Dissolution and Stability Enhancements of Trimethoprim by ${\beta}-Cyclodextrin$ Polymer Inclusion Complexation (트리메토프림과 ${\beta}$-시클로덱스트린 고분자 간의 포접복합체 형성에 의한 용출 및 안정성 향상)

  • To improve the solubility and dissolution rate of trimethoprim (TMP), which is slightly soluble drug, its inclusion complexes were prepared and studied in this experiment. Inclusion complexes of TMP with ${\beta}-cyclodextrin$ and ${\beta}-cyclodextrin$ polymer (CDPS) were prepared according to Fenyvesi method. These were compared with TMP and its physical mixture with CDPS. Water, diluted hydrochloric acid and phosphate buffer solution were used as dissolution media. And accelerated stability test was studied at $50,\;70\;and \;80^{\circ}C$. It was found that solubility and dissolution rate of inclusion complexes were increased in water. Especially, the solubility and dissolution rate of TMP was found to be markedly increased by inclusion complexation with CDPS. In stability test, ${\beta}-cyclodextrin$ inclusion complexes were more or less stable than TMP alone. This tendency was not led in CDPS. Consequently, CDPS was useful in increasing dissolution rate and stability of TMP.

Enhancement of Nitrendipine Bioavailability in Rats by its Solid Dispersion with $Hydroxypropyl-{\beta}-Cyclodextrin$ after Oral Administration (흰쥐에 경구 투여시 히드록시프로필-베타-시클로덱스트린과 니트렌디핀 고체분산에 의한 생체이용률 증가)

  • Nitrendipine, a slightly soluble calcium channel blocking agent forms a solid dispersion system with $hydroxypropyl-{\beta}-cyclodextrin$, which exhibits better dissolution characteristics than the uncomplexed drug. The dissolution rate of nitrendipine was markedly increased in solid dispersion system in pharmacopeial disintegration media at pH 1.2 and pH 6.8. Four different dosage forms of nitrendipine were administered to rats: (a) nitrendipine in the solution of PEG 400; (b) nitrendipine solid dispersion system with $hydroxypropyl-{\beta}-cyclodextrin$ in a molar ratio of 1:2 by solvent evaporation method and administered in capsule form; (c) physical mixture of nitrendipine with $hydroxypropyl-{\beta}-cyclodextrin$ in a molar ratio of 1:2 and administered in capsule form; (d) nitrendipine alone administered in capsule form. Relative bioavailability after the oral administration of various dosage forms to rats with a dose of 10 mg/kg equivalent to nitrendipine was compared with that of nitrendipine in the solution of PEG 400. The AUC of solid dispersion was significantly bigger than that of nitrendipine powder. $T_{max}$ of solid dispersion was significantly shorter and $C_{max}$ was higher than that of nitrendipine powder. These results indicate that the bioavailability of nitrendipine could be improved markedly by inclusion complexation. An interesting correlation also appears to exist between the in vitro dissolution data and the area under the plasma concentration-time curves.

Silymarin Inhibits Cytokine-Stimulated Pancreatic Beta Cells by Blocking the ERK1/2 Pathway

  • We show that silymarin, a polyphenolic flavonoid isolated from milk thistle (Silybum marianum), inhibits cytokine mixture (CM: TNF-${\alpha}$, IFN-${\gamma}$, and IL-$1{\beta}$)-induced production of nitric oxide (NO) in the pancreatic beta cell line MIN6N8a. Immunostaining and Western blot analysis showed that silymarin inhibits iNOS gene expression. RT-PCR showed that silymarin inhibits iNOS gene expression in a dose-dependent manner. We also showed that silymarin inhibits extracellular signal-regulated protein kinase-1 and 2 (ERK1/2) phosphorylation. A MEK1 inhibitor abrogated CM-induced nitrite production, similar to silymarin. Treatment of MIN6N8a cells with silymarin also inhibited CM-stimulated activation of NF-${\kappa}B$, which is important for iNOS transcription. Collectively, we demonstrate that silymarin inhibits NO production in pancreatic beta cells, and silymarin may represent a useful anti-diabetic agent.

Transformation Behaviour of High Temperature Thermoelectric $FeSi_2$ (고온열전재료 $FeSi_2$의 변태거동)

  • In the Fe-Si system, a mixture of a($Fe_{2}Si_5$) - and ${\epsilon}$(FeSi)-composition powders was sintered and heat-treated subsequently at various temperatures and time to get thermoelectric ${\beta}$-phase($FeSi_2$) compacts. The different transformational sequences depending on the heat treating temperature were found through the investigation into phase transformation and microstructural development. That is, a rapid eutectoid decomposition of ${\alpha}{\to}{\beta}+Si$ occurred together with a accompanying slow reaction between the dispersed Si formed by above decomposition and the preexisted ${\epsilon}$ phase at temperatures below $830^{\circ}C$. The unreacted Si and the micropores formed due to the density change upon the transformation coarsened as heat treating time elapsed. At temperatures above $880^{\circ}C$, however, transformation was proceeded by a peritectoid reaction of ${\alpha}+{\epsilon}{\to}{\beta}$. It took at least 200min. to achieve 90% volume fracion of transformed ${\beta}$ phase, and the growth of micro-pores was also observed in this transformational sequence with prolonged heat treating time.

  • $\beta$-SiC was chemically vapor deposited by pyrolysis of MTS+H2 gas mixture. The experiments were conducted in the temperature range of 1100~150$0^{\circ}C$ with a r.f. induction furnace under atmospheric pressure. The IR, XRD, EDS and AES analysis revealed that the free Si was always codeposited with SiC below 140$0^{\circ}C$, regardless of the total flow rate and MTS concentration, whereas $\beta$-SiC single phase was deposited at 150$0^{\circ}C$. C3H8 or CH2Cl2 as an excess C sources, was supplied with MTS in order to obtain stoichiometric SiC at low temperature. With the addition of C3H8 or CH2Cl2, the deposition rate was increased and $\beta$-SiC single phase could be deposited even at temperature as low as 110$0^{\circ}C$. Optimization of β-Cyclodextrin Recycling Process for Cholesterol Removal in Cream

  • This study was designed to find optimum conditions of four different factors (ratio of solvent to cholesterol-$\beta$-cyclodextrin complex, mixing speed, mixing temperature, and mixing time) for cholesterol dissociation in cream. Using the ratio of 6 to 1 (solvent to the complex) showed the highest cholesterol dissociation rate (82.50%) when mixed at 100 rpm at $50^{\circ}C$ for 1 h. Mixing speed did not significantly affect the cholesterol dissociation. Also, mixing time appeared to be insignificant. The optimum mixing temperature was $50^{\circ}C$ and mixing at $40^{\circ}C$ resulted in a significantly lower rate, compared with that at $50^{\circ}C$. In a subsequent experiment, using recycled $\beta$-cyclodextrin only showed 75.07% of cholesterol removal in cream, while the mixture of recycled to unused $\beta$-CD with the ratio of 6 to 4 increased cholesterol removal to 95.59%, which is highly close to that of 100% unused $\beta$-CD.

  • The mechanical synthesis of thermoelectric material $FeSi_2$ by planetary ball mill has been investigated. The homogeneous and amorphous mixture of Fe-Si has been obtained by mechanical alloying for 850 rpm-40 min. The $\beta-FeSi_2$ powder could be synthesized by 1123 K-3 hr annealing heat treatment after mechanical alloying for 850 rpm-10, 20, and 40 min. The ceramic samples doped with the maximum content up to $10\;at.\;\%$ Co have exhibited semiconduction phenomena and maximum thermoelectric powder at 440K.

  • ${\beta}$-SiC thin film was deposited on a Si substrate without buffer layer using a single precursor of Hexamethyldisilane (Si2(CH3)6) by chemical vapor deposition method. HCI gas was introduced into hexamethyldisilane /H2 gas mixture, and the feeding schedule of HCI and precursor gases was modified in order to enhance the selectivity of SiC deposition between a Si substrate and a SiO2 mask. The effect of HCI gas on the surface roughness of the SiC film was investigated and typical electrical properties of the SiC film were also investigated by Hall measurement.

Bulk Coordination Polymerization of Dicyclopentadiene (DCPD) by Pd Complexes Containing β-Ketoiminate or β-Diketiminate Ligands

  • Several palladium complexes containing ${\beta}$-ketoiminate and ${\beta}$-diketiminate ligands successfully produced poly(DCPD) possibly via vinyl addition. It was found that catalysts with ${\beta}$-diketiminate ligands containing bulkier aryl substituents showed the highest activity in the presence of MAO as a cocatalyst. Purity of DCPD is quite essential for the higher activity and small amount of organic solvent such as $CH_2Cl_2$ and toluene is required to reduce the viscosity of the reactant mixture for the higher activity. $^1H$ NMR spectra of produced polymers with N,N-dimethylanilinium tetra(pentafluorophenyl)borate (N,N-DAPFAr"$_4$) show that 5,6-double bond of DCPD is removed with 2,3-double bond remaining. Produced poly(DCPD) with MAO cocatalyst is quite rigid and insoluble in common organic solvents but rather brittle.
