경피제제로서 수종의 플루비프로펜 Vehicle과 O/W 마이크로에멀젼의 평가 (Evaluation or various vehicles and O/W Microemulsions of Flurbiprofen as Transdermal Delivery System)

  • In order to reduce systemic side effects following administration, flurbiprofen was formulated as O/W microemulsion consisting of the surfactant, oil phase and aqueous phase. Particle size distribution, apparent viscosity, solubility and skin permeation of flurbiprofen in various vehicles and microemulsion were evaluated. The domain of O/W microemulsion s phase diagram had difference between oil types and the area of O/W microemulsion was wide distributed by adding to PG and cosurfactant than that of water alone. As increasing 10, 15 and 20% of Brij 97 content and 1, 2.5, 5% of oil content, the solubility of flurbiprofen in O/W microemulsions and various vehicles was $400{\sim}1,000$ and $10{\sim}500$ times higher than that of control. Also, apparent viscosity of soybean oil microemulsions was higher than that of IPM microemulsions and that of vehicle were increased as increasing vehicle content. Since skin permeation of flurbiprofen decreased as increasing viscosity, in each vehicle, it was not affected 2% ${\beta}-CD$ and decreased as increasing PG content and to 2, 5 and 10% of $HP-{\beta}-CD$. In O/W microemulsion, 5% soybean oil. 20% Brij 97 and 75% water(A-1) with high viscosity showed low skin penetration.

형질전환 아르테미아(Artemia franciscana) 생산을 위한 리포터 유전자로서 lacZ 유전자의 유용성 검토 (Availability of the lacZ gene as a Reporter Gene for Production of Transgenic Artemia franciscana)

  • We examined the availability of the lacZ gene (${\beta}$-galactosidase gene) as a reporter of foreign gene transfer in the cysts of Artemia franciscana (A. franciscana) to conduct a risk assessment of living genetically modified organisms (LMOs) in the marine ecosystem. The LacZ gene was transferred to decapsulated cysts by particle bombardment, and its insertion and expression were assessed by means of polymerase chain reaction (PCR) and X-gal staining. X-gal staining indicated lacZ expression in all A. franciscana examined (including the control group), which exhibited not only negative but also positive PCR amplification. Endogenous ${\beta}$-galactosidase is highly active in the whole body of A. franciscana during all stages of the life cycle. Thus, the lacZ gene is unsuitable as a reporter for foreign gene transfer in A. franciscana cysts, because it is difficult to discriminate between exogenous and endogenous ${\beta}$-galactosidase activity.

에어로졸 중화기의 성능이 고하전 입자의 크기분포 측정에 미치는 영향 (Effect of Performance of Aerosol Charge Neutralizers on the Measurement of Highly Charged Particles Using a SMPS)

  • A SMPS(scanning mobility particle sizer) system measures the number size distribution of particles using electrical mobility detection technique. An aerosol charge neutralizer, which is a component of the SMPS, is a bipolar charger using a radioactive source to apply an equilibrium charge distribution to aerosols of unknown charge distribution. However, the performance of aerosol charge neutralizers is not well known, especially for highly charged particles. In this study, the effect of the particle charging characteristics of two aerosol charge neutralizers on the measurement using a SMPS system was experimentally investigated for highly charged polydisperse particles. One has radioactive source of $^{85}$ Kr (beta source, 2 mCi) and the other has $^{210}$ Po (alpha source, 0.5 mCi). The air flow rate passing through each aerosol charge neutralizer was changed from 0.3 to 3.0 L/min. The results show that the non-equilibrium character in particle charge distribution appears as the air flow rate increases although the particle number concentration is relatively low in the range of 1.5∼2x10$^{6}$ particles/㎤. The low neutralizing efficiency of the $^{85}$ Kr aerosol charge neutralizer for highly charged particles can cause to bring an artifact in the measurement using a SMPS system. However, the performance of the $^{210}$ Po aerosol charge neutralizer is insensitive to the air flow rate.

Asian Dust Particles Induce TGF-${\beta}_1$ via Reactive Oxygen Species in Bronchial Epithelial Cells

  • Background: Asian dust storms can be transported across eastern Asia. In vitro, Asian dust particle-induced inflammation and enhancement of the allergic reaction have been observed. However, the fibrotic effects of Asian dust particles are not clear. Production of transforming growth factor ${\beta}_1$ (TGF-${\beta}_1$) and fibronectin were investigated in the bronchial epithelial cells after exposure to Asian dust particulate matter (AD-PM10). Methods: During Asian dust storm periods, air samples were collected. The bronchial epithelial cells were exposed to AD-PM10 with and without the antioxidant, N-acetyl-L-cysteine (NAC). Then TGF-${\beta}_1$ and fibronectin were detected by Western blotting. The reactive oxygen species (ROS) was detected by the measurement of dicholorodihydrofluorescin (DCF), using a FACScan, and visualized by a confocal microscopy. Results: The expression of TGF-${\beta}_1$, fibronectin and ROS was high after being exposed to AD-PM10, compared to the control. NAC attenuated both TGF-${\beta}_1$ and fibronectin expression in the AD-PM10-exposed the bronchial epithelial cells. Conclusion: AD-PM10 may have fibrotic potential in the bronchial epithelial cells and the possible mechanism is AD-PM10-induced intracellular ROS.

다공질유리의 탄소 열적환원반응에 의한 Sialon의 합성에 관한 연구 (Synthesis of Sialon by Carbothermal Reduction of Porous Glass)

  • Synthesis of $\beta$-Sialon powder was attempted with carbothermal reduction of porous glass. The porous glass was prepared by heat and hydrothermal treatments of 9.32 Li2O.46.5B2O3.37.2SiO2.6.98Al2O3 glass. Carbon pyrolyzed from propane gas was deposited on the porous glass, thereafter activated carbon was added as reducing agents. The synthesized $\beta$-Sialon powder was pressureless sintered at 175$0^{\circ}C$ for 1hr in N2 atmosphere. The characterization of the $\beta$-Sialon powder was performed with XRD, BET, SEM and particle size analysis. The sinterability and mechanical properties of the sintered bodies were investigated in terms of bulk density, M.O.R., fracture toughness, morphology of microstructure and etc. The reduction effect of deposited carbon was better than that of activated carbon mechanically added. The formation of SiC was precominant over that of Si2ON2 and $\beta$-Sialon owing to low partial pressure of N2 inside the pore, wehreas on the surface of porous glass the formation of Si2ON2 and $\beta$-Sialon were predominant. Thereafter, SiC reduced unreacted glass to be $\beta$-Sialon. Single phase of $\beta$-Sialon(Z=1.92) was obtained from PGA porous glass having the largest pore radius by the simultaneous reduction and nitridation method at 145$0^{\circ}C$ for 5hrs. The bulk density, M.O.R., and KIC of the sitered body are 3.17g/cc, 434.4MPa and 4.1MPa.m1/2, respectively.

Manufacture and Physicochemical Properties of Chitosan Oligosaccharide/A2 β-Casein Nano-Delivery System Entrapped with Resveratrol

  • The purposes of this research were to form chitosan oligosaccharide (CSO)/A2 ${\beta}$-casein nano-delivery systems (NDSs) and to investigate the effects of production variables, such as CSO concentration levels (0.1%, 0.2%, and 0.3%, w/v) and manufacturing temperature ($5^{\circ}C$, $20^{\circ}C$, and $35^{\circ}C$), on the production and physicochemical characteristics of CSO/A2 ${\beta}$-casein NDSs to carry resveratrol. The morphological characteristics of CSO/A2 ${\beta}$-casein NDSs were assessed by the use of transmission electron microscopy (TEM) and particle size analyzer. High-performance liquid chromatography (HPLC) was applied to determine the entrapment efficiency (EE) of resveratrol. In the TEM images, globular-shaped particles with a diameter from 126 to 266 nm were examined implying that NDSs was successfully formed. As CSO concentration level was increased, the size and zeta-potential values of NDSs were significantly (p<0.05) increased. An increase in manufacturing temperature from $5^{\circ}C$ to $35^{\circ}C$ resulted in a significant (p<0.05) increase in the size and polydispersity index of NDSs. Over 85% of resveratrol was favorably entrapped in CSO/A2 ${\beta}$-casein NDSs. The entrapment efficiency (EE) of resveratrol was significantly (p<0.05) enhanced with an increase in manufacturing temperature while CSO concentration level did not significantly affect EE of resveratrol. There were no significant (p<0.05) changes observed in the size and polydispersity index of NDSs during heat treatments and storage in model milk and yogurt indicating that CSO/A2 ${\beta}$-casein NDSs exhibited excellent physical stability. In conclusion, the CSO concentration level and manufacturing temperature were the crucial determinants affecting the physicochemical characteristics of CSO/A2 ${\beta}$-casein NDSs containing resveratrol.

Particle Swarm Optimization을 이용한 비균일 급전, 비균등 간격의 선형 어레이 설계 (Design of a Randomly Excited and Randomly Spaced Linear Array Using the Particle Swarm Optimization)

  • 본 논문에서는 particle swarm optimization (PSO)을 사용하여 가장 낮은 SLL같을 갖거나 가장 좁은 빔폭을 가지는 비균일 급전, 비균등 간격의 선형 어레이를 설계하였다. 어레이 소자의 급전 크기와 급전 소자간의 간격을 조절하기 위해 변수로 지정하였다. 두 가지 변수를 동시에 무작위로 조절하여 빔패턴을 최적화하였다. 빔패턴의 널 포인트를 기준으로 나누어 각각의 사이드로브에 가중치를 부여함으로써 적합도 함수의 성능을 향상시켰고, 이를 이용하여 최적의 빔패턴을 얻을 수 있었다. 이 때, 가중치 값과 빔패턴을 나누는 각은 여러 번의 시도를 통해 얻을 수 있었다. SLL 뿐만 아니라 빔폭까지 고려하기 위해 fitness function에 추가적인 항목 ${\beta}{\times}BW$을 첨가하였다. 이로써, 가장 낮은 SLL값을 갖거나 가장 좁은 빔폭을 갖는 빔패턴을 갖는 어레이를 설계하였다. 10개의 어레이 소자를 이용하여 최적화 하였을 때, 전자는 -43dB의 SLL값과 $32.2^{\circ}$의 빔폭을 가졌고, 후자는 -26dB의 SLL값과 $24.2^{\circ}$의 빔폭을 가졌다.

Particle Inflow Gun을 이용한 벼 캘러스 내의 효율적 유전자 도입 (Efficient Gene Introduction into Rice Callus by Using Particle Inflow Gun System)

  • 본 연구실에서 개발된 particle inflow gun (PIG)은 조작이 간편하고, 사용비용도 저렴하며, 식물 세포 내의 유전자 도입효율이 높은 특징을 갖고 있다. PIG 장비를 이용하여 벼 캘러스 내로의 유전자 도입 조건을 검토하기 위해서 사용된 vector는 pIG121Hm으로서 T-DNA 내부에 intron GUS ($\beta$-glucuronidase)와 hygromycin 및 kanamycin 저항성 유전자를 포함하고 있다. 또한 벼 캘러스 내에 물리적으로 DNA를 도입할 때에 DNA 도입 효율과 관계가 높은 요인들을 GUS의 발현빈도를 통하여 조사하였다. 그 결과 gold particle에 DNA를 부착하는 과정에 사용되는 spermidine과 calcium chloride의 경우 무첨가구에 비해 16 mM의 spermidine과 1.5 M의 calcium chloride 첨가구에서 GUS 발현율이 각각 2배, 3배 증가하였다. 그리고 1회 분사되는 gold particles양이 2 mg의 경우 가장 높은 GUS 발현율을 보여주었으며, 또한 PIG장비의 분사거리와 헬륨의 압력은 벼의 배양세포의 경우 12cm의 분사거리에서 3.5 bar (50 psi)의 헬륨압력으로 분사하였을 때 GUS 발현율이 가장 높았다. 이상의 결과에서 PIG 장비를 이용한 유전자 도입은 본 연구에서 검토한 최적의 조건을 이용하였을 경우 기존에 많이 사용되고 있는 Biolistic Gun (Bio-Rad 사)과 거의 비슷한 유전자 도입효율을 보여 주었다. 특히 PIG 장비의 경우 조작이 매우 간편하고, 분사에 사용되는 일회용 부품이 필요하지 않기 때문에 대량의 반복실험을 필요로 하는 연구에서 손쉽게 사용되리라 기대된다.

베타-사이클로 덱스트린 중합체/신남산 복합체의 제조 및 특성 연구 (Preparation and Characterizations of Complex Composed of ${\beta}$-Cyclodextrin Polymer/Cinnamic Acid)

  • 베타-사이클로덱스트린($\beta$-cyclodextrin, $\beta$-CD) 중합체(polymer)는 가교제인 epichlorohydrin (EPI)과 $\beta$-CD의 몰 비가 10 : 1으로, 강한 염기조건에서 합성하였다. 합성한 $\beta$-CD 중합체 내의 $\beta$-CD contents는 52%였다. 광, pH반응성 복합체를 제조하기 위해 신남산을 첨가하였고 첨가된 신남산은 소수성 상호작용에 의해 $\beta$-CD 공동에 포접되었다. 형성된 $\beta$-CD 중합체와 신남산 복합체를 투과전자현미경을 이용하여 관찰하였을 때 복합체의 형상을 관찰하였다. 광 조사에 따른 이량화도는 $\lambda$ = 365 nm의 UV조사 시 증가하였으며 $\lambda$ = 254 nm의 UV조사 시 감소하였다. 또한 동적 광 산란(dynamic light scattering)장치를 이용하여 측정한 복합체의 크기는 광 조사 유무에 따라 큰 변화가 측정 되지 않았고, pH 반응성을 관찰한 실험에서도 복합체의 크기와 제타 전위(zeta potential) 모두 pH에 따른 변화가 나타나지 않았다.

Monte Carlo 계산에 의한 액체섬광계수기의 베타선 스펙트럼 Simulation (Simulation of Beta Ray Spectra in Liquid Scintillation Counting System by means of Monte Carlo Method)

  • 액체섬광 계수기에 대한 $^3H,\;^{14}C,\;^{36}Cl$의 베타선 스펙트럼의 시뮬레이션을 Monte Carlo 법으로 수행하였다. 베타선의 손실에너지는 'continuous slowing down approximation(CSDA)' 모형으로부터 구하였다. 순 베타방출 핵종이 균일하게 용해되어 있는 섬광용액은 $1.25cm{\times}3.5cm$의 원통형 geome try로 가정하였다. $^{14}C,\;^{36}Cl$에 대한 계산결과는 계수효율의 경우 실험 값과 2%의 범위 내에서 잘 일치하였으나 $^3H$에 대해서는 상당한 편차를 나타내었다. 이는 저 에너지 베타선에 대한 섬광용액 자체의 quenching 효과에 기인하는 것으로 판단된다.
