentation, the $\beta$-glucosidase activity in BL#1 soymilk, BL#2 soymilk, JP#l soymilk, and JP#2 soymilk increased to 59.528, 40.643, 70.844, and 56.962 mU/ml, respectively. The isoflavone glycosides, daidzin and genistin, in soymilks were hydrolyzed completely in the relatively short fermentation time of 18 h. These results show that Bifidobacterium sp. Int-57 can be used as a potential starter culture for developing fermented soymilk which has completely hydrolyzed isoflavone glycosides.
한국인의 분변으로부터 분리된 Bifidobacterium sp. Int-57은 다른 장내세균에 비해 ${\beta}-xylosidase$의 높은 역가를 보였으며, xylooligomer의 bifidogenic factor의 유용성을 고려하였을 때 효소생산에 초점을 둔 결과 최적 탄소원으로는 xylose였는데, 일반적으로 최종생산물은 feedback inhibition, feedback repression 등으로 효소 생산을 억제하는 일반 이론과는 상이한 결과를 보였다. 또한 xylose는 1.1%일 때 가장 높은 효소역가를 보였다. 질소원으로서는 yeast extract였고, 그 농도가 0.04%일 때, 무기염류 면에서는 $CoCl_2$였고, 농도가 0.0003%일 때 ${\beta}-xylosidase$의 최대 생산을 보였다.
${\alpha}-galactosidase$는 raffinose와 stachyose 등의 ${\alpha}-galactoside$ 결합을 분해하는 효소이다. Raffinose와 stachyose는 특히 두류에 다량 존재하는데 이들 당은 섭취후 복부팽만감의 원인 물질로 발표되었는가 하면 한편으로는 비피더스균 증식인자로 최근에 보고된 바 있기 때문에 이들 당의 인체 건강에 대한 재평가가 요구되고 있다. 본 연구에서는 이에 대한 기초 자료로 사용하기 위하여 한국인으로부터 분리된 Bifidobacterium sp. Int-57이 생산하는 ${\alpha}-galactosidase$의 효소 특성을 조사하였다. Bifidobacterium sp. Int-57은 여러 종류의 다른 장내 세균에 비하여 높은 ${\alpha}-galactosidase$ 활성을-보유하고 있고 raffinose에 의하여 효소 생산이 증진되었다. 세포의 음파 파쇄후 DEAE-celluose와 Sepharose 6B gell filtration을 거쳐 부분정제한 효소를 사용하여 PNP-${\alpha}-galactosidase$에 대한 작용을 분석한 결과 최적 pH는 7.0, 최적온도는 $40^{\circ}C$ 였고 금속이온을 5mM 농도로 처리하였을 때 효소활성을 $CoCl_{2}$는 33%, $CuCl_{2}$는 21% 저해하였다. 또한 얻어진 효소는 실제 기질로써 raffinose와 stachyose를 잘 분해하는 것으로 나타났다.
Ginsenosides have been regarded as the principal components responsible for the pharmacological and biological activities of ginseng. The transformation of ginsenosides with live lactic acid bacteria transformed ginsenosides Rb2 and Rc into Rd, but the reactions were slow. When the crude enzymes obtained from several lactic acid bacteria were used for transformation, those from Bifidobacterium sp. Int57 exhibited the most potent transforming activity of ginsenosides to compound K. In comparison, a relatively higher level of Rh2 was produced by the enzymes from Lactobacillus delbrueckii and Leuconostoc mesenteroides. These results suggest that it is feasible to develop a specific bioconversion process to obtain specific ginsenosides using the appropriate combination of ginsenoside substrates and specific microbial enzymes.
본 연구에서는 프로바이오틱스로 사용하는 유용 미생물과 식용으로 안전하게 사용 가능한 미생물 자원을 이용하여 배당체의 전환 연구를 실시하여 전환체의 새로운 생리활성 작용에 대하여 연구하였다. 연구에 사용한 주요 소재로는 인삼의 사포닌 배당체, 길경의 platycodin 배당체, 호로파의 사포닌 배당체, 칡과 두유의 이소플라본 배당체 등이었다. 사용 균주로는 bifidobacteria, lactobacilli, leuconostocs, yeasts, aspergilli 등이었다. 유산균 중에는 Bifidobacterium sp. Int-57의 전환능이 우수하였고 식용 곰팡이 중에는A. niger의 전환능이 우수하였다. 다양한 미생물 또는 효소를 이용한 전환체에 대하여 아질산 소거능, 항산화능, a-glucosidase와 a-amylase 저해능 등을 조사하였을 때 일반적으로 전환 후의 활성이 중가하였다. 길경 사포닌은 전환 후에 세포 독성이 줄어들었으며 관능성이 유의적으로 개선되었다. 결론적으로, 프로바이오틱스와 식용 미생물을 이용하여 천연물 중에 존재하는 배당체의 생리활성 및 관능성을 증가시킬 수 있어 새로운 기능성 식품의 개발에 활용할 수 있을 것이다.
The intestinal microflora of humans is an extraordinarily complex mixture of microorganisms, the majority of which are anaerobic microorganisms. The distribution of amylolytic microorganisms in the human large intestinal tract was investigated in various individuals of differing ages using anaerobic culture techniques. A large percentage of the amylolytic microorganisms present belonged to the Genus Bifidobacteria. The number of Bifidobacteria increased significantly at two years of age. Adults and children above 2 years old carried about $0.8{\times}10^9-2.0{\times}10^{10}$ colony forming units (CFU/gram) of amylolytic Bifidobacteria. Among these amylolytic Bifidobacteria, Int-57 was chosen for further studies. Between 65% and 85% of the amylase produced was secreted and the remaining amylase was bound to the cell wall facing the outside. Amylase production could be induced by starch in a stable form. When cells were grown on maltose or glucose, amylase production was much lower than on starch and amylase activity disappeared after 24 hours growth on these media. Partially purified enzymes showed optimum activity at a temperature of $50^{\circ}C$ and at an optimum pH of 5.5, respectively. Heat treatment at $70^{\circ}C$ for 30 minutes almost completely inactivated amylase. The hydrolysis products of starch were mainly maltose and maltotriose. Soluble starch, amylose, amylopectin, and $\gamma$-cyclodextrin($\gamma$-CD) were easily hydrolyzed. The rate of hydrolysis of $\alpha$-CD and $\beta$-CD was slower than that of $\gamma$-CD. Carboxymethyl cellulose, $\beta$-1, 3-glucan and inulin were not hydrolyzed.
