Objectives This study was performed to evaluate the effects of fermented lotus extracts on prediabetes and hyperlipidemia in high fructose diet rats. Methods Extracts of lotus leaf and lotus root were fermented using 4 different probiotics separately, including Lactobacillus plantarum, Lactobacillus rhamnosus, Bifidobacterium breve, and Bifidobacterium longum. Expressions of adipogenic transcription factors including Adiponectin, GLUT-4, Leptin, PPAR gamma, Resistin and Visfatin were analyzed by Real time PCR and Western blotting analysis. Results Fermented lotus extracts reduced blood glucose. Fermented lotus extracts inhibited adipogenic transcription factors by inhibiting preadipocytes differentiation. The level of gene expression of Adiponectin, GLUT-4, Leptin, PPAR gamma, Resistin and Visfatin in relation to that of GAPDH were increase or decrease significantly with the Fermented lotus formulation group. Conclusions Fermented lotus extracts showed hypoglycemic and hypolipidemic effects by inhibiting preadipocyte differentiation and controlling insulin sensitivity in high fructose diet rats.
Clostridium difficile infection (CDI) has become a significant threat to public health. Although broad-spectrum antibiotic therapy is the primary treatment option for CDI, its use has evident limitations. Probiotics have been proved to be effective in the treatment of CDI and are a promising therapeutic option for CDI. In this study, 4 strains of lactic acid bacteria (LAB), namely, Lactobacillus rhamnosus (LR5), Lactococcuslactis (SL3), Bifidobacterium breve (BR3), and Bifidobacterium lactis (BL3) were evaluated for their anti-C. difficile activity. Co-culture incubation of C. difficile ($10^6$ and $10^{10}$ CFU/ml) with each strain of LAB indicated that SL3 possessed the highest antimicrobial activity over a 24-hr period. The cell-free supernatants of the 4 LAB strains exhibited $MIC_{50}$ values between 0.424 mg/ml (SL3) and 1.318 (BR3) mg/ml. These results may provide a basis for alternative therapies for the treatment of C. difficile-associated gut disorders.
The antitumor activity of Bifidobacterium breve K-110, and K-I11, and B. infantis K-525 was investigated. These Bifidobacterial cells and their cell wail preparations (WPG) significantly increased the survival rate of mice who had been intraperitoneally implanted with sarcoma 180 cells. Solid tumor growth was inhibited even when the sarcoma 180 cells were implanted into the groins of the mice. However, the Bifidobacterial cells did not show in vitro cytotoxicity against tumor cell lines. Cell kinetic studies revealed that these WPGs induced neutrophils, which were followed by macrophages, at the site of peritoneal injection. The WPGs directly activated these cells to inhibit the growth of tumor cells in vitro assays. Our results suggest that Bifidobacterial WPGs induce and activate nonspecific phagocytes in situ to reject growing tumor cells in the mouse peritoneal cavity.
This study was performed to elucidate substrate specificity to the locust bean gum galactomannan by Trichoderma harzianum ${\beta}-mannanase$. The medium composition for enzyme production were determined 3% cellulose, 3% corn steep liquor, 1% $KH_2PO_4$, 0.2% $(NH_4){_2}SO_4$, and incubated for 115 hr at $28^{\circ}C$. The ${\beta}-mannanase$ exhibited maximum activity at pH 4.5 and $60^{\circ}C$. Locust bean gum galactomannan was hydrolyzed by the ${\beta}-mannanase$, and then hydrolysates separated by activated carbon column chromatography. The main hydrolysates were composed of D.P 4 and 7 galactosyl mannooligosaccharides by TLC. For the elucidate the structure of D.P 4 and 7 oligosaccharides, methylation analysis was performed. D.P 4 and 7 were identified as M-M-M-M and M-M-M-M-M (G- and M-represent ${\alpha-1,6-D-galactosidic\;and\;{\beta}-1,4-mannosidic$ linkages, respectively). //G-G To investigate the effects of locust bean gum galactosyl mannooligosaccharides on the in vitro growth of B. longum, B. bifidum, B. infantis, and B. breve, Bifidobacterium spp. were cultivated individually on the modified-MRS medium containing carbon source such as D.P 4 and 7 galactosyl mannooligosaccharides, respectively. B. longum grew up 3.4-fold and 4.3-fold more effectively by the replacement of D.P 4 and 7 galactosyl mannooligosaccharides as the carbon source in a comparasion of standard MRS.
The present study investigated the effects of Bisroot, that contains live bacteria (Bacillus polyfermenticus, Bacillus mesentericus, Streptococcus faecalis, & Bifidobacterium breve) and digestive enzymes (protease, lipase), on the growth, body composition and immune response of Nile tilapia fingerlings. One percent of the Bisroot was added to the experimenta feed. All exprimental fish were fed for 60 days. The weigh gains among the experimental fish were not significntly different (P>0.05). Hematocrit value, hemoglobin, total protein, glucose, GOT, and GPT were unaffected by Bisroot treatment. However, it was observed that glucose, GOT, and GPT value in the fish that were fed Bisroot, were lower than the control. The complement activity ($CH_50$) tended to be significantly increased by Bisroot treatment, but not lysozyme activity. Phagocytosis and respiratory burst activities of macrophages in the head kidney were enhanced by Bisroot. Therefore, the Bisroot diet enhances the cellular immune activities were enhanced by Bisroot. Therefore, the Bisroot diet enhances the cellular immune activities of non-specific immune responses. When fish were challenged with a virulent strain of Edwardsiella tarda, the Bisroot treated fish were more resistant than the control. The present results suggest that the introduction of Bisroot into the diet of Nile tilapia could increase their resistance against bacterial infection, reduce fish mortality, and offers economic benefits.
Alginate lyase from Streptomyces violaceoruber was purified by DEAE sephacel chromatography and SP sepharose chromatography. The specific activity of the purified enzyme was 14.6 units/mg protein, representing a 40.6-fold purification of the crude extract. The final preparation thus obtained showed a single band on Tricine-SDS polyacrylamide gel electrophoresis whose molecular weight was determined to be 23.3 kDa. The polyMG block of sodium alginate was hydrolyzed by the purified alginate lyase and then separated by activated carbon column chromatography and bio gel P-2 gel filtration. The main hydrolysates were composed of hetero type M/G-oligosaccharides with the degrees of polymerization (D.P.) being 6 and 8. To investigate the effects of hetero type M/G-oligosaccharides from the sodium alginate on the growth of some intestinal bacteria, cells were cultivated individually on the modified-MRS medium containing D.P. 6 and 8 M/G-oligosaccharides. B. longumgrew 4.25-fold and 6.44-fold more effectively by the treatment of D.P. 6 and 8 M/G-oligosaccharides compared with those of standard MRS medium. In addition, B. bifidumgrew 3.3-fold and 5.4-fold more effectively by the treatment of D.P. 6 and 8 M/G-oligosaccharides. In conclusion, D.P. 8 was more effective than D.P. 6 hetero M/G-oligosaccharides as regards the growth of Bifidobacteriumspp. and Lactobacillus spp.

In this study, we identified that the fermentation of Korean indigenous probiotics and red ginseng produced ginsenoside compound K (CK) from major ginsenosides. Based on whole genome sequencing of 19 probiotics species, β-glucosidase, α-arabinofuranosidase, β-xylosidase, and α-rhamnosidase related to bioconversion of ginsenosides are identified in the genome of 19 species, 3 species, 6 species, and 8 species, respectively. Among the 19 probiotics species, Bifidobacterium longum CBT BG7 converted from ginsenoside Rb1 to CK, and both B. breve CBT BR3 and B. lactis CBT BL3 converted ginsenoside Rb1 to Rd. The final concentration and yield of ginsenoside F2 and CK were higher in the fermentation with the nondisrupted cells than with disrupted cells. The combination of both CBT BG7 and BL3, and CBT BG7 and BR3 showed higher amounts of F2 than CBT BG7 only. CBT BG7 with adding α-amylase increased the amounts of F2. In this study, we identified that the fermentation of both Korean indigenous probiotic bacteria CBT BG7, BR3 and BL3, and red gingseng is able to produce CK, a bioactive compound that promotes health benefits.

The growth responses of a variety of human Intestinal bacteria to extracts of Pun(1.vKy'n.ieny and five other oriental medicinal Araliaceae were evaluated in vitro and in vivo. The extracts enhanced the growth of Bifidobncterilim breve and B. longum in Media with or without carbon sources, suggesting the bifid factor (5) might be involved in the phenomenon. This effect was most pronounced with water extract of p. ginseng, the growth of 27 bifidobacteria strains belonging to B. ndolexcentium, H. longlrm, and 1. breve and B. iniuntis being greatly stimurated, whereas seven B. bifidum strains and other bacteria such as clostridia and 5.fcherirhia coli had little or no ability to utilizes it (or growth. Methanol extracts of p, ginseng were found to selectively inhibit growth of various clostridia including C. perfringens and C. Paraputrificum, but this effect was not observed on other bacteria including bifidobacteria. The effect of ginseng extract intake(600 mg/day for two weeks) on the fecal microflora, pH, volatile fatty acids, ammonia, putrefactive products, and -glucuronidase, -glucosidase and nitroreductase activities, and on the blood components (triglyceride, total cholesterol and ammonia) were investigated using seven healthy human volunteers. The total concentration of fecal microflora including Bri'idobucterilim app. during the period of ginseng extract intake was significantly unaffected from the proceeding and sub sequent control periods. However, the frequency of occurrence of subjects having C. perfringens was significantly decreased. The fecal pH value was also significantly decreased, suggesting that the intake might increase the activity of Bifidobacterium spp. Other biochemical properties in faces did not changed significantly. The levels of ammonia and triglycerid in blood were decreased with ginseng extract intake. These results may be an indication of at least one of the pharmacological actions of P ginseng as an adaptogen.

The purpose of this study was to develop a simple detection method, possibly at the species-level, that allows for large-scale screening of bifidobacteria. Human fecal samples were plated on MRS-raffinose agar containing cysteine and neomycin sulfate, serving as selective pressure for bifidobacteria, and 0.003%(w/v) bromocresol green. All of the test strains grew well on this medium at $37{\pm}1^{\circ}C$, forming white colonies surrounded by yellow halos, which presented a sharp contrast against the green background. In this disc assay, the required incubation time to develop a yellowish zone varied with the species of Bifidobacterium that was tested, allowing for differential counts and easy identification at the species-level: 10-14 hr for B. bifidum, 20-22 hr for B. catenulatum and B. infantis. and 24-25 hr for B. longum and B. breve. No apparent color was observed for B. angulatum and B. adolescentis 28 hr after inoculation. To evaluate the results of pH indicator-based identification, individual isolates were subjected to a colony-PCR experiment with genus-specific primers. The amplified products from the isolates were in good accordance with those from the reference strains at a level of 95% agreement. These results suggest that the present method could be conveniently applied to cell counts, as well as to the preliminary identification of bifidobacteria from a variety of sample types including human feces, dairy products, and commercial probiotic supplements.

This study aimed to improve the physiological activity of onion juice via lactic acid bacterial fermentation. Seven types of lactic acid bacteria were used for the fermentation of onion juice. The pH and sugar content of the onion juice decreased, while its titratable acidity increased after lactic acid bacteria fermentation, and the cell count of lactic acid bacteria was 7.31-10.40 log CFU/mL. The total free sugar content decreased, while the total organic acid content increased in the fermented onion juice. Quercetin content of the fermented juice was 0.13-0.53 mg/kg. The total polyphenol and flavonoid contents increased after fermentation. Additionally, the 2,2-diphenyl-1-picryl-hydrazyl-hydrate free radical and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonate) radical cation scavenging activities were increased by fermentation. Overall, lactic acid bacteria fermentation of onion juice enhanced its physiological activity. Based on these findings, Bifidobacterium breve KCTC 3441 was selected as the onion juice fermentation strain.
