In 2015, stem blight of Rubus crataegifolius was observed in Pohang, Korea. The symptoms began as dark red spots in the stem, which led to stem blight, then leaf blight, and eventually resulted in death. A fungal isolate was obtained from a symptomatic stem and incubated on a potato dextrose agar plate. The isolated fungus produced white, cloudy mycelia turned black in 3 days. Based on the morphological characteristics, the causal fungus was assumed to be Botryosphaeria sp. A pathogenicity test was conducted according to Koch's postulates. To identify the causal agent, the combined sequence of the internal transcribed spacer, ${\beta}$-tubulin, and translation elongation factor $1{\alpha}$ genes were used for phylogenetic analysis. Approximately 1,200 bp of the combined sequence clearly suggested that the isolated pathogen was Botryosphaeria parva. This is the first report on stem blight in R. crataegifolius caused by B. parva in Korea.
To characterize benzimidazole-resistant and -sensitive Monilinia fructicola populations, the fungal isolates were obtained from peach plants showing brown rot and bloosom blight. Benzimidazole-sensitive isolates did not grow on potato dextrose agar(PDA) amended with $\geq1.0{\mu}g$ active ingredient(a.i.)/ml of the fungicides. However, benzimidazole-resistant isolates grew on PDA regardless of the tested concentrations of fungicides. Benzimidazole-resistant isolates did not grow on diethofencarb-PDA, but sensitive isolates grew on the same PDA. In the nucleotide sequences of $\beta$-tubulin gene, only codon 198(GAG: glutamic acid), a target site for benzimidazole, was replaced with GCG(alanine) in all of the resistant isolates, and this substitution seems to play an important role in the development of resistance. Other interesting codons such as 165(GCT), 200(TTC), and 241(GCT) were not changed among the isolates. Benzimidazole-resistant and -sensitive isolates were clustered clearly in random amplified polymerphic DNA analyses and the results revealed that low levels of genetic diversity between benzimidazole-sensitive and -resistant isolates of M. fructicola in the investigated regions.
During an investigation of the fungal diversity of Korean soils, four Penicillium strains could not be assigned to any described species. The strains formed monoverticillate conidiophores with occasionally a divaricate branch. The conidia were smooth or finely rough-walled, globose to broadly ellipsoidal and $2.5-3.5{\times}2.0-3.0{\mu}m$ size. Their taxonomic novelty was determined using partial ${\beta}$-tubulin gene sequences and the ribosomal internal transcribed spacer region. The phylogenetic analysis showed that the isolates belonged to section Lanata-Divaricata and were most closely related to Penicillium raperi. Phenotypically, the strains differed from P. raperi in having longer and thicker stipes and thicker phialides. Strain KACC $47721^T$ from bamboo field soil was designated as the type strain of the new species, and the species was named Penicillium koreense sp. nov., as it was isolated from various regions in Korea.
Eighteen endophytic fungi with different colony morphologies were isolated from the roots of Nymphoides peltata growing in the Dalsung wetland. The fungal culture filtrates of the endophytic fungi were treated to Waito-c rice seedling to evaluate their plant growth-promoting activities. Culture filtrate of Y2H0002 fungal strain promoted the growth of the Waito-c rice seedlings. This strain was identified on the basis of sequences of the partial internal transcribed spacer region and the partial beta-tubulin gene. Upon chromatographic analysis of the culture filtrate of Y2H0002 strain, the gibberellins (GAs: $GA_1$, $GA_3$, and $GA_4$) were detected and quantified. Molecular and morphological studies identified the Y2H0002 strain as belonging to Aspergillus clavatus. These results indicated that A. clavatus improves the growth of plants and produces various GAs, and may participate in the growth of plants under diverse environmental conditions.
A representative isolate KU53 of the predominant Penicillium species was obtained from rice samples from rice processing complexes of National Agricultural Cooperative Federation in Korea. In this study, isolate KU53 was identified by its morphological and molecular characteristics. The macro- and microscopic characteristics of isolate KU53 were compared with the P. fellutanum reference isolate KCTC16913 on different media; isolate KU53 was generally identical to those of the reference isolate KCTC16913. In a molecular-based identification, the ${\beta}$-tubulin and translation elongation factor 1-alpha sequences of isolate KU53 was most closely related to those of P. fellutanum. Thus, isolate KU53 from stored rice could be identified as P. fellutanum, some isolates of which are known to produce mycotoxin-related metabolites. To our knowledge, this is the first detection of P. fellutanum from stored rice in Korea.
In this study, we isolated endophytic fungal strains from the rhizoid and the leaf of Mnium heterophyllum and Hypnum plumaeforme, respectively. The isolated strains were identified based on morphological characters and molecular analysis of the internal transcribed spacer, large subunit rDNA, β-tubulin, and RNA polymerase II subunit regions. From the results, we confirmed two endophytic fungal species, Cercophora thailandica, and Biscogniauxia petrensis, which to the best of our knowledge, have not yet been reported in Korea. We further describe the morphological characteristics and the results of the molecular analyses of these isolated fungal strains.

Although action modes of equol and genistein have been extensively studied, their precise roles in tumor cells remain elusive. To address possible effects of these compounds on protein expression in mammary tumor cells, proteins modulated in MCF-7 mammary tumor cells when incubated in absence and presence of 10 uM equol or genistein were identified through 2-dimensional gel electrophoresis, MALDI-TOF MS/MS, and NCBInr database search using Mascot software. Most proteins differentially expressed in MCF-7 cells after treatment with 10 uM genistein or equol were identified as being the same. Exposure to both compounds caused decreased cellular expression of RNA-binding protein regulatory subunit and oncogene DJ1 tubulin beta-1 chain, and increased expression of heterogeneous ribonucleoproteins F and L, KH-type splicing regulatory protein, and translation elongation factor EF-Tu precursor. Genistein and equol at dose used in this study showed common action mechanism.
In the present work, a pair of primers specific to Tomato yellow leaf curl virus (TYLCV) was designed to allow specific amplification of DNA fragments from any TYLCV isolates using an extensive alignment of the complete genome sequences of TYLCV isolates deposited in the GenBank database. A pair of primers which allows the specific amplification of tomato ${\beta}$-tubulin gene was also analyzed as an internal PCR control. A duplex PCR method with the developed primer sets showed that TYLCV could be directly detected from the leaf crude sap of infected tomato plants. In addition, our developed duplex PCR method could determine PCR errors for TYLCV diagnosis, suggesting that this duplex PCR method with the primer sets is a good tool for specific and sensitive TYLCV diagnosis. The developed duplex PCR method was further verified from tomato samples collected from some farms in Korea, suggesting that this developed PCR method is a simple and reliable tool for rapid and large-scale TYLCV detections in tomato plants.
Pear skin stains on 'Niitaka' pears, which occur from the growing stage to the cold storage stage, reportedly negatively influence the marketing of pears. These stains on fruit skin are likely due to a pathogenic fungus that resides on the skin and is characterized by dark stains; however, the mycelium of this fungus does not penetrate into the sarcocarp and is only present on the cuticle layer of fruit skin. A pathogenic fungus was isolated from the skin lesions of infected fruits, and its pathogenicity was subsequently tested. According to the pathogenicity test, Mycosphaerella sp. was strongly pathogenic, while Penicillium spp. and Alternaria spp. showed modest pathogenicity. In this present study, we isolated the pathogenic fungus responsible for the symptoms of pears (i.e., dark brown-colored specks) and identified it as Mycosphaerella graminicola based on its morphological characteristics and the nucleotide sequence of the beta-tubulin gene. M. graminicola was pathogenic to the skin of 'Niitaka' pears, which are one of the most widely growing varieties of pears in South Korea.
Photoperiod sensitive genetic male sterile (PGMS) rice is sterile mutant controlled by photoperiod. A PGMS mutant 920S was sterile grown under long-day (LD) photoperiod (14 h light/10 h dark) but fertile grown under short-day (SD) photoperiod (10 h light/14 h dark). Proteome analysis revealed that 12 protein spots were differentially expressed in the spikelets of 920S plants either treated with LD or SD photoperiod. Among these proteins, three proteins including chlorophyll a/b binding protein, vacuolar ATPase ${\beta}-subunit,\;{\alpha}-tubulin$ and an unknown protein were more than three-fold abundant in the spikelet of the SD-treated plants than those of the LD-treated plants. On the other hand, eight proteins including acetyl transferase, 2, 3- biphosphoglycerate, aminopeptidase N, pyruvate decarboxylase, 60S acidic ribosomal protein and three unknown protein spots were more abundant in the spikelets of the LD-treated plants than those of the SD-treated plants. The results suggest that the observed proteins may be involved in sterile or fertile pollen development under LD or SD photoperiod respectively in the PGMS mutant rice.
