Synthesis of Silicon Nitride from Kimcheon Quartzite (김천규석으로부터 질화규소의 합성)

  • Silicon nitride powders were prepared by the simultaneous reduction and nitridation from powder mixtures of Kim cheon quartzite and carbon (graphite or carbon black) at1400$^{\circ}C$ for 10 hours in nitrogen atmosphere. The effects of the reaction variables on the yield of products and on the ${\alpha}$/${\beta}$ ratio were examined. The average particle size, density, and the ${\alpha}$/${\beta}$ ratio of the obtained si3N4 were 1.0$\mu\textrm{m}$, 3.10g/㎤ and 90/10, respectively. It was found that the Si3N4 powders obtained in this work were comparable to the foreign commercial products.

  • Blue phosphor of Ba-Mg-Al-O:Eu$^{2+}$ phase was fabricated by conventional firing techniques under reducing atmosphere and its photoluminescence properties are studied with varying Eu concentration and phost-annealing temperature under air atmosphere. This phosphors were well crystallized with particle size in the range of 3~5um and emitted a blue light at a dominent wavelength 450nm for 254nm UV irradiation. The concentration quenching wit Eu$^{2+}$ was that with increasing Eu concentration the energy transfer between the activator ions steadily improves, so that the excitation energy is transported over larger distances through the lattice before luminescence can occur. Thermal quenching also occurred in this phosphor means that in a host lattice with the $\beta$-alumina structure the bond of an Eu$^{2+}$ ion with the nearest-neighbour oxygen ion is much stronger than in a lattice with the magnetoplumbite structure.cture.

  • Mixtures of very small amounts of additive, carbon and silica(about 0.46${\mu}{\textrm}{m}$) which synthesized by the hydrolysis of ethyl silicate, the molar ratio of SiO2/C was fixed to 1/10, was nitrided at 145$0^{\circ}C$. It was considered that the optimum amount of additive to promote the nitridation reaction was below 2.0wt%. By the addition of additive, the nitridation reaction was promoted and formation of $\beta$-Si3N4 was promoted at 145$0^{\circ}C$ for 1hour, but, the nitridation reaction was decreased and the ratio of $\alpha$/$\beta$ of Si3N4, was increased at 145$0^{\circ}C$ for 5 hours. The crystal phase was $\alpha$ phase and the nitridation reaction was promoted and the particle size of silicon nitride was become smaller by the addition of $\alpha$-Si3N4, but silicon nitride of whisker-like form was produced by the addition of transition elements. There was a difference in the lattice constants of $\alpha$-Si3N4, but no difference in its of $\beta$-Si3N4 according to kinds of added substance and reaction time.

  • The strain of Serratia marcescens QM B1466 produces selectively large amount of chitinolytic enzymes (about 1mg/L medium). Enzymatic hydrolysis of chitin to N-acetyl-${\beta}$-D-glucosamine (NAG) was performed with a system consisting of two hydrolases (chitinase and chitobiase) produced by optimization of a microbial host consuming chitin particles. For the development of Large-scale biological process for the production of NAG from chitinaceous waste, the selection and optimization of a microbial host, particle size of crab/shrimp chitin sources and initial induction time using chitin as a sole carbon source on chitinase/chitobiase production and NAG production were examined. Crab-shell chitin(1.5%) treated by dilute acid and , ball-milled with a normal diameter less than 250m gave the highest chitinase activity over a 7 days culture. Crude chitinase/ chitobiase solution obtained in a 10 L fed-batch fermentation showed a maximum activities of 23.6 U/mL and 5.1 U/mL, respectively with a feeding time of 3 hrs, near pH 8.5 at 30$^{\circ}C$.

  • Stroke occurs when local thrombosis, embolic particle or the rupture of blood vessele interrupts the blood floe to the brain. $\beta$-estradiol 17-valerate has been reported to exert neuroprotective effects when administered before an ischemic insult. Recently, the pathophysiology of cerebral ischemia has been studied extensively in rat with various methods. In the present study, we investigates whether $\beta$-estrodiol 17-valerate can protect against brain injury. RNA sample were extracted from the hippocampus of female rat, reverse-transcription in the presence of [$\alpha$32p] dATP. Differential gene express-ion profiles were revealed (Bone morphogenetic protein type 1A receptor, Protein disulphide isomerase, Leukemia inhibitor factor receptor, cytochrome bc- 1 complex-x core P, thiol-specific antioxidant protein). RT-PCR was used to validate the relative expression pattern obtained by the cDNA array. The precise relationship between the early expression of recovery genes and stroke is a matter of luther investigation.   • The microstructural evolution during mechanical alloying of elemental Fe and Si powders, average composition $Fe_{30}Si_{70}$ and $Fe_{50}Si_{50}$, has been investigated by X-ray diffraction (XRD), Scanning electron microscopy (SEM) and Differential scanning calorimetry (DSC). Mechanical alloying was performed by using a SPEX 8000 Mixer/Mill under argon atmosphere with/without hexane as a process control agent (PCA). In the presence of PCA, the milling process was dominated by fracture resulting in the decrease in particle size to about $1{\mu}m$. The structural development with milling time depended on the average composition of starting powders. The mixture of $Fe_{50}Si_{50}$ and $Fe_{30}Si_{70}$ resulted in the formation of FeSi(${\varepsilon}$ - phase) and $FeSi_2$(${\beta}$ - phase), respectively. In the case of $Fe_{33.3}Si_{66.7}$, a mixture and $FeSi_2({\beta})$ was formed. These results were discussed by considering the thermodynamics and kinetics concerning the milling process.

  • Microsotructure designed porous ceramics of calcium hydroxyapatite $(Ca_{10}(PO_4)_6(OH)_2)$ were prepared by hydrothermal method. The particle size, shape, and the micro-pore size of the porous hydroxyapatite ceramics could becontrolled. The hydroxyapatite was non-stoichiometric apatite with calcium deficient compositions (Ca/P ratio < 1.67). The composition of non-stoichiometric hydroxyapatite ceramics could be controlled from 1.50 to 1.63 in Ca/P ratio. The hydroxyapatite ceramics preparedc at $105^{\circ}C$ under the saturated vapor pressure for 20h were composed of rod-shaped crystals with about 10$\mu\textrm{m}$ in length with the mean aspect ratio of 40. The porous ceramics of calcium deficient hydroxyapatite had about 45% porosity with the inter-connecting pore structure. Porous hydroxyapatite ceramics prepared hydrothermally had the compressive strength of about 10 to 30 MPa. In addition, porous ceramics of $\beta$-tricalcium phosphate ($\beta-Ca_3(PO_4)_2$) were prepared from the calcium deficient hydroxyapatite.

Establishment of a transformation protocol combination particle bombardment with Agrobacterium tumefaciens in different zoysiagrass cultivars (유전자총과 아그로박테리움을 이용한 여러 가지 한국 잔디류의 형질전환체계 확립)

  • In this report, several factors such as infection time and concentration of bacterial suspension, influencing on transient gene expression in Agrobacterium-mediated transformation were evaluated. An appropriate concentration (O.D 600nm = 1.0-1.2) of bateria and 30 min of infection time showed a higher level of GUS expression. To improve transformation efficiency (TE), friable embryogenic calli (FEC) were bombarded by tungsten particles without plasmid DNA, and then co-cultivated with A. tumefaciens LBA4404 which contains pTOK233 super binary vector, carrying neomycin phosphotransferase (NPTII), hygromycin phosphotransferase (hpt) and$\beta-glucuronidase$ (GUS) genes. Three days after co-cultivation with A. tumefaciens and particle bombardment, FEC cultures were transferred to the selection medium (SM: MS medium supplemented with BA 1mg/l, hygromycin 100mg/l, cefotaxime 250 mg/l and vancomycin 200mg/l). They were cultured for 2 weeks and then transferred to the second SM containing hygromycin 50mg/l, cefotaxime 200 mg/l and vancomycin 100mg/l. Later, stable GUS expression was detected 4 to 6 weeks after transfer to the SM. Further, TE from Agrobacterium-mediated transformation after particle bombardment increased to about 3-folds compared with Agrobacterium-mediated transformation without particle bombardment. In the present study, we established an efficient transformation protocol of zoysiagrass by using A. tumefaciens in the combination with particle bombardment for the first time.

Bone Cements in TTCP, DCPA, β-TCP and PHA System (TTCP-DCPA-β-TCP-PHA계 골 시멘트)

  • The effect of $\beta$-TCP and PHA as additives on initial setting time, compressive strength and surface micro-structure after in vitro test of bone cement in TTCP and DCPA system was investigated. The median particle sizes of TTCP, $\beta$-TCP, DCPA and PHA for bone cement were about 3, 5, 0.9 and 4${\mu}{\textrm}{m}$, respectively. Initial setting time and compressive strength of bone cement with various composition was measured by Vicat test and Universal Testing Machine, and surface morphology and crystalline phases of bone cements were observed and analyzed by SEM and x-ray diffractometer. Initial setting time was not affected by composition but by powder/liquid ratio, and cement with PHA required double amount of solution for paste as much as one without PHA, especially. It was thought that $\beta$-TCP and PHA in bone cements was not related to setting reaction. Thus, the addition of $\beta$-TCP and PHA in bone cements decreased compressive strength and inhabited HAP from being produced on surface in vitro test. In conclusion, it was not expected that $\beta$-TCP and PHA in TTCP-DCPD bone cements enhanced the strength and bioacitivity.

Development of Two-Step Temperature Process to Modulate the Physicochemical Properties of β-lactoglobulin Nanoparticles

  • The development of a new manufacturing process, a two-step temperature treatment, to modulate the physicochemical properties of nanoparticles including the size is critical. This is because its physicochemical properties can be key factors affecting the cellular uptake and the bioavailability of bioactive compounds encapsulated in nanoparticles. The aims of this study were to produce (beta-lactoglobulin) ${\beta}-lg$ nanoparticles and to understand how two-step temperature treatment could affect the formation and physicochemical properties of ${\beta}-lg$ nanoparticles. The morphological and physicochemical properties of ${\beta}-lg$ nanoparticles were determined using atomic force microscopy and a particle size analyzer, respectively. Circular dichroism spectroscopy was used to investigate the secondary structure of ${\beta}-lg$. The surface hydrophobicity and free thiol groups of ${\beta}-lg$ were increased with a decrease in sub-ambient temperature and an increase in mild heat temperature. As sub-ambient temperature was decreased, a decrease in ${\alpha}-helical$ content and an increase in ${\beta}-sheet$ content were observed. The two-step temperature treatment firstly involved a sub-ambient temperature treatment from 5 to $20^{\circ}C$ for 30 min, followed secondly by a mild heat temperature treatment from 55 to $75^{\circ}C$ for 10 min. This resulted in the production of spherically-shaped particles with a size ranging from 61 to 214 nm. Two-way ANOVA exhibited the finding that both sub-ambient and mild heat temperature significantly (p<0.0001) affected the size of nanoparticles. Zeta-potential values of ${\beta}-lg$ nanoparticles were reduced with increasing mild heat temperature. In conclusion, two-step temperature treatment was shown to play an important role in the manufacturing process - both due to its inducement of the conformational changes of ${\beta}-lg$ during nanoparticle formation, and due to its modulation of the physicochemical properties of ${\beta}-lg$ nanoparticles.
